        Wind characteristics of a strong typhoon in marine surface boundary layer

        High-resolution wind data were acquired from a 100-m high offshore tower during the passage of Typhoon Hagupit in September, 2008. The meteorological tower was equipped with an ultrasonic anemometer and a number of cup anemometers at heights between 10 and 100 m. Wind characteristics of the strong typhoon, such as mean wind speed and wind direction, turbulence intensity, turbulence integral length scale, gust factor and power spectra of wind velocity, vertical profiles of mean wind speed were investigated in detail based on the wind data recorded during the strong typhoon. The measured results revealed that the wind characteristics in different stages during the typhoon varied remarkably. Through comparison with non-typhoon wind measurements, the phenomena of enhanced levels of turbulence intensity, gust factors, turbulence integral length scale and spectral magnitudes in typhoon boundary layer were observed. The monitored data and analysis results are expected to be useful for the wind-resistant design of offshore structures and buildings on seashores in typhoon-prone regions.

        Screening and production of dsRNA molecules for protecting Cucumis sativus against Cucumber mosaic virus through foliar application

        Cucumber mosaic virus (CMV), which is one of the most prevalent plant viruses, harms many crops around the word. Although CMV causes severe economic losses to agriculture, successful prevention and treatment strategies are lacking. RNA interference (RNAi) is a promising strategy for the control of CMV because of its impressive specificity and sustain- ability. Here, a bacterial expression approach was conducted to produce hairpin RNA, and the best expression conditions to produce a high yield of dsRNA were explored. The highest expression of dsRNA in Escherichia coli HT115 was achieved at 37 °C using 2 mM IPTG and 17 h post-induction. For resistance analysis, six dsRNA molecules were constructed for CMV 1a, 2a, 3a and coat protein (CP). In cucumber (Cucumis sativus), foliar application of dsRNA preparations derived from CMV-1a and CMV-2a proteins conferred efficient protection against CMV. Our findings confirm that the exogenous application of dsRNA with a bacterial prokaryotic expression system could be a powerful biotechnological tool to protect plants against virus infections.

        Grape seed proanthocyanidins protect retinal ganglion cells by inhibiting oxidative stress and mitochondrial alteration

        Grape seed proanthocyanidins (GSP) are knownas condensed tannins and have been used as an anti-oxidantin various neurodegenerative diseases. In our study, GSPwas used as a daily dietary supplement and the neuroprotectiveeffects were evaluated on the retinal ganglion cells(RGCs) in the retinal tissues in glaucomatous DBA/2D (D2)mice. D2 mice and age-matched non-glaucomatous DBA/2JGpnmb+(D2-Gpnmb+) mice were fed with GSP or a controldiet for up to 6 months. The intraocular pressure (IOP), RGCsurvival, glial fibrillary acidic protein (GFAP), the levels ofapoptotic proteins, and the expression of oxidative stressmarkers in retinal tissues were determined. In our study, theneuroprotective effects of GSP on retinal tissues were confirmed,as evidenced by (a) GSP inhibited the IOP elevationin D2 mice; (b) GSP enhanced RGC survival and mediatedthe apoptotic protein expression; (c) GSP suppressed GFAPexpression; and (d) the oxidative stress and the levels ofmitochondrial reactive oxygen species were regulated byGSP. Our findings indicate that GSP has promising potentialto preserve retinal tissue functions via regulating oxidativestress and mitochondrial functions.

        The 16-year experience in treating low-risk gestational trophoblastic neoplasia patients with failed primary methotrexate chemotherapy

        Objective: To assess the outcomes and toxic effects of 5-day actinomycin D (Act-D) salvagetherapy and to explore the predictors of Act-D resistance in patients with low-risk gestationaltrophoblastic neoplasia (GTN)who failed 5-day methotrexate (MTX) chemotherapy. Methods: This retrospective study analyzed patients with low-risk GTN administered Act-Dsalvage therapy after failing MTX chemotherapy at Women's Hospital, School of MedicineZhejiang University between January 2000 and December 2015. The clinical parameters ofthese patients were collected and analyzed. Results: The final analysis included 89 cases. Of these, 73 cases (82.02%) responded tosalvage Act-D. The remaining 16 resistant cases were switched to etoposide, MTX, Act-D/cyclophosphamide, and vincristine chemotherapy and achieved complete remission. Serumhuman chorionic gonadotrophin levels before Act-D salvage therapy (hCGAct-D)in the Act-D resistant cases were significantly higher than those in the Act-D responders (median 605 vs. 103 IU/L, p=0.009). However, the range of hCGAct-D values in Act-D responders was wider thanthat in Act-D-resistant cases (5.76–16,664 IU/L vs. 11.43–6,732 IU/L). Thus, assigning a generalcut-off value was difficult considering the individual setting. Except for 2 cases requiring othersalvage regimens due to Act-D toxicity, 97.80% of cases (89/91) tolerated the toxicity. During atleast 1-year follow-up, the survival rate was 100.00% and no case developed recurrence. Conclusion: Based on the good therapeutic effect and tolerable toxicity, we recommendAct-D salvage therapy for all patients with low-risk GTN who fail primary MTX chemotherapy. The higher serum hCG levels before Act-D salvage therapy may be associated with resistanceto this treatment.

