      • Study of decreased melanin production through p53 by heme oxygenase-1 inhibitor in normal human melanocytes

        Background: Heme oxygenase (HO)-1 is induced by oxidative stress and plays important roles in anti-apoptosis and the rapid growth of several solid tumors. p53 has a central role in skin pigmentation and may impact onmelanoma at all stages. However, there has been little study of HO-1 in relation with p53 on melanin production Objectives: To know the effect of HO-1 on melanogenesis through p53 in normal human melanocytes Methods: Human melanocytes (hM) were primarily cultured from foreskin. After incubation, cells were rinsed twice with PBS then transfection with p53 siRNA Results: Melanin content was detected with ELISA and Western blot analysis and RT-PCR of tyrosinase, MITF were performed after ZnPP treatment and p53 transfection. After ZnPP treatment, melanin content was decreased, and tyrosinase and MITF protein levels were decreased. Tyrosinase and MITF mRNA levels were also decreased. However, melanin content and tyrosinase and MITF protein levels were increased after CoPP treatment. After p53 transfection, HO-1 expression was decreased when HO-1 stimulator was treated. In addition, melanin content was decreased, and tyrosinase expression was decreased in Western blot analysis Conclusion: These results suggest that melanogenesis is inhibited by ZnPP by decreased melanin production, tyrosinase and MITF protein and mRNA levels. Furthermore, those inhibitory effects of ZnPP may be dependent on p53 in normal human melanocytes. Therefore, HO-1 may play an important role in melanogensis

        현행 중등학교 과학 실험 , 실습 교육 실태 조사 및 그 운영 진단 (Ⅱ) : 고등학교 과학 실험 , 실습 교육을 중심으로

        본 연구는 96년도에 실시한 우리 나라 중학교 과학 실험·실습 교육의 실태 조사 연구에 따른 계속 연구의 일환으로 고등학교 과학 실험·실습 교육의 실태를 조사하기 위한 것이다. 연구내용은 고등학교 과학 교과서의 분석, 실험·실습 실시 현황 분석, 현행 실험·실습 교육의 문제점 분석, 현행 학교 구성원들의 실험·실습 교육 운영 실태 조사 및 현행 실험·실습 교육의 개선을 위한 연구과제 도출을 주요 연구내용으로 다루었다. 연구방법은 전국의 고등학교 80개교를 대상으로 이들 학교의 학생 1,977명, 교사 165명, 학교장 80명에게 적용하였다. 본 연구에서 밝혀진 결과를 중심으로 우리 나라 고등학교 과학 실험 실습 교육의 정상화를 위한 실험 내적인 연구과제를 제시하면 다음과 같다. (1)교재 개발에 대한 연구, (2)실험 킷트 개발에 대한 연구, (3)교사교육 및 재교육 프로그램 개발에 대한 연구, (4)교실 밖 실험·실습 활동 프로그램 개발에 대한 연구, (5)과학 실험·실습 소재의 개발에 대한 연구. This study was accomplished to analysis and survey on the experimental and practical science education of high school in korea for the consecutive study of the an analysis and survey on the experimental and practical science education of middle school in korea(Lee, Yoon-Jong et al., 1997). The status of facilities, management for the experiment, practices, teaching methods in high schools have been investigated. The present status and reasonable management of the high school science education have been grasped from the questionaires. To do this 165 high school science teachers, 1977 students and 80 principals of high schools in Korea are administered questionaires of Science Education Research Institute of Kyungpook National University(1997). The results of this study are as follows : The reasonable management for experiments and practices of science education were scanty in the high school around the urban and rural school owing to the shortage of facilities and equipments, crowded class, excessive class works for teacher, excessive contents of present textbooks and insufficiency of the administrative supports etc. The current teaching method of high school science has emphasized knowledge. This fact does not satisfy the objective of learning due to lack of the teaching method. Desirable directions for the improvement of present status of high school science education were proposed in this paper.

      • Anti-Inflammatory and Cytoprotective Effects of TMC-256C1 from Marine-Derived Fungus <i>Aspergillus</i> sp. SF-6354 via up-Regulation of Heme Oxygenase-1 in Murine Hippocampal and Microglial Cell Lines

