        한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

      • 한국인에서 Ataxia Telangiectasia 유전자의 다형성에 대한 연구

        Ataxia-telangiectasis(AT) is an autosomal recessive disorder characterized by berebellar ataxia, oulocutaneous telangiectaisa, immune defects, and a predisposition to malignancy. The gene for AT, deginated ATM(ataxia telangiectasia mutated), located on chromosome 11q22-q23, contains 66 exons spanning approximately 150 kb of genomic DNA and encodes a protein of 3056 amino acids. It is sell established that patiens with At have a strong predispositon to malignancy, particularly leukemias and lymphomas. An increase predisposition to malignancy has also been suggested to involve heterozygotes for ATM gene mutations. The genetic bases of carcinogenesis for various cancers in Korea have not been studies in detail and the study on the heterozygosity of AT in Korean population would be necessory. In this study, 150 healthy Koreans were studies for the polymorphism at AT using PCR of exon 39 of AT gene. The result was that 110 persons (73%) were heterozygote for wild and polymorphic type(A1/A2), and 40(27%) revealed homozygous polymorphic type A2/A2. The wild type homozygous tyep A1/A1 was not noted in Korean. Above results suggests that heterozygosity of AT is not infrequent in Koreans, and further studies on the carcinogenetic effects of AT in various cancers in Korea would be necessory.

      • Fotemustine, Teniposide and Dexamethasone in Treating Patients with CNS Lymphoma

        Purpose: We developed and evaluated a regimen including fotemustine, teniposide and dexamethasone (FTD) for treating patients with central nervous system (CNS) lymphoma based on pharmacokinetic properties of individual agents and in combination. Patients and Methods: In a comparison study, 8 patients with primary CNS lymphoma (PCNSL) and 8 with secondary CNS lymphoma (SCNSL) were treated with FTD (comprising fotemustine 100 mg/m2, 1h infusion, day 1; teniposide 60 mg/m2, >0.5 h infusion, on day 2, 3, 4; dexamethasone 40 mg, 1h infusion, on day 1, 2, 3, 4 and 5; and methotrexate 12 mg, cytosine arabinoside 50 mg plus dexamethasone 5 mg intrathecally, on day 2 and 7). Cycles were repeated every 3 weeks. After response assessment, patients received whole brain radiotherapy. Results: Of the 8 PCNSL patients, 4 (50%) achieved CR and 3 (38%) PR, an overall response rate of 88%. Four patients (50%) were in continuing remission at the end of this study after a median follow-up of 30 months (range 10 to 56 months). Of the 8 SCNSL patients the overall response rate was 63% (CR+PR: 38%+25%). All responses were achievable with predictable toxicity mainly reflecting reversible myelosuppression. Conclusion: This study suggests that FTD could be an effective treatment for CNS lymphoma, and is worthy of further evaluation.

        A Brief Review on Relaxor Ferroelectrics and Selected Issues in Lead-Free Relaxors

        Relaxor ferroelectricity is one of the most widely investigated but the least understood material classes in the condensed matter physics. This is largely due to the lack of experimental tools that decisively confirm the existing theoretical models. In spite of the diversity in the models, they share the core idea that the observed features in relaxors are closely related to localized chemical heterogeneity. Given this, this review attempts to overview the existing models of importance chronologically, from the diffuse phase transition model to the random-field model and to show how the core idea has been reflected in them to better shape our insight into the nature of relaxor-related phenomena. Then, the discussion will be directed to how the models of a common consensus, developed with the so-called canonical relaxors such as Pb(Mg1/3Nb2/3)O3 (PMN) and (Pb, La)(Zr, Ti)O3 (PLZT), are compatible with phenomenological explanations for the recently identified relaxors such as (Bi1/2Na1/2)TiO3 (BNT)-based lead-free ferroelectrics. This review will be finalized with a discussion on the theoretical aspects of recently introduced 0−3 and 2−2 ferroelectric/relaxor composites as a practical tool for strain engineering.

