        Optimization of the Extraction Parameters of Gardenia (Gardenia jasminoides Ellis) Fruits for the Maximum Antioxidant Capacity

        Response surface methodology (RSM) was used for the optimization of antioxidant capacity in gardenia extracts. The antioxidant capacities of gardenia fruit extracts were investigated by ferric reducing ability (FRA) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity (RSA) assays. The optimum extraction parameters for the strongest antioxidant capacity were the ethanol concentration (EtOH) of 48.9%, extraction temperature of 72.9℃, and extraction time of 29.9 min. Analysis of variance (ANOVA) showed that the quadratics of EtOH and extraction temperature had highly significant effect on the antioxidant capacity (p<0.001). The antioxidant capacity was correlated with contents of bioactive components [crocin, geniposide, and total phenolic (TP) compounds] in gardenia extracts and mainly attributed to the content of the TP compounds.

        Circulating Current Analysis for High-Speed Motors with Stranded Windings by Considering End and Temperature Effect

        This paper presents an improved circuit model for predicting additional copper loss caused by circulating current effect. Based on basic circuit equations and winding topology, this model takes account of both slot and end leakage inductance which are separately extracted by 2-D and 3-D finite element method. Several simplified windings are wounded in a stator to validate the accuracy of the proposed model. The results shows that this model has high accuracy and the maximum error of loss factor is about 1.5 %. In addition, it is found that the circulating current loss factor will decrease as the end winding length grows up or winding temperature increases.

        MGGC2.0: A preprocessing code for the multi-group cross section of the fast reactor with ultrafine group library

        How to generate the precise broad group cross section is important for the fast reactor design. In this study, a fast reactor multi-group cross-section generation code MGGC2.0 are developed in-house for processing ultrafine group MATXS format library. Validation and verification are performed for MGGC2.0 code by applying the benchmarks of ICSBEP handbook, and the results of MGGC2.0 agree well with that of MCNP. The consistent PN method with critical buckling search is in good agreement that condensed with TWODANT flux and flux moment for the inner core and outer core region. For the radial blanket and reflector, two region approximation method has been applied in MGGC2.0 by using collision Probability Method neutron flux solver. The RBEC-M benchmark was used to verify the power distribution calculation, and the relative error of power distribution comparison with the reference are less than 0.8% in the fuel region and the maximum relative error is 5.58% in the reflector region. Therefore, the precise broad cross section can be generated by MGGC2.0 for fast reactor

        FLOURY ENDOSPERM12 Encoding Alanine Aminotransferase 1 Regulates Carbon and Nitrogen Metabolism in Rice

        Starch is a major storage substance in cerealgrains, and starch biosynthesis is a complex process. In orderto elucidate regulation of the starch biosynthesis pathway, wescreened a series of rice (Oryza sativa L.) endospermmutants. In this study, we identified a floury white-coreendosperm mutant named floury endosperm12 (flo12). Theflo12 mutant exhibited loosely packed starch granules and alower thousand kernel weight compared to wild type. Semithinsections revealed that compound starch grains (SG) inflo12 interior endosperm cells were developed abnormally. Furthermore, amylose content was decreased, while totalprotein content was significantly increased in flo12 grains. Map-based cloning showed that FLO12 encodes rice alanineaminotransferase 1 (OsAlaAT1). OsAlaAT1 is highly expressedin developing endosperm. Subcellular localization showedthat OsAlaAT1 is localized in the cytosol. Moreover, theexpression of most starch synthesis-related genes wasdecreased, while most of the storage protein coding geneshad elevated expression levels in the flo12 mutant. Inaddition, overexpression of the OsAlaAT1 gene increasedgrain weight. In brief, we demonstrated that OsAlaAT1regulates carbon and nitrogen metabolism, which provides anew insight for the improvement of rice quality and yield.

        Loss of MicroRNA-137 Impairs the Homeostasis of Potassium in Neurons via KCC2

        Neuropsychiatric disorders are the leading cause of mental and intellectual disabilities worldwide. Current therapies against neuropsychiatric disorders are very limited, and very little is known about the onset and development of these diseases, and their most effective treatments. MIR137 has been previously identified as a risk gene for the etiology of schizophrenia, bipolar disorder, and autism spectrum disorder. Here we generated a forebrain-specific MIR137 knockout mouse model, and provided evidence that loss of miR-137 resulted in impaired homeostasis of potassium in mouse hippocampal neurons. KCC2, a potassium-chloride co-transporter, was a direct downstream target of miR-137. The KCC2 specific antagonist VU0240551 could balance the current of potassium in miR-137 knockout neurons, and knockdown of KCC2 could ameliorate anxiety-like behavior in MIR137 cKO mice. These data suggest that KCC2 antagonists or knockdown might be beneficial to neuropsychiatric disorders due to the deficiency of miR-137.

        Dronedarone Attenuates Ang II-Induced Myocardial Hypertrophy Through Regulating SIRT1/FOXO3/PKIA Axis

        Background and Objectives: Long-term pathological myocardial hypertrophy (MH) seriously affects the normal function of the heart. Dronedarone was reported to attenuate left ventricular hypertrophy of mice. However, the molecular regulatory mechanism of dronedarone in MH is unclear. Methods: Angiotensin II (Ang II) was used to induce cell hypertrophy of H9C2 cells. Transverse aortic constriction (TAC) surgery was performed to establish a rat model of MH. Cell size was evaluated using crystal violet staining and rhodamine phalloidin staining. Reverse transcription quantitative polymerase chain reaction and western blot were performed to detect the mRNA and protein expressions of genes. JASPAR and luciferase activity were conducted to predict and validate interaction between forkhead box O3 (FOXO3) and protein kinase inhibitor alpha (PKIA) promoter. Results: Ang II treatment induced cell hypertrophy and inhibited sirtuin 1 (SIRT1) expression, which were reversed by dronedarone. SIRT1 overexpression or PKIA overexpression enhanced dronedarone-mediated suppression of cell hypertrophy in Ang II-induced H9C2 cells. Mechanistically, SIRT1 elevated FOXO3 expression through SIRT1-mediated deacetylation of FOXO3 and FOXO3 upregulated PKIA expression through interacting with PKIA promoter. Moreover, SIRT1 silencing compromised dronedarone-mediated suppression of cell hypertrophy, while PKIA upregulation abolished the influences of SIRT1 silencing. More importantly, dronedarone improved TAC surgery-induced MH and impairment of cardiac function of rats via affecting SIRT1/FOXO3/PKIA axis. Conclusions: Dronedarone alleviated MH through mediating SIRT1/FOXO3/PKIA axis, which provide more evidences for dronedarone against MH.
