        Multi-channel analyzer based on a novel pulse fi tting analysis method

        A novel pulse fitting analysis (PFA) method is presented for the acquisition of nuclear spectra. Thecharging process of the feedback capacitor in the resistive feedback charge-sensitive preamplifier isequivalent to the impulsive pulse, and its impulse response function (IRF) can be obtained by non-linearfitting of the falling edge of the nuclear pulse. The integral of the IRF excluding the baseline representsthe energy deposition of the particles in the detector. In addition, since the non-linear fitting process inPFA method is difficult to achieve in the conventional architecture of spectroscopy system, a new multichannel analyzer (MCA) based on Zynq SoC is proposed, which transmits all the data of nuclear pulsesfrom the programmable logic (PL) to the processing system (PS) by high-speed AXI-Stream in order toimplement PFA method with precision. The linearity of new MCA has been tested. The spectrum of 137Cswas obtained using LaBr3(Ce) scintillator detector, and was compared with commercial MCA by ORTEC. The results of tests indicate that the MCA based on PFA method has the same performance as thecommercial MCA based on pulse height analysis (PHA) method and excellent linearity for g-rays withdifferent energies, which infers that PFA method is an effective and promising method for the acquisitionof spectra. Furthermore, it provides a new solution for nuclear pulse processing algorithms involvingregression and iterative processes

        Draft Genome Assembly and Annotation for Cutaneotrichosporon dermatis NICC30027, an Oleaginous Yeast Capable of Simultaneous Glucose and Xylose Assimilation

        The identification of oleaginous yeast species capable of simultaneously utilizing xylose and glucose as substrates to generate value-added biological products is an area of key eco- nomic interest. We have previously demonstrated that the Cutaneotrichosporon dermatis NICC30027 yeast strain is capable of simultaneously assimilating both xylose and glucose, resulting in considerable lipid accumulation. However, as no high-quality genome sequenc- ing data or associated annotations for this strain are available at present, it remains chal- lenging to study the metabolic mechanisms underlying this phenotype. Herein, we report a 39,305,439 bp draft genome assembly for C. dermatis NICC30027 comprised of 37 scaffolds, with 60.15% GC content. Within this genome, we identified 524 tRNAs, 142 sRNAs, 53 miRNAs, 28 snRNAs, and eight rRNA clusters. Moreover, repeat sequences totaling 1,032,129 bp in length were identified (2.63% of the genome), as were 14,238 unigenes that were 1,789.35 bp in length on average (64.82% of the genome). The NCBI non-redundant protein sequences (NR) database was employed to successfully annotate 11,795 of these unigenes, while 3,621 and 11,902 were annotated with the Swiss-Prot and TrEMBL databases, respectively. Unigenes were additionally subjected to pathway enrichment analyses using the Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), Cluster of Orthologous Groups of proteins (COG), Clusters of orthologous groups for eukaryotic com- plete genomes (KOG), and Non-supervised Orthologous Groups (eggNOG) databases. Together, these results provide a foundation for future studies aimed at clarifying the mech- anistic basis for the ability of C. dermatis NICC30027 to simultaneously utilize glucose and xylose to synthesize lipids.

        Optoelectronic Characteristics of UV Photodetector Based on GaN/ZnO Nanorods p-i-n Heterostructures

        We demonstrate an efficient ultraviolet (UV) photodetector operating at room temperature based on n-ZnO nanorods/i-ZnO/p-GaN heterojunctions. We employ x-ray diffraction and field-emission scanning electron microscopy to confirm the high quality of the ZnO nanorods using an undoped ZnO film as the interlayer. Then, we investigate the photoelectric properties of the fabricated photodetector with UV light illumination under a different reverse bias. Based on the current-voltage curve, the photocurrent to dark current ratio is approximately 73.3 at −4 V. At zerobias voltage, the peak responsivity was 138.9 mA/W at 362 nm under front-illumination conditions. Time-varying measurements indicate the reproducibility and stability of the heterojunction photodetector.

