        톳 에탄올 추출물이 알코올을 투여한 흰쥐의 항산화효소활성에 미치는 영향

        톳 에탄올 추출물이 알코올을 투여한 흰쥐의 간조직 손상에 미치는 영향을 알아보기 위해 정상군, 알코올 투여군(35% ethanol, 10 mL/kg B.W./day), 알코올 및 톳 에탄올 추출물 200 mg/kg 및 400 mg/kg병합투여군의 4군으로 나누어 6주간 사육 후 체중 증가율, 식이효율, 혈청중 ALT 및 ALP활성, 간조직의 손상억제효과를 검토하기 위해 SOD, catalase, XO 및 GSH-Px효소 활성을 측정하고 지질과산화물인 TBARS와 GSH함량을 측정한 결과는 다음과 같다. 1) 체중증가율은 알코올 투여군이 정상군에 비하여 약 38%가 감소되었으나 알코올과 톳 에탄올 추출물(kg당 200 mg 및 400 mg)을 병합 투여하여 알코올에 의해 둔화된 체중증가율이 정상군에 근접하도록 회복되었으며, 식이효율은 알코올 투여군이 정상군에 비하여 약 50%가 감소되었으나 톳 에탄올 추출물과 알코올의 병합투여로 증가되었다. 2) 간 손상지표 중의 하나인 혈청 ALT 및 ALP활성의 경우, 알코올 투여로 정상군에 비하여 160%정도 유의적으로 증가되었으나, 알코올과 톳 에탄올 추출물의 병합 투여로 알코올 투여군에 비하여 각각 63% 및 47%가 감소되었다. 3) 톳 에탄올 추출물은 알코올에 의해 증가된 세포질내의 XO활성을 감소시키지 못했으나 SOD, catalase 및 GSH-Px활성은 알코올 투여군에 비하여 각 56%, 38% 및 28%가 감소되었다. 4) 알코올만을 투여한 군의 TBARS함량은 정상군에 비하여 약 129%이상 증가되었으나, 알코올과 톳 에탄올 추출물 병합 투여로 알코올만을 투여한 군에 비하여 각각 34%, 56%의 감소를 나타냈는데 특히 400 mg 투여가 더 많은 감소 효과를 보였다. 5) 알코올 투여로 간조직중의 GSH함량은 정상군에 비해 약 38%가 감소되었으나, 알코올과 톳 에탄올 추출물 병합 투여로 알코올 투여군에 비하여 각각 33% 및 66%가 증가되었다. 특히 400 mg 투여군의 경우 GSH함량 상승 효과가 우수하였다. This study was designed to investigate the effects of Hijikia fusiforme (Harvey) Okamura ethanol extract on the ethanol-induced hepatotoxicity of rat administered orally experimental diets for 6 weeks. Sprague-Dawley rats weighing about 100 g were divided into 4 groups; normal group (NOR), ethanol (35% ethanol 10 mL/kg b.w./ day) treated group (CON), ethanol and Hijikia fusiforme ethanol extract 200 mg/kg (HE1) and 400 mg/kg (HE2) concomitantly treated group, respectively. Each group was examined for the growth rate, feed efficiency ratio (FER), activities of antioxidative enzymes and contents of TBARS and glutathione. Hijikia fusiforme ethanol extract showed increasing effects of the growth rate by 43%, and FER was gradually increased by Hijikia fusiforme ethanol extract treatment, compared with ethanol treatment. Ethanol elevated the activities of superoxide dismutase, catalase and glutathione peroxidase of rat liver markedly as compared to normal group, but those activities were significantly decreased in Hijikia fusiforme ethanol extract treatment by 56%, 38% and 25%, respectively. Xanthine oxidase activity elevated by ethanol was not affected by Hijikia fusiforme ethanol extract. The content of TBARS increased by ethanol treatment was signigicantly decreased in HE2, and the glutathione content depleted by ethanol treatment was increased by Hijikia fusiforme ethanol extract administration adjacent to normal level. These results suggest that Hijikia fusiforme ethanol extract is believed to be a possible protective effect for the ethanol-induced hepatotoxicity of rat liver.

