        Phenylalanine Ammonia Lyase and Cinnamic Acid 4-HydroxylaseActivities of Rice and Pepper in response to UV and Wounding

        The metabolites related to phenylpropanoid pathway play an important role in the self-defense of plants and induced by environmental stress like wounding, pathogen attack, UV-irradiation and so on. The mRNA level of rice phenylalanine ammonia lyase (PAL) was increased at 12 h to 48 h, however it was gradually decreased 48 h to 60 h after UV irradiation. The PAL enzyme activities in rice were peaked at the time of 24 h after UV irradiation, on the other hand, it was not affected by wounding. The PAL enzyme activities in pepper were raised high at 24 h and 10 h by UV irradiation and wounding respectively. The cinnamic acid 4-hydroxylase (C4H) activities were increased by wounding treatment and were detected from 12 h to end time point of experiment, while UV-irradiation didn't affect the C4H activity in rice and pepper. These results were assumed that the action of isoflavonid has an alternative effect on the defenses which include wounding and UV irradiation and on the diverse roles in rice and hot pepper.Key words - cinnamic acid 4-hydroxylase (C4H), pepper, phenylalanine ammonia lyase (PAL), rice 고추에서 UV와 상처가 PAL 및 C4H 효소 활성에 미치는 영향김미영·윤용휘·이정훈·김학윤1·신동현*·이인중·김달웅·김길웅(경북대학교 농대 농학과, 1계명대학교 환경학부)Phenylpropanoid pathway 생성물질은 식물의 self-defense에 관계하며 이러한 물질들은 UV뿐 아니라 wounding, pathogen과 같은 environmental stress에 의해 생성되는 것으로 알려져 있다. 벼에서 PAL mRNA 는 UV 조사 후 12시간에서 48시간까지는 증가하였으나 48시간부터 60시간까지는 점점 줄어드는 경향을 보였다. 한편 PAL의 활성은 UV 조사 후 24시간에서 가장 높았지만 상처에 의해서는 PAL의 활성이 벼에서는 증가하지 않았다. 그러나 고추에서는 UV조사와 상처를 준 후 24시간과 10시간에서 각각 높은 활성을 나타내었다. 벼와 고추 모두 cinnamic acid 4-hydroxylase의 활성은 상처를 준 후 12시간에서 증가하였지만 UV 조사는 C4H 활성에 영향을 주지 않았다. 이러한 결과로 볼 때 벼와 고추에서는 UV 조사와 상처가 모두 PAL, C4H 효소활성에 영향을 주는 것으로 나타났다.

        공과대학생의 온라인 학습에서 자기조절 학습 능력 및 학습 양식과 학습참여도와의 관계

        The Relationship among Self-regulated Learning Ability and Learning Style and Degree of Learning Participation in On-line Learning of Engineering University Student 이 연구의 목적은 4년제 공과대학 학생들의 자기조절학습능력과 학습유형을 조사하고, 자기조절학습능력과 학습유형간의 관계를 분석하며, 이들이 온라인 학습환경에서 학습자들의 참여도, 만족도에 어떠한 영향을 미치는지를 파악하는 것이다. 연구의 대상으로 4년제 대학교 정규과정 1개반을 선정하여 온라인 학습 환경을 설계한 후 한학기 동안 운영하고 학습참여도를 확인하기 위해 시스템 로그 파일과 게시판 게시횟수를 분석하였다. 연구 결과로서 자기 주도 학습 능력과 학습 참여도간에는 정적인 상관관계가 나타나고 학습양식과 학습참여도간에는 통계적으로 유의하지 않았다. 또한 자기조절학습 능력과 학습양식 사이에는 통계적으로 유의미 하지 않았다. 학습양식은 연구자가 기대한 수렴자와는 달리 분산자가 63.4%로 높게 나타났다.

        고령자용 저작 용이 가래떡 제품의 개발 및 특성 연구

        This study was conducted to develop easily chewable Korean rice cake (Garaedduk) for the elderly while maintaining its original form and flavor. We developed two types of easy-to-chew Garaedduk products by adding starch-degrading enzyme or trehalose, respectively. Characteristics of Garaedduk products were investigated and comparative analysis was carried out between control and experimental groups. The water content of control and enzyme-added Garaedduk was 43.55% and 44.11%, respectively, which was significantly higher than trehalose-added Garaedduk (40.30%) as free water content was reduced by the formation of hydrogen bonds between trehalose and water molecules. Due to the browning of reducing sugar produced by the decomposition of rice starch, Hunter b-value of enzyme-added Garaedduk was significantly higher compared to others. Hardness, adhesiveness, gumminess, and chewiness of experimental groups were lower than the control group. Consumer test scores showed significant differences with respect to overall liking, chewiness, and swallowing between control and experimental groups. Elderly preferred experimental Garaedduks over control and the experimental groups were evaluated to be softer, easily chewable, and swallowable.

