      • 위암환자의 복강내에 투여한 Mitomycin C-Carbon Particle의 Mitomycin 용출에 관한 연구

        Locoregional recurrence is the most common type of recurrence in surgical operation of gastric adenocarcinoma, and peritoneal dissemination is one of the most difficult problems in advanced gastric adenocarcinoma treatment. Because the peritoneal cavity is the most common site of the first recurrence after gastric cancer resection, intraperitoneal chemotherpy seems a logical choice for cancer chemotherapy. The Mitomycin C(MMC) adsorbed by the activated charcoal particles(CH) is relatively released when the drug concentration surrounding the carbon particles becomes low in the peritoneum of the peritoneal cavity. For the intraperitoneal chemotherapy on the advanced gastric adenocarcnoma, mitomycin C adsorbed on activated carbon particles was administered in the peritoneal cavity just before abdominal wall closure. The closed drainage tubes were inserted in the peritoneal cavity and clamped for tuo hours after completion of operation. MMC concentrations were serially measured in peritoneal fluid, plasma and urine at 2hour, 48 hour, 72 hour and 168 hour following its administration in order to study the efficacy of the MMC-CH as a drug delivery system. There were minimal toxicities in born marrow, liver, and gastrointestinal system after intraperitoneal MMC-CH administration. The data of this study suggested that MMC-CH may have a somewhat more beneficial effect than surgery alone when administered in optimal dose and schedules, but the MMC concentration of the peritoneal fluid was not sufficient to eradicate remnant cancer cells, and effective duration of maintenance was only below 24 hours in the peritoneal fluid and plasma.

      • Rat의 복강내에 투여한 5-fluorouracil-polyglycolic acid 제형이 장기에 미치는 영향에 관한 연구

        For more effective intraperitoneal chemotherapy, the authers made 5-fluorouracil-polyglycolic acid composite disks(FU-PGA disks) with 5-fluorouracil and polyglycolic acid, a biodegradable polymer. We inserted the FU-PGA disk into the peritoneal cavites of Sprague-Dawley male rats, the control and three experimental groups; one FU-PGA disk insert group(300mg/kg), two FU-PGA disk insert group(600mg/kg), and three FU-PGA disk insert group(900mg/kg). The control group received a similar number of PGA disk inserts. A pharmacokinetic study was performed to measure the 5-fluorouracil concentration in the peritoneal fluid, blood, and tissues(liver, kidney and heart) at 24 hours, 72hours and 168 hours after insertion of the FU-PGA disk. Light and electron microscopic studies were done. The results were as follows: 1) The number of white blood cells and platelets decreased after FU-PGA insertion. The degree was proportional to the duration and amounts inserted. The change in the number of red blood cells varied slightly. 2) Light microscopically, slight changes were noted at 168 hours in the 3 disk insert group (900mg/kg). Mild fatty change and hepatocyte degeneration around the central veins of the liver were noted, with vacuolar degeneration in the proximal convoluted tubules of the kidney. 3) Electron microscopically,the liver showed focal increases of lipid droplets in the hepatocytes, and irregular nuclear membrane with focal nucleolar segregation of the fibrillar and granular elements. Also the double membranous structure of the hepatocytes and Kupffer cells was widened with bleb formation. The kidney showed wide separation of the nuclear double membrane in the proximal convoluted tubular cells and mesangial cells, with vacuolation and myelin figure formation in the mitochondria of the proximal convoluted tubular cells, at 72 hours and 168 hours in the 2 to 3 FU-PGA insert groups. The heart showed focal loss of cristae,vacuolation and myelin figure formation in the mitochondria of the 2 to 3 FU-PGA insert groups. In conclusion, despite the large amont of FU-PGA inserted, the histopathological changes in the liver, kidney and heart were slight, and consonant with the very low amount of 5-fluorouracil concentrations detected in the liver, kidney and heart. The above results suggest that the FU-PGA composite can serve as a new device for releasing drugs in a controlled manner and easily targeted to intraperitoneal organs. This device can improve the efficacy of intraperitoneal chemotherapy for gastric cancer.

        To evaluate the in-vivo effects of mitomycin-C-polyglycolic acid composite(MMC-PGA) the disk: shaped composite were implanted into the peritoneal cavity of the Sprague-Dawley rats. The pathologic changes were examined at various time points up to 12 weeks. Initially the soft tissue around the inserted disks showed capillary congestion at 3 days. Inflammatory infiltrates with foreign body giant cells appeared from the 2nd week and reached peak response at 6-8 weeks. These reaction diminished prominently at 12 weeks. No specific pathologic change was found in the liver, the kidneys, and the heart. The above results suggest that the MMC-PGA composite can serve as a new device for intraperitoneal chemotherapy of various types of cancers.

