        Enhanced Catalytic Ability of Candida rugosa Lipase Immobilized on Pore-enlarged Hollow Silica Microspheres and Cross-linked by Modified Dextran in Both Aqueous and Non-aqueous Phases

        Candida rugosa lipase (CRL) is one of the most widely used lipases. To enhance the catalytic abilities of CRL in both aqueous and non-aqueous phases, hollow silica microspheres (HSMSs) with a pore size of 18.07 nm were used as an immobilization support, and aldehydecontaining dextrans were employed to further cross-link the adsorbed CRL. In the experimental ranges examined,the loading amount of lipase linearly increased to 171 ±3.4 mgprotein/gsupport with the CRL concentration and all the adsorption equilibriums were reached within 30 min. After simple cross-linking, the tolerance to pH 4.0 ~ 8.0 as well as the thermal stability of immobilized CRL at 40 ~ 80oC were both substantially increased, and 82 ± 2.1% activity remaining after the sixth reuse. The immobilized CRL was successfully applied to the resolution of racemic ibuprofen in non-aqueous phase. The initial reaction rate increased by 1.4- and 3.6-fold compared with the rates of adsorbed and native lipases, respectively. Furthermore, the R-ibuprofen was obtained at ee > 93%, and the enantiomeric ratio reached E > 140 at the conversion of 50 ± 1.5% within 48 h.

        Effects of zearalenone on the localization and expression of the growth hormone receptor gene in the uteri of post-weaning piglets

        Objective: In this study, we investigated the adverse effects of dietary zearalenone (ZEA) (0.5 to 1.5 mg/kg diet) on the localization and expression of the growth hormone receptor (GHR) in the uteri of post-weaning gilts and explored alternative mechanism of the reproductive toxicity of ZEA on piglets. Methods: A total of forty healthy piglets (Duroc×Landrace×Large White) aged 28 d were selected for study. Piglets were transferred to single cages after 10 days’ adaptation on an obstetric table. The animals were allocated to one of four treatments: a normal basal diet supplemented with 0 (Control), 0.5 (ZEA0.5), 1.0 (ZEA1.0), or 1.5 (ZEA1.5) mg/kg purified ZEA, and fed for 35 d after the 10-d adaptation. Analyzed ZEA concentrations in the diets were 0, 0.52±0.07, 1.04±0.03, and 1.51±0.13 mg/kg, respectively. At the end of the feeding trial, piglets were euthanized after being fasted for 12 h. Two samples of uterine tissue from each pig were rapidly collected, one of which was stored at –80°C for analysis of the relative mRNA and protein expression of GHR, and the second was promptly fixed in Bouin’s solution for immunohistochemical analysis. Results: The relative weight of the uteri and thickness of the myometrium and endometrium increased linearly (p<0.001) and quadratically (p<0.001) with an increasing level of ZEA. The results of immunohistochemical analysis indicated that GHR immunoreactive substance was mainly localizated in the cytoplasm of uterine smooth muscle, glandular epithelial, luminal epithelial, stromal, and vascular endothelial cells. In contrast, nuclear staining was rarely observed. The immunoreactive integrated optic density of GHR in the myometrium, luminal epithelium, glandular epithelium, and whole uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. The mRNA and protein expression of GHR in the uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. Conclusion: In conclusion, ZEA at a concentration of 0.5 mg/kg was sufficient to significantly thicken the myometrium and endometrium, and at a concentration of 1.0 mg/kg induced a high level of GHR expression to promote growth and development of the uteri. This revealed an alternative molecular mechanism whereby ZEA induces growth and development of the uteri and provides a theoretical basis for the revision of Chinese feed hygiene standards.

      • Arena Simulation of Multi-Level Medicine Inventory Control In Hospital Pharmacy

        Precise control of hospital pharmacy inventory is an important guarantee for hospital. The optimal inventory policy for a multi-level medicine inventory control in hospital pharmacy was considered in this paper, and modeling simulation technology and intelligent optimization has been used. Analyzed with large-scale hospitals data, this paper has discussed the following problems: random fluctuations of demand in hospital departments, total volume limits and uncertainty response time of suppliers’ delivery. Establish multilevel pharmaceutical inventory simulation model, then use PSO method for intelligent optimization. Examples of simulation models and PSO algorithms verified by numerical results show that: when each department was tested randomly varying demand of the patients, multi-level inventory management must be overall coordination in order to achieve optimum system.

        A Single-phase Buck-boost AC-AC Converter with Three Legs

        This paper proposes a single-phase buck-boost AC-AC converter. It consists of three legs with six switching units (each unit is composed of an active switch and a diode) and its input and output ports share a common ground. It can provide buck-boost voltage operation and immune from shoot-through problem. Since only two switching units are involved in the current paths, the conduction losses are low, which improves the system efficiency. The operation principle of the proposed circuit is firstly presented, and then, various operation conditions are introduced to achieve different output voltages with step-changed frequencies. Additionally, the parameters design and comparative analysis of the power losses are also given. Finally, experimental results verify the correctness of the proposed converter.