        Multifunctional wool fiber treated with ε-polylysine

        A creative method for fabricating environmentally-benign multifunctional wool fibers was established and reported. Through coating the wool fibers with ε-polylysine, the surface morphology and biochemical properties of the fibers were altered, enhancing their antimicrobial, hygroscopic and finished properties. The process of ε-polylysine coating was dependent on the solution environment, which influenced the electrostatic interactions between ε-polylysine molecules and wool fibers. The results showed that a maximum ε-polylysine coating (23.60 mg/g) on the surface of wool fibers was reached when wool fibers were soaked at 50 oC for 2 h in the solution with 10% on weight of fabric (owf) ε-polylysine and pH 8.0. The coated wool fiber showed promising antimicrobial rates of 96.98% and 97.93%against Escherichia coli and Micrococcus luteus, respectively. The wool fiber coated with the ε-polylysine was more hydrophilic than the uncoated wool fabrics. The functional wool fibers after water scrubbing for two times still have good antibacterial efficiency against Escherichia coli and Micrococcus luteus, and antimicrobial rates were 96.77%and 97.33%, respectively. This study shows that wool fibers modified by the nontoxic ε-polylysine have a great potential to be used in constructing multifunctional textiles.

      • High Resolution Melting Analysis for Epidermal Growth Factor Receptor Mutations in Formalin-fixed Paraffin-embedded Tissue and Plasma Free DNA from Non-small Cell Lung Cancer Patients

        Background:The aim of the research was to explore a cost effective, fast, easy to perform, and sensitive method for epidermal growth factor receptor (EGFR) mutation testing. Methods: High resolution melting analysis (HRM) was introduced to evaluate the efficacy of the analysis for dectecting EGFR mutations in exons 18 to 21 using formalin-fixed paraffin-embedded (FFPE) tissues and plasma free DNA from 120 patients. Results: The total EGFR mutation rate was 37.5% (45/120) detected by direct sequencing. There were 48 mutations in 120 FFPE tissues assessed by HRM. For plasma free DNA, the EGFR mutation rate was 25.8% (31/120). The sensitivity of HRM assays in FFPE samples was 100% by HRM. There was a low false-positive mutation rate but a high false-negative rate in plasma free DNA detected by HRM. Conclusions: Our results show that HRM analysis has the advantage of small tumor sample need. HRM applied with plasma free DNA showed a high false-negative rate but a low false-positive rate. Further research into appropriate methods and analysis needs to be performed before HRM for plasma free DNA could be accepted as an option in diagnostic or screening settings.

        Glabridin attenuates atopic dermatitis progression through downregulating the TLR4/MyD88/NF-κB signaling pathway

        Background Glabridin (GB), a bio-available phytoestrogen, displays various biological properties such as anti-infammatory, antibacterial, and antiviral. Objective To explore the role of GB in the process of atopic dermatitis (AD). Methods CCK8 was used to detect the therapeutic efect of Glabridin in HaCat and NHEK cell infammatory models. And evaluated the efect on cell proliferation and cell viability. The expression of TLR4, MyD88, P65 and P50 in HaCat and NHEK cell tissues was detected by qRT-PCR and PCR. At the same time, an AD animal model was constructed, and the cell experiment results were verifed by hematoxylin–eosin (HE) and Immunohistochemistry staining (IHC). Results Enzyme-linked immunosorbent assay (ELISA) demonstrated that IL-1β, IL-6, and TNF-α upregulated by lipopolysaccharide (LPS) was decreased by treatment with GB. AD progression was further confrmed to be regulated by GB by inhibiting the TLR4/MyD88/NF-κB signaling pathway through real-time PCR and Western blot analyses. An AD-like mouse model demonstrated that GB considerably alleviated epidermal injury, relieve edema, and reduced infammatory cell infltration by H&E staining. Concurrently, IHC staining exhibited GB to reduce AD progression by impeding TLR4 expression. Conclusion GB was observed to decrease the AD progression by suppressing the TLR4/MyD88/NF-κB signaling pathway, which may likely serve as a novel therapeutic drug for AD management.