        <P>In the course of searching for bioactive secondary metabolites from marine fungi, TMC-256C1 was isolated from an ethyl acetate extract of the marine-derived fungus <I>Aspergillus</I> sp. SF6354. TMC-256C1 displayed anti-neuroinflammatory effect in BV2 microglial cells induced by lipopolysaccharides (LPS) as well as neuroprotective effect against glutamate-stimulated neurotoxicity in mouse hippocampal HT22 cells. TMC-256C1 was shown to develop a cellular resistance to oxidative damage caused by glutamate-induced cytotoxicity and reactive oxygen species (ROS) generation in HT22 cells, and suppress the inflammation process in LPS-stimulated BV2 cells. Furthermore, the neuroprotective and anti-neuroinflammatory activities of TMC-256C1 were associated with upregulated expression of heme oxygenase (HO)-1 and nuclear translocation of nuclear factor-E2-related factor 2 (Nrf2) in HT22 and BV2 cells. We also found that TMC-256C1 activated p38 mitogen-activated protein kinases (MAPK) and phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathways in HT22 and BV2 cells. These results demonstrated that TMC-256C1 activates HO-1 protein expression, probably by increasing nuclear Nrf2 levels via the activation of the p38 MAPK and PI3K/Akt pathways.</P>

        수산화세륨(Ⅲ)으로부터 질산암모늄세륨(Ⅳ)의 제조

        본 연구에서는 수산화세륨으로부터 질산세륨 용액 제조 후 질산암모늄 첨가에 의한 질산암모늄세륨 제조시, 출발용액의 산도와 농도 그리고 질산암모늄 첨가량, 반응온도 및 반응시간이 질산암모늄세륨 합성에 미치는 영향을 고찰하였으며, XRD, SEM 그리고 TGA 분석을 통하여 결정의 형태 및 구조에 대한 고찰을 수행하였다. 4가의 수산화세륨을 질산에 용해한 질산세륨(Ⅳ)수용액으로부터 질산암모늄 첨가에 의한 질산암모늄세륨 제조시 회수율을 고려한 최적 합성조건은 출발물질의 세륨 농도 70-100g/l, 출발용액의 산도 7 mol/l, 질산암모늄 첨가량 2.5당량, 감압·가열온도 65℃ 및 반응시간 15-30min.이었으며, 제조된 질산암모늄세륨의 결정 화학식은 (NH_4)_2[Ce(NO_3)_6) · ×H_2O(x=6-8)인 것을 알 수 있었다. Synthetic process of ammonium cerium nitrate from cerium hydroxide was presented and influences of concentration and acidity of starting solution, addition quantity of ammonium nitrate, reaction temperature, reaction time on the yield of ammonium cerium nitrate are discussed. The shape and composition of crystal were determined by SEM, thermogravimetry differential thermal, x-ray diffraction and chemical analysis. The optimum synthesizing conditions of ammonium cerium nitrate were discribed as follows; rare earth concentration of starting solution 70-100 g/l. acidity of starting solution 7 mol/l, addition quantity of ammonium nitrate 2-3 equivalent, the condition of rotary evaporation 65℃ and reaction time 15-30 min. It was also known that the chemical formulae of ammonium cerium nitrate was (NH_4)_2[Ce(NO_3)_6] ·×H_2O(x=6-8) from TGA.

      • 인터넷 통신에서 자바 바이트코드를 이용한 애플릿 보안

        Java applets are downloaded from web server through internet and executed in Java Virtual Machine of clients' browser. Before execution of java applets, JVM checks bytecode program with bytecode verifier and performs runtime tests with interpreter. However, these tests will not protect against undesirable runtime behavior of java applets, such as denial of service attack, email forging attack, URL spoofing attack, or annoying sound attack. In order to protect malicious applets, a technique used in this paper is java bytecode modification. This technique is used to restrict applet behavior or insert code appropriate to profiling or other monitoring efforts. Java byte modification is divided into two general forms, class-level modification involving subclassing non-final classes and method-level modification used when control over objects from final classes or interface. This paper showed that malicious applets are controlled by java bytecode modification using proxy server. This implementation does not require any changes in the web sever, JVM or web browser.

        This paper has tried to get hold of spatial distribution of land value in Pusan and an influence of land value on the various kinds of urban functions. Conclusions by the research are as follows. First, points of peak land value in Pusan are Kwangbok-Dong and Bujeon-Dong, and Kwangbok-Dong is greater than Bujeon-Dong in the influential zone according to just spatial present condition of land value. Second, specialized functional agglomeration and regional differentiation functionally are explicit in the region of Kwangbok-Dong. Third, functions having great correlation with land value are high quality retail, food and entertainment, cultural, and service function. Finally, functions having interdependent relation with many other ones are official, food and entertainment, service, and cultural function.

