        Correlation between DNA methylation and Thymic Stromal Lymphopoietin expression in asthmatic airway epithelial cells

        Background: The overexpression of TSLP and DNA methylation in asthma were both risk factors the relationship was not clear. Objective: This study aimed to investigate the relationship between methylation status of TSLP promoter and mRNA/protein expression in asthmatic airway epithelial cells. Methods: Human bronchial epithelial cells were cultured in vitro and divided into: Control group, treated with PBS, model group, sensitized with LPS (10 μg/mL) for 12 h (37 °C, 5% CO2). Other groups were cultured with the pCMV3 plasmid (M + NC/pCMV), pGPH1 plasmid (M + NC/pGPH), DNMT1/pCMV3 plasmid (M + DNMT1/pCMV), and DNMT1/pGPH1 plasmid (M + DNMT1/pGPH) for 48 h. The expression of DNA methyltransferase 1 and TSLP were measured using real-time PCR and western blotting. Results: Compared with the control group, TSLP mRNA (1.00 ± 0.00 vs. 2.82 ± 0.81 vs. 1, P < 0.001) and protein (1.07 ± 0.04 vs. 1.46 ± 0.11, P < 0.01) were significantly greater, and the methylation of promoter was lower (92.75 ± 1.26 vs. 58.57 ± 3.34, P < 0.05) in the model group. Compared with the model group, TSLP mRNA (2.82 ± 0.81 vs. 1.17 ± 0.10, P < 0.001) decreased, but TSLP promoter methylation increased (58.57 ± 3.34 vs. 92.58 ± 7.30, P < 0.05) in M + DNMT1/pCMV. TSLP mRNA and protein were higher (2.82 ± 0.81 vs. 5.32 ± 0.21, P < 0.001; 1.46 ± 0.11 vs. 1.94 ± 0.11, respectively, P < 0.01), TSLP promoter methylation was lower (58.57 ± 3.34 vs. 33.57 ± 4.29, P < 0.05) in M + DNMT1/pGPH. Conclusions: Overexpression of TSLP in asthmatic airway epithelial cells may be regulated by DNA demethylation.

        Removal of cadmium from aqueous solution by organic-inorganic hybrid sorbent combining sol-gel processing and imprinting technique

        An organic-inorganic hybrid sorbent with high adsorption capacity was prepared by surface imprinting technique combined with sol-gel processing with 3-[2-(2-aminoethylamino)ethylamino]propyl-trimethoxysilane as a functional precursor and silica as the support for the removal of Cd(II) ion from aqueous solution, and was characterized by Fourier-transform infrared spectroscopy, scanning electron microscopy, nitrogen gas sorption and thermogravimetric analysis. The influences of different adsorption parameters, such as pH value of solution, contact time and the initial concentrations of Cd(II) ions on the adsorption amount of Cd(II), were examined. The optimum pH for adsorption was found to be in the range of 4-8. The adsorption rate of Cd(II) on the imprinted hybrid sorbent was rapid. The relative selectivity coefficients of the imprinted hybrid sorbent were higher than those of the non-imprinted sorbent. Ho’s pseudosecond-order model best described the kinetics of the adsorption reaction. The adsorption process of metals followed Redlich-Peterson and Langmuir isotherms models, and the experimental value of maximum adsorption capacity for Cd(II) was 77.2mg·g−1. The positive values of ΔHo suggested endothermic nature of Cd(II) adsorption on the imprinted hybrid sorbent. Increase in entropy of adsorption reaction was shown by the positive values of ΔSo and the negative values of ΔGo indicating that the adsorption was spontaneous.

