        사료원료 공급량 및 스크류 회전속도를 달리하여 제조한 배합사료가 넙치(Paralichthys olivaceus)의 성장 및 체조성에 미치는 영향

        The aim of this study was to investigate the effect of diet extruder conditions, such as feed-loading rate and screw speed, on growth performance and biochemical responses in olive flounder Paralichthys olivaceus. Over 8 wks, we used four identical diets (triplicated per treatment) with differing ratios of feed-loading rate (kg/h):screw speed (rpm/min) in a laboratory-scaled twin-screw extruder of 50:640, 80:640, 120:640, and 80:400, designated as EP1, EP2, EP3, and EP4, respectively. Screw speed impacted the buoyancy of experimental diets. Diets produced at a screw speed of 640 rpm/min floated for > 24 hrs, whereas those produced at a speed of 400 rpm/min sank between 10 s and 5 min. Fish that were fed EP1 and EP4 diets grew significantly faster than those fed EP2 and EP3 diets. Fish fed EP1 diets ate and gained weight most efficiently among treatments, a result that is likely to be related to feed-loading rate, i.e., ingredients extruded at a low feed-loading rate may have more time to cook in the pre-conditioner of the extruder. A cooked diet may be easier to digest in fish. Fish fed EP4 diets also showed significant weight gain, as compared to those fed EP2 and EP3 diets. However, we found no differences among treatments in proximate compositions of dorsal muscle, liver, and viscera of fish. Our results suggest that extruder conditions, may influence feed quality, impacting feed efficiency and growth of fish.

      • 황놀래기,Pseudolabrus japonicus (HOUTTUYN)의 生殖周期와 性轉換

        1990년 12월부터 1991년 10월까지, 그리고 1992년 7월부터 11월까지 제주도 북방합덕연안을 중심으로 채집된 황놀래기, Pseudolabrus japonicus를 대상으로 그들의 生殖生態를 파악하기 위하여 體色變化와 生殖巢熟度指數, 그리고 生殖巢의 조직학적 방법에 의한 生殖週期, 性轉換 過程등을 조사한 결과는 다음과 같다. 1. 생식소 숙도지수(GSI)는 암수 다같이 8월에 접어들면서 상승하기 시작하며 10월에 최대 치를 나타내고, 12월에 접어들면서 하강하기 시작하여 1월부터 낮은 값을 나타내고 있다. GSI최대값은 암컷 2.32(±0.35), 수컷 0.38(±0.18)을 나타냈다. 2. 生殖週期는 成長期(8∼9월), 成熟期(9∼10월), 完熟 및 産卵期(10∼12월), 回復 및 休止期(12∼7월)등의 연속적인 주기를 나타냈다. 3. 體長 11.5∼16.0㎝되는 개체들에서 雌性에서 雄性으로 性轉換 단계의 生殖巢를 갖고 있다. 4. 본 種은 1차정소와 2차정소가 혼합되어 나타나는 複雄性의 雌性完熟魚에 속하는 것으로 나타났다. 5. 암컷에서 수컷으로 性轉換 함에 따라 적색에서 청색으로 體色變化가 나타나고 있다 Reproductive cycle and sex reversal of Pseudolabrus japonicu s were investigated histologically on the gonadal development, sex change of the gonad and studied by gonadosomatic index(GSI), color patterns. Sample were collected at the costal area of Ham duck in Cheju-do, from December, 1990 to November 1992. In male and female, GSI began to increase from August and reached it's maximum value in October, respectively. It began to decrease from December, the low temperature season, thereafter, maintained relatively low values until July of next year. The annual reproductive cycle of this species could be classified into four sucessive developmental stage : growing stage(August-September), mature stage(September-October), ripe and spent stage(October-December), degenerative and resting stage(December-July). The species is a diandric, protogynous hermaphrodite, and has two color patterns: red-yellow (A) and dark-green (B). A-fish were less than 14.5㎝ SL and consisted of females (92.7%), secondary male(80.2%), primary male(5.5%), female(5.5%) and fish with transitional gonad(8.8%). In P.japonicus, individuals with hermaphrodic gonad were observed in 11.5∼16.0㎝ SL. Sex reversal of this species could be occured from 11.5 to 16.0㎝ SL.

