Background/Aims: Healthcare-associated infections (HAIs) in critically ill patients with prolonged length of hospital stay and increased medical costs. The aim of this study is to assess whether daily chlorhexidine gluconate (CHG) bathing will significantly reduce the rates of HAIs in adult intensive care units (ICUs). Methods: PubMed, EMBASE, and the Cochrane Central Register of Controlled Trials were systematically searched until December 31, 2014 to identify relevant studies. Two authors independently reviewed and extracted data from included studies. All data was analyzed by Review Manager version 5.3. Results: Fifteen studies including three randomized controlled trials and 12 quasi- experimental studies were available in this study. The outcomes showed that daily CHG bathing were associated with significant reduction in the rates of primary outcomes: catheter-related bloodstream infection (risk ratio [RR], 0.44; 95% confidence interval [CI], 0.32 to 0.63; p < 0.00001), catheter-associated urinary tract infection (RR, 0.68; 95% CI, 0.52 to 0.88; p = 0.004), ventilator-associated pneumonia (RR, 0.73; 95% CI, 0.57 to 0.93; p = 0.01), acquisition of methicillin-resistant Staphylococcus aureus (RR, 0.78; 95% CI, 0.68 to 0.91; p = 0.001) and vancomycin-resistant Enterococcus (RR, 0.56; 95% CI, 0.31 to 0.99; p = 0.05). Conclusions: Our study suggests that the use of daily CHG bathing can signifi- cantly prevent HAIs in ICUs. However, more well-designed studies are needed to confirm these findings.

Waste glass is a global solid waste with huge reserves. The discarded waste glass has caused a series of problems such as resource waste and environmental pollution, so it is urgent to recycle waste glass with high replacement level. Glass powder (GP), as a supplementary cementitious material (SCM), used in cement-based materials has already become one of the important ways to recycle waste glass mainly attributed to its pozzolanic reaction and filling effect, especially to the suppressed ASR expansion. This paper demonstrates an overview of the properties of GP and its effect on the fresh and mechanical properties of cement-based materials. The study found that the influence of GP on the performance of cement-based materials mainly depends on its content, particle size, color and type, curing conditions, and other SCMs. Finally, based on the problems involved in the investigation of concrete containing GP, some corresponding suggestions and efforts are given to further guide the utilization of GP in cement-based materials.

Purpose Current variability in methods for tumor mutational burden (TMB) estimation and reporting demonstrates the urgent need for a homogeneous TMB assessment approach. Here, we compared TMB distributions in different cancer types using two customized targeted panels commonly used in clinical practice. Materials and Methods TMB spectra of 295- and 1021-gene panels in multiple cancer types were compared using targeted next-generation sequencing (NGS). The TMB distributions across a diverse cohort of 2,332 cancer cases were then investigated for their associations with clinical features. Treatment response data were collected for 222 patients who received immune-checkpoint inhibitors (ICIs) and their homologous recombination DNA damage repair (HR-DDR) and programmed death-ligand 1 (PD-L1) expression were additionally assessed and compared with the TMB and response rate. Results The median TMB between gene panels was similar despite a wide range in TMB values. The highest TMB was eight and 10 in patients with squamous cell carcinoma and esophageal carcinoma according to the classification of histopathology and cancer types, respectively. Twenty-three out of 103 patients (22.3%) were HR-DDR–positive and could benefit from ICI therapy; out of those 23 patients, seven patients had high TMB (p=0.004). Additionally, PD-L1 expression was not associated with TMB or treatment response among patients receiving ICIs. Conclusion Targeted NGS assays demonstrated the ability to evaluate TMB in pan-cancer samples as a tool to predict response to ICIs. In addition, TMB integrated with HR-DDR–positive status could be a significant biomarker for predicting ICI response in patients.

