To investigate effects of water temperature on immune response of flounder, Paralichthys olivaceus, against Edwardsiella tarda, fish were immunized with formalin killed E. tarda antigen, and humoral immune response of these fish were observed. At lower water temperature (12 and 15℃), the antibody appeared 2 to 3 weeks after injection of formalin killed E. tarda antigen and the maximum agglutination titer was 16 and 32, respectively. However at higher water temperature (20 and 23℃), the antibody appeared one week after injection and the maximum agglutination titer was about 2,048. Once produced agglutination titer was sensitively responsed to variation of water temperature and showed that this phenomenon had also a similar tendency under natural condition. And it showed that agglatination titer of flounder immunized with formalin killed E. tarda maintained above 19 months.

An epidemic was occurred in Stephanolepis cirrhifer during acclimation in laboratory tanks (water temperature was about 17℃). Diseased fish showed an unique external sign, large cloudy skin surface. We investigated the cause of the disease, and isolated one pure cultured bacterium, that was identified as Vibrio anguillarum by biochemical analysis and sequence analysis of the 16S rRNA and recA genes. During the outbreak, about 30% of S. cirrhifer showed the large cloudy skin surface, and approximately 10% of the stocked fish died. Fish were effectively cured by treatment with oxolinic acid.

We studied two research items that can be undetermined issues in the artificial seed production of the Japanese scallops, Patinopecten yessoensis, batch-specific quality of the reproductive outputs and nursery acclimation, in the embayed waters of Yangyang, Gangwon, Korea. The first batch of the spawner showed better results in terms of survival, growth, and resistance against parasitic ciliate infection over the second batch that was obtained in 5 days after first batch from the same spawner. The early attached spats directly placed in the farming ground (the unacclimated) were resistant against the water current of the open environment, by showing survival of about 80% for a month, a normal survival compared with other results. However, the spat survival during the nursery acclimation was significantly lowered in comparison with that in the unacclimated condition (P<0.01). We discussed the research results of the two items, particularly focusing on their availability for mass seed production for aquaculture in the Korean coastal waters of the East Sea (Sea of Japan).

The barren ground is an abnormal phenomenon of coastal ecosystem in which seaweeds, are destroyed and mostly replaced by the coralline algae containing the calcium carbonate components. To restore the seaweed forest, We have exerted an effort in the local areas, Samchuck, Korea, where barren phenomena are profound. Two methods of seaweed forest construction developed in the present study are underwater longline and seed transplantation for the brown seaweed Costaria costata, a fast growing edible seaweed. The sizes of C. costata attached on the underwater longline were $96.7{\pm}2.2mm$ of blade length and $83.6{\pm}7.7g$ of blade weight in April. Thereafter the sizes declined from May. Similar pattern was obtained from in the transplantation method with maxima of $90.4{\pm}15.8mm$ and $70.1{\pm}31.7g$ for blade length and weight, respectively in April. It appeared totality maturation from two methods in May. This maturation time is the same like that of wild C. costata.
양식넙치에서 분리된 Edwardsiella tarda 의 생화학적 및 혈청학적 특성에 관한 연구

This study was carried out in order to identify the biochemical and serological characteristics of Edwardsiella tarda isolated from cultured flounder, Paralichthys olivaceus in the east and south coast of Korea. During the year of 1990 and 1991, the number of isolated E. tarda were 131 strains. To identify the biochemical characteristics of them kinds of tests were conducted. The results represented that all the strains had the same biochemical characteristics, and their biochemical characteristics were no differences among strains. A serological analysis was carried out based on agglutination test with antiserum belonging to E. tarda serotype a (E-22), b (SU138), c (SU-100) classfied in japan. The selected 10 isolates showed agglutinin titer of 51202560, 40-80 and below 40 against E. tarda serotype a, b and c, respectively. Sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE) profiles of cell proteins of selected 10 isolates were showed no differences in kinds and volumes of proteins among strains.