        Identification and Characterization of a Bacteriocin Produced by an Isolated Bacillus sp. SW1-1 that Exhibits Antibacterial Activity against Fish Pathogens

        The selected isolate, Bacillus sp. SW1-1 showed antibacterial activity against both Gram-positive and Gram-negative bacteria involved in fish diseases, including Edwardsiella tarda, Streptococcus iniae, S. parauberis, Vibrio anguillarum, and V. harveyi. The Maximum bacteriocin production was observed at $30^{\circ}C$ after 24 h with brain heart infusion medium (pH 7.0). The bacteriocin SW1-1 was purified by 50% ammonium sulfate precipitation, followed by HiPrep diethylaminoethyl 16/10 FF and Sephacryl S-100 High resolution column chromatography. The substance was characterized as a bacteriocin-like inhibitory substance with a molecular mass of 38 kDa. Bacteriocin SW1-1 was sensitive to the proteolytic action of pepsin, trypsin, chymotrypsin, and protease types I and XIV, and relatively heat labile, despite the fact that bacteriocin activity was still detected after heating at $100^{\circ}C$ for 30 min. The activity of bacteriocin SW1-1 was stable in the pH range of 2.0-11.0, and relatively unaffected by organic chemicals. The bacteriocin SW1-1 had a bacteriolytic mechanism, resulting in cell wall degradation of E. tarda. These characteristics indicate that this bacteriocin may be a potential candidate for alternative agent to control important pathogens of fish diseases in aquaculture.

        물가자미 ( Eopsetta grigorjewi ) 의 정자변태 및 정자 미세구조

        The roundnose flounder`s (Eopsetta grigorjewi) spermiogenesis and fine structure of the spermatozoon were examined by means of the scanning and transmission electron microscopy. During the spermiogenesis, the chromatin of the spermatid became fine granular form, and progressively condensed into many large globules, finally homogeneously condensed in the spermatozoan head. The main characteristics of the spermiogenesis were the disappearance of Golgi complex, the appearance of microfilament, the reduction of mitochondria and the appesrance of lysosome in the cytoplasm. A spermatozoon consisted of head and tail, but the acrosome was absent The cytoplasmic collar containing seven mitochondria was observed in the posterior part of the head: The well-developed axonemal lateral fins were observed in the tail. The cross section of the axial filament showed 9+2 axonemal structure of microtubules, and the numerous vesicles were observed in the cytoplasm.

        사료 내 미역 (Undaria pinnatifida) 당단백질의 첨가가 넙치 (Paralichthys olivaceus) 치어의 성장 및 면역 증강에 미치는 영향

        This study evaluated the effects of adding sea mustard Undaria pinnat 4/ida glycoprotein to the diet of juvenile olive flounder Paralichthys olivaceus on its growth, and levels of insulin-like growth factor I (IGF-l), IGF binding proteins (IGFBPs), and interleukins. Three experimental diets (U0, UO.5, and U 1.0) were formulated that contained different amounts of an extract of U. pinnat4flda (0, 0.5, and 1.0%, respectively), Experimental groups were established in triplicate (30 fish/group) and fed for 12 weeks, The experimental group fed 1.0% added U pinnatjflda glycoprotein had the greatest rate of weight gain, which differed significanily from the other experimental groups. SDS-PAGE of the plasma IGF-I and muscle protein showed that the experimental groups taking U pinnatjflda glycoprotein had significantly more IGF-1 and a ca. 200kDa protein, as compared to the control group. In addition, the amount of IGF`BP-3 at ca. 43kDa increased in the group given the U. pinnatiiflda extract, as compared to the control group. The interluckin-2, -4, -6, and-12 levels paralleled the level of growth factor in the groups given the U. pinnat i/Ida extract. In conclusion, supplementing the diet of olive flounder with U. pinnatifida glycroprotein improved its growth and immunity.

