        Performance validation and application of a mixed force-displacement loading strategy for bi-directional hybrid simulation

        Hybrid simulation (HS) is a versatile tool for structural performance evaluation under dynamic loads. Although real structural responses are often multiple-directional owing to an eccentric mass/stiffness of the structure and/or excitations not along structural major axes, few HS in this field takes into account structural responses in multiple directions. Multi-directional loading is more challenging than uni-directional loading as there is a nonlinear transformation between actuator and specimen coordinate systems, increasing the difficulty of suppressing loading error. Moreover, redundant actuators may exist in multi-directional hybrid simulations of large-scale structures, which requires the loading strategy to contain ineffective loading of multiple actuators. To address these issues, lately a new strategy was conceived for accurate reproduction of desired displacements in bi-directional hybrid simulations (BHS), which is characterized in two features, i.e., iterative displacement command updating based on the Jacobian matrix considering nonlinear geometric relationships, and force-based control for compensating ineffective forces of redundant actuators. This paper performs performance validation and application of this new mixed loading strategy. In particular, virtual BHS considering linear and nonlinear specimen models, and the diversity of actuator properties were carried out. A validation test was implemented with a steel frame specimen. A real application of this strategy to BHS on a full-scale 2-story frame specimen was performed. Studies showed that this strategy exhibited excellent tracking performance for the measured displacements of the control point and remarkable compensation for ineffective forces of the redundant actuator. This strategy was demonstrated to be capable of accurately and effectively reproducing the desired displacements in large-scale BHS.

        A Genetic Linkage Map Construction for Sesame (Sesamum indicum L.)

        Sesame (Sesamum indicum L.) is one of the oldest oilseed crops with high seed oil quality. The first sesame genetic linkage map based on F2 segregating population of an intraspecific cross between two cultivars was constructed. Using three types of PCR-based markers, 284 polymorphic loci including 10 EST-SSR marker, 30 AFLP marker and 244 RSAMPL marker, respectively, had been screened. Subsequently, a total of 220 molecular markers were mapped in 30 linkage groups covering a genetic length of 936.72 cM, and the average distance between markers was 4.93 cM. In this map, the linkage groups contained from 2 to 33 loci each and ranged in distance from 6.44 cM to 74.52 cM. Based on map information, sesame genome length was estimated to be approximately 1,232.53 cM, and genome coverage of this map was about 76.0%. As a starting point of sesame genome study, the genetic linkage map will be hopeful to tag traits of breeding interest and further aid in the sesame molecular breeding. Furthermore, RSAMPL marker had been also appreciated in this paper, for its first usage in genetic map construction and higher utilization potential in some crop species lacking much genome information.

        Effect of surface roughness on the flotation separation of hematite from fine quartz

        Although the effect of surface roughness on the floatability of a single mineral has been investigatedextensively, a systematic study for the effect of surface roughness on the flotation separation of differentminerals, e.g., flotation recovery, has never been dwelled. To fulfill this gap, in this study, the effect of surfaceroughness on the flotation separation of hematite from quartz was investigated with a series ofexperiments and detections. Micro-flotation experiments demonstrated that hematite particles withhigher roughness resulted in higher recoveries for both hematite and quartz, but the improvement ofhematite recovery was more significant, which promoted the flotation separation of hematite fromquartz with a higher separation index (SI). Adhesion force measurements revealed that after interactingwith 125 mg/L sodium oleate at pH 9, increasing nano-scaled roughness improved the hydrophobicity ofhematite surface, and reduced the adhesion force of water droplet on hematite surface. Scanning ElectronMicroscope – Energy Dispersive Spectrometer observations confirmed that increasing nano-scaled roughnessincreased the number of fine quartz particles coating on hematite surface. Combined with the analysisof adhesion of water droplet and observation of fine particles coating on hematite surface, the effectof surface roughness on minerals flotation separation could be better understood.