        The detection efficiency study of NaI(Tl) scintillation detector with the different numbers of SiPMs

        SiPMs are generally coupled into whole columns in gamma energy spectrum measurement, but the relationship between the distribution of whole SiPM columns and the energy resolution of the measured energy spectra is rarely reported. In this work, ∅ 3 3 inch NaI scintillator is placed on an 8 8 SiPM array, and the energy resolution of the 137Cs peak at 662 keV corresponding to the g-ray is selected as a reference. Each SiPM is switched to explore the influence of the number of SiPM arrays, distribution position, and reflective layer on the energy resolution of SiPMs. Results show that without coupling, the energy resolution is greatly improved when the number of SiPMs ranges from 4 to 32. However, after 32 slices (the area covered by SiPMs relative to the scintillator reaches 25.9%), the improvement in energy resolution and total pulse count is not obvious. In addition, the position of SiPMs relative to the scintillator does not exert much impact on the energy resolution. Results also indicate that by adding a reflective film (ESR), the energy resolution of the tested group increases by 10.38% on average. This work can provide a reference for the design and application of miniaturized SiPM gamma spectrometers

        Genistein inhibit the proliferation induced by zearalenone in MCF-7 cells

        Zearalenone (ZEA) is a nonsteroidal estrogenic mycotoxin and exists widely in the moldy feeds and food. Genistein (GEN) is a natural isoflavone phytoestrogen. ZEA has adverse effects on animal's reproduction performance and immunologic function, also can induce the proliferation of MCF-7 cells. However, the interactive effect of ZEA and GEN on proliferation and cell cycle in MCF-7 cells is remain poorly understood. Here, the interactive effect of ZEA and GEN on proliferation and cell cycle in MCF-7 cells were determined through MTT assay and flow cytometry, respectively. Results demonstrated that ZEA(10 and 20 nM) could stimulate the proliferation of MCF-7 cells and significantly increase S phase distribution (P< 0.05). RT-PCR analysis revealed that ZEA could significantly increase estrogen receptor (ER) $\alpha$ mRNA expression (P < 0.05), while GEN (16 and $32\;{\mu}M$) could inhibit proliferation and lessen S phase distribution of MCF-7 cells (P < 0.05) which were induced by ZEA. In the co-administration treatments of GEN and ZEA, $ER\alpha$ mRNA expression decreased, bcl-2 decreased and bax increased at both mRNA and protein levels compared to those with ZEA alone. These results showed that the proliferative activity of ZEA could be repressed by GEN through regulating the expression of ER, bax and bcl-2 in MCF-7 cells.

        Expression and Purification of Lacticin Q by Small Ubiquitin-Related Modifier Fusion in Escherichia coli

        Lacticin Q is a broad-spectrum class II bacteriocin with potential as an alternative to conventional antibiotics. The objective of this study was to produce recombinant lacticin Q using a small ubiquitin-related modifier (SUMO) fusion protein expression system. The 168-bp lacticin Q gene was cloned into the expression vector pET SUMO and transformed into Escherichia coli BL21(DE3). The soluble fusion protein was recovered with a Ni-NTA Sepharose column (95% purity); 130 mg protein was obtained per liter of fermentation culture. The SUMO tag was then proteolytically cleaved from the protein, which was re-applied to the column. Finally, about 32 mg lacticin Q (≥96% purity) was obtained. The recombinant protein exhibited antimicrobial properties similar to that of the native protein, demonstrating that lacticin Q had been successfully expressed by the SUMO fusion system.

        The aim of this paper is to show that there exists a weakly quasisymmetric homeomorphism $f:(X,d){\rightarrow}(Y,d^{\prime})$ between two locally compact non-complete metric spaces such that $f:(X,d_h){\rightarrow}(Y,d^{\prime}_h)$ is not quasi-isometric, where dh denotes the Gromov hyperbolic metric with respect to the metric d introduced by Ibragimov in 2011. This result shows that the answer to the related question asked by Ibragimov in 2013 is negative.

