      • Developmental competence of porcine parthenogenetic embryos relative to embryonic chromosomal abnormalities

        <P>Parthenogenetically activated (PA) embryos exhibit delayed development, a lower blastocyst rate, and less successful development in vitro compared to in vitro fertilized (IVF) embryos. To investigate the possible mechanisms for unsuccessful parthenogenetic development, this study analyzed the chromosome abnormalities and developmental potential of porcine PA embryos. Mature oocytes were electrically activated and cultured in Porcine Zygote Medium-3 (PZM<SUB>3</SUB>) supplemented with 3 mg/ml BSA for 6, 7, or 8 days. The percentage of PA blastocysts was lower than that of IVF embryos on days 6 and 7 (16.4 ± 7.4 vs. 28.7 ± 3.7; 10.9 ± 2.8 vs. 21.5 ± 4.7, P < 0.05; respectively), and the PA blastocysts had significantly fewer nuclei than IVF blastocysts (23.2 ± 1.8 vs. 29.7 ± 0.8; 29.7 ± 3.3 vs. 32.0 ± 2.4, P < 0.05). The percentage of abnormal PA embryos (including embryos with condensed nuclei, arrested embryos and fragmented embryos) was higher than that of IVF embryos (PA: 52.9 ± 12.8 vs. 16.4 ± 7.4 on day 6), and increased with culture time (71.9 ± 12.1 vs. 10.9 ± 2.8. on day 7,and 75.0 ± 22.6 vs. 12.1 ± 2.3 on day 8, P < 0.05). The Day-6 PA blastocysts (n = 147) were divided into three classes according to the total number of nuclei (<20, 20–39, >40) and into three groups according to the morphological diameter (<150, 150–180, >180 µm). Of the haploid blastocysts, 56.1% had less than 20 nuclei, and 71.5% were less than 150 µm in diameter. Of all (114) blastocysts suitable for analysis, 55.5% displayed chromosomal abnormalities. Among chromosomal abnormalities in PA blastocysts, haploid blastocysts were most prevalent (43.6%), while polyploidy (4.4%) and mixoploidy (7.7%) embryos were less prevalent. Chromosomal abnormalities of porcine PA embryos might contribute to a higher rate of abnormal embryonic development. We suggest that a careful consideration should be given when using the blastocysts with smaller size, and establishing the optimum culture condition for PA embryos development in vitro. Mol. Reprod.         Function identification of bovine Nramp1 promoter and intron 1

        The Nramp1/Slc11a1 locus encodes a proton-coupled divalent cation transporter, expressed in late endosomes/lysosomes of macrophages, that constitutes a component of the innate immune response to combat intracellular pathogens and it was shown to play an important role in regulating inherent immunity. The previously identified Z-DNA forming polymorphic repeat(GT)n in the promoter region of the human Nramp1 gene does act as a functional polymorphism influencing gene expression. Research has shown that INF-${\gamma}$, TNF-${\alpha}$, IL-$1{\beta}$ and bacteria LPS increase the level of Nramp1 expression. However, the molecular mechanism for Nramp1 gene regulation is unclear. In this research, bovine Nramp1 5'-flanking region (-1748~+769) was cloned and analyzed by bioinformatics. Then to find the core promoter and the cis-acting elements, deletion analysis of promoter was performed using a set of luciferase reporter gene constructs containing successive deletions of the bovine Nramp1 5'-flanking regions. Promoter activity analysis by the dual luciferase reporter assay system showed that the core promoter of Nramp1 was located at +58~-89 bp. Some positive regulatory elements are located at -89~-205 bp and -278~-1495 bp. And the repressor elements were in region -205~-278 bp, intron1 and -1495~-1748 bp. LPS-responsive regions were located at -1495~-1748 bp and -278~-205 bp. The present study provides an initial effort to explore the molecular mechanism of transcriptional activation of the bovine Nramp1 gene and should facilitate further studies to decode the complex regulatory process and for molecular breeding for disease resistance in bovines.