        The aim of this study is to report 2 cases of acute promyelocytic leukemia who died from intracranial hemorrhage following leukapheresis and to provide proper preventive measures against hemorrhage following leukapheresis. From 1994 to 1997, a total of twenty-six patients with leukemia underwent leukapheresis to control hyperleukocytosis at Ajou University Hospital. Two patients with acute promyelocytic leukemia received all-trans retinoic acid but developed drug-induced hyperleukocytosis. Shortly after leukapheresis, they died from intracranial hemorrhage. The risk factors for fatal hemorrhage are thought to be coexisting disseminated intravascular coagulation(DIC), thrombocytopenia aggravated by leukapheresis, exacerbated coagulopathy related to mechanical trauma through leukapheresis and the excess use of citrate during leukapheresis. To reduce the risk of the bleeding associated with leukapheresis, it is necessary to replace platelet sufficiently before and after leukapheresis and to give calcium to correct coagulopathy induced by excess citrate which is used as anticoagulant as well as the correction of disseminated intravascular coagulation.

        This study was performed to characterize the seasonal fluctuation of water quality and sediment environment from April, 2001 to November, 2001 in Gamak bay. Which was a productive area, but recently its fisheries conditional have been deteriorated because of increasing waste-water from municipal area, industry and autochthonous pollution by farm industry. In the survey of seawater quality, DO and COD concentrations were lowest in bottom layer at summer, but DIN and DIP contents were higher than those in surface layer. Eutrophication indices mean that eutrophication area move from south to north entrance via northern-west inner area with seasonal change from summer to autumn. In the survey of sediment environment, ignition loss(IL), chemical oxygen demand(COD), DIN and Dip were in the range of 3.8~15.7%, 10.3~60.6mg/g-dry, 5.053~1.810mg/ℓ and 0.065~0.324mg/ℓ , respectively. COD in sediment were highest at winter and spring. And, they were higher than sediment standard in the south inner area, which mean heavily polluted conditions with organic constituents.

        As a part of efforts for conformity of demand in regard to high quality of concrete, blast-furnace slag has been utilized by means of cement replacement. Superior performance can be ensured, environmental pollution can be prevented and economical advantage can be obtained with utilization by cement placement. But the study on the blast-furnace slag is not systematic and reasonable. So, it is planed that basic data in regard to technique of manufacturing and economic improvement of high quality concrete is showed with experimental comparison and investigation of engineering properties of high quality concrete utilized by cement placement of blast-furnace as industry by-product in this study.

      • 於靑島 海松林의 植生構造에 關한 硏究

        This study was carried out to understand the vegetation structure of Pinus thunbergii forest on O˘cho˘ng island and the results were summarized as following: 1. The most dominant species on O˘chong island was Pinus thunbergii and the Ⅰ.Ⅴ. of Pinus thunbergii in the northern aspect showed higher than that of southeren aspect. Quercus acutissima was emerged to the Pinus thunbergii forest and the Ⅰ.Ⅴ. of Quercus acutissima was higher than that of Pinus thunbergii in shrub layer. In addition, the Ⅰ.Ⅴ. of Eurya japonica was the highest among shrubs. The Ⅰ.Ⅴ. of Castanopsis cuspidata var. sieboldii was the highest among evergreen broad leaved trees. 2. The sites investigated were grouped by Cluster Analysis into Pinus thunbergii forest and mixed-species forest in which Castanopsis cuspidata var. sieboldii, Carpinus coreana, and Quercus acuta are dominant. The factor affecting the most remarkable difference might be the silt between Pinus thunbergii forest and the mixed-species forest. 3. As results of COA Odination, community ordination was appeared to Pinus thunbergii area and mixed-species area. In case of the species ordination all the woody species were closely coordinated except Quercus acuta, Meliosma oldhamii, Quercus variabilis. Sand and organic matter were appeared as the most influencing factors to Ⅱ and Ⅲ axes, respectively. 4. The Ⅰ.Ⅴ. of Pinus thunbergii might be decreased by scarce reproduction in the shrub layer. On the other hand, Quercus acutissima, Castanopsis cuspidata var. sieboldii, and Carpinus coreana would increase and succession would be completed in some area by those species. 5. The content of NaCl in soil was much higher than that of land. Soil pH showed significant difference at 5% between the northern and the southern aspect and also sand, silt, Avail. P_2O_5. K^+, and Na^+ were showed significance 5% between Pinus thunbergii forest and mixed-species forest in which Castanopsis cuspidata var. sieboldii, Carpinus coreana, Quercus acuta are dominant. 6. The mainly emergent woody species in O˘chong island - Pinus thunbergii, Quercus acutissima, Q.acuta, Castanopsis cuspidata var. sieboldii, Carpinus coreana. Eurya japonica, Callicarpa japonica, Vibumum dilatatum, and so on - are strong in salinity. Threrfore, the vegetation of O˘chong island was severely influenced by sea breeze and NaCl in soil.
