Embryonic germ (EG) cells are undifferentiated stern cells isolated from cultured primordial germ cells (PGC). These cells share many characteristics with embryonic stem cells including morphology and pluripotency. Undifferentiated porcine EG cell lines demonstrating capacities of differentiation both in vitro and in vivo have been established. Since EG cells can be cultured indefinitely in an undifferentiated state, whereas somatic cells in primary culture are often unstable and have limited lifespan, EG cells may provide inexhaustible source of karyoplasts in nuclear transfer (NT). In this study the efficiencies of NT using porcine EG and fetal fibroblast cells were compared. Two different techniques were used to perform NT. With conventional NT procedure (Roslin method) involving fusion of donor cells with enucleated oocytes, the rates of development to the blastocyst stage in EG and somatic cell NT were 16.8% (59/351) and 14.5% (98/677), respectively. In piezo-driven microinjection (Honolulu method) of donor nuclei into enucleated oocytes, the rates of blastocyst formation in EG and somatic cell NT were 11.9% (15/126) and 9.4% (9/96), respectively. Regardless of NT methods used in this study, EG cell NT gave rise to comparable rate of blastocyst development to somatic cell NT. Overall, EG cells can be used as karyoplast donor in NT procedure, and embryos can be produced by EG cell NT that may be used as an alternative to conventional somatic cell NT.

Development of Commercial Waxy-Super Sweet Corn Hybrids in Korea

Super-sweet corn controlled by shrunken2 (sh2) was first introduced into Korea three decade ago, but its cultivation and uses have not been popularized. Main reasons of the un-popularity are considered as lack of locally bred adapted cultivars, low germination, too sweet and less chewing of the green ears. Considering traditional and favorable eating habits of glutinous waxy corn by Koreans, we initiated to develop a double mutant wx/sh corn that has 3:1 segregation in an ear from 1996. First we formed several double cross hybrids using commercial single crosses of wx and sh2 hybrid considering seed production difficulties. The results showed that a double mutant corn hybrid showed high germination, and high palatability score with tolerance to major diseases and insects in Korea. In 2008, we conducted single and double cross trials in Gokseong, South Jeolla-do. The yield of waxy-super sweet was higher than both waxy and super sweet corn with good agronomic traits of ear height, ear weight, ear length and plant height. Among waxy-super sweet corn single cross was even better than double cross in all parameters. This double mutant approach shall be an outstanding option to change of traditional food habits into a new eating pattern. This attempt has potential success of commercial wx/sh hybrid corn development.
마우스 초대배양 대식세포에서 20(S)-Protopanaxadiol에 의해 발현 변화하는 염증 관련 유전자 탐색

Background : Ginseng has been commonly used as a traditional oriental medicine for its wide spectrum of medicinal properties, including anti-inflammatory, antitumorigenic, adaptogenic and anti-aging properties. 20(S)-Protopanaxadiol (PPD), a intestinal metabolite of ginsenosides, is one of the active ingredients in ginseng. In this study, we have found inflammation-related genes regulated by 20(S)-PPD in mouse bone marrow-derived macrophage (BMDM) to elucidate the role of 20(S)-PPD in inflammatory signaling pathways. Methods and Results : We examined cell viability of BMDM cells after treatment of 20(S)-PPD and found that 20(S)-PPD has no cytotoxicity up to 10 uM. BMDM cells treated with none or 10uM 20(S)-PPD were used for RNA extraction and microarray analysis. It was found that 2 inflammation-related genes are upregulated and 4 genes are downregulated by 20(S)-PPD. Conclusion : These results can give clues to elucidate the role of 20(S)-PPD in inflammatory signaling pathways.
