        Stem-leaf saponins from Panax notoginseng counteract aberrant autophagy and apoptosis in hippocampal neurons of mice with cognitive impairment induced by sleep deprivation

        Backgroud: Sleep deprivation (SD) impairs learning and memory by inhibiting hippocampal functioningat molecular and cellular levels. Abnormal autophagy and apoptosis are closely associated with neurodegenerationin the central nervous system. This study is aimed to explore the alleviative effect and theunderlying molecular mechanism of stemeleaf saponins of Panax notoginseng (SLSP) on the abnormalneuronal autophagy and apoptosis in hippocampus of mice with impaired learning and memory inducedby SD. Methods: Mouse spatial learning and memory were assessed by Morris water maze test. Neuronalmorphological changes were observed by Nissl staining. Autophagosome formation was examined bytransmission electron microscopy, immunofluorescent staining, acridine orange staining, and transienttransfection of the tf-LC3 plasmid. Apoptotic event was analyzed by flow cytometry after PI/annexin Vstaining. The expression or activation of autophagy and apoptosis-related proteins were detected byWestern blotting assay. Results: SLSP was shown to improve the spatial learning and memory of mice after SD for 48 h,accomanied with restrained excessive autophage and apoptosis, whereas enhanced activation of phosphoinositide3-kinase/protein kinase B/mammalian target of rapamycin signaling pathway in hippocampalneurons. Meanwhile, it improved the aberrant autophagy and apoptosis induced by rapamycinand re-activated phosphoinositide 3-kinase/Akt/mammalian target of rapamycin signaling transductionin HT-22 cells, a hippocampal neuronal cell line. Conclusion: SLSP could alleviate cognitive impairment induced by SD, which was achieved probablythrough suppressing the abnormal autophagy and apoptosis of hippocampal neurons. The findings maycontribute to the clinical application of SLSP in the prevention or therapy of neurological disordersassociated with SD.

      • Aqueous gelcasted ZrB<sub>2</sub>-SiC foams derived from composite poring mechanisms

        ZrB<SUB>2</SUB>-SiC foams were prepared from composite poring mechanisms of foaming and solid poring (PAA beads) by the application of a gelcasting technique. The porosities and microstructures of sintered bulks were tailored through changing the solids loading of slurries and the amount of added PAA beads. The increase in solid loading caused high viscosities of slurries, and thus resulted in the reduction of porosity and a tendency to form close pores. With increasing the amount of PAA beads, high porosities (up to 91.4%) and obvious connectivity improvement between bubble-derived pores emerged owing to efficient foaming and presence of bead-derived pores. As the porosity increased, the compressive strength (0.4-8.2MPa) decreased, and the gas permeability (9.7-62.1x10<SUP>-12</SUP>m<SUP>2</SUP>) augmented because of the effects of bead-derived pores and the increase in window number and size of bubble-bead pores.

        CHMP1A suppresses the growth of renal cell carcinoma cells via regulation of the PI3K/mTOR/p53 signaling pathway

        Background: CHMP1A, a member of the ESCRT-III complex family, has been indicated as a brand-new inhibitor gene of tumors. Our previous research has revealed that CHMP1A plays a vital role in the development and progression of renal cell carcinoma (RCC). Objective: To investigate the potential target pathway of the regulation of the tumor cell growth by CHMP1A. Methods: The effect of CHMP1A on mTOR pathway was elucidated by western blotting. The effect of CHMP1A on the expression of p53 was evaluated, and A498 cell growth was assessed by colony formation and MTT assays. The expression of p53 was knocked down by shRNA-p53, and the effect of CHMP1A on mTOR after knockdown of p53 was evaluated. The effect of CHMP1A on apoptosis and its relationship with MDM2 pathway were detected by western blotting and FCM. Finally, the relationship between the regulation of p53 by CHMP1A and the PI3K/mTOR pathway was detected. Results: This study showed that the mTOR pathway was suppressed significantly in CHMP1A-overexpressing A498 and 786-0 cells; moreover, the enhanced expression of p53 and the reduced proliferation were shown in CHMP1A-overexpressing A498 cells. Furthermore, CHMP1A was able to regulate the PI3K/PTEN/mTOR and MDM2/p53 pathways in order to suppress RCC. In addition, CHMP1A regulated Bax and Bcl-2 via MDM2/p53 to induce the apoptosis of tumor cells and upregulated the expression of p53 via the PI3K/mTOR pathway. Conclusions: The results convey that CHMP1A-related suppression of RCC is closely related to the PI3K/mTOR/p53 pathway.