      • Hemalin, a thrombin inhibitor isolated from the midgut EST database of hard tick Haemaphysalis longicornis

        A full-length sequence of a thrombin inhibitor (designated as hemalin) from the midgut of pathenogenetic Haemaphysalis longicornis has been identified. Sequence analysis shows that this gene belongs to a Kunitz-type family, containing two Kunitz domains with high homology to boophilin, the thrombin inhibitor from Rhipicephalus (Boophilus) microplus. The recombinant protein expressed in insect cells delays bovine plasma clotting time and inhibits both thrombin-induced fibrinogen clotting and platelet aggregation. A 20-kDa protein was detected from the midgut lysate with antiserum against recombinant hemalin. The gene expresses at all stages of the tick except for the egg stage and mainly in the midgut of the female adult tick. Real-time PCR analysis shows that this gene has a distinctly high expression level in the rapid bloodsucking period of the larvae, nymphs, and adults. Disruption of the hemalin gene led to a 2-day extension of the tick blood feeding period, and 27.7% of the ticks did not successfully complete the blood feeding. These findings indicate that the newly indentified thrombin inhibitor from the midgut of H. longicornis might play an important role in tick blood feeding.

        Cortical Depression and Potentiation: Basic Mechanisms for Phantom Pain

        People experience the feeling of the missing body part long after it has been removed after amputation are known as phantom limb sensations. These sensations can be painful, sometimes becoming chronic and lasting for several years (or called phantom pain). Medical treatment for these individuals is limited. Recent neurobiological investigations of brain plasticity after amputation have revealed new insights into the changes in the brain that may cause phantom limb sensations and phantom pain. In this article, I review recent progresses of the cortical plasticity in the anterior cingulate cortex (ACC), a critical cortical area for pain sensation, and explore how they are related to abnormal sensory sensations such as phantom pain. An understanding of these alterations may guide future research into medical treatment for these disorders.

        진로장벽과 진로결정 자기효능감 및 진로준비행동 간의 관계- 중국 랴오닝 지역 대학생을 중심으로 -

        This study attempts to reveal the causal relationship between certain variables, with the objective being toinvestigate the influence of career disability on self-efficacy and career preparation behavior of Chinese college students. In terms of data collection, online surveys were conducted during the period spanning September 1 to September 20,2022 targeting fourth-year university students living in Liaoning, China. Atotalof536completedsurveyswere used for thefinal analysis, and the SPSS 25.0 program was employed in this regard. The results of the study confirmed that the careerbarriers of Chinese college students had both positive and negative effects on career decision-making self-efficacy andcareer preparation behavior, signaling a partial causal relationship. This indicates that career barriers are not only negativefactors in career decision-making or career preparation. The current study is meaningful in that the results can serve asthe basis for future studies, as the work itself investigated the influence of career barriers on career decision self-efficacyand career preparation behavior; additionally, the results were quantified. It is anticipated that there will be further studieson such influences considering the diverse variables of career barriers, career decision self-efficacy, and career preparationbehavior. The results of this study are expected to provide useful information on career preparation to Chinese collegestudents – information which can be used as empirical data.

      • Recreational Physical Activity and Risk of Ovarian Cancer: a Meta-analysis

        Our aim was to access the association between recreational physical activity (RPA) and risk of ovarian cancer (OC). The studies were retrieved from the PubMed and Embase databases up to February 20th, 2014. Risk ratios (OR) and 95% confidence intervals (CI) were used to estimate effect sizes. Random-effects or fixed-effects models were used to pool the data. The trim and fill method was applied for sensitivity analysis. Begg's rank correlation test and Egger's regression asymmetry test were employed to assess the publication bias. A total of 6 studies (435398 participants including 2983 OC patients) were included in this meta-analysis. The overall estimate indicated that there was weakly inverse association between RPA and OC risk (RR=0.90, 95%CI: 0.72-1.12, p=0.335). Meanwhile, for prospective cohort studies, a result consistent with the overall estimate was obtained (RR=1.12, 95% CI: 0.88-1.42, p=0.356). However, for case control studies, the pooled estimate of RR was 0.76 (95%CI: 0.64-0.90, p=0.002), indicating a clear significant association between RPA and OC risk. In addition, the sensitivity analysis indicated a significant link between RPA and risk of OC after removing Lahmann's study (RR=0.80, 95% CI: 0.68-0.93, p=0.004). No significant publication bias was found (Begg's test: p=1.00; Egger's test: p=0.817). In conclusion, our meta-analysis indicated a weakly inverse relationship between RPA and the occurrence of OC.
