        Cytological, genetic, and proteomic analysis of a sesame (Sesamum indicum L.) mutant Siyl‑1 with yellow–green leaf color

        Background Both photosynthetic pigments and chloroplasts in plant leaf cells play an important role in deciding on the photosynthetic capacity and efficiency in plants. Systematical investigating the regulatory mechanism of chloroplast development and chlorophyll (Chl) content variation is necessary for clarifying the photosynthesis mechanism for crops. Objective This study aims to explore the critical regulatory mechanism of leaf color mutation in a yellow–green leaf sesame mutant Siyl-1. Methods We performed the genetic analysis of the yellow-green leaf color mutation using the F2 population of the mutant Siyl-1. We compared the morphological structure of the chloroplasts, chlorophyll content of the three genotypes of the mutant F2 progeny. We performed the two-dimensional gel electrophoresis (2-DE) and compared the protein expression variation between the mutant progeny and the wild type. Results Genetic analysis indicated that there were 3 phenotypes of the F2 population of the mutant Siyl-1, i.e., YY type with light-yellow leaf color (lethal); Yy type with yellow-green leaf color, and yy type with normal green leaf color. The yellowgreen mutation was controlled by an incompletely dominant nuclear gene, Siyl-1. Compared with the wild genotype, the chloroplast number and the morphological structure in YY and Yy mutant lines varied evidently. The chlorophyll content also significantly decreased (P < 0.05). The 2-DE comparison showed that there were 98 differentially expressed proteins (DEPs) among YY, Yy, and yy lines. All the 98 DEPs were classified into 5 functional groups. Of which 82.7% DEPs proteins belonged to the photosynthesis and energy metabolism group. Conclusion The results revealed the genetic character of yellow-green leaf color mutant Siyl-1. 98 DEPs were found in YY and Yy mutant compared with the wild genotype. The regulation pathway related with the yellow leaf trait mutation in sesame was analyzed for the first time. The findings supplied the basic theoretical and gene basis for leaf color and chloroplast development mechanism in sesame.

        J Wave Syndromes: History and Current Controversies

        The concept of J wave syndromes was first proposed in 2004 by Yan et al for a spectrum of electrocardiographic (ECG) manifestations of prominent J waves that are associated with a potential to predispose affected individuals to ventricular fibrillation (VF). Although the concept of J wave syndromes is widely used and accepted, there has been tremendous debate over the definition of J wave, its ionic and cellular basis and arrhythmogenic mechanism. In this review article, we attempted to discuss the history from which the concept of J wave syndromes (JWS) is evolved and current controversies in JWS.

        Catalytic graphene-mediated oxygen blocking and depletion for enhancing dual passive/active anticorrosion of epoxy coatings

        Doping graphene to epoxy resins can improve the protective ability of the coating, but the lack of active anticorrosion function greatly limits its application in the field of anticorrosion. Herein, N/S-rich few-layer-graphene (N/S-FLG) was prepared and adopted to endow epoxy coating with dual passive/active corrosion protection. The obtained amphiphilic N/S-FLG is highly dispersed in the epoxy coating, giving rise to the enhanced hosting effect for graphene defects, avoiding the interface corrosion and blocking the penetration of corrosive species. Furthermore, the doping of N and S endows graphene sheets favourable catalytic ability for corrosive oxygen, actively eliminating its contribution to metal corrosion. Under this dual effect, the passive and active anticorrosion properties of epoxy coating are simultaneously enhanced. The coating with 1 wt% N/S-FLG reduces the corrosion rate of metal to 6.5 × 10–5 mm/a, exhibiting almost no corrosion. The proposed concept of introducing nanocatalytic N/S-FLG is facile and eco-friendly, and will undoubtedly promote the practical application of anticorrosion coatings.

