      • Clinical Significance of BCR-ABL Fusion Gene Subtypes in Chronic Myelogenous and Acute Lymphoblastic Leukemias

        Background: Some reports have suggested that chronic myeloid leukemia (CML) patients have a higher prevalence of M-bcr than acute lymphoblastic leukemia (ALL) patients, which show a higher prevalence of m-bcr. However, the relationship between BCR-ABL subtypes and progression of CML and ALL remains unclear. Materials and Methods: 354 CML chronic phase (CML-CP) patients, 26 CML blastic phase (CML-BP) patients and 72 ALL patients before treatment with BCR-ABL positive were recruited for blood routine examination and bone marrow smear cytology. Some 80 CML-CP and 32 ALL patients after imatinib (IM) treatment were followed-up for BCR-ABL relative concentrations detected after treatment for 3, 6 and 9 months and 1 year. Results: Before treatment, CML-CP patients showed lower BCR-ABL relative concentrations with a higher proportion of M-bcr (42.7%) compared to CML-BP and ALL patients while ALL patients had a higher BCR-ABL relative concentration with high expression of m-bcr (51.4%). Patients with M-bcr demonstrated higher WBC counts than those with m-bcr and the mixed group and higher PLT counts were noted in the CML-CP and ALL groups. After imatinib (IM) treatment, patients with m-bcr showed higher BCR-ABL relative concentrations in both CML-CP and ALL groups. Conclusions: This study identified the BCR-ABL gene as an important factor in CML and ALL cases. The M-bcr subtype was associated more with CML while the m-bcr subtype was associated more with ALL. Patients with m-bcr seem to have a poorer response to IM in either CML or ALL patients compared to M-bcr patients.

        The detection efficiency study of NaI(Tl) scintillation detector with the different numbers of SiPMs

        SiPMs are generally coupled into whole columns in gamma energy spectrum measurement, but the relationship between the distribution of whole SiPM columns and the energy resolution of the measured energy spectra is rarely reported. In this work, ∅ 3 3 inch NaI scintillator is placed on an 8 8 SiPM array, and the energy resolution of the 137Cs peak at 662 keV corresponding to the g-ray is selected as a reference. Each SiPM is switched to explore the influence of the number of SiPM arrays, distribution position, and reflective layer on the energy resolution of SiPMs. Results show that without coupling, the energy resolution is greatly improved when the number of SiPMs ranges from 4 to 32. However, after 32 slices (the area covered by SiPMs relative to the scintillator reaches 25.9%), the improvement in energy resolution and total pulse count is not obvious. In addition, the position of SiPMs relative to the scintillator does not exert much impact on the energy resolution. Results also indicate that by adding a reflective film (ESR), the energy resolution of the tested group increases by 10.38% on average. This work can provide a reference for the design and application of miniaturized SiPM gamma spectrometers

        Effect of nano-carbon addition on color performance of polystyrene superstructure film

        Polystyrene superstructure films show faint rainbow color, and this low color saturation limits its wide application. In thispaper, polystyrene superstructure films with single bright blue color were prepared by vertical deposition self-assemblymethod using polystyrene microspheres with average diameter of 310 ± 10 nm as raw material. Polystyrene superstructurefilms were modified by adding nano-carbon powder, and effect of the amount of nano-carbon powde on color performancewas studied. The results showed that without addition of nano-carbon powder, the superstructure films showed a faint rainbowcolor, while with addition of nano-carbon power, the superstructure films exhibited a single bright blue under the same naturallight source. Changing the amount of nano-carbon powder addition could adjust color saturation of the film. With increasingthe amount of nano-carbon powder addition from 0.008 wt% to 0.01 wt%, color saturation of the superstructure filmincreased gradually. Further increasing the amount of nano-carbon powder addition to 0.011wt%, color saturation of thesuperstructure film didn’t increase anymore and tended to get dark.

        Schisandrin B Improves the Hypothermic Preservation of Celsior Solution in Human Umbilical Cord Mesenchymal Stem Cells

        BACKGROUND: Human umbilical cord mesenchymal stem cells (hUCMSCs) have emerged as promising therapy for immune and inflammatory diseases. However, how to maintain the activity and unique properties during cold storage and transportation is one of the key factors affecting the therapeutic efficiency of hUCMSCs. Schisandrin B (SchB) has many functions in cell protection as a natural medicine. In this study, we investigated the protective effects of SchB on the hypothermic preservation of hUCMSCs. METHODS: hUCMSCs were isolated from Wharton’s jelly. Subsequently, hUCMSCs were exposed to cold storage (4 C) and 24-h re-warming. After that, cells viability, surface markers, immunomodulatory effects, reactive oxygen species (ROS), mitochondrial integrity, apoptosis-related and antioxidant proteins expression level were evaluated. RESULTS: SchB significantly alleviated the cells injury and maintained unique properties such as differentiation potential, level of surface markers and immunomodulatory effects of hUCMSCs. The protective effects of SchB on hUCMSCs after hypothermic storage seemed associated with its inhibition of apoptosis and the anti-oxidative stress effect mediated by nuclear factor erythroid 2–related factor 2 signaling. CONCLUSION: These results demonstrate SchB could be used as an agent for hypothermic preservation of hUCMSCs.

