        Self-assembly and Mechanism of L-Alanine-based Dihydrazide Derivative as Excellent Gelator of Organic Solvents

        A new organogelator, L-Alanine dihydrazide derivative can self-assemble in various organic solvents and turned them into thermally reversible physical supramolecular organogels at extremely low concentrations (< 2 wt %). The gel-sol phase transition temperatures ($T_{GS}$) were determined as a function of gelator concentration and the corresponding enthalpies (${\Delta}H_g$) were extracted. Scanning electron microscopy (SEM) measurements revealed that the interspaces of fiber-like network structures were diminished with the increasing of the LMOG concentration. FT-IR spectroscopy studies revealed that hydrogen-bonding and hydrophobic interaction were the driving forces for the formation of the gels. Based on the data of XRD and molecular modeling, the possible packing modes for the formation of organogelator aggregates were proposed.

      • Preventive Effect of Actinidia Valvata Dunn Extract on N-methyl-N'-nitro-N-nitrosoguanidine-induced Gastrointestinal Cancer in Rats

        Purpose: This study was conducted to assess the preventive effect of Actinidia valvata Dunn (AVD) extract on an animal model of gastrointestinal carcinogenesis on the basis of changes in tumor incidence, cell proliferation, and apoptosis. Materials and Methods: Seventy-five male Wistar rats were divided into five different treatment groups with 15 rats in each group. Group I was given normal feed, whereas Groups II to IV were treated with 10% sodium chloride in the first six weeks and 100ug/mL of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in drinking water for 24 weeks. Group II was then given normal feed, whereas Group III was given AVD extract (0.24g/kg/day) for 12 weeks. Group IV was given AVD extract from the first week to the 36th week, whereas Group V was treated with AVD extract alone for 36 weeks. All rats were sacrificed at the end of the 36-week experiment and assessed for the presence of gastrointestinal tumors. The occurrence of cancer was evaluated by histology. Bax, Bcl-2, Caspase-3, and cyclinD1 were determined by immunohistochemical staining and Western blotting. Results: The incidences of gastric cancer were 0% in Group I, 73.3% in Group II, 33.3% in Group III, 26.7% in Group IV, and 0% in Group V. Bcl-2 and cyclinD1 expression was decreased in AVD extract treated groups, whereas Bax and Caspase-3 expression was increased. Comparison with group II revealed significant differences (p<0.01). Conclusions: AVD extract exhibits an obvious preventive effect on gastrointestinal carcinogenesis induced by MNNG in rats through the regulation of cell proliferation and apoptosis.

        Remarkable impact of amino acids on ginsenoside transformation from fresh ginseng to red ginseng

        Background: Amino acids are one of the major constituents in Panax ginseng, including neutral aminoacid, acidic amino acid, and basic amino acid. However, whether these amino acids play a role in ginsenosideconversion during the steaming process has not yet been elucidated. Methods: In the present study, to elucidate the role of amino acids in ginsenoside transformation fromfresh ginseng to red ginseng, an amino acids impregnation pretreatment was applied during thesteaming process at 120 C. Acidic glutamic acid and basic arginine were used for the acid impregnationtreatment during the root steaming. The ginsenosides contents, pH, browning intensity, and free aminoacids contents in untreated and amino acidetreated P. ginseng samples were determined. Results: After 2 h of steaming, the concentration of less polar ginsenosides in glutamic acidetreatedP. ginseng was significantly higher than that in untreated P. ginseng during the steaming process. However,the less polar ginsenosides in arginine-treated P. ginseng increased slightly. Meanwhile, free aminoacids contents in fresh P. ginseng, glutamic acid-treated P. ginseng, and arginine-treated P. ginsengsignificantly decreased during steaming from 0 to 2h. The pH also decreased in P. ginseng samples at hightemperatures. The pH decrease in red ginseng was closely related to the decrease in basic amino acidslevels during the steaming process. Conclusion: Amino acids can remarkably affect the acidity of P. ginseng sample by altering the pH value. Theywere the main influential factors for the ginsenoside transformation. These results are useful in elucidatingwhy andhowsteaming induces the structural change of ginsenoside in P. ginseng and also provides an effectiveand green approach to regulate the ginsenoside conversion using amino acids during the steaming process.