      • Implication of Polymorphisms in DNA Repair Genes in Prognosis of Hepatocellular Carcinoma

        XRCC1 genetic polymorphisms could be associated with increased risk of various cancer, including hepatocellular carcinoma (HCC), the fifth most common cancer. We here conducted a study to explore the role of selective SNPs of the XRCC1 and XPD genes in the prognosis of HCC. A total of 231 cases were collected, and genotyping of XRCC1 Arg194Trp, XRCC1 Arg399Gln, XPD Lys751Gln and XPD Asp312Asn was performed by duplex polymerase-chain-reaction with the confronting-two-pair primer method. Our findings indicated XRCC1 399Gln/Gln genotype was associated with a significant difference in the median survival time compared with patients carrying Arg/Trp and Arg/Arg genotypes, and individuals with XPD 751 Gln/ Gln genotype had a significantly greater survival time than patients carrying Lys/Lys and Lys/Gln genotypes. The Cox's regression analysis showed individuals carrying XRCC1 399Trp/Trp genotype had 0.55 fold risk of death from HCC than Arg/Arg genotype. Similarly, XPD 751Gln/Gln had a strong decreasein comparison to XPD Lys/Lys carriers with an HR of 0.34. These results suggest that polymorphisms in XRCC1 and XPD may have functional significance in the prognosis of HCC.

        Dehydration reaction of bio-ethanol to ethylene over modified SAPO catalysts

        Mnmodified SAPO-11 (Mn-SAPO-11), Zn-SAPO-11, Mn-SAPO-34 and Zn-SAPO-34 were first synthesized with hydrothermal method in the laboratory. Dehydration of ethanol to ethylene over SAPO-11, SAPO-34and four materials above as catalysts was carried out and Mn-SAPO-34 exhibited the best conversion and selectivity (99.35% and 98.44%, respectively) at 340 8C. The introduction of Mn2+ or Zn2+ into the SAPO channel generated in Mn-SAPO or Zn-SAPO samples was proved by X-ray diffraction (XRD), scanning electron microscopy (SEM) and nitrogen adsorption. NH3-TPD study revealed that modification of Mn2+or Zn2+ in the SAPO framework led to increase the weak acid strength and give rise to weak acid sites. The effects of operation parameters, such as loading amount, modification methods, reaction time, reaction temperature, mass space velocity and concentration of ethanol have also been investigated experimentally. 2010 The Korean Society of Industrial and Engineering Chemistry.         Zearalenone regulates key factors of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1-nuclear factor erythroid 2-related factor 2 signaling pathway in duodenum of post-weaning gilts

        Objective: This study explored the mechanism of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway under conditions of zearalenone (ZEA)-induced oxidative stress in the duodenum of post-weaning gilts. Methods: Forty post-weaning gilts were randomly allocated to four groups and fed diets supplemented with 0, 0.5, 1.0, or 1.5 mg/kg ZEA. Results: The results showed significant reductions in the activity of the antioxidant enzymes total superoxide dismutase and glutathione peroxidase and increases the malondialdehyde content with increasing concentrations of dietary ZEA. Immunohistochemical analysis supported these findings by showing a significantly increased expression of Nrf2 and glutathione peroxidase 1 (GPX1) with increasing concentrations of ZEA. The relative mRNA and protein expression of Nrf2, GPX1 increased linearly (p<0.05) and quadratically (p<0.05), which was consistent with the immunohistochemical results. The relative mRNA expression of Keap1 decreased linearly (p<0.05) and quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet. The relative mRNA expression of modifier subunit of glutamate-cysteine ligase (GCLM) increased quadratically (p<0.05) in all ZEA treatment groups and the relative mRNA expression of quinone oxidoreductase 1 (NQO1) catalytic subunit of glutamate-cysteine ligase decreased linearly (p<0.05) and quadratically (p<0.05) in the ZEA1.0 group and ZEA1.5 group. The relative protein expression of Keap1 and GCLM decreased quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet, respectively. The relative protein expression of NQO1 increased linearly (p<0.05) and quadratically (p<0.05) in all ZEA treatment groups in the duodenum. Conclusion: These findings suggest that ZEA regulates the expression of key factors of the Keap1-Nrf2 signaling pathway in the duodenum, which enables resistance to ZEA-induced oxidative stress. Further studies are needed to examine the effects of ZEA induced oxidative stress on other tissues and organs in post-weaning gilts.