      • Association of Oral Contraceptives Use and Lung Cancer Risk among Women: an Updated Meta-analysis Based on Cohort and Case-control Studies

        Background: Previous studies on the association of oral contraceptives (OC) use and lung cancer generated inconsistent findings. The aim of this study was to confirm any definite correlation between OC use and lung cancer risk. Methods: Publications were reviewed and obtained through PubMed and EMBASE databases literature search up to November, 2013. Reference lists from retrieved articles were also reviewed. The language of publication was restricted to English. A meta-analysis was performed to evaluate the association by calculating pooled odds ratios (ORs) and 95% confidence intervals (CIs). Results: A total of 14 studies consisting of 9 case-control studies and 5 cohort studies were finally included in this meta-analysis. There was no significant association observed between OC use and lung cancer risk in the overall analysis (OR=0.91; 95% CI=0.81-1.03). There was a significant protective effect in Europe (OR=0.74; 95% CI=0.60-0.91) and a borderline significant protective effect with an adenocarcinoma histology (OR=0.90; 95% CI=0.80-1.01) in subgroup analyses. No association was observed for methodological quality of study, study design, smoking status and case number of study. Conclusion: This meta-analysis suggests that OC use is not likely to be associated with the risk of lung cancer at all. While a significant protective effect of OC use on lung cancer was observed in Europe, interpretation should be cautious because of the potential biases of low-quality studies. At the same time, more attention should be paid to the possible association of OC use with adenocarcinoma of lung. Our findings require further research, with well-conducted and large-scale epidemiological studies to confirm effects of OC use on lung cancer.

        Dietary Supplementation with Acanthopanax senticosus Extract Modulates Gut Microflora in Weaned Piglets

        This study was conducted to investigate the effects of Acanthopanax senticosus extract (ASE) as a dietary additive on gut microflora in weaned piglets. A total of sixty pigs were weaned at 21 d of age (BW = $5.64{\pm}0.23kg$) and allocated on the basis of BW and litter to three dietary treatments in a randomized complete block design. The dietary treatments were: control group (basal diet), antibiotics group (basal diet+0.02% colistin), and ASE group (basal diet+0.1% ASE). On d 7, 14 and 28 after consuming the experimental diets, five piglets per group were sacrificed and then the contents from the jejunum, ileum and cecum were collected to determine changes in the microbial community by using a polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) technique and estimating the contents of Lactobacillus and E. coli by in vitro culturing methods. The results showed that the ASE promoted the microflora diversity in the cecum. Enumeration of bacteria in the gut contents showed that the number of Lactobacillus increased (p<0.05), while that of E. coli decreased (p<0.05) when compared with the other 2 groups as the days of age progressed post-weaning. These findings suggested that the ASE, as a substitute for dietary antimicrobial products, could improve the development of the normal gut microflora and suppress bacterial pathogens, and effectively promote a healthy intestinal environment.

      • Understanding Breast Cancer Screening Practices in Taiwan: a Country with Universal Health Care

        While the incidence of breast cancer (BC) has been relatively low in Asian countries, it has been rising rapidly in Taiwan. Within the last decade, it has replaced cervical cancer as the most diagnosed cancer site for women. Nevertheless, there is a paucity of studies reporting the attitudes and practices of breast cancer screening among Chinese women. The aim of this study is to assess Taiwanese women's knowledge of and attitudes toward BC screening and to identify potential factors that may influence screening behavior. The study population consisted of a sample of 434 Taiwanese women aged 40 and older. Despite access to universal health care for Taiwanese women and the fact that a majority of the women had heard of the breast cancer screening (mammogram, clinical breast exams, etc.), the actual utilization of these screening modalities was relatively low. In the current study, the majority of women had never had mammograms or ultrasound in the past 5 years. The number one most reported barriers were "no time," "forgetfulness," "too cumbersome," and "laziness," followed by the perception of no need to get screened. In addition, the results revealed several areas of misconceptions or incorrect information perceived by study participants. Based on the results from the regression analysis, significant predictors of obtaining repeated screening modalities included age, coverage for screening, barriers, self-efficacy, intention, family/friends diagnosed with breast cancer. The findings from the current study provide the potential to build evidence-based programs to effectively plan and implement policies in order to raise awareness in breast cancer and promote BC screening in order to optimize health outcomes for women affected by this disease.