In the genome of Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus, open reading frame 39 (Ha39) is the only gene predicted to encode an RNA recognition protein. Computer analysis revealed that Ha39 homologues were found in 15 NPVs, but not in GVs. Its transcripts were detected from 3 through 72 hours post infection (h p.i.) using RT-PCR and Northern blot analysis. The protein was detected in infected-cell lysates from 6 h p.i. Western blot assay of ODV and BV preparations revealed that Ha39 encodes a structural protein associated with BVs. Additionally, immunofluorescence microscopy demonstrated that the protein was present within cytoplasm in virus-infected cells, but not in the nuclear region.

A 3 factor, 3 level Box-Behnken factorialdesign combined with response surface methodology wasused to optimize the fermentation process for production ofextracellular polysaccharide (EPS) from Hirsutella sinensis. The 3 independent variables were temperature (X1), initialpH (X2), and the glucose concentration (X3). Experimentaldata were fitted to a 2nd order polynomial equation usingmultiple regression. The optimal fermentation conditionsof the production of EPS were a temperature of 18.21oC,an initial pH of 5.81, and a glucose concentration of 7.39g/L. The maximum predicted EPS production of 2.41 g/Lwas close to the actual experimental EPS yield (2.42±0.038%), demonstrating the model validity. EPS from H. sinensis had a direct in vitro immuno-stimulating activityusing murine macrophage RAW264.7 cells, and stimulatedrelease of several major cytokines (IL-1β, NF-κB, TNF-α,and iNOS) in a dose-dependent manner.

Aquaporin 1 (AQP1) is expressed in most microvascula-ture endothelial cells and forms water channels that play major roles in a variety of physiologic processes. This study aimed to delineate the transcriptional regulation of AQP1 by Mef2c in endothelial cells. Mef2c cooperated with Sp1 to activate human AQP1 transcription by binding to its proximal promoter in human umbilical cord vein endothelial cells (HUVEC). Over-expression of Mef2c, Sp1, or Mef2c/Sp1 increased HUVEC migration and tube-forming ability, which can be abolished AQP1 knockdown. These data indicate that AQP1 is a direct target of Mef2c in regulating angiogenesis and vasculogenesis of endothelial cells.

This study evaluated the effects of ZD1839, an orally active, selective epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, on nasopharyngeal carcinoma (NPC) both in vitro and in vivo. Influence of ZD1839 alone or combined with cisplatin on the NPC cell line CNE2 was detected by MTT assay with flow cytometry assessment of cell cycle distribution and apoptosis rates. Nude mice NPC xenografts were also used to evaluate the effects of ZD1839 alone or combined with cisplatin. The Student's t test evaluated statistical significance. ZD1839 alone or combined with cisplatin inhibited CNE2 cell line proliferation. ZD1839 induced CNE2 cell cycle arrest in the G1 phase, and higher concentrations induced apoptosis. Xenograft tumors were significantly smaller when treated with 200 mg/kg ZD1839, cisplatin, or cisplatin combined with 100 mg/kg ZD1839 than untreated controls. ZD1839 (200 mg/kg) alone showed good tumor inhibition effects, reduction of tumor weights, and smaller tumor volume without loss of body weight. ZD1839 (200 mg/kg) might provide a good and effective therapeutic reagent for NPC.

Drug-induced diabetes is widely reported inclinical conditions, and it is becoming a global issuebecause of its potential to increase the risk of severe cardiovascularcomplications. However, which drug mechanismsexert their diabetogenic effects and why the effectspresent significant inter-individual differences remain largelyunknown. Pharmacogenomics, which is the study ofhow genomic variation influences drug responses, providesan explanation for individual differences in drug-induceddiabetes. We highlight that pharmacogenomics can beinvolved in regulating the expression of genes in signalingpathways related to the pharmacokinetics or pharmacodynamicsof drugs or the pathogenesis of diabetes, contributingto the differences in drug-induced glucoseimpairment. The pharmacogenomics studies of the majordiabetogenic drugs are reviewed, including calcineurininhibitors, antipsychotics, hormones, and antihypertensivedrugs. We intend to elucidate the genetic basis of druginduceddiabetes and pave the way for the precise use ofthese drugs in the clinic.
