저장조건이 딸기 "미녀봉"의 저장 중 품질 및 미생물학적 변화에 미치는 영향을 연구하기 위하여 저장 중 중량감소율, 부패율, pH 및 적정산도, 총균수 및 곰팡이와 효모 수의 변화가 측정되었다. 딸기 시료는 LDPE로 포장하여 4℃ 와 20℃에서 각각 저장하였다. LDPE 포장은 대조구에 비해 중량감소율 및 부패율을 감소시켰다. 또한 포장방법은 저장중 미생물학적 변화에 영향을 끼쳤는데 저장온도와 상관없이 대조구와 비교해서 효모와 곰팡이뿐만 아니라 총균수의 감소를 야기시켰다. 본 연구결과는 딸기 "미녀봉"의 유통기한을 고려할 때 4℃에서 LDPE 포장으로 저장하는 것이 바람직하다는 것을 나타냈다. These results indicate that storage of strawberry fruits wrapped with LDPE at 4℃ should be recommended in terms of quality as well as its shelf-life.

Patulin, a mycotoxin mainly produced by Penicillium and Aspergillus, is found in various foods. In the present, a maximum acceptable level for patulin is established at 50 μg/kg(ppb) in apple juices and apple concentrates in Korea. But patulin may be detected in foods produced with other fruits. In the present study, patulin contamination was analyzed in 520 samples of fruit juices and beverages, and 50 samples of fruit juice concentrates. High performance liquid chromatography(HPLC) was applied to quantitatively analyze patulin levels in samples and liquid chromatography-mass spectrometry(LC/MS/MS) was used to remove false positive results. The results showed that three samples of 520 fruit juices and beverages and five samples of 50 fruit juice concentrates were contaminated by patulin, 9.8 - 18.0 μg/kg and 4.7 - 18.2 μg/kg respectively. Contaminated samples were produced with apple, orange or pear. This indicates that it is necessary to extend the regulatory range of patulin. In the other hands, the present study confirmed the effectiveness of LC/MS/MS analytical method to remove false positive results.

Thr, Trp을 overproduce하는 효모 균주를 분리하기 위하여 Saccharomyces cerevisiae를 UV와 EMS로 mutagenesis한 후 screen하였다. Thr analogue인 hydroxynorvaline이 UV mutagenesis 후 Thr overproducing 하기 위해 사용되었다. 31 mutant 중 아미노산 분석 결과에 의해 TC 5-1이 선정되었고 다시 Trp overproducing 위해 EMS mutagenesis 하였다. 8개의 mutant가 flurotryptophan을 이용하여 Thr, Trp을 overproduce하는 mutant로 선정되었다. 아미노산결과에 의해 그 중 TC 6-1이 최종 효모 균주로 선정되었다. To isolate a mutant which overproduces threonine and tryptophan, mutants of Saccharomyces cerevisiae were screened after UV and EMS mutagenesis. Hydroxynorvaline, a Thr analogue was used for selection of a Thr-overproducing mutant after UV mutagenesis. Among 31 mutants, TC 5-1 was selected as the strain candidate, based on amino acid analysis. TC 5-1 was then treated by EMS mutagenesis for Trp overproduction. Eight mutants were selected using fluorotryptophan for Thr and Trp overproducing strains. Amino acid analysis results showed that TC 6-1 was the best strain since it had the highest amount of Thr and Trp among mutants.

플루메퀸의 분석은 시료를 ethyl acetate로 추출 농축한 후 이 농축액에 acetonitrile과 n-hexane을 가하여 분리하고, 분리된 acetonitrile층을 농축하였다. 이를 이동상에 녹여 여과한 후 C₁? 컬럼을 이용한 HPLC-FLD로 분석하였다. 분석법은 표준물질 0.1-1.0㎎/㎏의 농도범위에서 직선성(r²= 0.9979)을 나타냈으며, 검출한계는 0.005 ㎎/㎏, 정량한계는 0.017 ㎎/㎏이었다. 또한 회수율은 쇠고기에서 90.8-96.7%, 돼지고기에서 96.4-96.6%, 닭고기에서 94.0-101.1%이었으며, 상대표준편차(RSD)는 쇠고기, 돼지고기, 닭고기에서 각각 5.3-8.6%, 3.0-6.8%, 3.1-4.1%이었다. 확립된 분석방법으로 유통 중인 식육(쇠고기, 돼지고기, 닭고기) 150건을 전국지역(서울, 부산, 대구, 대전, 광주)별로 수거하여 플루메퀸의 잔류 실태를 조사한 결과, 쇠고기 1건(0.080 ㎎/㎏), 돼지고기 1건(0.048 ㎎/㎏)에서 검출되었으며, 조사한 식육 150건의 플루메퀸 잔류량은 모두 잔류허용기준 이하의 수준이었다. An analytical method for the determination of flumequine in meats was developed and validated using high-performance liquid chromatography with fluorescence detection. The samples were mixed with sodium sulfate and extracted with ethyl acetate. After clean-up, the residues were dissolved in mobile phase. The calibration curves showed high linearity (r²= 0.9979) within the concentration range of 0.1-1.0 ㎎/㎏. The limit of detection and limit of quantification were validated at 0.005 and 0.017 ㎎/㎏, respectively. The recoveries in fortified meats ranged from 90.8 to 101.1%. The method was then validated in correspondence with the CODEX guidelines for flumequine residue in meats. Herein we monitored 150 samples of meats that were purchased in Korea (Seoul, Busan, Daegu, Daejeon, and Gwangju). Among the tested samples, flumequine was detected in 1 of beef and 1 of pork at levels in the range of 0.048-0.080 ㎎/㎏. Overall, the flumequine residues in the tested samples were within the Maximun residue limit.