        Protective strategy for the caudate lobe bile duct during left hemihepatectomy based on imaging data analysis

        Purpose: This study was performed to analyze the rule of confluence of the caudate lobe bile duct (CLD) into the left hepatic duct (LHD) and to discuss the protective strategy during left hemihepatectomy. Methods: MRI of 400 patients and T-tube angiography images of 100 patients were collected, and the imaging rules of the confluence of the CLD into the LHD were summarized. The clinical data of 33 patients who underwent left hemihepatectomy using the protective strategy were analyzed. Results: MRI and T-tube angiography images showed that the length from the confluence point of the CLD into the LHD to the confluence of the left and right hepatic ducts was 1.19 ± 0.40 cm and 1.26 ± 0.39 cm, respectively. The average angle between the longitudinal axis of the 2 bile ducts was 68.27° ± 22.59° and 66.58 ± 22.88°, respectively. Coronal and cross-sectional images showed that inflow from the foot side to the cranial side was noted in 79.8% and 82.0% of patients, respectively, and inflow from the dorsal to the ventral side was observed in 84.5% and 88.0%, respectively. Based on these imaging rules, the safe transection length and plane were summarized, and the CLD was effectively protected in 33 cases of left hemihepatectomy. Conclusion: In left hemihepatectomy, the LHD should be transected at least 1.5 cm away from the confluence of the left and right hepatic ducts, and the plane of transection should be oblique to the dorsal side at an angle of 45° with the LHD, these parameters represent an effective strategy to protect the CLD.

        Effect of naphthalene quinoline and H2S on DBT hydrodesulfurization over unsupported NiMoW catalyst

        Unsupported catalysts have attracted much attention for high activity in comparison with the traditional supported catalyst. Meanwhile, the clear structure of unsupported catalysts is helpful for the recognition of active phase for conducting the industry production. The NiMoW unsupported catalyst was prepared by hydrothermal synthesis and characterized by BET, XRD and HRTEM. The effects of naphthalene, quinoline and H2S on the hydrodesulfurization reactivity of dibenzothiophene (DBT) were investigated in both a batch autoclave and a continuous 10 ml fixed bed micro-reactor over NiMoW and supported catalyst for comparison. The results showed that the hydrogenation reaction and the hydrogenolysis reaction occurred on different active sites. For supported catalyst, the inhibition was relatively weaker, and the inhibition of the hydrodesulfurization pathway was much higher than the direct desulfurization pathway. Although unsupported catalyst was very sensitive to quinoline and H2S in this experiment, the HDS ratio on the unsupported catalyst was maintained at a high level above 99.7%, which is attributed to the very high active site density of unsupported catalysts.

        Preparation of Graphene–Polypyrrole Hollow Sphere by Pickering Emulsion Method and Their Electrochemical Performances as Supercapacitor Electrode

        Graphene–polypyrrole hollow sphere composite was synthesized for the first time via Pickering emulsion polymerization using graphene oxide as Pickering stabilizer. It was found that polypyrrole and graphene were well hybrid and all graphene–polypyrrole composites had a uniform hollow sphere structure, whose diameters gradually decreased as the pyrrole content decreased. The electrochemical properties of the composites as supercapacitor electrode were investigated. The test results displayed that the specific capacitance of the optimal ratio of composite could reach 238 F/g at a current density of 1 A/g, and a 90.7% capacitance retention could be achieved after 1500 cycles, which was a significant improvement compared with the 62.2% retention of pure polypyrrole.