      • 노화가 인체 중간엽 줄기세포로부터 조골세포로의 증식 및 분화에 미치는 영향

        연구배경: 일반적으로 골다공증과 연관된 위험인자로는 연령, 폐경, 약물, 불충분한 칼슘섭취, 만성질환 및 운동부족 등이 있는데, 특히 노화가 진행할수록 골밀도가 감소하는 것은 잘 알려져 있다. 노화와 관련하여 진행되는 골소실은 조골세포 및 전구조골세포의 기능적 결핍에 의한 골형성의 감소가 주요한 요인으로 여겨지고 있다. 그 동안 연령이 조골모 세포의 양과 조골모 세포로부터 성숙조골세포로의 분화 및 증식에 미치는 영향에 대한 일부 보고들이 있었으나 아직 일치된 견해는 없는 형편이다. 방법: 다양한 연령의 사람으로부터 골수를 채취, 중간엽 줄기세포가 포함된 단핵세포를 분리한 후 조골세포로 분화하기 좋은 조건하에서 배양하였다. 대상군은다시 젊은군과 노령군으로 구분하여 다양한 변수를 비교 분석하였다. 일차배양에서는 CFU-F를 계수하여 골수내 중간엽 줄기세포의 수를 추산하였고, 칼슘측정을 통하여 기질의 무기화 정도를 비교하였다. 계대배양후 이차배양에서는 시기별로 알카리성 포스파타제 활성도를 측정하고 오스테오칼신 mRNA의 발현을 관찰하여 젊은군과 노령군 사이의 증식능 차이를 비교하였다. 또한 이차배양 시기별로 MTT 측정을 하여 양군간에 증식능 차이가 있는지 알아보았다. 결과: 1. 일차배양 15일째에 평균 CFU-F의 수는 젊은군에서 유의하게 많았다(젊은군 148.3±28.9, 노령군 54.3±9.1, p=0.02). CFU-F의 평균면적은 젊은군에서 넓은 경향을 보였으나 통계적으로 유의하지는 않았다. 2. 일차배양 17일 경과 후 양군간에 기질 칼슘 침착정도는 유의한 차이를 보이지 않았다(젊은군 103.6±50.6, 노령군: 114.0±56.5, p=NS). 3. 이차배양 10일째에 젊은군에서 알카리성 포스파타제 활성도가 고령군에 비해 유의하게 높았다(젊은군: 935.5±115.0 U/mg, 노령군: 578.4±115.7U/mg,p.0.05). 고령군에서는 시간 경과에 따른 변화가 미약했으며 전반적으로 알카리성 포스파타제의 활성도가 젊은군에 비해 낮았다. 4. 이차배양도중 오스테오칼신 mRNA의 발현은 배양시기별로 젊은군에 비해 고령군에서 더 낮은 경향을 관찰할 수 있었다. 5. 이차배양 10일과 15일에 젊은군에서 노령군보다세포증식이 유의하게 증가된 양상을 보였다(10앓 젊은군 0.73±0.05, 노령군 0.58±0.04, p=0.05, 15일; 젊은군 0.80±0.05, 노령군 0.70±0.03, p=0.05).결론: 이상의 연구에서 저자들은 노령군에서 젊은군보다 골수 내 중간엽줄기세포의 수가 적고, 노령군에서 유래한 전구조골세포의 성숙조골세포로의 증식 및 분화가 젊은군 보다 감소해 있는 것을 관찰할 수 있었다. Background: Osteoblasts originate from osteoprogenitor cells in bone marrow stroma, termed mesenchymal stem cells (MSCs) or bone marrow stromal cells. Each MSC forms colonies (colony forming units-fibroblasts [CFL-Fs]) when cultured ex vivo. There are some reports about the age-related changes of the number and osteogenic potential of osteoprogenitor cells, but any relationship has not been clearly established in humans. In this study, we counted MSCs using CFU-Fs count and examined the proliferative capacity and differentiation potential of osteoprogenitor cells. Finally, we analyzed how these parameters varied with donor age. Methods: Bone marrow was obtained from the iliac crest of young (n=6, 27.2±8.6 years old) and old (n= 10, 57.4k6.7 years old) healthy donors. Mononuclear cells, including MSCs, were isolated and cultured in osteogenic medium. In primary culture, we compared the colony-forming efficiency of MSCs between the two groups and determined the matrix calcification. When primary culture showed near confluence, the cells were subcultured. Alkaline phosphatase activity, osteocalcin expression by RT-PCR and proliferative potential by MTT assay were examined by the time course of secondary culture. Results: At the 15th day of primary culture, the mean number of CFU-Fs was significantly higher in the younger donors (young: 148.3±28.9, old: 54.3±9.1, p=0.02) and the mean size of CFL-Fs was also larger in the younger donors than the older donors. However, matrix calcification was not different between the two groups (young: 103.6±50.6, old: 114.0±56.5, p=NS). In secondary culture, alkaline phosphatase activities were significantly lower in the older donors. The younger donors showed peak alkaline phosphatase activity at day 10, while the older donors didn't showed a remarkable peak (young: 935.5±115.OU/mg, old: 578.4±115.7U/mg, p<0.05). Total cell number as a proliferative index increased progressively during the secondary culture and a significantly greater cell number was noted in the younger donors. Osteocalcin expression was generally upregulated in the younger donors, but this was not statistically significant. Conclusion: Our study shows that the number of osteoprogenitor cells is decreased during aging and that the proliferative capacity and differentiation potential of osteoprogenitor cells seem to be reduced during aging (J Kor SOC Endocrinol 18:296-305, 2003).

