Brucellosis is the most common zoonosis worldwide, which is caused by Brucella spp. In humans, it can be mainly occurred by direct contact with infected animals or consumption of contaminated dairy products. This study focused on human brucellosis caused by B. melitensis discovered from Chinese worker in Korea in 2015. We investigated molecular epidemiological evidence to find the infection source. We first performed several PCR methods including 16S rRNA PCR, multiplex PCR and real-time PCR to identify Brucella species. We also conducted MLVA typing for epidemiological trace-back analysis. The isolate from the patient was confirmed to B. melitensis through Brucella-specific PCR. In clustering analysis with B. melitensis from foreign countries, this human isolate was correlated with B. melitensis isolates from humans and sheep in China by 99.9% similarity. Thus, we assumed the brucellosis patient has been already infected in China followed by migration to Korea according to molecular epidemiological analysis with history evidence. Moreover, we suggest it needs to take measures to reduce the risk for intercountry transmission of brucellosis due to the influx of infected people from abroad.

This study investigated the quality variation characteristics of Ssamjang containing Cheongkukjang, in order to improve the degree of preference of Cheongkukjang. The amount of Cheongkukjang was set at 1% based on sensory evaluation. Ssamjang containing Cheongkukjang was stored at 37℃ for 13 weeks, after which quality variation characteristics were weekly. During storage for 13 weeks, physicochemical quality characteristics, moisture content, and pH of Ssamjang containing Cheongkukjang slightly decreased, whereas salt content did not change. Amino nitrogen content slightly increased by 1 week but decreased by 3 weeks and then increased by 5 weeks. For microbiology quality characteristics, viable cell counts and total cell counts of B. cereus were unchanged. For sensory quality characteristics, shape quality was poor after 13 weeks while mold, drying phenomenon, and swelling phenomenon were not observed. Therefore, physicochemical quality and microbiology quality of Ssamjang containing Cheongkukjang were unchanged during storage for 13 weeks, and the storage limit was determined to be 12 weeks according to sensory quality evaluation.

The microbial composition in Nuruk, a Korean cereal fermentation starter, is a critical factor for the quality and organoleptic properties of traditional alcoholic beverages. This study was aimed at monitoring the compositional change and enzyme activity of culturable lactic acid bacteria (LAB) in two types of Nuruk fermented at different temperatures. All culturable LAB were isolated at various time points (0, 3, 6, 10, 20, and 30 days) and identified by 16S rRNA sequencing. In traditional Nuruk type A (TN-A), which was fermented at 36℃, the population of total culturable LAB during the fermentation period was between 104 and 105 log CFU/mL. On the other hand, the LAB population in traditional Nuruk type B (TN-B) fermented at 45℃ (primary fermentation for 10 days) and 35℃ (secondary fermentation for 20 days) was 102 log CFU/mL; however, these bacteria could not be detected after 6 days. Major LAB strains were identified in both Nuruk types: (1) from the MRS-culture of TN-A, Pediococcus pentosaceus at 3-30 days; (2) from MRS-culture of TN-B, P. pentosaceus at 3 days and Enterococcus hirae at 6 days. The protease activities of the dominant LAB isolated from the TN-A and TN-B cultures were within the ranges of 0.64~1.03 mg/mL and 0.74~0.81 mg/mL (tyrosine content), respectively, whereas the α-amylase activities were 0.75~0.98 mg/mL and 0.78~0.79 mg/mL (amylose content), respectively.

This study was conducted to evaluate the quality characteristics of pickled radish wrap supplemented with calcium chloride (CaCl2) at different concentrations. With CaCl2 supplementation, initial pH and acidity were 3.8~4.0 and 2.7~3.0% (w/v), respectively. During the 28 day-preservation at 4℃ or 50℃, the initial pH and acidity were maintained. However, when the pickled radish wrap was preserved at 25℃, it had lower pH (by 0.5) and higher acidity (by 1.9~2.2%) when compared with the initial values. The difference in color (△E-value) of the pickled radish wrap preserved at 50℃ was significant (△E-value 23~27), though the hardness of the sample prepared at 4℃ or 25℃ showed no distinguishable differences at various concentrations of CaCl2 supplementation (0.1, 0.2, 03, 0.4, and 0.5%). However, once preserved at 50℃, it resulted in the greater hardness with the higher concentrations of CaCl2 supplementation. In terms of softness, the overall trend in changes was similar to the hardness. A sensory evaluation using the samples preserved at 4℃ for 28 days demonstrated that the preparation with 0.4% CaCl2 ranked highest in taste, hardness and overall acceptability. It wass suggested that supplementation with 0.4% CaCl2 would be most appropriate to improve the textural properties of pickled radish wrap while maintaining its organoleptic quality.

In Korea livestock farms breeding cattle have been suffering from re-emerging problems of Brucella (B.) abortus infection while steady decline of bovine brucellosis. Therefore, this study underscored the identification and association of etiological agent of brucellosis in cattle in South Korea. The incidence of brucellosis in cattle was analyzed by bacteriological and molecular methods in 187 brucellosis-suspicious farms of 11 regions between 2018 and 2020. Brucella isolates from various specimens were identified by Brucella-specific multiplex PCR. Epidemiological data were collected by local official veterinarians through history taking from farmers and animal data systems. In 230 of 560 cattle (40.9%) and 94 of 187 farms (50.3%), a total of 313 B. abortus were isolated from various specimens, the majority of isolates were from supramammary lymph node (41%). In epidemiological findings, the majority of positive cases were mainly caused by resurgence (43.7%) and unknown (37.2%). Of 94 positive cases isolated B. abortus, abortion in cattle infected by B. abortus occurred in 51 farms (54.3%) where led to resurgence in 30 farms and environmental survival of B. abortus in 9 farms. Consequently, these findings revealed the existence of etiological agent of bovine brucellosis in Korea, which still occurred at low levels in distinct regions where are allowed to call for persistent biosecurity. Thus, we highlight that brucellosis in Korea needs to take more effective control strategies with potential evidence. Moreover it is ultimately important to maintain a constant monitoring for eradication of brucellosis.
