This study combines the feedback linearization method and our recent researches of Chen electrical unifying approach to propose a global tracking controller design for nonlinear uncertain diff erential-diff erence systems with time delay. One example, which cannot be solved by the fi rst paper on the almost disturbance decoupling problem, is presented in this study to show the essential points that the tracking performance is easily solved by the proposed approach. The study applies Chen electrical unifying approach to take the place of using the disorganized virtual ground technique and the Kirchhoff ’s law for those traditional approaches. In order to show its signifi cant practicability, the study has fi rstly designed an easy-to-implement adder and a time-delay circuit, and an unstable time-delay operational amplifi er control system based on the Chen electrical unifying approach, and then has proposed a stable controller.

Time-dependent density-functional theory molecular dynamics reveals an unexpected effect of optical excitation in the experimentally observed rhombohedral-to-cubic transition of GeTe. The excitation induces coherent forces along [001], which may be attributed to the unique energy landscape of Peierls-distorted solids. The forces drive the A(lg) optical phonon mode in which Ge and Te move out of phase. Upon damping of the A(1)g mode, phase transition takes place, which involves no atomic diffusion, defect formation, or the nucleation and growth of the cubic phase.

Chen Cai-Ling,Wang Yu-Ting,Yao Yin,Pan Li,Guo Bei,Zhu Ke-Zhang,Ma Jin,Wang Nan,Li Xue-Li,Deng Yi-Ke,Liu Zheng 대한천식알레르기학회 2021 Allergy, Asthma & Immunology Research Vol.13 No.6
Purpose: The pathogenic mechanisms of antrochoanal polyps (ACPs) remain largely unknown. This study aimed to characterize inflammatory patterns and tissue remodeling features in ACPs. Methods: Inflammatory cell infiltration and tissue edema severity as well as fibrin deposition in ACPs and bilateral eosinophilic and noneosinophilic nasal polyps (NPs) were studied with immunohistochemical and immunofluorescence staining. Cytokine levels in sinonasal tissues were detected with the Bio-Plex assay. The expression of coagulation and fibrinolytic markers was measured using reverse-transcription polymerase chain reaction and enzyme-linked immunosorbent assays. Results: Compared to control tissues and bilateral eosinophilic and noneosinophilic NPs, ACPs had higher levels of neutrophil infiltration and expression of myeloperoxidase (MPO), interleukin (IL)-8 and interferon (IFN)-γ. In total, 94.4% of ACPs demonstrated an eosinophil cationic protein/MPO ratio of < 1, compared to 79.0% of noneosinophilic and 26% of eosinophilic NPs. Principle component and multiple correspondence analyses revealed a neutrophilic and type 1 inflammation pattern in ACPs. Compared to control tissues, edema scores and fibrin deposition were increased, whereas d-dimer and tissue plasminogen activator (tPA) levels were decreased in ACPs and bilateral NPs, with more prominent changes in ACPs even than in eosinophilic NPs. The tPA levels were negatively correlated with IFN-γ, IL-8, and MPO levels in ACPs. Neutrophils were the major cellular source of IFN-γ in ACPs, and the number of IFN-γ+ neutrophils was elevated in ACPs than in control tissues and bilateral eosinophilic and noneosinophilic NPs. Conclusions: ACPs are characterized by the neutrophilic and type 1 inflammation endotype. Neutrophil-derived IFN-γ is associated with reduced tPA production in ACPs.

Multiplex PCR with the Blunt Hairpin Primers for Next Generation Sequencing
Ke Chen,Yuxun Zhou,Kai Li,Junhua Xiao 한국생물공학회 2017 Biotechnology and Bioprocess Engineering Vol.22 No.3
The multiplex PCR is one of the important methods to enrich the target DNAs for next generation sequencing. The non-specific amplification and interaction between the primers are the pivotal challenges of multiplex PCR. Here, we introduce the novel blunt hairpin primers for effective reducing the primer dimers and mispriming events. We also used a pair of auxiliary primers to enhance PCR efficiency. We simultaneously amplified 89 target regions from 44 samples and sequenced all amplicons on ion torrent PGM platform. Among all the filtrated amplicons (3438 different amplicons), 99.7, 97.6, 90.1 and 72.8% had sequencing depths fell within 200, 100, 50 and 25-fold range. The sequencing depth variations among all the samples were less than 27-fold. We also amplified multiplex regions with blunt hairpin, stick hairpin and normal linear primers, and the blunt hairpin primers could significantly reduce the amount of primer dimers and unspecific products. These results show that multiplex PCR with the blunt hairpin primers is a flexible, specific and economical target-region captured approach for the next generation sequencing.

Alectinib, an inhibitor of anaplastic lymphoma kinase (ALK), was approved by the Food and Drug Administration (FDA) for the treatment of patients with ALK-positive non-small cell lung cancer (NSCLC). Here we investigated the reversal effect of alectinib on multidrug resistance (MDR) induced by ATP-binding cassette (ABC) transporters, which is the primary cause of chemotherapy failure. We provide the first evidence that alectinib increases the sensitivity of ABCB1- and ABCG2-overexpressing cells to chemotherapeutic agents in vitro and in vivo. Mechanistically, alectinib increased the intracellular accumulation of ABCB1/ABCG2 substrates such as doxorubicin (DOX) and Rhodamine 123 (Rho 123) by inhibiting the efflux function of the transporters in ABCB1- or ABCG2-overexpressing cells but not in their parental sensitive cells. Furthermore, alectinib stimulated ATPase activity and competed with substrates of ABCB1 or ABCG2 and competed with [125I] iodoarylazidoprazosin (IAAP) photolabeling bound to ABCB1 or ABCG2 but neither altered the expression and localization of ABCB1 or ABCG2 nor the phosphorylation levels of AKT and ERK. Alectinib also enhanced the cytotoxicity of DOX and the intracellular accumulation of Rho 123 in ABCB1-overexpressing primary leukemia cells. These findings suggest that alectinib combined with traditional chemotherapy may be beneficial to patients with ABCB1- or ABCG2-mediated MDR.

Objective: To investigate the changes of serum vascular endothelial growth factor (VEGF), soluble interleukin-2 receptor (SIL-2R) and hepatocyte growth factor (HGF) contents in patients with primary hepatocellular carcinoma (HCC) before and after percutaneous microwave coagulation therapy (PMCT) and determine their clinical significance. Materials and Methods: Fasting venous blood (3 mL) from 81 patients with primary HCC diagnosed by pathology was collected in the mornings 1 day before PMCT, and 1 day, 7 days and 1 month after PMCT, and then the serum was separated and stored in $-70^{\circ}C$. The contents of VEGF, SIL-2R and HGF were detected by enzyme linked immunosorbent assay (ELISA). Results: The serum VEGF, SIL-2R and HGF contents in 81 patients with primary HCC had obviously dynamic changes before and after PMCT. By comparison to 1 day after PMCT with pre-operation, there was no statistical significance regarding VEGF and SIL-2R contents (P>0.05), but HGF content showed significant difference (P<0.01). Compared with pre-operation, VEGF, SIL-2R and HGF contents 7 days and 1 month after PMCT all manifested significant differences (P<0.01). By comparison to 7 days with 1 month after PMCT, there was no statistical significance regarding the VEGF content (P>0.05), whereas SIL-2R and HGF contents showed significant change (P<0.01). Conclusions: The contents of serum VEGF, SIL-2R and HGF have obviously dynamic changes in primary HCC before and after PMCT, and their joint detection is expected to be an effective hematologic evaluation index of PMCT for primary HCC.