        Hydrogels with diffusion-facilitated porous network for improved adsorption performance

        Porous alginate-based hydrogel beads (porous ABH) have been prepared through a facile and sustainable template-assisted method using nano-calcium carbonate and nano-CaCO3 as pore-directing agent for the efficient capture of methylene blue (MB). The materials were characterized by various techniques. The sorption capacities of ABH towards MB were compared with pure sodium alginate (ABH-1:0) in batch and fixed-bed column adsorption studies. The obtained adsorbent (ABH-1:3) has a higher BET surface area and a smaller average pore diameter. The maximum adsorption capacity of ABH-1:3 obtained from Langmuir model was as high as 1,426.0mg g1. The kinetics strictly followed pseudo-second order rate equation and the adsorption reaction was effectively facilitated, approximately 50minutes to achieve adsorption equilibrium, which was significantly shorter than that of ABH-1:0. The thermodynamic parameters revealed that the adsorption was spontaneous and exothermic. Thomas model fitted well with the breakthrough curves and could describe the dynamic behavior of the column. More significantly, the uptake capacity of ABH-1:3 was still higher than 75% of the maximum adsorption capacity even after ten cycles, indicating that this novel adsorbent can be a promising adsorptive material for removal of MB from aqueous solution under batch and continuous systems.

        Enhanced Oral Bioavailability of Curcumin <i>via</i> a Solid Lipid-Based Self-Emulsifying Drug Delivery System Using a Spray-Drying Technique

        <P>In this study, a novel liquid self-emulsifying drug delivery system (SEDDS) containing curcumin was formulated and further developed into a solid form by a spray drying method using Aerosil 200 as the solid carrier. The optimum liquid SEDDS consisted of Lauroglycol Fcc, Labrasol and Transcutol HP as the oil phase, the surfactant and the co-surfactant at a weight ratio of 15.0 : 70.8 : 14.2 (w/w/w), respectively. There was no difference in droplet size between the emulsions obtained from the liquid and solid forms of SEDDS. Solid state characterization of the solid SEDDS was performed by scanning electron micrograph (SEM), differential scanning calorimetry (DSC), and X-ray powder diffraction (XRPD). The drug formulated in the solid SEDDS was quickly and completely dissolved within 5 min, both in 0.1 <SMALL>N</SMALL> HCl and phosphate buffer pH 6.8 dissolution media, whereas crude curcumin powder was significantly less dissoluble. The solid SEDDS formulation was stable for at least 3 months at 40°C with 75% relative humidity. After oral administration to rats, curcumin in the solid SEDDS resulted in significant improvement in <I>in vivo</I> absorption compared with that of curcumin powder. As the dose of curcumin formulated in solid SEDDS increased from 25 to 100 mg/kg, the <I>C</I><SUB>max</SUB> and area under the drug concentration time curve (<I>AUC</I>) of curcumin were increased by 4.6 and 7.6 times, respectively. However, the over-proportional increase in the <I>AUC</I> in the higher dose group might be due to underestimation of <I>AUC</I> in the lower dose group. In conclusion, this solid SEDDS is a promising solid dosage form for poorly water-soluble curcumin.</P>

        Predictive Value of Exercise Blood Pressure Changes for Orthostatic Hypotension in Patients With Parkinson’s Disease

        Background and Purpose Orthostatic hypotension (OH) is common in patients with Parkinson’s disease (PD). Early recognition OH is required with sensitive assessments. The purpose of this study was to determine whether blood pressure (BP) changes during exercise can predict the occurrence of OH in PD. Methods This prospective cohort study included 80 consecutive patients with PD. All patients agreed to participate in a baseline evaluation and cardiopulmonary exercise test (CPET). According to the initial active standing test (AST), those without OH (PD-nonOH) at baseline had their AST results followed up for 6 months. The main outcome was defined as whether patients without OH at baseline would develop OH after 6 months. Logistic regression analysis was applied to identify the relevant variables. A nomogram was constructed based on clinical features and identified variables. The concordance index (C-index) and area under the receiver operating characteristic curve (AUC) were used to evaluate the accuracy and predictive ability of the nomogram, respectively. Results CPET results indicated that peak load, peak heart rate, heart rate recovery at 1 min, and systolic BP change (ΔSBP) were lower in those with OH than in the PD-nonOH group (p<0.05) at baseline. Logistic regression analysis indicated that peak load and ΔSBP during CPET had significant effects on OH (p<0.05). Age, sex, peak load, and ΔSBP were used to construct the nomogram model (C-index=0.761). The prediction model had an AUC of 0.782 (95% confidence interval=0.649–0.889) and a specificity and sensitivity of 70.0% and 81.8%, respectively. Conclusions This study has identified predictive factors for OH development in patients with PD. CPET could be used as a complementary examination to identify patients at a high risk of OH.

