        Intragranular and Intergranular Crack Propagation in Nanocrystalline Ni Under Single-Cycle Mode I Loading

        As the first step to study the fatigue crack propagation, the molecular simulations were performed to investigate theopening, propagation and closure behaviors of intragranular and intergranular cracks in nanocrystalline Ni under single-cyclemode I loading. The atomic crack angle was proposed to characterize the crack-tip blunting and sharpening quantitatively. There are obvious different mechanisms between intragranular and intergranular crack propagation. Intragranular crack propagationcould be induced by stacking faults and can change its direction very easily, but the intergranular crack perpendicularto the loading direction would propagate along the grain boundary and encounter strong resistance at the triple junction. New cracks could form in the grain boundaries ahead of the original crack. Different from the traditional understanding forintragranular and intergranular cracks, however, the dislocation density increases even in the unloading process and the cracktip is possibly sharpened temporarily during loading.

        Molecular and cytological analyses of A and C genomes at meiosis in synthetic allotriploid Brassica hybrids (ACC) between B.napus (AACC) and B.oleracea (CC)

        Success of interspecific hybridization relies mostly on the adequate similarity between the implicated genomes to ensure synapsis, pairing and recombination between appropriate chromosomes during meiosis in allopolyploid species. Allotetraploid Brassica napus (AACC) is a model of natural hybridization between Brassica rapa (AA) and Brassica oleracea (CC), which are originally derived from a common ancestor, but genomic constitution of the same chromosomes probably varied among these species through time after establishment, giving rise to cytogenetic difference in the synthetic hybrids. Herein we investigated meiotic behaviors of A and C chromosomes of synthetic allotriploid Brassica hybrids (ACC) at molecular and cytological levels, which result from the interspecific cross between natural B. napus (AACC) and B.oleracea (CC), and the results showed that meiosis course was significantly aberrant in allotriploid Brassica hybrids, and chromosomes aligned chaotically at metaphase I, chromosome bridges and lags were frequently observed from later metaphase I to anaphase II during meiosis. Simultaneously, we also noticed that meiosis-related genes were abruptly down-regulated in allotriploid Brassica hybrids, which likely accounted for irregular scenario of meiosis observed in these synthetic hybrids. Therefore, these results indicated that inter-genomic exchanges of A and C chromosomes could occur frequently in synthetic Brassica hybrids, and provided an efficient approach for genetic changes of homeologous chromosomes during meiosis in polyploid B.napus breeding program.

        Nodule Rich Protein 2 modulates nodule number in Medicago truncatula

        Symbiotic nitrogen fxation is beneft to sustainable agriculture and global nitrogen cycle. Many small peptides were identifed as regulators involving in the interaction between rhizobia and legume. Here we reported Nodule Rich Protein 2 (MtNRP2) encoding a small peptide in Medicago truncatula, belonged to a group of nodule rich protein restricted in legume species. MtNRP2 expressed highly in root nodule and its promoter was active during the initiation and development of root nodule and lateral root. To investigate the function of MtNRP2 in nodulation, we generated MtNRP2-overexpression and MtNRP2- knockdown transgenic Medicago. MtNRP2-overexpression transgenic lines performed normal nodulation phenotype compared with vector control. However, in the MtNRP2-RNAi transgenic plants, the decrease of MtNRP2 expression lead to the increase of infection threads number (7 day post inoculation) and nodules number (3 week post inoculation); meanwhile, the expression of MtRGF3 and MtPUB1 was inhibited. These results suggested that MtNRP2 negatively regulated nodulation in Medicago truncatula.

