        Surgical Options for Appropriate Length of J-Pouch Construction for Better Outcomes and Long-term Quality of Life in Patients with Ulcerative Colitis after Ileal Pouch-Anal Anastomosis

        Background/Aims: Total proctocolectomy with ileal pouch-anal anastomosis (IPAA) is widely accepted as a radical surgery for refractory ulcerative colitis (UC). Definite results on the appropriate pouch length for an evaluation of the risk-to-benefit ratio regarding technical complications and long-term quality of life (QOL) are still scarce. Methods: Data on UC patients who underwent IPAA from 2008 to 2022 in four well-established pouch centers affiliated to China UC Pouch Center Union were collected. Results: A total of 208 patients with a median follow-up time of 6.0 years (interquartile range, 2.3 to 9.0 years) were enrolled. The median lengths of the patients’ short and long pouches were 14.0 cm (interquartile range, 14.0 to 15.0 cm) and 22.0 cm (interquartile range, 20.0 to 24.0 cm), respectively. Patients with a short J pouch configuration were less likely to achieve significantly improved long-term QOL (p=0.015) and were prone to develop late postoperative complications (p=0.042), such as increased defecation frequency (p=0.003) and pouchitis (p=0.035). A short ileal pouch was an independent risk factor for the development of late postoperative complications (odds ratio, 3.100; 95% confidence interval, 1.519 to 6.329; p=0.002) and impaired long-term QOL improvement (odds ratio, 2.221; 95% confidence interval, 1.218 to 4.050, p=0.009). Conclusions: The length of the J pouch was associated with the improvement in long-term QOL and the development of late post-IPAA complications. A long J pouch configuration could be a considerable surgical option for pouch construction.

        Expression of three neuropeptide genes (SfEH, SfETH, and SfCCAP) in response to insecticide stress on the white-backed planthopper Sogatella furcifera (Hemiptera: Delphacidae)

        Insect neuropeptides are involved in many important developmental processes. Here, we cloned three neuro peptide genes (SfEH, SfETH, and SfCCAP) from Sogatella furcifera (Horváth) and analyzed the relative expression levels of these genes through Real-time Quantitative polymerase chain reaction (RT-qPCR) during different developmental stages and in different wing types of S. furcifera following exposure to sublethal concentrations (LC 10 and LC 25 ) of methylaminoavermectin benzoate. The results revealed that the three neuropeptide genes (SfEH, SfETH, and SfCCAP) of S. furcifera contained open reading frame sequences of 222, 636, and 408 bp, encoding 73, 211, and 135 amino acids, respectively. Moreover, the expression of SfEH and SfETH reached its peak on day 3 of the 5 th instar nymph stage, while the SfCCAP expressed pinnacle on 40 min after eclosion. Furthermore, the relative expression levels of the three genes were in the following order: SfEH, short-winged female > long-winged female > long-winged male; SfETH, long-winged female > short-winged female > longwinged male; and SfCCAP, short-winged female > long-winged male > short-winged female. Upon calculating the relative expression of the three peptide genes after 24 and 48 h of treatment with sublethal concentrations of methylaminoavermectin benzoate, it was found that the relative mRNA expression levels of SfEH and SfETH declined upon LC 10 treatment but increased upon LC 25 treatment. Meanwhile, the relative mRNA expression level of SfCCAP declined upon both LC 10 and LC 25 treatments. This result will help us understand the effects of pesticides on insect growth and development at the molecular level.

        A modified heteropoly acid catalyst with cetyltrimethylammonium bromide for methacrolein to methacrylic acid

        A novel type of heteropolyacids, CTAB-CsH3PMo11VO40 (CTA-CsPAV), was developed by a method of emulsion synthesis using cetyltrimethyl ammonium bromide (CTAB) as the structure promoter, which was applied as a catalyst in the oxidation of methacrolein (MAL) to methacrylic acid (MAA) with fixed-bed reactor. The structure and property of CTA-CsPAV were characterized by FT-IR, XRD, nitrogen adsorption-desorption, NH3-TPD, SEM, XPS, and so on. The structure-performance relationship of CTA-CsPAV was systematically studied, wherein the influence of surface area, acidity, redox property of CTA-CsPAV were focused and discussed. Importantly, the long-term performance of CTA-CsPAV was also investigated.

        3D heat transfer, lubrication and friction coupled study for piston ring-liner on diesel engines

        The piston ring and liner of internal combustion engines are generally used under increasingly harsh conditions. To investigate the mechanisms affecting the work ineffective, the coupled heat transfer, lubrication and friction model between the piston ring and liner is proposed by coupling the gas, piston set-liner, lubrication film and coolant. In this model, the temperature variation of lubricant, the gas blowby, the surface roughness, the rupture position and the non-axial symmetry of oil film are considered. The coupled heat transfer, lubrication and friction simulation is carried out for 6110 diesel engine, and the temperature fields of piston set-liner along with the corresponding lubrication and friction curves are obtained. The simulated results are verified by comparing with the temperature experiment of piston.

        Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

        Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

        Two groups of S-Layer proteins, SLP1s and SLP2s, in Bacillus thuringiensis co-exist in the S-Layer and in parasporal inclusions

        We screened four B. thuringiensis strains whose parasporal inclusions contained the S-layer protein (SLP), and cloned two slp genes from each strain. Phylogenetic analysis indicated these SLPs could be divided into two groups, SLP1s and SLP2s. To confirm whether SLPs were present in the S-layer or as a parasporal inclusion, strains CTC and BMB1152 were chosen for further study. Western blots with isolated S-layer proteins from strains CTC and BMB1152 in the vegetative phase showed that SLP1s and SLP2s were constituents of the S-layer. Immunofluorescence utilizing spore-inclusion mixtures of strains CTC and BMB1152 in the sporulation phase showed that SLP1s and SLP2s were also constituents of parasporal inclusions. When heterogeneously expressed in the crystal negative strain BMB171, four SLPs from strains CTC and BMB1152 could also form parasporal inclusions. This temporal and spatial expression is not an occasional phenomenon but ubiquitous in B. thuringiensis strains. [BMB reports 2011; 44(5): 323-328]

      • HOXB7 Predicts Poor Clinical Outcome in Patients with Advanced Esophageal Squamous Cell Cancer

        Background: Esophageal squamous cell carcinoma (ESCC) accounts for most esophageal cancer in Asia, and is the sixth common cause of cancer-related deaths worldwide. Previous studies indicated HOXB7 is overexpressed in ESCC tissues, but data on prognostic value are limited. Methods: A total of 76 advanced ESCC cases were investigated. Immunohistochemistry (IHC) was used to detect the expression levels of HOXB7 and Kaplan-Meier curves and Cox regression models to determine prognostic significance. Stratified analysis was also performed according to lymph node (LN) status. Results: Kaplan-Meier curve analysis indicated that HOXB7 positive patients had significantly shorter overall survival (OS) than HOXB7 negative patients. Multivariate analysis using the Cox proportional hazards model indicated only TNM stage and HOXB7 expression to be independent predictors of overall survival of advanced ESCC patients. HOXB7 indicated poor OS in both lymph node negative (LN-) and lymph node positive (LN+) patients. Conclusion: HOXB7 predicts poor prognosis of advanced ESCC patients and can be applied as an independent prognostic predictor.

        Effects of Ring Pack Friction Heat on Temperature Fields of Piston Set-Liner

        To investigate the temperature fields of piston set-liner under the friction heat of ring pack, it is established a friction heat allocation model of ring pack based on the heat transfer theory of semi-infinite body. In this model, the piston ring, lubrication film and liner are treated as a three-layer medium. Furthermore, the distribution model of ring pack friction heat, the unsteady heat-mixed lubrication and friction model of piston ring-liner, and the heat transfer model of piston assembly-liner are also built and coupled with the allocation model to develop a coupled heat transfer model of piston set-liner. Employing the model for 6110 diesel engine, the effects of ring pack friction heat on the temperature fields of piston set-liner are simulated. Numerical results demonstrate that the friction heat allotted to the piston ring, lubrication film and liner accounts for nearly 47 %, 21 % and 32 % of ring pack friction heat, respectively. Due to the effects of friction heat, the maximum temperature rises on the piston ring, piston and liner surfaces run approximately up to 8.5 ℃, 5 ℃ and 5 ℃, separately. The established allocation and distribution models related to friction heat in this paper can be applied to any type of piston set-liner and provide the theoretical foundation for the optimal design of piston ring-liner.

        Comparative transcriptome analysis to reveal key ethylene genes involved in a Lonicera macranthoides mutant

        Background Lonicera macranthoides Hand.-Mazz. is an important medicinal plant. Xianglei-type (XL) L. macranthoides was formed after many years of cultivation by researchers on the basis of the natural mutant. The corolla of L. macranthoides XL remains unexpanded and its flowering period is nearly three times longer than that of wild-type (WT) plants. However, the molecular mechanism behind this desirable trait remains a mystery. Objective To understand the floral phenotype differences between L. macranthoides and L. macranthoides XL at the molecular level. Methods Transcriptome analysis was performed on L. macranthoides XL and WT. One DEG was cloned by RT-PCR amplification and selected for qRT-PCR analysis. Results Transcriptome analysis showed that there were 5603 differentially expressed genes (DEGs) in XL vs. WT. Enrichment analysis of DEGs showed that pathways related to plant hormone signal transduction were significantly enriched. We identified 23 key genes in ethylene biosynthesis and signal transduction pathways. The most abundant were the ethylene biosynthesis DEGs. In addition, the open reading frames (ORFs) of WT and XL ETR2 were successfully cloned and named LM-ETR2 (GenBank: MW334978) and LM-XL-ETR2 (GenBank: MW334978), respectively. qRT-PCR at different flowering stages suggesting that ETR2 acts in the whole stage of flower development of WT and XL. Conclusions This study provides new insight into the molecular mechanism that regulates the development of special traits in the flowers of L. macranthoides XL. The plant hormone ethylene plays an important role in flower development and flowering duration prolongation in L. macranthoides. The ethylene synthesis gene could be more responsible for the flower phenotype of XL. The genes identified here can be used for breeding and improvement of other flowering plants after functional verification.

        Carbon nanofibers supported Ru catalyst for sorbitol hydrogenolysis to glycols: Effect of calcination

        Carbon nanofiber (CNFs) supported Ru catalysts for sorbitol hydrogenolysis to ethylene glycol and propylene glycol were prepared by incipient wetness impregnation, calcination and reduction. The effect of calcination on catalyst properties was investigated using thermal gravimetry analysis, temperature-programmed reduction, X-ray diffraction,X-ray photoelectron spectroscopy, transmission electron microscopy and N2 physisorption. The results indicated that calcination introduced a great amount of surface oxygen-containing groups (SOCGs) onto CNF surface and induced the phase transformation of Ru species, but slightly changed the texture of Ru/CNFs. The catalytic performance in sorbitol hydrogenolysis showed that Ru/CNFs catalyst calcined at 240 oC presented the highest glycol selectivities and reasonable glycol yields. It was believed that the inhibition and confinement effect of SOCGs around Ru particles as well as the high dispersion of Ru particles was the key factor for the catalytic activity.