        당뇨가 유발된 햄스터 창상치유에 미치는 벼세포 유래 GM-CSF의 효과

        상처치유는 growth factor와 cytokine들의 복합적인 작용에 의해 일어나는 과정이다. GM-CSF는 호중구와 대식세포를 분화시키고 각질세포를 증식시키는 다면발현성 사이토카인으로 여러 세포에서 재조합되어 사용되고 있다. (주)핸슨바이오텍에서 개발한 rice cell 유래의 재조합 rrhGM-CSF는 다른 세포에서 유래된 GM-CSF들보다 탄수화물 측쇄가 많은 것이 특징이다. rrhGM-CSF의 창상치유효과를 알아보기 위하여 햄스터에 만성 당뇨를 유도하여 상처치유가 어려운 동물모델을 만든 후, 등쪽에 인위적으로 피부 전 층을 제거하여 창상을 유발하였다. 실험군에는 rrhGM-CSF가 포함된 연고를 적용하고, 대조군에는 base cream만을 적용하여 재상피화되는 과정과 피부조직의 구조 및 PCNA 반응을 통한 세포증식 정도를 비교하였다. 육안 및 조직학적 관찰 결과, rrhGM-CSF를 적용한 그룹의 재상피화 속도가 더 빨랐으며, PCNA 면역조직화학적 조사에서도 증식하는 각질세포의 수가 대조군보다 상대적으로 많은 것으로 나타남으로써, rrhGM-CSF에 의한 피부상처 치유 효과를 관찰할 수 있었다. 이러한 결과들로 미루어 보아 벼세포 유래의 rrhGM-CSF는 국소적인 피부 창상 치유에 효과적인 것으로 보이며, rrhGM-CSF의 풍부한 당화 사슬은 이 단백질이 상처부위에 안정된 상태로 기능을 나타냄으로써 효과를 오랫동안 지속시킬 수 있었던 것으로 사료된다. GM-CSF is a multipotent growth factor, which also plays an important role during the process of wound healing. rrhGM-CSF was specifically produced from rice cell culture in our laboratory (Hanson Biotech Co., Ltd, Daejeon). The rrhGMCSF contains more oligosaccharide side chains than any other types of GM-CSF. This work was taken to evaluate the influence on wound healing of rrhGM-CSF in male golden hamsters. Full thickness skin defects of 9 mm in diameter were made in the back of hamsters, and 100 ${\mu}L$ ointment containing rrhGM-CSF 50 ${\mu}g/mL$ was applied. Control groups were given ointment without rrhGM-CSF. The wound sizes were relatively reduced and skin was well regenerated in the experimental group compared with the control group. Structurally, reepithelialization and architecture of the skin following injury were well accomplished in the experimental group. And also, positive reaction of PCNA of the skin following injury was more prominent in rrhGM-CSF containing ointment treatment group. Since this type of GM-CSF has highly glycosylated side chains, the effectiveness might be retain longer and stable, regarding acceleration of wound healing in the animal model. The present study has important implications for further development of the therapeutic manipulation of wound healing using rrhGM-CSF.