        The Investigation and 3D Numerical Simulation of Herb Roots in Reinforcing Soil and Stabilizing Slope

        This paper investigated and simulated the mechanical effect of root reinforcing soil and stabilizing slope. The unconfined compression test was used to investigate the reinforcement soil behavior of herb roots. This paper used the 3D finite element to quantify the mechanical effect of the herb root for slope’s stability in terms of the safety factor. The soil was simulated by an elasticplastic model, and the root was simulated by a linear elastic model. Results show that using the Root Content (RC) to estimate the additional cohesion cr of Setaria sphacelata and Orchardgrass was a simple and reliable method. Lateral fine roots can play a more significant role in reinforcing soils and slopes. Compared to various slope angles, the steep slope was less stable. The slope existed a slope angle threshold αthr. When the slope angle α was smaller than αthr, the herb root system can improve the stability of slope. The α existed an optimal angle αopt for the root system. When α = αopt and α < αthr, the herb roots can exert its maximum stabilization capacity. Moreover, we can select herb species with lateral fine roots and more roots in deeper soil layer. Results can better estimate the cr for the herb of lateral fine roots and select the herb and the slope’s geometry in the engineering.

        Synthesis of acid-activated reversible conversion supramolecular nanoplatform: application in drug delivery and anti-tumor activity

        Supramolecular nanoplatforms with stimuli-responsive behavior feature sensitive performance and effective drug delivery, which are desirable as intelligent drug delivery systems. Generally, tumor cells are characterized by excessive acid production, resulting in a lower pH in the tumor microenvironment (pH < 6.5) than in normal tissues (pH ≈7.4) and providing the possibility for the drug delivery system to exploit this decrease in pH as a trigger for drug release. Here, an acid-sensitive supramolecular nanoplatform (CM-β-CD/FC12 +Br− NPs) with assembly/disassembly properties was designed and constructed, which was exploited to capture, deliver, and release anti-tumor compound CSL. 2D NOESY was utilized to examine the host–guest interaction and the potential mechanism for CM-β-CD/FC12 +Br− NPs loading CSL. CM-β-CD/FC12 +Br− NPs present good blood compatibility. Cytotoxicity assay revealed that CSL-loaded NPs display minimal toxicity against normal cells BEAS-2B and good anticancer ability against five human cancer cell lines, especially for Human hepatoma cell line SMMC-7721. In addition, cell apoptosis and cycle assay further verified that CSL-loaded NPs-induced apoptosis in SMMC- 7721 cells up to ~ 93%, as well as blocking the cells in G0/G1 phase and inhibiting the proliferation of SMMC-7721 cells in a dose-dependent manner. We expect that CM-β-CD/FC12 +Br− NPs will be potential acid-answered drug delivery candidates.

        Effects of Artemisia argyi flavonoids on growth performance and immune function in broilers challenged with lipopolysaccharide

        Objective: This research aimed to study the effects of Artemisia argyi flavonoids (AAF) supplemented in diets on the growth performance and immune function of broiler chickens challenged with lipopolysaccharide (LPS). Methods: A total of one hundred and ninety-two 1-d-old broiler chicks were assigned into 4 treatment groups, which were, respectively, fed a basal diet (control), fed a diet with 750 mg/kg AAF, fed a basal diet, and challenged with LPS, fed a diet with 750 mg/kg AAF, and challenged with LPS. Each treatment had six pens with 8 chicks per pen. On days 14, 16, 18, 20 (stress phase I) and 28, 30, 32, 34 (stress phase II), broilers were injected with LPS (500 ㎍/kg body weight) or an equivalent amount of saline. Results: The results demonstrated that dietary AAF significantly improved the body weight (d 21) and alleviated the decrease of average daily gain in broilers challenged with LPS on d 21 and d 35 (p<0.05). Dietary AAF increased bursa fabricius index, and dramatically attenuated the elevation of spleen index caused by LPS on d 35 (p<0.05). Furthermore, serum interleukin-6 (IL-6) concentration decreased with AAF supplementation on d 21 (p<0.05). Diet treatment and LPS challenge exhibited a significant interaction for the concentration of IL-1β (d 21) and IL-6 (d 35) in serum (p<0.05). Additionally, AAF supplementation mitigated the increase of IL-1β, IL-6 in liver and spleen induced by LPS on d 21 and 35 (p<0.05). This study also showed that AAF supplementation significantly reduced the expression of IL-1β (d 21) and nuclear transcription factor kappa-B p65 (d 21 and 35) in liver (p<0.05), and dietary AAF and LPS treatment exhibited significant interaction for the gene expression of IL-6 (d 21), toll like receptor 4 (d 35) and myeloid differentiation factor 88 (d 35) in spleen (p<0.05). Conclusion: In conclusion, AAF could be used as a potential natural immunomodulator to improve growth performance and alleviate immune stress in broilers challenged with LPS.