        의사의 진료설명 이해 용이성에 영향을 미치는 환자의 특성

        Purpose:The purpose of this study was to identify the patient's characteristics influencing patient's comprehensibleness of doctor's explanation. Methods: This study was conducted as a secondary data analysis using the raw data of The 6th Korea National Health and Nutrition Examination Survey (KNHANES VI) conducted in 2015. The survey sampled 22,948 adults, and 4,469 of them were included in this analysis. Multiple regression analysis was performed to determine the patient's characteristics influencing patient's comprehensibleness of doctor's explanation. Results: The patient's comprehensibleness of doctor's explanation were more higher as age (t=5.65, p<.001), female (t=4.40, p<.001), subjective good health status (t=3.48, p=.001) were higher. On the other hand, the patient's comprehensibleness of doctor's explanation were more higher education level (t=-6.80, p<.001), not-recent outpatient experience (t=-6.04, p<.001), pain/discomfort (t=-2.64, p=.008), anxiety/depression (t=-2.58, p=.010) were lower. Conclusion:It is necessary to be provided the patients who are higher education level, pain/discomfort and anxiety/depression with applying the doctor's detailed explanation and intervention programs.

        王陽明의 心學 이론으로 본 한국전통춤의 私欲과 天理體認+

        이 글의 목적은 전통춤의 실천적 행위에서 강조되는 ‘마음[心]’과 ‘체인’의 특징을 왕양명(王陽明)의 심학(心學) 이론을 바탕으로 설명하는 것이다. 전습록(傳習錄)에 전하는 ‘심’과 ‘체인’ 및 그의 예술 이론을 전통춤계의 대가들이 강조했던 말들과 교차적으로 살펴보며 그 특징을 공론화할 수 있는 언어 로 드러내고자 한다. 왕양명의 심학이론으로 보면, 전통춤의 실천적 행위의 테제인 “마음이 고와야 춤이 곱다”에서 마음 이 고운 것은 ‘마음에서 정명(精明)한 양지(良知)가 발현된 상태’이고, ‘춤이 곱다’는 것은 움직임의 ‘중 (中)’과 희노애락의 ‘미발지중(未發之中)’이 구현된 상태라고 말할 수 있다. 특히 홀로 알고 있는[獨] 마음의 역에서 늘 ‘진정성[誠意]’을 추구해야 한다. 이러한 언어들을 통해 한국전통춤의 내외적 가치 및 의미를 드러낼 수 있다. “유효한 실천은 그것에 관한 이론에 선행한다.”라는 말이 있다. 전통춤의 유효한 실천은 이미 오랜 옛날부터 있어왔다. 그러나 그 실천에 관한 이론은 부족하다. 오랜 옛날부터 늘 우리 곁에 있어 온 유효 한 실천을 이론적으로 설명하기 위해 이 글을 통해 한 땀의 시침질을 해 보았다. 이 글은 최초로 전통춤계 대가들이 강조했던 춤 행위자의 마음과 체인을 연구의 대상으로 삼아, “마 음이 고와야 춤이 곱다”는 전통춤의 테제를 이론적으로 설명하고자 했다는 데에 의의가 있다. 특히 왕 양명의 심학을 바탕으로 전통춤의 마음 바탕론과 체인론에 접근했다는 점에서 큰 의미가 있다. The purpose of this article is to explain the characteristics of mind-heart and acknowledgement, emphasized in the practice of traditional korea dance, based on the theory of Wang Yangmyung's psychology. We examine the mind-heart, acknowledgement, and the art theory in JJeonseubrok(the tradition records)along with the words emphasized by the masters of the traditional dance, and express them with a language that can be publicized. According to Wang Yangmyung's theory of psychology, we can say that what good mind stands for in "Good dance comes from good mind" is the state where the rightness is embodied in mind and that 'good dance' means the state where 'non-embodiment' of movement and 'non-embodiment' of emotion is realized. In particular, we must always pursue 'sincerity' in the domain of mind-heart, which we know alone. Through these languages, the internal and external value and meaning of Korean traditional dance can be revealed. There is a saying that "effective practice precedes theories of it." Effective practice of traditional dance has existed for a long time. However, the theory of its practice is not sufficient. This article attempts to theoretically explain the effective practice that has always been with us since a long time ago. This article has its significance in that it tries for the first time to theoretically explain the thesis of traditional dance, "Good dance comes from good mind" by using the mind-heart and the acknowledgement of dancer which was emphasized by traditional dancing masters. In particular, it is meaningful that it approached the theory of mind and acknowledgement theory of traditional dance based on the study of Wang Yangmyung.