        FLOURY ENDOSPERM12 Encoding Alanine Aminotransferase 1 Regulates Carbon and Nitrogen Metabolism in Rice

        Starch is a major storage substance in cerealgrains, and starch biosynthesis is a complex process. In orderto elucidate regulation of the starch biosynthesis pathway, wescreened a series of rice (Oryza sativa L.) endospermmutants. In this study, we identified a floury white-coreendosperm mutant named floury endosperm12 (flo12). Theflo12 mutant exhibited loosely packed starch granules and alower thousand kernel weight compared to wild type. Semithinsections revealed that compound starch grains (SG) inflo12 interior endosperm cells were developed abnormally. Furthermore, amylose content was decreased, while totalprotein content was significantly increased in flo12 grains. Map-based cloning showed that FLO12 encodes rice alanineaminotransferase 1 (OsAlaAT1). OsAlaAT1 is highly expressedin developing endosperm. Subcellular localization showedthat OsAlaAT1 is localized in the cytosol. Moreover, theexpression of most starch synthesis-related genes wasdecreased, while most of the storage protein coding geneshad elevated expression levels in the flo12 mutant. Inaddition, overexpression of the OsAlaAT1 gene increasedgrain weight. In brief, we demonstrated that OsAlaAT1regulates carbon and nitrogen metabolism, which provides anew insight for the improvement of rice quality and yield.

        Effects of temperature and humidity on immature development of Lygus pratensis (L.) (Hemiptera: Miridae)

        Lygus pratensis distributes worldwide and is one of the common insect pests on cotton in Xinjiang Uygur Autonomous Region, the largest cotton growing region of China. This study assessed the effects of six constant temperatures (10, 15, 20, 25, 30 and 35 °C) at 60% relative humidity (RH) on egg and nymphal development of L. pratensis. Eggs failed to hatch and the newly-hatched nymphs could not normally develop at 10 °C. The developmental duration of egg and nymph decreased as temperature increased from 15 to 30 except 35 °C for egg. Based on the linear model, the lower developmental threshold and effective accumulated temperature were 10.68 °C and 150.2 DD for egg, and 12.08 °C and 208.3 DD for nymph, respectively. Among three non-linear models (Briere-1, Logan-6 and Lactin), Logan-6 provided the most accurate estimate for the mean optimum and lethal maximum temperatures (33.6 and 40.9 °C for egg, and 34.0 and 37.4 °C for nymphs). The interactive effects of three RH levels (45, 60 and 75%) and two temperatures (25 and 35 °C) on the immature developmental stages were tested. Temperature, RH and their interaction showed significant effects on egg and nymphal development. High relative humidity (75% RH) shortened the developmental duration of egg at 25 °C and nymph at 25 and 35 °C,whereas no significant difference was found for egg duration between different RH levels at 35 °C. The present study is useful for further predicting the phenology of L. pratensis and developing forecast and management strategies for this emerging mirid bug in China.

        Supplementation of guanidinoacetic acid and rumen-protected methionine increased growth performance and meat quality of Tan lambs

        Objective: Tan lambs (n = 36, 3 mo old, 19.1±0.53 kg) were used to assess effects of dietary guanidinoacetic acid (GAA) and rumen-protected methionine (RPM) on growth performance, carcass traits, meat quality, and serum parameters. Methods: Lambs were randomly assigned to three treatment groups, with 6 pens per group and 2 lambs per pen. Dietary treatments were: basal diet alone (I); basal diet supplemented with 0.08% GAA+0.06% RPM (II); and basal diet supplemented with 0.08% GAA+0.08% RPM (III). Diets were provided three times a day for 90 d. Intake per pen was recorded daily and individual lamb body weight (BW) was measured monthly. Carcass traits were measured after slaughter and meat quality at the end of the experiment, blood samples were taken on a subgroup of lambs for analysis of indicators mostly related to protein metabolism. Results: Final BW and average daily gain for the first and second month, and for the entire experiment were greater in Treatment II compared to Treatment I (p<0.05), whereas feed to gain ratio was lower (p<0.05). Treatment II had the optimal dressing percentage and net meat weight proportion, as well as crude protein and intramuscular fat concentrations in muscles. Treatment II improved meat quality, as indicated by the greater water holding capacity, pH after 45 min and 48 h, and lower shear force and cooking loss. Dietary supplementation of GAA and RPM also increased the meat color a* and b* values at 24 h. Finally, Treatment II increased total protein, and serum concentrations of albumin and creatinine, but decreased serum urea nitrogen concentrations, indicating improved protein efficiency. Conclusion: In this study, 0.08% GAA+0.06% RPM supplementation improved growth performance and meat quality of Tan lambs.         Drug-induced diabetes is widely reported inclinical conditions, and it is becoming a global issuebecause of its potential to increase the risk of severe cardiovascularcomplications. However, which drug mechanismsexert their diabetogenic effects and why the effectspresent significant inter-individual differences remain largelyunknown. Pharmacogenomics, which is the study ofhow genomic variation influences drug responses, providesan explanation for individual differences in drug-induceddiabetes. We highlight that pharmacogenomics can beinvolved in regulating the expression of genes in signalingpathways related to the pharmacokinetics or pharmacodynamicsof drugs or the pathogenesis of diabetes, contributingto the differences in drug-induced glucoseimpairment. The pharmacogenomics studies of the majordiabetogenic drugs are reviewed, including calcineurininhibitors, antipsychotics, hormones, and antihypertensivedrugs. We intend to elucidate the genetic basis of druginduceddiabetes and pave the way for the precise use ofthese drugs in the clinic.