        단백질 및 지질함량이 다른 배합사료와 생사료로 사육한 넙치의 육질평가

        This study was conducted to investigate the effect of extruded pellets (EP) containing different levels of protein (51%-55%) and lipid (9%-15%) on flesh quality of flounder (Paralichthys olivaceus) comparing with raw fish-based moist pellet (MP). Two replicate groups of 40 fish per each tank (initial mean weight 106 g) were fed one of three experimental EP (EP1, EP2 and EP3) containing different protein and lipid levels, a commercial EP (EP4) and MP for 16 weeks. Moisture content of fish fed MP was significantly higher than that of fish fed all EP. Significantly higher contents of 16:1n-7, 20:4n-6, 20:5n-3 and lower 18:2n-6 were observed in fish fed EP4 and MP compared with fish fed EP1, EP2, EP3. The 22:6n-3 content was not significantly different among all groups. Taurine content of fish fed MP was significantly higher than that of fish fed all EP. Asparagine content of fish fed EP1 was significantly higher than that of fish fed EP2, EP, EP4 and MP. Significant difference were observed in lysine and serine contents of fish fed experimental diets. In nucleotides and their related compounds, ATP and AMP content was not affected by diets. Significant difference were observed in IMP contents of fish fed experimental diets. Textural properties, no significant difference was observed among the fish groups fed different diets.

        Four Members of Heat Shock Protein 70 Family in Korean Rose Bitterling (Rhodeus uyekii)

        Heat shock protein (HSP) 70, the highly conserved stress protein families, plays important roles in protecting cells against heat and other stresses in most animal species. In the present study, we identified and characterized four Hsp70 (RuHSP4, RuHSC70, RuHSP12A, RuGRP78) family proteins based on the expressed sequence tag (EST) analysis of the Korean rose bitterling R. uyekii cDNA library. The deduced RuHSP70 family has high amino acid identities of 72-99% with those of other species. Phylogenetic analysis revealed that RuHsp70 family clustered with fish groups (HSP4, HSC70, HSP12A, GRP78) proteins. Quantitative RT-PCR analysis showed the specific expression patterns of RuHsp70 family members in the early developmental stages and several tissues in Korean rose bitterling. The expression of 4 groups of Hsp70 family was detected in all tested tissue. Particularly, Hsp70 family of Korean rose bitterling is highly expressed in hepatopancreas and sexual gonad (testis and ovary). The expression of Hsp70 family was differentially regulated in accordance with early development stage of Rhodeus uyekii.

        각시붕어 , Rhodeus uyekii 의 생식주기

        1991년 8월부터 1993년 7월까지 경상남도 김해군 상동면 매리마을의 낙동강 지류에서 채집된 한국 특산어인 각시붕어, Rhodeus uyekii를 대상으로 생식생태를 파악하기 위하여 연간 생식소숙도지수(GSI)와 난경조성, 산란관 길이 변화를 조사하였고, 이들 생식소 발달과정을 광학현미경적 방법에 의해 조사하였다. GSI는 수온이 상승하기 시작하는 2월부터 증가하기 시작하여 5월에 연중 최대값을 나타내고, 7월 부터 하강하여 8월에는 연중 최저값을 나타내었다. 이후 9월부터 11월까지 서서히 증가하다가 12월에는 일시적으로 증가를 멈추고 11월과 비슷한 수준을 유지하였다. 생식연주기는 7∼8월의 퇴화 및 휴지기, 9∼11월의 1차 성장기, 12월의 정체기, 1,2월의 2차 성장 및 성숙기, 3∼6월의 완숙 및 산란기 등 연속적인 주기로 구분할 수 있었으며, 산란성기는 4월 하순에서 5월이었다. The reproductive cycle of the bitterling, Rhodeus uyekii was studied to observe the annual variations of gonadosomatic index(GSI), size frequency distribution of egg, ovipositor length and histological changes of gonad. GSI began to increase from February when the water temperature started to increase, and reached the maximum value in May, whereas it began to decrease from July and reached the minimum value in August which in the highest water temperature season. It began to incerase again but showed low value from September to November. The GSI remained stable thereafter. Monthly changes in GSI, ovipositor length, frequency of egg diameter and gonadal histology showed that the annual reproductive cycle was classified into the following successive phases : primary growing phase from September to November, quiescent phase in December, secondary growing and mature phase from January to February, ripe and spawning phase from March to June, and recovery and resting phase from July to August.