        A New On-line Power Flow Calculation Method with Computation Loads Separation Technique in DC Power Systems

        The speed of one-time power fl ow calculation is hard to be further improved with satisfi ed accuracy and stability after decades of researches. However, the improvement on many-times calculation speed may be possible and is important for practical application. This paper proposes a novel method which can dramatically improve the many-times calculation speed of power fl ow without sacrifi ce of accuracy, convergence and stability in DC power systems. To achieve the purpose, this paper adopts a novel basic technique to separate the computation loads of grid topology analysis and bus types analysis from power fl ow equations. The technique is based on the analysis of linear relationships-based nonlinear problems and strict mathematical derivations. Combined with the basic technique, a correctness method is also proposed to deal with the specifi c nonlinear factors in power fl ow calculation. Thus, a complete novel power fl ow calculation method is proposed and the detailed mathematical derivation is presented. With the novel ant lion optimizer, the proper DC test systems are constructed. Compared with classical Newton–Raphson method, the proposed method is more stable and the initial values requirement is lower. At last, the numerical verifi cation validates that the speed of the proposed method can be more than 10 times faster than Newton–Raphson method without the sacrifi ce of other performance.

        Study on mechanical behaviors of column foot joint in traditional timber structure

        Column is usually floating on the stone base directly with or without positioning tenon in traditional Chinese timber structure. Vertical load originated by the heavy upper structure would induce large friction force and compression force between interfaces of column foot and stone base. This study focused on the mechanical behaviors of column foot joint with consideration of the influence of vertical load. Mechanism of column rocking and stress state of column foot has been explored by theoretical analysis. A nonlinear finite element model of column foot joint has been built and verified using the full-scale test. The verified model is then used to investigate the mechanical behaviors of the joint subjected to cyclic loading with different static vertical loads. Column rocking mechanism and stress distributions of column foot were studied in detail, showing good agreement with the theoretical analysis. Mechanical behaviors of column foot joint and the effects of the vertical load on the seismic behavior of column foot were studied. Result showed that compression stress, restoring moment and stiffness increased with the increase of vertical load. An appropriate vertical load originated by the heavy upper structure would produce certain restoring moment and reset the rocking columns, ensuring the stability of the whole frame.

        Relationship of mineral elements in sheep grazing in the highland agro-ecosystem

        Objective: Minerals are one of the important nutrients for supporting the growth of sheep grazing in the highland, northwest of China. The experiment was conducted to investigate the relationship of both macro and micro minerals in sheep grazing in the highlands of six districts located in the Qilian Mountain of China. Methods: Samples of herbage (n = 240) and soil (n = 240) were collected at random in a "W" shape across the area designated for harvesting from 24 farms, where the sheep commonly graze in October (winter) for mineral analyses. In addition, serum samples were taken via jugular vein from 20 sheep per farm from 24 farms (n = 480 samples in total) for serum minerals analyses. Mean values of macro and micro minerals were statistically compared among districts and the correlations among soil-plant-animal were statistically analyzed and correlations were regressed, as well. Results: The results revealed that there were variations for both macro and micro minerals among districts. Statistical analysis of the correlation coefficients between herbage and sheep were significantly different for most of the minerals but not for P, Cu, and Se. Many correlation regression coefficients were found significantly different among minerals of herbage, soil, and sheep serum especially those of K, Na, Fe, Mn, and Zn (between herbage and sheep serum), and Fe and Mn (between herbage and soil), Na, Fe, Mn, and Zn (between soil and sheep serum), respectively. The regression coefficient equations derived under this experiment for prediction of Ca (R<sup>2</sup> = 0.618), K (R<sup>2</sup> = 0.803), Mg (R<sup>2</sup> = 0.767), Na (R<sup>2</sup> = 0.670), Fe (R<sup>2</sup> = 0.865),Zn (R<sup>2</sup> = 0.950), Mn (R<sup>2</sup> = 0.936), and Se (R<sup>2</sup> = 0.630), resulted in significant R<sup>2</sup> values. Conclusion: It is inferred that the winter herbage minerals in all the districts were below the recommended levels for macro minerals which indicated there would be some mineral deficiencies in sheep grazing the herbage in these regions. Supplemental minerals may therefore play an important role in balancing the minerals available from the herbage in winter and would lead to increased productivity in sheep on the highland areas of China. These findings could be potentially applied to the other regions for improving the livestock productivity.
