      • Streptozotocin 유도 당뇨쥐에서의 Phospholipase A₂, Cyclooxygenase 활성과 Thromboxane 및 Prostacyclin합성

        당뇨병에서의 혈전생성 기전에 관련된 주된 요인을 관찰코져 흰쥐를 4주간 사육한 후 streptozotocin(STZ)으로 당뇨를 유발한 후 6일째에 희생하여 당뇨쥐에 있어서의 phospholipase A₂ 활성변화에 따른 조직의 과산화적 손상과 혈소판 응집능 등의 변화를 관찰한 결과는 다음과 같다. 체중증가는 STZ를 투여한 후에는 STZ 투여군에서 현저하게 감소하였다. 식이섭취에 있어 STZ injection 후, DM군은 16% 증가하였다. 식이효율은 STZ injection 후 DM군은 정상군에 비해 유의적으로 감소하였다. 혈청 중의 총 지질과산화물가는 정상군에 비해 DM군은 2.5배 높았다. HDL 중의 TBARS는 정상군에 비해 DM군은 약 1.6배 정도 높았고 LDL 중의 TBARS는 정상군에 비해 DM군에서 4.8배의 높은 증가를 보였다. 혈소판 phospholipase A₂ 활성은 DM군은 정상군에 비해 42% 증가하였다. 혈소판 cyclooxygenase 활성은 정상군에 비해 DM군이 2배 정도 높은 수준이었다. 혈소판 중의 TXA₂ 농도는 정상군과 비교하여 DM군에서 169%로 현저하게 높았고 대동백 PGI₂의 농도는 정상군에 비해 DM군은 낮은 수준을 보였으며 대동맥 PGI₂와 혈소판 TXA₂의 비는 DM군에서 정상군에 비해 55% 낮았다. 결론적으로 STZ 유발 당뇨쥐에 있어서는 혈청 지질과산화물 수준의 증가와 더불어 phospholipase A₂ 활성이 증가되었으며 따라서 cyclooxygenase가 유도하는 AA cascade 활성화에 의해 TXA₂ 생성증가 및 PGI₂/TXA₂ ratio의 감소가 관찰되었다. 이러한 결과는 당뇨병 질환에서 나타나는 동맥경화증, 심혈관계 질환 등의 여러 혈관계의 병리적 현상들이 당뇨 상태에서의 지질대사 이상으로 인한 지질과산화물의 증가와 그의 독성으로 인한 여러 혈소판 관련 인자들의 활성화와 밀접하게 연관되어 나타나는 것을 시사하는 것이라 하겠다. The relation between lipid peroxidation and thrombotic reaction were investigated in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 100±10gm were randomly assigned to normal and STZ-induced diabetic group(DM). Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of STZ in citrate buffer(pH 4.3) after 4 weeks feeding of basal diet. Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in diabetic group after STZ injection. Serum levels of thiobarbituric acid reacting substances(TBARS) that were markedly increased in DM group compared with of normal group. TBARS levles of HDL and LDL were similar patterns to total TBARS of serum. Activities of platelet phospholipase A₂(PLA₂) were higher in diabetic group than those of normal group. Activities of platelet cyclooxygenase were 106% in DM group than normal group. Platelet thromboxane A₂(TXA₂) formation was increased in DM group than normal group. Production of aortic prostacyclin(PGI₂) was lower in diabetic group than that of normal group. PGI₂/TXA₂ ratios were decreased by 55% in DM groups than those of normal group. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stess which leads to acceleration of lipid peroxidation and platelet aggregability. In conclusion, accelerating effect of lipid peroxidation and thrombogenesis in diabetic state is regarded to be resulted from enhancement of PLA₂ activity and arachidonic acid metabolism, inhibition of antiaggregating agent and aortic PGI₂ formation.

        Bronchial carcinoid tumors are relatively uncommon neoplasms that are considered to be malignant tumors of low to intermediate grade. They are classified by pathologic features as typical or atypical carcinoids and have distinctly different prognoses and therapeutic options. Surgery is the treatment of choice in typical and atypical carcinoid tumors but the approach has been changing. Recently, several studies have described experiences using other technologies as adjuncts to bronchoscopic resection, technologies such as laser and cryotherapy with curative intent in endoluminal typical carcinoids. Here we present a case of atypical bronchial carcinoid that was treated with bronchoscopic cryotherapy.