      • 금형공장의 협업적 전자주문처리 시스템 개발

        현재 금형공장에서는 변화하는 시장에 빠른 반응을 보이고, 생산하고 있는 제품의 높은 품질과 낮은 가격을 보장하기 위하여 기존 시스템과 인터넷을 접목하려는 많은 시도를 하고 있다. 금형공장에는 ETO(Engineering-To-Order) 방식의 제조방식과 사출금형의 특수성으로 인하여 표준견적 시스템의 개발이 어렵다. 그리고 영업과 수주에 반영할 객관적인 정보가 없으며, 고객의 요청 시 실시간으로 확인 가능한 정보가 존재하지 않는다. 이러한 금형공장의 특징을 반영하고, 문제점을 극복할 수 있는 협업적 전자주문 처리 시스템의 프레임윅(framework)을 설계하고, 또한 각 세부 모듈인 견적가격 산정기능(Cost Estimation Tool), 납기 가능일자 산정기능(Leadtime Tool), 주문상황 응답처리 기능(Order Status Tool), 작업상황 파악기능(Shop Status Tool)을 설계한다. 그리고 현장 적용이 가능하도록 하기 위하여 금형공장의 업무 프로세스를 분석한다. 그리고 주문처리 프로세스의 UMM방법론 모델링을 통하여 윈형(prototype) 시스템을 설계 및 개발하고자 한다.

      • Bovine Leukemia Virus에 인공감염된 한국 재래흑염소의 체액성 면역반응

        Korean native goats were experimentally infected with bovine leukemia virus(BLV), and the changes of humoral immune responses were investigated by agar-bel immunodifusion test(AGID), complement dependent antibody cytotoxicity(CDAC), complement fixation test(CF) and virus neutralization test(VN) using syncytium inhibition assay. AGID antibody titers of Group Ⅰ inoculated with FLK-BLV cells were 2 to 4 at 1 month post - inoculation(pi), and persisted for leucocytes from BLV-infected cattle showed the lower AGID antibody titer than GroupⅠ. Cytotoxicity of rabbit complement and normal goat serum against FLK-BLV cells was investigated. Four out of 6 rabbit complements were revealed cytotoxicity index under 5.0%. Range of cytotoxic index of 17 normal goat sera were 11.8 ± 28.6 to 28.6 ± 9.7. CDAC antibody titers of Group Ⅰ were 8 to 16 at 1 month pi and then increased gradually showing maximum titer of 128 at 6 months pi. At 12 months pi CDAC titer were 32 to 128. Group Ⅱ showed relatively lower CDAC titer than Group Ⅰ. CF antibody titer of Group Ⅰ were 4 to 8 at 1 months pi, and then increased up to 32 to 128 at 8 to 10 months pi. GroupⅡ showed the similar patterns of CF titers as Group Ⅰ, but No 3 revealed particularly lower CF titer throughout the experiments. In syncytium assay for detection of BLV in the peripheral lymphocytes, all six heads of goats in Group Ⅰ and Group Ⅱ revealed the positive SA through 12 months showing the maximum SA values of 148. The SA values appeared declining tendency as the observation period passed. VN antibody titer of Group Ⅰ were 16 to 32 1 month pi, and then increased gradually by the maximum titer of 128. Through 12 month experiment, Group Ⅱ manifested the lower VN antibody titer than Group Ⅰ. throughout 12 month period, the control groups that are Group Ⅲ and Group Ⅳ, showed no antibody responses in the above tests.

        The porcine reproductive and respiratory syndrome virus was inoculated into swine tracheal organ culture. The ballooning degeneration of the tracheal ciliated epithelium and a decrease of ciliary activity from the tracheal organs infected with PRRS virus were observed one day post-infection(PI). Tracheal epithelium showed degeneration and focal necrosis 2 days PI. Epithelial necrosis and loss of cilia and epithelial cells became more severe 3 and 4 days PI. An entire loss of cilia and epithelial cells were recognized 4 days PI, however, control tracheal organs were normal during the period of the experiment except a little decrease of ciliary activities with time..

