        Group Differences in Individual and Organizational Capabilities and Operational Performance in Implementing DX: Focusing on Nepalese SMEs

        Purpose: The purpose of this study is to examine differences between excellent and following groups based on individual and organizational capabilities and operational performance, focusing on Nepalese SMEs. Design/methodology/approach: The proposed research model and a set of hypotheses were developed and tested using T-test analysis based on data collected from 154 (70.32%) responses in Nepal. Findings: From the T-test results showed a significant mean difference between excellent and following groups on change acceptance and technology acquisition on individual capabilities. In addition, the T-test results showed a significant mean difference between excellent and following groups on organizational infrastructure and organizational support, and operational performance showed the or a difference between the two groups. Research limitations/implications: The limitations of this study are the sample size and target respondents. The results of this study suggested practical insights for DX implementation for SMEs. Originality/value: The value of this study will be used as basic guidelines not only for companies that want to implement DX effectively but also for organizations that consider implementing DX.

        Competencies Matter for Digital Transformation and Operational Performance in Nepalese SMEs

        본 연구의 목적은 네팔 중소기업을 중심으로 개인 및 조직역량이 디지털 전환 구현 및 운영성과에 미치는 영향을 분석하고자 하였다. 네팔 중소기업 직원 154명으로부터 수집된 데이터를 활용하여 회귀분석을 기반으로 연구 모델과 가설검증을 실시하였다. 분석결과, 정보통신 기술 능력(H1), 변화 수용(H2), 기술 습득(H3) 등 개인역량은 네팔 중소기업의 디지털 전환 구현에 양의 영향을 미치는 것으로 나타났으며, 조직역량 중 조직 인프라(H4), 최고 경영진의 의지(H5), 조직 지원(H6)은 디지털 전환 구현에 긍정적인 영향을 미치는 것으로 나타났다. 또한 디지털 전환 구현(H7)은 운영성과에 긍정적인 영향을 미치는 것으로 분석되었다. 본 연구결과에서 디지털 전환을 구현하기 위해서는 개인 및 조작 역량의 중요성이 실증분석결과에서 제시된 만큼 관리자나 최고경영진은 자사에서 개인과 조직의 역량이 디지털 전환 구현의 동인임을 인지하고 구성원(종업원)의 업무 프로세스를 바탕으로 업무 효율성을 향상시키기 위한 매뉴얼과 지침을 개발해야 할 것이다. 이러한 관점에서 구성원은 업무 프로세스 단계에서 디지털 디바이스를 적용하고 자신의 업무 역량을 향상시키기 위해 필요한 교육훈련 기회에 참여해야 할 것이다. 본 연구 결과는 디지털 전환을 고려하고 있는 조직이나 기업 또는 디지털 전환 전략을 효과적으로 채택하려는 조직이나 기업에게 기초적인 자료로 활용될 수 있다. This study investigates the effect of individual and organizational competencies on digital transformation implementation and operational performance in small and medium-sized enterprises in Nepal. Regression analysis with a small sample size is used to test the proposed research model and hypotheses utilizing data collected from 154 employees of SMEs in Nepal. The results of this study showed that individual capabilities measure of information and communication technology skills, change acceptance, and technology acquisition have positive relationships with digital transformation implementation in Nepal’s SMEs. In addition, the study’s results indicated that organizational infrastructure, top management’s willingness, and organizational support as a part of organizational capabilities positively affect digital transformation implementation. As individual and organizational capabilities are confirmed to be the drivers of DX implementation, firms should develop manuals and guidelines to understand employees’ work processes and improve work efficiency. Digital transformation implementation positively affects operational performance in Nepal’s small and medium-sized enterprises. The significance and value of this study lies in its potential to serve as fundamental guidance for corporations seeking to implement digital transformation and organizations contemplating adopting digital transformation strategies effectively.

        Integrating Digital Transformation and Servitization into Digital Servitization: A Case Study on Domino's

        Purpose: The purpose of this study is to suggest a digital servitization model which can be implemented by the restaurant industry and to explore the opportunities and challenges of digital servitization. Design/methodology/approach: The case study analysis method was applied for this research. Findings: This study reveals that pizza firms can gain competitive advantages and create value by implementing the digital servitization business model. In addition, firms can understand the opportunities and challenges of digital servitization and prepare themselves before implementing the business model. Research limitations/implications: Digital servitization model was proposed to analyze a single pizza company but a comparative study with other firms can be conducted to identify differences and create a common strategy. Originality/value: Firms commonly use an integrated digital transformation and servitization model to create customer value, but few case studies on integrated case models are available. This study investigates this research gap using a case study. The findings of this study have meaningful implications for restaurant managers to implement a digital servitization strategy.

        Human mesenchymal stem cells (MSCs) are currently being evaluated as a cell-based therapy for tissue injury and degenerative diseases. Recently, several methods have been suggested to further enhance the therapeutic functions of MSCs, including genetic modifications with tissue- and/or diseasespecific genes. The objective of this study was to examine the efficiency and stability of transduction using an adenoviral vector in human MSCs. Additionally, we aimed to assess the effects of transduction on the proliferation and multipotency of MSCs. The results indicate that MSCs can be transduced by adenoviruses in vitro, but high viral titers are necessary to achieve high efficiency. In addition, transduction at a higher multiplicity of infection (MOI) was associated with attenuated proliferation and senescence-like morphology. Furthermore, transduced MSCs showed a diminished capacity for adipogenic differentiation while retaining their potential to differentiate into osteocytes and chondrocytes. This work could contribute significantly to clinical trials of MSCs modified with therapeutic genes.