        Tiny-DB와 MySQL을 이용한 유비쿼터스 센서 네트워크 기반의 실시간 정보 서비스 설계 및 구현

        무선 센서 네트워크는 컴퓨팅 기술 능력과 무선 통신 능력을 지난 센서 노드들을 이용하여 자율적인 네트워크를 형성하고, 서로 간에 정보를 전달한다. 최근 센서 네트워크의 노드의 초저전력, 초소형, 저비용에 대한 연구와 더불어 수집된 센싱 데이터의 활용 방안에 대한 연구가 진행되고 있다. 이에 본 논문에서는 Tiny-DB와 MySQL를 연동하여 센서 네트워크에 수집된 실시간 대용량 데이터를 효과적으로 저장하고, 산불이나 화재 등의 재난을 방지하기 위해 IPv4/IPv6 기반의 인터넷을 이용하는 실시간 경보 서비스의 프로토타입을 설계하고 구현한다. 이를 위해 TinyOS를 이용하여 센싱 쿼리 프로세싱을 통해 정보를 추출하는 모률, MySQL 데이터베이스 관리 시스템, JDBC를 이용하여 정보를 서버의 데이터베이스의 연동 모듈, IPv4/IPv6 기반에 인터넷에서 ODBC를 이용하여 실시간으로 센싱 데이터를 수신하여 사용자에게 제공하는 클라이언트 모듈 등을 개발한다. Wireless sensor network forms the self-organization network, and transfers the information among sensor nodes that have computing technology ability and wireless communication ability. The recent sensor network is study for low-power, micro, low cost of node is proceeded. Recently, the research of application services in wireless sensor networks is proceeded. Therefore, in this paper, we design the prototype of the real-time information service that support a user the information of temperature, illumination etc. And, we implement the alarm application service for the disaster prevention on Internet base on IPv4/IPv6. We develop the module of the extract information using the query processing based on TinyOS, and the module of the server's database using MySQL data base management system and JDBC. Additionally, we develop the client module that receive the real-time sensing data using ODBC in Internet based on IPv4/IPv6.

        Changes in hippocalcin expression in cortical neurons and glial cells by epigallocatechin gallate administration in an animal model of stroke

        Ischemic stroke causes brain damage and neuronal cell death by depriving oxygen and nutrients and releasing excessive levels of glutamate and intracellular calcium. Epigallocatechin gallate (EGCG) is a polyphenolic compound present in green tea. It has antioxidant, anti-inflammatory, and neuroprotective effects. Hippocalcin is a calcium binding protein that regulates calcium concentration, neuronal differentiation, neuronal excitability, and neuronal cell death. In this study, we investigated whether EGCG regulates the expression of hippocalcin in neurons and astrocytes after focal cerebral ischemia. Cerebral ischemia was induced by meddle cerebral artery occlusion (MCAO). EGCG (50 mg/kg) or PBS was injected into the abdominal cavity just before MCAO surgery. Neurobehavioral tests were performed to evaluate the effect of EGCG on neurological behavioral deficits 24 h after MCAO surgery. Immunofluorescence staining was performed to evaluate the positive response to hippocalcin in the cerebral cortex after MCAO surgery. We also detected the positive reactions of neuronal nuclear protein (NeuN) and glial fibrillary acidic protein (GFAP) as markers of neuron and astrocyte, respectively. MCAO caused severe neurological impairment and EGCG treatment attenuated these impairments. MCAO damage reduced the number of NeuN-positive cells and increased the number of GFAP-positive cells. This result indicates a decrease in neurons and an increase in astrocytes. However, EGCG alleviated these changes caused by MCAO damage. MCAO reduced the number of hippocalcin-positive cells in neurons and astrocytes, and EGCG treatment attenuated these reductions. Hippocalcin exerts neuroprotective effect through regulating intracellular calcium concentration. In conclusion, EGCG regulates the expression of hippocalcin in neurons and astrocytes and has neuroprotective effects in focal cerebral ischemia.

        <P>Infinite rectangular-tubular helices, [MX2L](M = Zn(II), Hg(II); X- = Cl-, Br-; L = bis(3-pyridyl)cyclotetramethylenesilane), have been efficiently constructed via the combined effects of the potential flexible conformers of L and the tetrahedral geometry of M(II) ions. This helical molecular system affords a racemic mixture of P- and M-helices in a crystal. The helical pitches (7.8934(4)-8.1560(2) angstrom) that are sensitive to the nature of M(II) ions and halide anions are attributable to subtle change in the flexible dihedral angles between the two pyridyl groups around Si and the M(II) hinges. Their photoluminescence intensities, correspondingly, are in the order [ZnCl2L] > [ZnBr2L] >> [HgCl2L] > [HgBr2L]. Zinc(II) complexes show recyclable catalytic effects on the transesterification reaction in the order [ZnCl2L] > [ZnBr2L]. Calcination of [ZnCl2L] and [ZnBr2L] at 500 degrees C produces uniform hexagonal tubular spire crystals of 1.2 x 1.2 x 4.0 mu m(3) dimensions and spheres, respectively.</P>