      • Efficacy and Safety of Sorafenib for Advanced Non-Small Cell Lung Cancer: a Meta-analysis of Randomized Controlled Trials

        Background: Many clinical trials have been conducted to evaluate sorafenib for the treatment of advanced NSCLC, but the results for efficacy have been inconsistent. The aim of this study was to evaluate the efficacy and safety of sorafenib in patients with advanced NSCLC in more detail by meta-analysis. Methods: This meta-analysis of randomized controlled trials (RCTs) was performed after searching PubMed, EMBASE, ASCO Abstracts, ESMO Abstracts, and the proceedings of major conferences for relevant clinical trials. Two reviewers independently assessed the quality of the trials. Outcomes analysis were disease control rate (DCR), progression- free survival (PFS), overall survival (OS) with 95% confidence intervals (CI) and major toxicity. Subgroup analysis was conducted according to sorafenib monotherapy, in combination with chemotherapy or EGFR-TKI to investigate the preferred therapy strategy. Results: Results reported from 6 RCTs involving 2, 748 patients were included in the analysis. Compared to sorafenib-free group, SBT was not associated with higher DCR (RR 1.31 (0.96- 1.79), p=0.09), PFS (HR 0.82 (0.66-1.02), p=0.07) and OS (HR 1.01 (0.92-1.12), p=0.77). In terms of subgroup results, sorafenib monotherapy was associated with significant superior DCR and longer PFS, but failed to show advantage with regard to OS. Grade 3 or greater sorafenib-related adverse events included fatigue, hypertension, diarrhea, oral mucositis, rash and HFSR. Conclusions: SBT was revealed to yield no improvement in DCR, PFS and OS. However, sorafenib as monotherapy showed some activity in NSCLC. Further evaluation may be considered in subsets of patients who may benefit from this treatment. Sorafenib combined inhibition therapy should be limited unless the choice of platinum-doublet regimen, administration sequence or identification of predictive biomarkers are considered to receive better anti-tumor activity and prevention of resistance mechanisms.

        Impact of cable sag on the efficiency of an inertial mass damper in controlling stay cable vibrations

        Passive negative stiffness dampers (NSDs) that possess superior energy dissipation abilities, have been proved to be more efficient than commonly adopted passive viscous dampers in controlling stay cable vibrations. Recently, inertial mass dampers (IMDs) have attracted extensive attentions since their properties are similar to NSDs. It has been theoretically predicted that superior supplemental damping can be generated for a taut cable with an IMD. This paper aims to theoretically investigate the impact of the cable sag on the efficiency of an IMD in controlling stay cable vibrations, and experimentally validate superior vibration mitigation performance of the IMD. Both the numerical and asymptotic solutions were obtained for an inclined sag cable with an IMD installed close to the cable end. Based on the asymptotic solution, the cable attainable maximum modal damping ratio and the corresponding optimal damping coefficient of the IMD were derived for a given inertial mass. An electromagnetic IMD (EIMD) with adjustable inertial mass was developed to investigate the effects of inertial mass and cable sag on the vibration mitigation performance of two model cables with different sags through series of first modal free vibration tests. The results show that the sag generally reduces the attainable first modal damping ratio of the cable with a passive viscous damper, while tends to increase the cable maximum attainable modal damping ratio provided by the IMD. The cable sag also decreases the optimum damping coefficient of the IMD when the inertial mass is less than its optimal value. The theoretically predicted first modal damping ratio of the cable with an IMD, taking into account the sag generally, agrees well with that identified from experimental results, while it will be significantly overestimated with a taut-cable model, especially for the cable with large sag.

        Halomonas alkalitolerans sp. nov., a Novel Moderately Halophilic Bacterium Isolated from Soda Meadow Saline Soil in Daqing, China

        A moderately halophilic bacterial strain 15-13^T, which was isolated from soda meadow saline soil in Daqing City, Heilongjiang Province, China, was subjected to a polyphasic taxonomic study. The cells of strain 15-13^T were found to be Gram-negative, rod-shaped, and motile. The required growth conditions for strain 15-13^T were: 1-23% NaCl (optimum, 7%), 10-50°C (optimum, 35°C), and pH 7.0-11.0 (optimum, pH 9.5). The predominant cellular fatty acids were C18:1 ω7c (60.48%) and C16:0 (13.96%). The DNA G+C content was 67.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequence comparisons indicated that strain 15-13^T clustered within a branch comprising species of the genus Halomonas. The closest phylogenetic neighbor of strain 15-13^T was Halomonas pantelleriensis DSM 9661^T (98.9% 16S rRNA gene sequence similarity). The level of DNA-DNA relatedness between the novel isolated strain and H. pantelleriensis DSM 9661^T was 33.8%. On the basis of the phenotypic and phylogenetic data, strain 15-13^T represents a novel species of the genus Halomonas, for which the name Halomonas alkalitolerans sp. nov. is proposed. The type strain for this novel species is 15-13^T (=CGMCC 1.9129^T =NBRC 106539^T).