      • A real-time hybrid testing based on restart-loading technology for viscous damper

        Real-Time Hybrid Testing (RTHT) requires the numerical substructure calculations to be completed within the defined integration time interval due to its real-time loading demands. For solving the problem, A Real-Time Hybrid Testing based on Restart-Loading Technology (RTHT-RLT) is proposed in this paper. In the proposed method, in case of the numerical substructure calculations cannot be completed within the defined integration time interval, the experimental substructure was returned back to the initial state statically. When the newest loading commands were calculated by the numerical substructure, the experimental substructure was restarted loading from the initial state to the newest loading commands so as to precisely disclosing the dynamic performance of the experimental substructure. Firstly, the methodology of the RTHT-RLT is proposed. Furthermore, the numerical simulations and experimental tests on one frame structure with a viscous damper are conducted for evaluating the feasibility and effectiveness of the proposed RTHT-RLT. It is shown that the proposed RTHT-RLT innovatively renders the nonreal-time refined calculation of the numerical substructure feasible for the RTHT. The numerical and experimental results show that the proposed RTHT-RLT exhibits excellent performance in terms of stability and accuracy. The proposed RTHT-RLT may have broad application prospects for precisely investigating the dynamic behavior of large and complex engineering structures with specific experimental substructure where a restarting procedure does not affect the relevant hysteretic response.

        Effects of zearalenone on the localization and expression of the growth hormone receptor gene in the uteri of post-weaning piglets

        Objective: In this study, we investigated the adverse effects of dietary zearalenone (ZEA) (0.5 to 1.5 mg/kg diet) on the localization and expression of the growth hormone receptor (GHR) in the uteri of post-weaning gilts and explored alternative mechanism of the reproductive toxicity of ZEA on piglets. Methods: A total of forty healthy piglets (Duroc×Landrace×Large White) aged 28 d were selected for study. Piglets were transferred to single cages after 10 days’ adaptation on an obstetric table. The animals were allocated to one of four treatments: a normal basal diet supplemented with 0 (Control), 0.5 (ZEA0.5), 1.0 (ZEA1.0), or 1.5 (ZEA1.5) mg/kg purified ZEA, and fed for 35 d after the 10-d adaptation. Analyzed ZEA concentrations in the diets were 0, 0.52±0.07, 1.04±0.03, and 1.51±0.13 mg/kg, respectively. At the end of the feeding trial, piglets were euthanized after being fasted for 12 h. Two samples of uterine tissue from each pig were rapidly collected, one of which was stored at –80°C for analysis of the relative mRNA and protein expression of GHR, and the second was promptly fixed in Bouin’s solution for immunohistochemical analysis. Results: The relative weight of the uteri and thickness of the myometrium and endometrium increased linearly (p<0.001) and quadratically (p<0.001) with an increasing level of ZEA. The results of immunohistochemical analysis indicated that GHR immunoreactive substance was mainly localizated in the cytoplasm of uterine smooth muscle, glandular epithelial, luminal epithelial, stromal, and vascular endothelial cells. In contrast, nuclear staining was rarely observed. The immunoreactive integrated optic density of GHR in the myometrium, luminal epithelium, glandular epithelium, and whole uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. The mRNA and protein expression of GHR in the uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. Conclusion: In conclusion, ZEA at a concentration of 0.5 mg/kg was sufficient to significantly thicken the myometrium and endometrium, and at a concentration of 1.0 mg/kg induced a high level of GHR expression to promote growth and development of the uteri. This revealed an alternative molecular mechanism whereby ZEA induces growth and development of the uteri and provides a theoretical basis for the revision of Chinese feed hygiene standards.

      • Expression of C4.4A is a Potential Independent Prognostic Factor for Patients with Gastric Cancer

        C4.4A, a metastasis-associated gene, encodes a glycolipid-anchored membrane protein which is overexpressed in several human malignancies. However, there are few data available on C4.4A expression and its relationship with progression in gastric cancer. Our study was designed to explore the expression of C4.4A in gastric cancer and to correlate it with clinical outcome. C4.4A expression was studied by quantitative real-time RT-PCR and immunohistochemistry for assessment of correlations with clinicopathological factors. C4.4A mRNA expression was significantly up-regulated in gastric cancer as compared with noncancerous tissue (p<0.05)., being observed in 107 (88.4%) of the 121 gastric cancer cases by immunohistochemistry. We found that the expression of C4.4A mRNA was correlated with size of the tumor, depth of invasion, lymph node metastasis, distant metastasis and TNM stage. Moreover, patients with overexpression of C4.4A has a significantly worse survival (p<0.05). Further multivariable analysis indicated that the expression of C4.4A was an independent prognostic indicator for gastric cancer (p<0.05). In conclusion, overexpression of C4.4A correlates with metastatic potential of gastric cancer and C4.4A could be a novel independent prognostic marker for predicting outcome.