        Analgesic Activity of Myricetin Isolated from Myrica rubra Sieb. et Zucc. Leaves

        Myrica rubra Sieb. et Zucc. leaves are commonly used as an astringent, antidiarrheic, and analgesics in folk medicine in China. In the present study, the analgesic activity of myricetin, a major compound in Myrica rubra Sieb. et Zucc. leaves was evaluated in vivo. The analgesic effect of myricetin was tested by a serial of models, such as acetic acid-induced writhing response, formalin-induced paw licking and hot plate test. The sedative activity was evaluated by pentobarbital-induced sleep time. Platelet aggregation induced by collagen and arachidonic acid was also performed in vitro. Myricetin showed a significant inhibition on chemical nociceptive models such as the acetic acid-induced writhing response and the licking time on the late phase in the formalin test in a dose-dependent manner, but did not manifest a signicant effect in hot plate test. Myricetin was also not able to increase the sleeping time induced by pentobarbital, which further indicated that the analgesic effect of myricetin was unrelated to sedation. In addition, myricetin inhibited the content of PGE2 in the peritoneal fluid and platelet aggregation induced by collagen and arachidonic acid in vitro. These results collectively demonstrated that myricetin possessed potent analgesic activity, which was related with peripheral analgesia, but, not with the opioid system. Myricetin may be a potent COX-1 inhibitor with anti-platelet activity.

        Comparison of pre-indwelling double-J stents versus ureteral catheters for artificial hydronephrosis in percutaneous nephrolithotomy: A retrospective cohort study

        Purpose: To compare the clinical efficacy and safety of pre-indwelling double-J stents versus ureteral catheters for artificial hydronephrosis in percutaneous nephrolithotomy (PCNL). Materials and Methods: We retrospectively analyzed the data of 1,258 patients who underwent PCNL for kidney stones from August 2017 to July 2020 in our hospital. Among them, 682 patients had double-J stents inserted (DJ group) and 576 patients had ureteral catheters (UC group). We analyzed baseline patient characteristics, perioperative outcomes, and complications in both groups. Results: The puncture success rate was 97.9% and 97.4% in the DJ and UC groups, respectively (p>0.05). The operation time was 74.5±37.8 minutes in the DJ group compared with 80.8±38.5 minutes in the UC group (p=0.004). The total stone-free rate in the DJ and UC groups was 80.5% and 78.7%, respectively (p>0.05). The incidence of perioperative complications was relatively low in both groups and showed no obvious differences. In the subgroup analysis, the operation time for patients with no obvious or mild hydronephrosis preoperatively was significantly shorter in the DJ group than in the UC group (p<0.05). However, there were no significant differences among patients who had moderate or severe hydronephrosis preoperatively. Conclusions: It is feasible, safe, and effective to create artificial hydronephrosis by insertion of pre-indwelling double-J stents in PCNL surgery. Furthermore, the operation time was significantly shorter in the DJ group than in the group with pre-indwelling ureteral catheters, especially in patients who had no obvious or mild hydronephrosis preoperatively.

        A fiber beam element model for elastic-plastic analysis of girders with shear lag effects

        This paper proposes a one-dimensional fiber beam element model taking account of materially non-linear behavior, benefiting the highly efficient elastic-plastic analysis of girders with shear-lag effects. Based on the displacement-based fiber beam-column element, two additional degrees of freedom (DOFs) are added into the proposed model to consider the shear-lag warping deformations of the slabs. The new finite element (FE) formulations of the tangent stiffness matrix and resisting force vector are deduced with the variational principle of the minimum potential energy. Then the proposed element is implemented in the OpenSees computational framework as a newly developed element, and the full Newton iteration method is adopted for an iterative solution. The typical materially non-linear behaviors, including the cracking and crushing of concrete, as well as the plasticity of the reinforcement and steel girder, are all considered in the model. The proposed model is applied to several test cases under elastic or plastic loading states and compared with the solutions of theoretical models, tests, and shell/solid refined FE models. The results of these comparisons indicate the accuracy and applicability of the proposed model for the analysis of both concrete box girders and steel-concrete composite girders, under either elastic or plastic states.