      • UN 국제지수 평가 모델에 근거한 한-중 전자정부 비교연구

        전자정부의 발전에 따라 세계에 주목을 받고 있다. 국가가 전자정부를 통해서 정부를 제공하고, 통합적으로 관리하고, 국민들이 전자정부를 통해서 더 편하게 국가 정보를 얻을 수 있고, 민원 처리 등 구청이나 행정부문을 갈 필요 없고 인터넷으로 신속하게 처리할 수 있게 된다. 본 연구는 UN 국제지수 평가 모델 기준으로 전자정부의 발전은 어떤 단계를 있는지 파악하고, 그 단계에 따라서 국가 전자정부 평가에 대한 의미가 있다. 이를 위한 한국과 중국의 국제지수, 입법 현황, 관련 조직 등을 조사하여 차이를 분석하였다. 한국과 중국은 비슷한 시기에 전자정부를 시작했지만, 두 나라에 차이가 큰다. 한국에서는 전자정부를 구축하면서 이를 통해 가장 효율, 효과적이고 체계적인 방법으로 전자정부를 추진할 수 있게 된 결과를 볼 수 있고, 중국은 한국보다 아직 많이 뒤떨어지는 상태였다. 중국과 한국은 왜 이렇게 많은 차이가 나는지에 의문을 가지고, 본 연구는 국제지수 평가 모델을 통해서 중국과 한국의 차이를 확인하고자 하였다. With the development of e-government, the world is attracting attention. The government can provide the government through e-government, manage it collectively, allow citizens to obtain national information more easily through the e-government, and do not need to go to the ward office or administrative department such as complaints handling, do. This study is meaningful for national e - government evaluation according to the stage of development of e - government based on UN International Index Assessment Model. To do this, we analyzed the differences between Korea and China by examining the international index, legislative status, and related organizations. Korea and China started e-government in a similar period, but there is a big difference between the two countries. In Korea, e-government has been constructed and it has been shown that e-government can be implemented in the most efficient, effective and systematic way, and China is far behind in Korea. The question of why there are so many differences between China and Korea, this study tried to identify the differences between China and Korea through the international indexing model.

      • Analysis of Indoleamine 2-3 Dioxygenase (IDO) and EGFR Co-expression in Breast Cancer Tissue by Immunohistochemistry

        Background: To determine the amount of co-expression of IDO and EGFR in breast cancer patients. Materials and Methods:In order to obtain the distribution of co-expression of IDO and EGFR in breast cancer, we tested 110 breast cancer paraffin tissue blocks with immunohistochemical methods. Then we investigated the relationship between the diagnostic and pathologic characteristics (tumor size, lymph node status, histologic grade, the gene expression of ER, PR, HER2, p53, Ki67 and PCNA) with the situation of co-expression of IDO and EGFR by reviewing the medical records of 32 breast cancer patients. Results: Among 110 breast cancers, 32 cases demonstrated IDO and EGFR co-expression (29.1%), IDO and EGFR synchronous co-expression being found in 19.1% and asynchronous in 10.0%. Conclusions: IDO and EGFR were co-expressed in breast cancer, including synchronous and asynchronous co-expression. The results suggest that considering IDO and EGFR as two indicators for breast cancer treatment or prognosis analysis provides a potential option of individual treatment for the portion of breast cancer patients with co-expression of IDO and EGFR.

        Targeting treatment of bladder cancer using PTK7 aptamer-gemcitabine conjugate

        Gemcitabine (GEM) is one of the first-line chemotherapies for bladder cancer (BC), but the GEMs cannot recognize cancer cells and have a low long-term response rate and high recurrence rate with side effects during the treatment of BC. Targeted transport of GEMs to mediate cytotoxicity to tumor and avoid the systemic side effects remains a challenge in the treatment of BC.Based on a firstly confirmed biomarker in BC-protein tyrosine kinase 7 (PTK7), which is overexpressed on the cell membrane surface in BC cells, a novel targeting system protein tyrosine kinase 7 aptamer-Gemcitabine conjugate (PTK7-GEMs) was designed and synthesized using a specific PTK7 aptamer and GEM through auto-synthesis method to deliver GEM against BC. In addition, the antitumor effects and safety evaluation of PTK7-GEMs was assessed with a series of in vitro and in vivo assays.PTK7-GEMs can specifically bind and enter to BC cells dependent on the expression levels of PTK7 and via the macropinocytosis pathway, which induced cytotoxicity after GEM cleavage from PTK7-GEMs respond to the intracellular phosphatase. Moreover, PTK7-GEMs showed stronger anti-tumor efficacy and excellent biosafety in three types of tumor xenograft mice models.These results demonstrated that PTK7-GEMs is a successful targeted aptamer-drug conjugates strategy (APDCs) to treat BC, which will provide new directions for the precision treatment of BC in the field of biomarker-oriented tumor targeted therapy.