        ZNF424, a novel human KRAB/C2H2 zinc finger protein, suppresses NFAT and p21 pathway

        Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]

        Metastasis associated genomic aberrations in stage II rectal cancer

        Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

        One new tirucallane-type triterpenoid, alianthusaltinin A (1), one new $C_{29}$ sterone, alianthaltone A (2), and 12 known compounds have been isolated from the stem bark of Ailanthus altissima. The structures of new compounds were identified by means of spectroscopic methods. Compound 3 was isolated from natural sources for the first time, and compounds 4, 5, and 9 were isolated from this plant for the first time.

        Numerical simulation Analysis of Tip Clearance Flow in a Centrifugal Compressor

        In order to research the relationship between the tip clearance and leakage flow of centrifugal compressor, a high speed centrifugal compressor was investigated by using CFD. A numerical study on the effect of four different rotor tip clearance sizes of centrifugal compressor, which were 0.5times, 1 times, 1.5times and 2.0times of the design tip clearance, was carried out. Efficiency and pressure ratio curves were obtained under different mass flow. The reasons of the clearance vortex and the factors of vortex size were analyzed. The result indicated that with the increase of tip clearance size, the performance of the compressor changed obviously, the performance parameters such as efficiency and pressure ratio tended to decrease obviously. While, the leakage flow does not always lead to leak vortex. The strength of the vortex increased with the tip clearance. The size of leak vortex was affected by the pressure difference between the suction side and the pressure side of blade tip.

      • Tall Buildings as Urban Habitats: A Quantitative Approach for Measuring Positive Social Impacts of Tall Buildings' Lower Public Space

        After decades of high-speed development, designing tall buildings as critical components of urban habitat, rather than simply standing aloof from their environments, has become an important concern in many Asian cities. Nevertheless, the lack of quantitative understanding cannot support efficient architectural design or urban renewal that targets better place-making. This study attempts to fill the gap by providing a typological approach for measuring the social impact of tall buildings' ground conditions: that is, public space, podiums, and interfaces. The central business districts (CBD) of three Asian cities, Shanghai, Hong Kong, and Singapore, were selected as cases. Typical patterns and categories of lower-level public spaces among the three CBDs were abstracted via typological analyses and field study. The following evaluation is achieved through the analytic hierarchy process (AHP). This quantified approach helps to provide a visualization of high or low positive social impacts of tall buildings' lower-level public spaces among the three cases. This study also helps to suggest a design code for tall buildings aimed at a more human-oriented urban habitat.

      • XRCC1 and ADPRT Polymorphisms Associated with Survival in Breast Cancer Cases Treated with Chemotherapy

        Aim: To investigate whether XRCC1 and ADPRT polymorphisms might be associated with outcomes of breast cancer. Methods: A prospective study was conducted with a total of 335 breast cancer patients undergoing chemotherapy consecutively collected from Jan. 2005 to Jan. 2008. Genotyping of XRCC1 and ADPRT polymorphisms was conducted by PCR-RFLP assay. Results: All 335 patients were followed up until death or the end of Jan. 2012, with a median follow-up period of 38.8 (2-64) months. It was shown that the variant genotype of XRCC1 399Gln/Gln was strongly significantly associated with a decreased risk of death from breast cancer, with an HR (95% CI) of 0.52 (0.28-0.91). Similarly, individuals carrying the ADPRT 762Ala/Ala demonstrated longer survival compared to ADPRT 762 Val/Val, with an HR (95% CI) of 0.58 (0.31-0.97). Individuals with combination genotypes of XRCC1 399Gln allele and ADPRT 762Ala/Ala presented with a longer survival, the HR (95% CI) being 0.56 (0.32-0.97). Conclusion: We found a significant association between XRCC1399Gln/Gln and ADPRT 762Ala/Ala polymorphisms and clinical outcomes. These two genotypes could be used as a surrogate markers of clinical outcome in glioma cases receiving chemotherapy.