        결명자 에탄올추출물이 알코올 투여 흰쥐의 항산화물질 및 지질대사에 미치는 영향

        결명자 에탄올추출물이 알코올을 투여한 흰쥐의 간손상에 미치는 영향을 알아보기 위해, 정상군, 결명자 에탄올추출물(200 mg/kg)투여군, 알코올 투여군(35% ethanol 10 mL/kg, b.w./day), 알코올 및 결명자 에탄올추출물(200 mg 및 400mg/kg) 병합투여군 등 5군으로 나누어 6주간 사육후 체중증가율, 식이효율 및 혈액중 총 콜레스테롤, HDL-콜레스테롤 및 LDL-콜레스테롤농도, 혈청중 ALT 및 AST활성, 간손상 억제 효과를 확인하기 위해 유리기 해독계 효소인 SOD, catalase, GSH-Px 등의 활성과 유리기 생성계 효소인 XO활성을 측정하고, 지질과산화물인 TBARS 및 GSH함량에 미치는 영향을 관찰하였다. 1) 각 분획구의 항산화력은 n-핵산>클로르포름>에틸아세테이트>물>메탄올 순이었다. 2) 결명자 에탄올추출물과 알코올을 병합투여하므로서 체중증가율은 알코올만을 투여한 군에 비하여 유의적인 변화는 아니었으나 증가되는 경향을 보였으며 식이효율은 정상군의 치에 근접하게 증가되었다. 3) 간 손상의 지표의 하나인 혈청 중 ALT 및 AST활성의 경우, 알코올 투여로 정상군에 비하여 유의적으로 증가되었으나 결명자 에탄올추출물을 병합투여하므로서 정상군보다 오히려 낮았다. 4) 알코올 투여로 총 콜레스테롤 및 LDL-콜레스테롤함량은 정상군에 비하여 많이 증가되었으나 결명자 에탄올추출물 병합투여로 투여량이 증가됨에 따라 더욱 우수한 감소효과를 나타내었다. 반면 HDL-콜레스테롤함량은 알코올 투여로 정상군에 비하여 유의적으로 감소되었으나 결명자 에탄올추출물 병합투여로 정상군의 농도에 근접하게 증가되었는데, 특히 결명자 에탄올추출물만을 투여한 군의 함량이 정상군보다 높았다. 5) 결명자 에탄올추출물이 알코올 투여로 상승된 유리기 생성계효소인 XO활성은 알코올만을 투여한 군에 비하여 약 42%정도 현저하게 감소시켰으나, 유리기 해독계 효소인 SOD, catalase 및 GSHPx 활성에는 유의한 영향을 미치지 못했다. 6) 알코올 투여로 증가된 간 중의 TBA반응성 산물량을 결명자 에탄올추출물이 유효하게 감소시켰으며, 특히 결명자 에탄올추출물만을 투여한 군의 간 TBA 반응성 산물량이 정상군보다 오히려 낮았다. 7) 알코올 섭취로 간조직 중의 GSH함량이 정상군에 비하여 약 51%정도 감소되었으나 결명자 에탄올추출물투여로 정상군의 함량에 근접하도록 증가되었다. 이상의 실험 결과에서 결명자 에탄올추출물의 항산화작용은 주로 알코올투여로 증가된 유리기 생성계 효소인 XO활성억제와 비효소적 항산화작용을 나타내는 GSH함량을 증가시키므로서 지질과 산화물에 대한 방어력이 증강되어 나타난 결과로 여겨지고, 또한 혈청중 지질함량, ALT 및 AST의 활성을 유의성있게 감소시켰음은 결명자 에탄올추출물이 알코올에 의한 지방간 또는 손상된 간세포를 회복시키는 작용이 있는 것으로 추정된다. This study was done to investigate the effects of ethanol extract of Cassia semen (Cassia tora L.) on the activities of hepatic oxygen free radicals metabolizing enzymes and blood lipid profile in rats of hepatotoxicity induced by ethanol. Sprague-Dawley male rats weighing 100~160 g were divided into 5 groups; control group (CON), Cassia semen ethanol extracts (200 mg/kg) treated group (CEL), ethanol (10 mL/kg, 35%) treated group (ETH), Cassia semen ethanol extracts (200 mg/kg) and ethanol treated group (CE1) and Cassia semen ethanol extracts (400 mg/kg) and ethanol treated group (CE2), respectively. Compared with ETH, the growth rate of CE1 and CE2 were to be increased tendency, and in blood levels of total cholesterol, LDL-cholesterol and the activities of alanine aminotranferase and asparate aminotransferase elevated by ethanol were significantly decreased (p<0.05). It was observed that the activities of superoxide dismutase, catalase, xanthine oxidase and glutathione peroxidase of rat liver increased by ethanol, were more decreased by the treatment of Cassia semen ethanol extract than the only ethanol-treated group. The content of glutathione depleted by ethanol treatment was increased in CE1 and CE2. TBA-reactants of liver increased by ethanol were decreased in CE1 and CE2, compared with ethanol-treated group. These results suggested that ethanol extract of Cassia semen may influence upon the ability of oxygen free radical detoxication and lowering of blood lipid level on ethanol-induced hepatotoxicity in rat.

      • 핸드볼 選手의 投力과 體格의 相關關係 考察 : 長投, 定確投 , Pass 能力과의 相關을 中心으로 With the Relation Betwen Long Throw a Degree of Accuracy and Pass Skill in the Center

        This study is on the basis on the statistics that is measured by 25 female/male players in university as an object of study, to consider the correlation between throwing power and physique for Handball player. The results of study is as follows. 1. Physique of a testee is found that for male, body-height is 179.8±5.15㎝, body weight is 76.06±2.86㎏ While for female, body height is 164.30±3.68㎝ and body weight is 61.20±4.70㎏. Both female and male showed some less value than the mean value of national representative player. 2. In the correlation among such measuring items as run, jump, and throw; For male, the connection with jumping is showed that 20m run higher relation than 100m run, as r = -0.406. ( r is the connection with long jump) Female presents the highest correlation in the connection between run and throw. So she presents the same high relation as r = -0.570 in the conncetion between 100m run and bashetball throwing. 3. Both female and male find that standing throw is the method which can throw the farthest. Male throw about 10m farther than female on an average. 4. The measurement of pass individually and sexually is similar to value between female and male. Male find that the method of shoulder pass is the fastest. 5. The measurement of shoot per location is showed similar tendency each of female and male. But the method of Jump shoot is found that female is more accurate than male. 6. In the connection between physique and long throw; While Circumference of hand, tip of fingers of physique is showed that r = 0.437, r = -0.439 each, a connection with sitting height has low relation as r = -0.236 even though standing throw. And other items is showed much lower correlation. Therefore, the longer length of the legs and arms is, the more profitable long throw is. 7. In the connection between physique and pass in general; Male player has a contrary connection with height size of body height, higher limbs, lower limbs, tip of fingers and so on. While female player presents high rational connection with chest-girth.