      • Proposals for flexural capacity prediction method of externally prestressed concrete beam

        Flexural capacity prediction is a challenging problem for externally prestressed concrete beams (EPCBs) due to the unbonded phenomenon between the concrete beam and external tendons. Many prediction equations have been provided in previous research but typically ignored the differences in deformation mode between internal and external unbonded tendons. The availability of these equations for EPCBs is controversial due to the inconsistent deformation modes and ignored secondorder effects. In this study, the deformation characteristics and collapse mechanism of EPCB are carefully considered, and the ultimate deflected shape curves are derived based on the simplified curvature distribution. With the compatible relation between external tendons and the concrete beam, the equations of tendon elongation and eccentricity loss at ultimate states are derived, and the geometric interpretation is clearly presented. Combined with the sectional equilibrium equations, a rational and simplified flexural capacity prediction method for EPCBs is proposed. The key parameter, plastic hinge length, is emphatically discussed and determined by the sensitivity analysis of 324 FE analysis results. With 94 collected laboratory-tested results, the effectiveness of the proposed method is confirmed, and comparisons with the previous formulas are made. The results show the better prediction accuracy of the proposed method for both stress increments and flexural capacity of EPCBs and the main reasons are discussed.

        Combination of Tumor Volume and Epstein-Barr Virus DNA Improved Prognostic Stratification of Stage II Nasopharyngeal Carcinoma in the Intensity Modulated Radiotherapy Era: A Large-Scale Cohort Study

        Purpose Little is known about combination of the circulating Epstein-Barr viral (EBV) DNA and tumor volume in prognosis of stage II nasopharyngeal carcinoma (NPC) patients in the intensity modulated radiotherapy (IMRT) era. We conducted this cohort study to evaluate the prognostic values of combining these two factors. Materials and Methods By Kaplan-Meier, we compare the differences of survival curves between 385 patients with different EBV DNA or tumor volume levels, or with the combination of two biomarkers mentioned above. Results Gross tumor volume of cervical lymph nodes (GTVnd, p < 0.001) and total tumor volume (GTVtotal, p < 0.001) were both closely related to pretreatment EBV DNA, while gross tumor volume of nasopharynx (GTVnx, p=0.047) was weakly related to EBV DNA. EBV DNA was significantly correlated with progress-free survival (PFS, p=0.005), locoregional-free survival (LRFS, p=0.039), and distant metastasis-free survival (DMFS, p=0.017), while GTVtotal, regardless of GTVnx and GTVnd, had a significant correlation with PFS and LRFS. The p-values of GTVtotal for PFS and LRFS were 0.008 and 0.001, respectively. According to GTVtotal and pretreatment EBV DNA level, patients were divided into a low-risk group (EBV DNA 0 copy/mL, GTVtotal < 30 cm3; EBV DNA 0 copy/mL, GTVtotal  30 cm3; or EBV DNA > 0 copy/mL, GTVtotal < 30 cm3) and a high-risk group (EBV DNA > 0 copy/mL, GTVtotal  30 cm3). When patients in the low-risk group were compared with those in the high-risk group, 3-year PFS (p=0.003), LRFS (p=0.010), and DMFS (p=0.031) rates were statistically significant. Conclusion Pretreatment plasma EBV DNA and tumor volume were both closely correlated with prognosis of stage II NPC patients in the IMRT era. Combination of EBV DNA and tumor volume can refine prognosis and indicate for clinical therapy.

        Defining Peace: Peace vs. Security

        One of the most popular beliefs in the modern world is that peace means security. Meanwhile many wars have been fought for the sake of security at the price of peace. The article argues that the state of security differs in terms of the degree of security. Peace is only one state of security, along with the other two states of security: war and neither-peace-nor-war. Peace comprises four degrees of security: no security, partial security, fair security and absolute security. These inherent characteristics of peace, explain why people substitute peace for war. As a social phenomenon, peace shares some common characteristics with war. Peace is an objective sought by man as well as a means used by man. Therefore, the justice of peace is determined by human concepts rather than by peace per se. These social characteristics of peace explain why peace has no superior justice over war. In terms of the social qualities of peace, it is clear that peace is an objective that people hope to achieve, but it is not the highest one for national security. Therefore, we should not confuse "peace" with "security." Peace is one way to resolve conflict but not necessarily the most efficient way. Accordingly, we should take care not to regard "maintaining peace" as the essential approach to strengthening national security. Peace as a criterion for justice is true only in some situations; peace achieved through power politics is an injustice.