      • IEEE 802.11e MAC 계층에서의 QoS 제공을 위한 스케줄링 방안 연구

        The future information service paradigm will move from the concept that one should search in the information space to the new concept that one gets more accurate information just where there are. Within this paradigm, a new group of information device, called next generation mobile terminal will take a major trend instead of the fixed legacy terminal. The main difference of the next generation mobile terminal is to provide the multimedia service efficiently with various bandwidths at any time, at any place and with any person. Thus the next generation mobile terminal should be a common mobile platform to provide the multimedia service such as VoIP(Voice of IP), video streaming service and e-mail. These multimedia services require various channel resources according to the traffic class, and can make differences in performances according to the channel resource management mechanism And mobile data service terminal can provide multimedia service in the wire and wireless network environments. These requirements mean to provide a minimum QoS (Quality of Service) in case of the terminal roving the wireless networks of different bandwidths and different error rates. To support the end-to-end QoS services in the next generation mobile terminal, the wireless access part takes into account first of all due to insufficient resources and technical limitations. In this research, we study the scheduling methods at the medium access control (MAC) layer to support QoS in IEEE 80211e.

      • 뿜칠시공에 의한 단면복구재의 시공성 및 공학적 특성에 관한 실험적 연구

        These days, serious concerns of reinforced concrete have been raised recently over steel-bar corrosion, cracking, the deterioration of durability and structural stress resistance and so on, all due to the effects of exterior factors. Such being the case, in domestic, various repair materials and method systems to keep up with these reinforced concrete deteriorated due to salt damage, carbonation, chemical decay et. developed and applied. However, on-site quality control of various repair materials and method systems isn't achieved desirably because it is depend completely on a men of experience' opinions above all else regardless of various on - site environments. In this background, mock up test with due regard to real on-site environments was performed to secure fundamental data for establishment of desirable on-site quality control. Mock up test using repair mortar analyzed from angles of construction methods, mechanical spraying pressures, W 1M. Construction methods were designed manpower method and spraying method, spraying pressures were designed 32, 42, 52 psi, W/M were designed 14.4, 15.4, 16.4 %. And compressive strength, Chloride ion diffusion coefficient, bond strength, SEM of mock up test specimens were evaluated. In conclusion, we confirmed excellency of mechanical spraying pressures, fined extremely excellency of condition of spraying pressure 42 ps, W/M 14.4% within this study. therefore the results of this study will be useful to provide fundamental data for establishment of desirable on-site quality control.

      • 전단력 하에서 유도된 광민감군이 첨가된 HDPE 필름의 광분해

        융용 혼합기를 이용하여 공기 분위기 하에서 강한 전단력을 혼합물에 가하여 광분해성 M/B(Master Batch)를 제조하였다. 제조된 M/B가 45 중량 % 첨가된 광분해성 HEPE 얇은 필름과 분해성 M/B가 첨가되지 않은 일반 HDPE 필름을 인플레이션 법으로 제조하였다. 두 필름의 광분해성을 시험하기 위하여 필름을 자외선 조사기와 옥외에 노출시켜 기계적 물성 및 필름의 표면 변화를 살펴보았다. 자외선 조사 시험에서 분해성 필름은 120시간 이내에 처음 신도의 95%를 잃고 매우 취약한 상태로 되었으나, 비분해성 필름은 150시간 조사에도 처음 신도의 80%를 유지하고 있었다. 옥외 노출 시험에서는 분해성 필름의 경우 옥외 노출 21일 후 처음 신도의 약 98%를 상실하였으나, 비분해 필름은 같은 노출 시간에 원래 신도의 98%를 유지하고 있었다. SEM을 이용하여 필름 표면을 관찰한 결과, 특이한 표면 변화를 보여주었다. Photodegradable master batch was prepared by using mechanical oxidation during melt mixing process with high shear and in the presence of air. High density polyethylene (HDPE) thin film containing 45 wt % photodegradable M/B (Master Batch) and common high density polyethylene (HDPE) thin film were prepared by inflation film processing. The films were exposed under both mercury-vapor lamp light and sunlight. The photodegradable film lost about 95 % of its original elongation for 120 hours irradiation and was very brittle. The common film without photodegadability retained 80 % of its original elongation for 150 hours irradiation. Under sunlight, photodegradable HDPE film lost about 98 % of its original elongation at break after 21 days exposure and was very brittle. The common film still retained 88 % of its original elongation at the same period. Surface morphology of the films was examined by using scanning electron microscopy (SEM). SEM micrographs revealed the interesting change of the surface morphology of photodegradable thin film during outdoor exposure.