        Effects of Glucagon-Like Peptide-2-Expressing Saccharomyces cerevisiae Not Different from Empty Vector

        Saccharomyces cerevisiae (S. cerevisiae) and glucagon-like peptide-2 (GLP-2) have been employed to improve the intestinal development of weaned animals. The goal of this study was to determine whether either exogenous S. cerevisiae or GLP-2 elicits major effects on fecal microbiotas and cytokine responses in weaned piglets. Ninety-six piglets weaned at 26 days were assigned to one of four groups: 1) Basal diet (Control), 2) empty vector-harboring S. cerevisiae (EV-SC), 3) GLP-2-expressing S. cerevisiae (GLP2-SC), and 4) recombinant human GLP-2 (rh-GLP2). At the start of the post-weaning period (day 0), and at day 28, fecal samples were collected to assess the bacterial communities via sequencing the V1-V2 region of the 16SrRNA gene, and piglets’ blood was also sampled to measure cytokine responses (i.e., IL-1β, TNF-α, and IFN-γ). This study revealed that, on the one hand, although S. cerevisiae supplementation did not significantly alter the growth of weaned piglets, it induced increases in the relative abundances of two core genera (Ruminococcaceae_norank and Erysipelotrichaceae_norank) and decreases in the relative abundances of two other core genera (Lachnospiraceae_norank and Clostridiale_norank) and cytokine levels (IL-1β and TNF-α) (p < 0.05, Control vs EV-SC; p < 0.05, rh-GLP2 vs GLP2-SC). On the other hand, GLP-2 supplementation had no significant influence on fecal bacterial communities and cytokine levels, but it produced better body weight and average daily gain (p < 0.05, Control vs EV-SC; p < 0.05, rh-GLP2 vs GLP2-SC). Therefore, altered fecal microbiotas and cytokine response effects in weaned piglets were due to S. cerevisiae rather than GLP-2.

        Wind farm resonance characteristics analysis based on harmonic impedance measurement method

        Recently, subsynchronous oscillation (SSO) has been captured in direct-drive permanent magnetic synchronous generator (PMSG) based wind farms in many areas without series compensation in nearby power grids. However, there are fewer reports related to this new type of SSO. In addition, an analysis of the new SSO based on existing methods is computationally cumbersome, and the control effect of the converter was generally simplified in previous studies, which lead to serious errors. This paper proposes a method for analyzing the resonance characteristics of wind farms based on impedance measurement to simplify calculations and improve accuracy. The correctness and effectiveness of the proposed method are validated through the IEEE first benchmark model. A time-domain simulation model of a direct-drive PMSG is established and analyzed by harmonic impedance measurement. The results show that SSO exists when direct-drive PMSGs are connected to a weak AC system. The converter of the direct-drive PMSG exhibits a small negative value at the oscillation mode frequency. This means that the converter behaves like a capacitor with negative impedance. The capacitive impedance of the converter forms a resonant loop with the inductance of the AC grid and causes unstable SSO.

        Early potential effects of resveratrol supplementation on skeletal muscle adaptation involved in exercise-induced weight loss in obese mice

        Exercise and resveratrol supplementation exhibit anti-obesity functions in the long term but have not been fully investigated yet in terms of their early potential effectiveness. Mice fed with high-fat diet were categorized into control (Cont), exercise (Ex), resveratrol supplementation (Res), and exercise combined with resveratrol supplementation (Ex + Res) groups. In the four-week period of weight loss, exercise combined with resveratrol supplementation exerted no additional effects on body weight loss but significantly improved whole-body glucose and lipid homeostasis. The combined treatment significantly decreased intrahepatic lipid content but did not affect intramyocellular lipid content. Moreover, the treatment significantly increased the contents of mtDNA and cytochrome c, the expression levels of peroxisome proliferator-activated receptor gamma coactivator-1 alpha and its downstream transcription factors, and the activities of ATPase and citrate synthase. However, exercise, resveratrol, and their combination did not promote myofiber specification toward slow-twitch type. The effects of exercise combined with resveratrol supplementation on weight loss could be partly due to enhanced mitochondrial biogenesis and not to fiber-type shift in skeletal muscle tissues.         Interpretability Analysis of Shear Capacity in Reinforced Recycled Aggregate Concrete Beams Using Tree Models

        Recycled aggregate concrete is an effective solution for efficiently managing municipal construction waste on a large scale. Shear bearing capacity (SBC) is significant for reinforced concrete structures, and it is essential to develop trustworthy calculation models for structural design. This paper proposes a tree model-based SBC assessment system that considers eight design parameters of reinforced recycled concrete beams (RRCBs). Evaluation results revealed that the extreme gradient boosting model yielded the highest prediction accuracy with an R2of 0.960 and a mean absolute percentage error of 7.343%. To reduce the risk of black-box models, this study conducted feature importance calculations, sensitivity analyses and reliability validation of the prediction results. The findings demonstrated that increasing the hoop reinforcement ratio and beam width significantly improved the SBC of RRCB. The compressive strength and longitudinal reinforcement ratio had positive effects on the SBC, while longitudinal steel bar yield strength had no effect on the SBC. These analyses can be combined with physical mechanisms to better refine the performance design. Furthermore, a comparative study utilizing two commonly used standard formulas was conducted. The results indicated that the SBCs estimated using the tree model are more accurate than those calculated by the standard formulas.
