The purpose of this experiment is to construct an in vitro model of rabbit penis enlargement using tissue engineering method, and to investigate the efficacy and safety of autologous fibroblast cell seeded to biodegradable scaffold and also to provide a new perspective of clinical application. We observed the growth of GFP transgenic fibroblast cell from SD mouse after transplantation with the biodegradable scaffold by fluorescence microscope. At the second hand, 160 male New Zealand rabbits were divided into four groups; Group A for control; Group B for acellular matrix implanted; Group C for scaffold implanted only; Group D for autologous cultured fibroblast seeded scaffold. Rabbit scrotal skin (4×4 mm) was detached under general anesthesia, and fibroblast (FB) was cultured and proliferated for 2weeks. Cultured cells were seeded into the scaffold and inserted on the between buck`s and dartos fascia of rabbit penis. To assess the efficacy and safety of the cells, we observed for 4months. The FB of rabbit were successfully cultured and confirmed by immunohistochemistry method. In the results of group D, dermal cell seeding group, the penile girth was significantly increased to 22.3% compared to other groups (Group A 0.3%, Group B 12.7%, Group C 14.6% and Group D 22.3%). We checked the results in 2~4 months later. We observed that moderate penile girth increase was gained using the biodegradable scaffold without other side-effects. Therefore, we carefully suggest a new guidance for penile augmentation by the tissue engineering method.

With the rapid development of multiple functional nanozymes, the research on the unique characteristic,contributing to the enzyme-like activity, is essential for us to construct mimic enzymes with outstandingactivity. Herein, 2D Co-V mixed metal oxides (MMOs) are prepared through Co-V layered double hydroxide(LDH) precursor. The single phase of Co-V LDHs is prepared by controlling the dosage of hexamethylenetetramineaccurately in the coprecipitation system. After calcinated at 400 C, the obtainedCo-V MMOs, composed of Co3V2O8 and Co3O4, still inherite the microtopography of nanosheets fromthe precursor, which have superior surface properties to enhance the peroxidase/oxidase (POD/OXD)-like activity. 2D Co-V MMO nanosheets have excellent bactericidal properties in the presence of H2O2as low as 1 lM. The enhanced catalytic performances are attributed to unique hybrid nanosheets architectureand synergistic effect of two active components, ranking the hybrid nanostructure as a promisingantibacterial material.

A novel sulfatase gene, ary423 (1,536 bp ORF), encoding a protein of 511 amino acids with a calculated molecular mass of 56 kDa, was identified from Flammeovirga pacifica, which was isolated from deep-sea sediments of west Pacific Ocean. Amino acid sequence analysis revealed that Ary423 possessed a conserved C-X-A-X-R motif, which was recognized as the sulfatase signature. Phylogenetic analysis suggested that Ary423 belonged to arylsulfatases. After heterologous expression in Escherichia coli cells, the recombinant Ary423 was purified with a N i+ affinity column, and was shown to be highly active at a broad range of temperatures from 30° to 70°C, with maximum activity at 40°C. Furthermore, recombinant Ary423 retained more than 70% and 40% of its maximum activity after 12 h of incubation at 50°C and 60°C, respectively, exhibiting good thermostability at high temperatures. The optimal pH for Ary423 was determined to be 8.0 and the activity of Ary423 could be slightly enhanced by Mg2+. The recombinant enzyme could hydrolyze sulfate ester bonds in pnitrophenyl sulfate (NPS) and Asparagus crude polysaccharides with a specific activity of 64.8 U/mg and 25.4 U/mg, respectively. These favorable properties could make Ary423 attractive for application in the desulfating process of agar production.

In order to obtain the cellulolytic bacterial consortia, sediments from Great Basin hot springs (Nevada, USA) were sampled and enriched with cellulosic biomass as the sole carbon source. The bacterial composition of the resulting anaerobic ethanol-producing celluloytic bacterial consortium, named SV79, was analyzed. With methods of the full-length 16S rRNA librarybased analysis and denaturing gradient gel electrophoresis, 21 bacteria belonging to eight genera were detected from this consortium. Clones with closest relation to the genera Acetivibrio, Clostridium, Cellulosilyticum, Ruminococcus, and Sporomusa were predominant. The cellulase activities and ethanol productions of consortium SV79 using different agricultural residues (sugarcane bagasse and spent mushroom substrate) and energy crops (Spartina anglica, Miscanthus floridulus, and Pennisetum sinese Roxb) were studied. During cultivation, consortium SV79 produced the maximum filter paper activity (FPase, 9.41 U/ml), carboxymethylcellulase activity (CMCase, 6.35 U/ml), and xylanase activity (4.28 U/ml) with sugarcane bagasse, spent mushroom substrate, and S. anglica, respectively. The ethanol production using M. floridulus as substrate was up to 2.63 mM ethanol/g using gas chromatography analysis. It has high potential to be a new candidate for producing ethanol with cellulosic biomass under anoxic conditions in natural environments.