      • 靑少年의 比體重과 體脂肪量의 相關과 回歸分析에 關한 從斷的 考察

        By the method of the Longitudinal study, this research were measureed the body weight and height through the 15,441 numbers of the high class students of Primary, Junior and Senior High school as the subject of this study, and get for the body fat amount and calculated Relative Body Weight related body height and weight and the regression equation which can get for the body fat amount utilizing the relationship with body fat amount. In conclusion, we can see the following results. 1. Measurement of Body Height It showed the rectilineal growth in the body height and weight of bots sexes, and the ages for in tersection of the bots sexes appeared in 10.5-13.5 for the body height, 10.5-14.0 for the body weight. 2. Relative Body Weight It showed highest amount in 34.47 for boys, 32.61 for girls when they are 17 years old. But it can't arrive the regular exponent and the ages for intersection of the both sexes appeared in 10.5-14.5. 3. Body fat amount by the method of the Vital Measurement The body fat amount increased continuously according to that of ages. It shows 8.68㎏(14.8%) for boy, 12.91㎏(25.15%) for girls when they are 17 years old. It appeared much more body fat amount in girls compared with body through the whole ages. 4. Regrssion Analysis about the Relative body weight and Indirect Body Fat 1) Correlation to Relative Body Weight and Body Fat Amount. The corelative to the body surface(area) showed high degree relation through the whole ages, it appeared climax as r = + 0.908 (P 0.001) in 13 yrs. for boy, 12 years for girls. The corelative to the body capacity showed high degree relation and it appeared climax in 13.15 years. as r = + 0964 for boys, 12 years as r = + 968 for girls. The corelative to the body density are getting lower according to the increase of ages. It appeared reverse relation as r = - 0.160 in the 17 yrs. for boy, and middle degree of corelative as r = + 0.574 for girls. The corelative to the body fat ratio showed reverse relation in 14 yrs. (r = -0.0071) for the boys, and through the whole ages for girls. The corelative to the body fat amount showed reverse relation until 12 yrs. (r = -0008) for boys, 10 yrs. (r = -0160) for girls, but after that ages it showed middle degree relation in 17 yrs. (r = +0.695) for the body and lower relation as r = -0.393 for the girls. 2) Regression Analysis of Relative Body Weight and Body Fat Amount The ages which appeared high ratio of the body fat amount through the regression analysis of Relative Body Weight are appeared as follwings. Body Surface : b = 0.038 in 13 yrs. for boy, b = 0.035 in 11 yrs. for girls. Body Capacity ; b = 1.896 in 15 yrs. for boy, b = 1.630 in 12 yrs. for girls. Body Density : b = 0.008 b = 0.010 in 6 yrs. for both sexes. The rate of the Body Fat : b = 0.126 in 16 yrs. for boys, b = -0464 in 17 yrs. for girls. Body Fat Amount : b = 0.416 b = 0.193 in 17 yrs. for both sexes. The formular which calculated the regression equation were presented in the Table 4-8.

      • Hand Ball Game에서의 Running method 의 類型과 要因分析

        For the purpose of this study, we investigated and analyzed the running - speed, running-distance, running-method, and interval - time of running - speed in Handball games. In order to investigate and analyze, we had utilized the high - speed camera to pursue all activities of 18 players during the game. The results were as follows; 1) Male and female players had run about 6 ㎞, 4.5㎞, through one Handball game respectively. 2) He, who was R.O.S of Won-Kwang University team, had the maximum speed and distance that were 7.1∼8m/sec., 54.96 m. 3) The maximum speed of the male's and female's fast attack was 6.04 m/sec., 5.08m/sec. respectively. 4) The returning time of male and female after attacking was 5.53m/sec., 4.27m/sec. respectively. 5) The maximal running-speed of male and female during the game was 82.45%, 81.12% of the maximum speed of the 100m dash. 6) The frequency of the male and female curve was 6-10, 1-5 times and the angle was 31°-40° respectively. 7) In male, the interval time of maximum running was 1'14", who R.O.S of Chung-Nam University team. In female, 2'13", who was a member of Inchon City Hall team.

        급성심근경색에 병발한 심실중격결손증의 수술적 치료 1 예

        Perforalion of the ventricular septum is a rare but serious complication following myocardial infarction. It usually results in progressive intractable congestive heart failure and death; for that reason, it requires aggressive surgical intervention. Since the first report of surgical treatment for this serious complication by Cooley and colleagues, many reports decribed their complications. Here We report a case of postinfarction septal rupture treated by patch repair of the rupture and single coronary bypass graft.