        휴면아 경정 배양법을 통한 포도 '거봉' 에서 Grapevine fleck virus의 제거

        본 연구진은 세계 최초로 GFkV에 단독 감염된 포도 품종 거봉의 휴면아로부터 직접 경정 분열조직을 절취하여 배양함으로써 별도의 열처리나 화학처리 없이 GFkV가 제거된 신초 재생에 성공하였다. GFkV는 포도 식물체의 체관에만 한정적으로 존재하고 접목으로 감염되는 포도 바이러스이다. 경정조직은 $4^{\circ}C$에서 일정기간 저장된 1년생 경화가지의 휴면아로부터 0.3 mm (73 절편체)와 0.8 mm (5절편체)를 절취하였는데, 절취 부위는 생장점(apical meristem)을 포함하여 2 ~ 5개의 엽원기(leaf primordia)와 미분화 원기(uncommitted primordia) 1 ~ 4개를 포함하였다. 절취된 경정조직을 BA 3.0 mg/L와 IBA 0.1 mg/L가 혼합된 배지에 16주간 배양한 결과, 0.3 mm와 0.8 mm의 경정조직에서 각각 4.1%와 40.0%의 신초 재생률이 관찰되었고, 재생된 신초에서의 바이러스 제거율(재생된 신초수에 대한 RT-PCR negative 신초수의 백분율)은 0.3 mm의 경정조직에서는 100%를, 0.8 mm에서는 50%를 나타내었다. 신초를 증식시킨 후 감염바이러스를 다시 진단한 결과, 신초가 처음 재생되었을 때의 진단결과와 동일하였다. 휴면아로부터 분열조직을 배양하여 바이러스가 제거된 신초를 확보한 예는 세계적으로 보고된 바가 없는 것으로서, 본 연구진의 새로운 바이러스 제거방법은 향후 포도 뿐만 아니라 과수를 포함한 낙엽성 수목의 무병묘 생산에 매우 유용하게 활용될 수 있을 것이다. Herein, we report the meristem-tip culture from dormant buds of grape 'Kyoho' single-infected with Grapevine fleck virus (GFkV), which is phloem-limited and transmitted by graft inoculation. We produced GFkV-free shoots without thermo- or chemotherapy using meristem-tip explants approximately 0.3 mm (73 explants) and 0.8 mm long (five explants) including shoot apical meristem, 2-5 leaf primordia, and 1-4 uncommitted primordia from dormant buds of the infected woody cuttings (stored at $4^{\circ}C$). Explants were cultured on Murashige and Skoog (MS) medium supplemented with 3% sucrose, 3.0 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-butyric acid (IBA). After 16 weeks of culture, shoot (10-mm long) regeneration frequency achieved from 0.3-mm explants was 4.1% and that obtained from 0.8-mm explants was 40.0%. Virus-free efficiency (expressed as the percentage of RT-PCR negative shoots regenerated) from 0.3- and 0.8-mm explants was 100% and 50%, respectively. Following in vitro multiplication, RT-PCR assays revealed identical results to assays of the first regenerated shoots. Our new methodological approach could be applied for eliminating other viruses in grapevines, as well as for producing virus-free plants in many other deciduous tree species, including fruit trees.

        CER 가격에 따른 도서지역용 MG 발전사업자의 연평균수익률 분석

        Islands that is far away from onshore have used internal-combustion engine driving alternators to secure electric power. However, with Paris convention on climate change, there is a growing interest in eco-friendly energy independent island that replaces microgrid(MG) consisting of photovoltaic, wind power, and energy storage system with alternators for internal combustion engines. And also, national emission trading has been implemented, and the price of certified emission reduction(CER) has a influence on system marginal price(SMP). Because, the low cost generation source decides SMP when CER price is low, while the more expensive generation source decides SMP when CER price is high. Therefore, the increasing of CER price has a strong influence on the annualized return of MG independent power producer. Moreover, the fixed RPS price is implemented from 2017, which the increasing of CER price under the fixed RPS price or the variable RPS price affects annualized return differently. In this paper, the annualized return according to CER price is analyzed for large, middle, and small scaled islands, the effect of CER price on the annualized return was confirmed.

        Metabolic activities affect femur and lumbar vertebrae remodeling, and anti-resorptive risedronate disturbs femoral cortical bone remodeling

        Metabolic activities are closely correlated with bone remodeling and long-term anti-resorptive bisphosphonate treatment frequently causes atypical femoral fractures through unclear mechanisms. To explore whether metabolic alterations affect bone remodeling in femurs and lumbar vertebrae and whether anti-osteoporotic bisphosphonates perturb their reconstruction, we studied three mouse strains with different fat and lean body masses (BALB/c, C57BL6, and C3H mice). These mice displayed variable physical activity, food and drink intake, energy expenditure, and respiratory quotients. Following intraperitoneal calcein injection, double calcein labeling of the femoral diaphysis, as well as serum levels of the bone-formation marker procollagen type-I N-terminal propeptide and the bone-resorption marker C-terminal telopeptide of type-I collagen, revealed increased bone turnover in mice in the following order: C3H > BALB/c ≥ C57BL6 mice. In addition, bone reconstitution in femurs was distinct from that in lumbar vertebrae in both healthy control and estrogen-deficient osteoporotic mice with metabolic perturbation, particularly in terms of femoral trabecular and cortical bone remodeling in CH3 mice. Interestingly, subcutaneous administration of bisphosphonate risedronate to C3H mice with normal femoral bone density led to enlarged femoral cortical bones with a low bone mineral density, resulting in bone fragility; however, this phenomenon was not observed in mice with ovariectomy-induced femoral cortical bone loss. Together, these results suggest that diverse metabolic activities support various forms of bone remodeling and that femur remodeling differs from lumbar vertebra remodeling. Moreover, our findings imply that the adverse effect of bisphosphonate agents on femoral cortical bone remodeling should be considered when prescribing them to osteoporotic patients.