      • The Transgenic Poplar as an Efficient Bioreactor System for the Production of Xylanase

        <P>Plants are attractive expression systems for large-scale, low-cost production of high-value proteins. The xylanase 2 gene (Xyn2), encoding an endo-β-1,4-xylanase from Trichoderma reesei, was cloned and expressed in Escherichia coli and the poplar (Populus spp.). The optimal temperature and pH of the recombinant xylanase were 50 C and 5.0 respectively when expressed in E. coli. The purpose of this study was to produce recombinant xylanase in poplar. The Xyn2 gene was transferred into poplars by Agrobacterium-mediated transformation. The transgenic status and transgene expression of the transformed poplar were confirmed by polymerase chain reaction (PCR) genotyping and reverse transcription (RT)-PCR analysis. The poplar-expressed xylanase was biologically active, with an expression level of up to 14.4% of total leaf soluble protein. In the leaves, the average xylanase content was 1.016 mg per g of leaf fresh weight in the transgenic poplar. We found that the poplar might make possible the large-scale production of commercially important recombinant proteins.</P>

        High Cadmium-Binding Ability of a Novel <i>Colocasia esculenta</i> Metallothionein Increases Cadmium Tolerance in <i>Escherichia coli</i> and Tobacco

        <P>Experimental evidence <I>in vivo</I> as to the functional roles and binding properties to cadmium (Cd) of type-2 plants metallothionein (MT) has been limited thus far. We investigated the biological role of metallothionein from <I>Colocasia esculenta</I> (CeMT2b) in <I>Escherichia coli</I> and tobacco, and developed a new model for the relationship between Cd tolerance and Cd-binding ability. Heterologous expression of CeMT2b in <I>Escherichia coli</I> greatly enhanced Cd tolerance and accumulated Cd content as compared to control cells. The molecular weight of CeMT2b increased with Cd, and CeMT2b bound up to 5.96±1 molar ratio (Cd/protein). Under Cd stress, transgenic tobacco plants displayed much better seedling growth and high Cd accumulation than the wild type. The presence of an extra CXC motif in CeMT2b contributed to the enhanced Cd-tolerance. The present study provides the first insight into the ability of type-2 plant MT to bind physiological Cd.</P>

        쇄골두개 이형성증 환아의 증례보고

        쇄골두개 이형성증은 골격과 치아 이상을 동반하는 선천성 질환으로서 다수의 조직과 장기에 직접적인 영향을 주는 중배엽성 기능장애이다. 쇄골두개 이형성증에서 일반적인 골격 결함은 두개 봉합과 천문의 지연 폐쇄, wormian bone, clavicle aplasia가 나타날 수 있다. 또한 장골, 지골, 척추, 골반 등의 전체 골격 뿐 아니라 근육, 중앙 신경계 등에 영향을 줄 수 있다. 구강내 특징으로 유치의 만기잔존, 다수의 과잉치, 미맹출된 영구치, 낭 형성 등이 나타난다. 그러나 쇄골두개 이형성증 환아의 경우 정신적 및 신체적으로 특별한 이상을 나타내지 않아 스스로 질환을 인식하지 못하는 경우가 많으며 이의 결과 유치 탈락 후 영구치의 미맹출로 인하여 저작 및 심리적인 문제가 초래되는 경우가 많다. 따라서 쇄골두개 이형성증은 조기진단이 필요하며, 적절한 시기에 과잉치 제거 및 영구치 맹출을 유도하는 치료를 시행하여 저작기능의 회복 뿐 아니라 환자의 안모도 개선시킬 수 있어야 한다. Cleidocranial dysplasia(CCD) is a congenital disorder of skeletal and dental abnormality, which is mesodermal dysfunction influencing many tissues and organs. Skeletal abnormalities in CCD are delayed closure of cranial suture and fontanelle, presence of wormian bone and clavicle aplasia. And CCD has an effect on the long bones, phalanges, spine, pelvis, muscles, and central nervous system. Dental manifestations include retention of deciduous teeth, multiple supernumerary teeth and formation of cysts around nonerupted teeth. But as a result in common with a lack of medical and physical disability, patient may have no substantive complaint, there are many masticatory and psychological problem by absent eruption of permanent teeth after exfoliation of deciduous teeth. For this reason CCD is necessary fo early diagnosis and must be improvement of the patient's appearance as well as provision of a functioning masticatory mechanism by treatment of surgical removal of supernumerary teeth, followed orthodontically eruption of the natural permanent teeth at adequate time.