Curcumin Analogue A501 induces G2/M Arrest and Apoptosis in Non-small Cell Lung Cancer Cells

Curcumin and its analogues have been reported to exert anti-cancer activity against a variety of tumors. Here, we reported A501, a new curcumin analogue. The effect of A501 on cell viability was detected by MTT assay, the result showed that A501 had a better inhibiting effect on the four non-small cell lung cancer (NSCLC) cells than that of curcumin. Moreover, Colony forming experiment showed A501 significant restrained cell proliferation. Flow cytometry displayed A501 can cause G2/M arrest and induce apoptosis. Western blotting showed that A501 decreased the expression of cyclinB1, cdc-2, bcl-2, while increased the expression of p53, cleaved caspase-3 and bax. In conclusion, curcumin analogues A501 played antitumor activity by inhibiting cell proliferation and inducing apoptosis of NSCLC cells. And it was likely to be a promising starting point for the development of curcumin-based anticancer drugs.

<P>Polydiacetylenic complex films were prepared using 10,12-pentacosadiynoic acid (PCDA) and para-xylenediamine (<I>p</I>XDA) upon acid-base interactions. The thermochromatic reversibility of the complex films was modulated by changing the mixed molar ratio (3 : 1, 2 : 1, and 1 : 1) of the two molecules. The corresponding conformational changes of the complex films were studied by ex situ FTIR analysis upon thermal stimulation for the first time. In addition, the binding specificities of <I>α</I>-, <I>β</I>-, and <I>γ</I>-cyclodextrins (CDs) with the films were studied, where the <I>α</I>-CDs can induce stronger red fluorescent emission of the films. These fundamental results may be useful for platforms that use these polydiacetylenic complex films as optoelectronic devices or chemical/biological sensors.</P>

Background: Abnormal glucose metabolism is a risk factor for colorectal cancer (CRC). However, association of glycosylated hemoglobin (HbA1c) with CRC risk remains under-reported. We examined the association between glycemic indicators (HbA1c, fasting plasma glucose, fasting insulin, 2-hour glucose, 2-hour insulin, and homeostasis model of risk assessment-insulin resistance index) and CRC risk using prospective analysis and meta-analysis.Methods: Participants (<i>n</i>=1,915) from the Guangzhou Biobank Cohort Study-Cardiovascular Disease Substudy were included. CRC events were identified through record linkage. Cox regression was used to assess the associations of glycemic indicators with CRC risk. A meta-analysis was performed to investigate the association between HbA1c and CRC risk.Results: During an average of 12.9 years follow-up (standard deviation, 2.8), 42 incident CRC cases occurred. After adjusting for potential confounders, the hazard ratio (95% confidence interval [CI]) of CRC for per % increment in HbA1c was 1.28 (95% CI, 1.01 to 1.63) in overall population, 1.51 (95% CI, 1.13 to 2.02) in women and 1.06 (95% CI, 0.68 to 1.68) in men. No significant association of other measures of glycemic indicators and baseline diabetes with CRC risk was found. Meta-analyses of 523,857 participants including our results showed that per % increment of HbA1c was associated with 13% higher risk of CRC, with the pooled risk ratio being 1.13 (95% CI, 1.01 to 1.27). Subgroupanalyses found stronger associations in women, colon cancer, Asians, and case-control studies.Conclusion: Higher HbA1c was a significant predictor of CRC in the general population. Our findings shed light on the pathology of glucose metabolism and CRC, which warrants more in-depth investigation.
CCDC26 Gene Polymorphism and Glioblastoma Risk in the Han Chinese Population

Background: Glioblastoma (GBM) is an immunosuppressive tumor whose median survival time is only 12-15 months, and patients with GBM have a uniformly poor prognosis. It is known that heredity contributes to formation of glioma, but there are few genetic studies concerning GBM. Materials and Methods: We genotyped six tagging SNPs (tSNP) in Han Chinese GBM and control patients. We used Microsoft Excel and SPSS 16.0 statistical package for statistical analysis and SNP Stats to test for associations between certain tSNPs and risk of GBM in five different models. ORs and 95%CIs were calculated for unconditional logistic-regression analysis with adjustment for age and gender. The SHEsis software platform was applied for analysis of linkage disequilibrium, haplotype construction, and genetic associations at polymorphism loci. Results: We found rs891835 in CCDC26 to be associated with GBM susceptibility at a level of p=0.009. The following genotypes of rs891835 were found to be associated with GBM risk in four different models of gene action: i) genotype GT (OR=2.26; 95%CI, 1.29-3.97; p=0.019) or GG (OR=1.33; 95%CI, 0.23-7.81; p=0.019) in the codominant model; ii) genotypes GT and GG (OR=2.18; 95%CI, 1.26-3.78; p=0.0061) in the dominant model; iii) GT (OR=2.24; 95%CI, 1.28-3.92; p=0.0053) in the overdominant model; iv) the allele G of rs891835 (OR=1.85; 95%CI, 1.14-3.00; p=0.015) in the additive model. In addition, "CG" and "CGGAG" were found by haplotype analysis to be associated with increased GBM risk. In contrast, genotype GG of CCDC26 rs6470745 was associated with decreased GBM risk (OR=0.34; 95%CI, 0.12-1.01; p=0.029) in the recessive model. Conclusions: Our results, combined with those from previous studies, suggest a potential genetic contribution of CCDC26 to GBM progression among Han Chinese.