      • Fabrication and evaluation of pH-modulated solid dispersion for telmisartan by spray-drying technique

        The present study was undertaken to overcome the problems associated with solubility, dissolution and oral bioavailability of a poorly water-soluble ionizable drug, telmisartan (TMS). For these purposes, a solubility test was carried to select the appropriate formulation composition from various carriers and alkalizers. Solid dispersions (SDs) of TMS were prepared at different drug-to-carrier ratios by the spray-drying technique, and were characterized by dissolution and aqueous solubility studies. The optimum formulation was investigated by dissolution studies at different pH and water media and its solid state characterisations were performed by scanning electron microscopy (SEM), differential scanning calorimetry (DSC) and X-ray diffraction (XRD) studies. In solubility and dissolution tests, all TMS-loaded pH-modulated SDs (pH(M)-SDs) exhibited marked improvement in the dissolution behavior when compared with crystalline TMS powder. The optimum formulation of pH(M)-SD consisted of TMS/PVP (polyvinylpyrrolidone) K30/Na(2)CO(3) at a weight ratio of 2/0.5/3 and showed significant improvement in the aqueous solubility and dissolution rate by approximately 40,000- and 3-fold, respectively, compared to TMS powder. Solid-state characterization revealed the changed in crystallinity of TMS into amorphous state. Furthermore, area under the drug concentration time-curve (AUC) of TMS from the pH(M)-SD increased by 13.4- and 2.1-fold, compared with TMS powder and commercial product, respectively. According to these observations, taken together with dissolution and pharmacokinetic behaviors, pH-modulated SD in the presence of an alkalizer for a poorly water-soluble ionizable drug, TMS, appeared to be efficacious for enhancing its bioavailability. ⓒ2012 Elsevier B.V.All rights reserved.

        A Novel Paramagnetic Nanoparticle T2 Magnetic Resonance Imaging Contrast Agent With High Colloidal Stability: Polyacrylic Acid-Coated Ultrafine Dysprosium Oxide Nanoparticles

        Until now, there have been limited studies on T2 magnetic resonance imaging (MRI) contrast agents (CAs) using paramagnetic nanoparticles (NPs). In this study, ultrafine dysprosium oxide (Dy2O3) NPs were synthesized as a T2 MRI CA. To obtain high biocompatibility and colloidal stability in aqueous medium for in vivo applications, they were grafted with polyacrylic acid. They displayed paramagnetism with an appreciable magnetization value at 300?K and consequently, an appreciable transverse water proton spin relaxivity (r2) value which increased as the applied MR field increased. Their potential as a T2 MRI CA was proved by taking in vivo T2 MR images in mice.

      • Statistical Modeling, Optimization and Characterization of Spray-Dried Solid Solid Self-Microemulsifying Drug Delivery Sytem Using Design of Experiments

        The present study systematically and simultaneously investigates the influence of process variables of spray-drying on the properties of solid self-microemulsifying drug delivery system (SMEDDS) using design of experiment (DOE) and optimizes them in order to produce solid-SMEDDS satisfying pre-defined powder quality attributes. Flurbiprofen-loaded liquid-SMEDDS was dispersed in dextran and spray-dried. After preliminary screening, the independent factors selected according to three-factor, three-level Box-Behnken design were inlet temperature (X(1)), feed rate (X(2)) and carrier concentration (X(3)). The responses used to compute the effects of independent factors were moisture content (Y(1)), yield (Y(2)), drug content (Y(3)) and droplet size (Y(4)) of the micro-emulsion. SMEDDS powder characteristics such as morphology, thermal behavior, crystallinity and flowability were also considered. Models were developed and model fitting analysis showed an adequate fit for all responses, indicating good predictability. Significant effects of processing parameters on powder characteristics were observed. The spray-drying process parameters were optimized as inlet temperature (134°C), feed rate (5%) and carrier concentration (0.6%) to produce solid-SMEDDS with acceptable moisture content (0.72±0.02%), yield (58.5±2.9%), drug content (70.1±2.7 mg/g) and droplet size (166.8±13.8 nm). Validation of the optimization process in five batches showed experimental value very close to the predicted one, ensuring the reproducibility of the developed models. Furthermore, optimized parameters resulted a highly crystalline flurbiprofen changed to an amorphous form. In conclusion, this study demonstrated the applicability of the DOE approach for optimizing the process parameters to manufacture solid-SMEDDS with desired quality attributes.

      • Development and Optimization of Self-Nanoemulsifying Drug Delivery System with Enhanced Bioavailability by Box-Behnken Design and Desirability Function

        The aim of our study was to characterize and optimize a self-nanoemulsifying drug delivery system (SNEDDS) formulation by a three-factor, three-level Box-Behnken design (BBD) combined with a desirability function. The independent factors were the amounts of Capryol PGMC (X(1)), Tween 20 (X(2)), and Transcutol HP (X(3)). The dependent variables were droplet size (Y(1)), equilibrium solubility (Y(2)), and cumulative percentage ofdrug released in 15 min (Y(3)) from the SNEDDS formulation. The responses were fitted to a second-order quadratic model and statistical validation of the fitted models was carried out by analysis of variance. Various response surface graphs and contour plots were constructed to understand the effects of different factor level combinations on the responses. The optimized SNEDDS formulation consisting of Capryol PGMC-Tween 20-Transcutol HP at proportions of 5:58.4:40 (w/w) was prepared and a comparison of the predicted values and experimental values was found to be in close agreement. Furthermore, an in vivo pharmacokinetic study of the optimized SNEDDS formulation showed a 2.2-fold increase in relative oral bioavailability compared with that of the suspension. In conclusion, the BBD demonstrated its effectiveness in optimizing the SNEDDS formulation and in understanding the effects of formulation variables on the performance of SNEDDS.ⓒ2012 Wiley Periodicals, Inc. and the American Pharmacists Association J Pharm Sci 101:4584_4596,2012
