5-Fluorouracil이 백서의 섬유모세포에 미치는 세포 독성에 관한 연구

Present study was carried out to investigate the cytotoxicity of 5-Fluorouracil(5-FU) on cultured rat fibroblasts. The colorimetric assay of neutral red(NR) and tetrazolium MTT, the measurement of the rate of DNA synthesis and sister chromatid exchange(SCE) analysis were performed on the fibroblasts cultured in the media containing various concentrations of 5-FU. The obtained results are as follows: 1. The NR and MTT values were increased dose-dependently. 2. The NR90, NR50, MTT90, and MTT50 values of 5-FU were 5ug/ml, 35ug/ml, 7ug/ml and 47ug/ml, respectively. 3. The rate of DNA synthesis at concentrations of 5-FU, 5×10??ug/ml, 5×10??ug/ml, 10ug/ml and 35ug/ml values were 90%, 50%, 20% and 10%, respectively. 4. The frequency of sister chromatid exchange increased by 2 times at concentration, 10ug/ml of 5-FU. 5. 5-FU showed the toxic effect in the orders of DNA, lysosome, mitochondria. 6. 5-FU did not show drug-specific morphological changes in fibroblasts. From above the results, it suggests that 5-FU induced the DNA damage and chromosome damage at below concentrations which dose not affect lysosomal function and mitochondrial enzyme activity.