      • Esophageal/Gastric Cancer Screening in High-risk Populations in Henan Province, China

        Objective: To summarize the endoscopic screening findings in high-risk population of esophageal and gastric carcinoma and analyze influential factors related to screening. Methods: In seven selected cities and counties with high incidences of esophageal carcinoma, people at age of 40-69 were set as the target population. Those with gastroscopy contradictions were excluded, and all who were voluntary and willing to comply with the medical requirements were subjected to endoscopic screening and histological examination for esophageal, gastric cardia and gastric carcinoma in accordance with national technical manual for early detection and treatment of cancer. Results: In three years, 36,154 people were screened, and 16,847 (46.60%) cases were found to have precancerous lesions. A total of 875 cases were found to have cancers (2.42%), and among them 739 cases had early stage with an early diagnosis rate is 84.5%. Some 715 patients underwent prompt treatment and the success rate was 81.8%. Conclusions: In a high-risk population of esophageal and gastric carcinoma, it is feasible to implement early detection and treatment by endoscopic screening. Screening can identify potential invasive carcinoma, early stage carcinoma and precancerous lesions, improving efficacy through early detection and treatment. The exploratory analysis of related influential factors will help broad implementation of early detection and treatment for esophageal and gastric carcinoma.

        Ribosome-binding Sequences (RBS) Engineering of Key Genes in Escherichia coli for High Production of Fatty Alcohols

        Fatty alcohols are widely used in the fields of food, pharmaceutical and renewable energy and so on, thus biosynthesis of fatty alcohols has attracted wide attention in recent years. However, the concentration of fatty alcohols is still at a low level. In this paper, we constructed an Escherichia coli strain with high yield of fatty alcohols through the combinatorial regulation of three key genes of fabH, fabZ, and far. Firstly, we designed new ribosomebinding sequences (RBS) of fabH and fabZ with 10-fold strengths compared to the original ones by RBS calculator, separately. Secondly, we engineered the starting strain MGL1 by replacing the original RBS with designed ones, which resulted in MGLHS (fabH-RBS1), MGLZS (fabZRBS2), and MGLHZS (fabH-RBS1/fabZ-RBS2), respectively. Thirdly, the plasmid pL1 harboring far gene was transferred into above strains to test fatty alcohols production under the induction of different isopropyl-β-D-thiogalactoside concentrations. The results showed that the concentration of fatty alcohols in strains MGLHS-far, MGLZS-far and MGLHZS-far reached 2.75 g/L, 2.54 g/L and 3.82 g/L, respectively, which increased 35%, 25% and 88% than that of the starting strain MGL1-far. The fatty alcohols reached 9.35 g/L in fed batch fermentation with MGLHZS-far, of which the main components were two saturated fatty alcohols (C14:0 and C16:0) and two unsaturated fatty alcohols (C16:1 and C18:1). The results demonstrated that combinatorial regulation of three key genes of fabH, fabZ, and far in E. coli can achieve high-yield production of fatty alcohols.

        Propagation characteristics of longitudinal wave, shear wave and bending wave in porous circular nanoplates

        On the basis of nonlocal strain gradient theory, considering the material properties of porous FGM changing with thickness and the influence of moment of inertia, the wave equation of FG nano circular plate is derived by using the first-order shear deformation plate theory, by introducing dimensionless parameters, we transform the equations into dimensionless wave equations, and the dispersion relations of bending wave, shear wave and longitudinal wave are obtained by Laplace and Hankel integral transformation method. The influence of nonlocal parameter, porosity volume fraction, strain gradient parameters and power law index on the propagation characteristics of bending wave, shear wave and longitudinal wave in FG nano circular plate.