        An Innovative Collaborative Design of Shape and Performance for Super-High-Contact-Ratio Spiral Bevel Gear

        For upgrading the transmission performance of spiral bevel gear and extending the time between overhauls of the helicopter reducer, an innovative multi-objective collaborative optimization strategy of shape and performance is put up for the super-high-contact-ratio spiral bevel gear transmission with the contact path along tooth width based on ease-off. Firstly, according to the preset contact path curve along tooth width and the symmetrical transmission error curve, the pinion optimal surface satisfying the preset functions is obtained based on ease-off. Secondly, based on the basic principles of simulated annealing and gravitational search, the simulated annealing-gravity search cooperative intelligent optimization algorithm is set up for the calculation of the multi-objective collaborative optimization model of the spiral bevel gear transmission, and the modified surface of pinion satisfying the function requirements is obtained based on the collaborative optimization of shape and performance. Finally, combined with the tooth contact analysis method, meshing impact analysis method and dynamic analysis method, the loaded transmission error, meshing impact and dynamic performance of the super-high-contact-ratio spiral bevel gear transmissions are discussed. The simulation result shows the contact path of the collaborative designed spiral bevel gear transmission is along the tooth width, and the contact ratio reaches the maximum. The loaded transmission error, meshing impact, dynamic load factor and root mean square of vibration acceleration of the collaborative design spiral bevel gear transmission are all smaller than the original design and optimized design spiral bevel gear transmissions. The feasibility of the innovative multi-objective collaborative optimization strategy of shape and performance for the spiral bevel gear is verified, and the super-high-contact-ratio design can greatly cut down the contact vibration of the spiral bevel gear transmissions and better the helicopter’s performance.

        Genome-wide identification and analysis of long noncoding RNAs in longissimus muscle tissue from Kazakh cattle and Xinjiang brown cattle

        Objective: In recent years, long noncoding RNAs (lncRNAs) have been identified in many species, and some of them have been shown to play important roles in muscle development and myogenesis. However, the differences in lncRNAs between Kazakh cattle and Xinjiang brown cattle remain undefined; therefore, we aimed to confirm whether lncRNAs are differentially expressed in the longissimus dorsi between these two types of cattle and whether differentially expressed lncRNAs regulate muscle differentiation. Methods: We used RNA-seq technology to identify lncRNAs in longissimus muscles from these cattle. The expression of lncRNAs were analyzed using StringTie (1.3.1) in terms of the fragments per kilobase of transcript per million mapped reads values of the encoding genes. The differential expression of the transcripts in the two samples were analyzed using the DESeq R software package. The resulting false discovery rate was controlled by the Benjamini and Hochberg's approach. KOBAS software was utilized to measure the expression of different genes in Kyoto encyclopedia of genes and genomes pathways. We randomly selected eight lncRNA genes and validated them by quantitative reverse transcription polymerase chain reaction (RT-qPCR). Results: We found that 182 lncRNA transcripts, including 102 upregulated and 80 downregulated transcripts, were differentially expressed between Kazakh cattle and Xinjiang brown cattle. The results of RT-qPCR were consistent with the sequencing results. Enrichment analysis and functional annotation of the target genes revealed that the differentially expressed lncRNAs were associated with the mitogen-activated protein kinase, Ras, and phosphatidylinositol 3-kinase (PI3k)/Akt signaling pathways. We also constructed a lncRNA/mRNA coexpression network for the PI3k/Akt signaling pathway. Conclusion: Our study provides insights into cattle muscle-associated lncRNAs and will contribute to a more thorough understanding of the molecular mechanism underlying muscle growth and development in cattle.