        Various amide prodrugs of salicylic acid were synthesised, and their physicochemical propertiesincluding lipophilicity, chemical stability and enzymatic hydrolysis were investigated. In vivo skinpermeation and accumulation profiles were also evaluated using a combination of common permeationenhancing techniques such as the use of a supersaturated solution of permeants in an enhancer vehicle, a lipophilic receptor solution, removal of the stratum corneum and delipidisation of skin. Their capacity factor values were proportional to the degree of carbon-carbon saturation in the side chain. All these amides were highly stable in acetonitrile and glycerine. Amide prodrugs were converted tosalicylic acid both in hairless mouse liver and skin homogenates. N-dodecyl salicylamide (C12SM) showed the lowest permeation of salicylic acid in skin compared to the other prodrugs, probably due to its low aqueous solubility. It had a high affinity for the stratum corneum and its accumulation was restricted to only the uppermost layer of skin. Thus, this amide prodrug could be a safer topicalsunscreen agent with minimum potential for systemic absorption.ⓒ2011 Elsevier B.V.All rights reserved.

      • Effect of dose and dosage interval on the oral bioavailability of docetaxel in combination with a curcumin self-emulsifying drug delivery system (SEDDS).

        <P>The present study investigated the effects of a curcumin self-emulsifying drug delivery systems (SEDDS) on the pharmacokinetics of orally administered docetaxel in rats. A single dose of docetaxel was orally administered (30 mg/kg) alone or after oral curcumin SEDDS (25, 50, 100 and 150 mg/kg) administration with time intervals of 0, 15 and 30 min, respectively. After oral administration, the C (max) and the area under the plasma concentration-time curve (AUC) of docetaxel were significantly increased (0 min, p < 0.05; 15 and 30 min, p < 0.01) by 2.2, 4.7 and 4.6 times and 2.0, 3.8 and 4.1 times compared to that of control group, respectively, after treatment with curcumin SEDDS (100 mg/kg) for each interval. Moreover, The C (max) of docetaxel was increased by 2.6 and 4.4 times in response to 25 and 50 mg/kg curcumin SEDDS treatment, respectively, the corresponding AUC was increased by about 2.4 and 3.1 times, and consequently the absolute bioavailabilities of docetaxel in these two treatment groups were 7.9 and 10.4%, respectively, which showed a significant increase of about 2.4- and 3.2-fold in comparison to the control value (3.3%). However, no further increase in either AUC or C (max) values of docetaxel was observed as the curcumin SEDDS dose was increased from 50 to 150 mg/kg. It is worth noting that the presence of curcumin SEDDS did not significantly decrease the systemic clearance, which was shown by the almost unchanged terminal half-life (t (1/2)) of docetaxel in all treatment groups. Thus, the enhanced bioavailability of oral docetaxel by curcumin SEDDS seemed to be likely due to an inhibition function of cytochrome P450 (CYP) 3A and P-glycoprotein (Pgp) in the intestines of the rats. However, further in vivo studies are needed to verify these hypotheses.</P>

      • Computer vision-based displacement measurement with m-sequence target

        The development of image sensors enables the application of vision-based techniques to the non-contact displacement measurement of large-scale structures. The features of the physical targets are critical to the accuracy, stability and anti-interference of the displacement measurement results. In this study, a novel m-sequence target and the associated circular correlation processing technique are developed for real-time displacement measurement. The properties of the m-sequence as a pseudo-random sequence are introduced. The vision-based displacement calculation method is then derived from the correlation property of the m-sequence. The algorithms and measurement systems are integrated in the LabVIEW environment. To verify the anti-interference performance of the developed system, static and dynamic experimental tests are carried out with various forms of interference, such as partial occlusion, uneven illumination, out of focus and smoke effect. Experimental results indicate that the developed system cannot only accurately measure structural displacement, but also has outstanding antiinterference performance, even if 30% of the target is masked.
