      • Prognostic Significance of Desmoglein 2 and Desmoglein 3 in Esophageal Squamous Cell Carcinoma

        Objective: Desmogleins (DSGs) are major members among the desmosomal cadherins critically involved in cell-cell adhesion and the maintenance of normal tissue architecture in epithelia. Reports exploring links of DSG family member expression with cancers are few and vary. The aim of this study was to investigate the ratio of DSG2 and DSG3 mRNA expression in esophageal squamous cell carcinoma (ESCC) tissue to normal tissue (T/N ratio) and evaluate correlations with clinical parameters. Methods: The mRNA expression of DSGs, as well as ${\gamma}$-catenin and desmoplakin, was detected by real-time quantitative RT-PCR in 85 cases of ESCC tissue specimens. Results: The expression level of DSG3 mRNA was significantly higher than that of DSG2 in ESCC specimens (p=0.000). DSG3 mRNA expression highly correlated with histological grade (p=0.009), whereas that of DSG2 did not significantly relate to any clinicopathologic parameter. Kaplan-Meier survival analysis showed that only DSG3 expression had an impact on the survival curve, with negative DSG3 expression indicating worse survival (p=0.038). Multivariate Cox regression analysis demonstrated DSG3 to be an independent prognostic factor for survival. Furthermore, correlation analysis demonstrated the mRNA level of DSG3 to highly correlate with those of ${\gamma}$-catenin and desmoplakin in ESCC samples (p=0.000), implying that the expression of desmosomal components might be regulated by the same upstream regulatory molecules. Conclusions: Our findings suggest that DSG3 may be involved in the progression of ESCC and serve as a prognostic marker, while expression of DSG2 cannot be used as a predictor of ESCC patient outcome.

        Knockdown of Chloride Channel-3 Inhibits Breast Cancer Growth In Vitro and In Vivo

        Purpose: Chloride channel-3 (ClC-3) is a member of the chloride channel family and plays a critical role in a variety of cellular activities. The aim of the present study is to explore the molecular mechanisms underlying the antitumor effect of silencing ClC-3 in breast cancer. Methods: Human breast cancer cell lines MDAMB- 231 and MCF-7 were used in the experiments. Messenger RNA and protein expression were examined by quantitative realtime polymerase chain reaction and western blot analysis. Cell proliferation was measured by the bromodeoxyuridine method, and the cell cycle was evaluated using fluorescence-activated cell sorting. Protein interaction in cells was analyzed by co-immunoprecipitation. Tumor tissues were stained with hematoxylin- eosin and tumor burden was measured using the Metamorph software. Results: Breast cancer tissues collected from patients showed an increase in ClC-3 expression. Knockdown of ClC-3 inhibited the secretion of insulin-like growth factor (IGF)-1, cell proliferation, and G1/S transition in breast cancer cells. In the mouse xenograft model of human breast carcinoma, tumor growth was significantly slower in animals injected with ClC- 3-deficient cells compared with the growth of normal human breast cancer cells. In addition, silencing of ClC-3 attenuated the expression of proliferating cell nuclear antigen, Ki-67, cyclin D1, and cyclin E, as well as the activation of extracellular signalregulated protein kinases (ERK) 1/2, both in vitro and in vivo. Conclusion: Together, our data suggest that upregulation of ClC- 3 by IGF-1 contributes to cell proliferation and tumor growth in breast cancer, and ClC-3 deficiency suppresses cell proliferation and tumor growth via the IGF/IGF receptor/ERK pathway.

        Synthesis and Antibacterial Activity of 1,3-Diallyltrisulfane Derivatives

        A series of novel 1,3-diallyltrisulfane analogues were synthesized and assayed in vitro for antimicrobial activity against Gram positive, Gram negative bacteria and fungi. The antimicrobial activity of the 1,3-diallyltrisulfane derivatives showed, on the whole, very potent towards all the tested Gram positive, Gram negative and fungi (MIC ranging from 4 to 256 μg/mL). 1,3-Di(pent-4-enyl)trisulfane 3b and 1,3-bis(3-methylbut-2-enyl)trisulfane 3e exhibited the strongest antibacterial activity among all the compounds, and both of them were more active than 1,3-diallyltrisulfane (DATS). Results indicated the relationship of either carbon number or lipophilicity with antimicrobial activity presented “V” shape. These observations provided some predictions in order to further design 1,3-diallyltrisulfane derivatives with antimicrobial activity.

        A Study on the Influence of Personality Traits of College Students on the Way of Coping with Employment Stress and Entrepreneurial Intention in the Post-Epidemic Era

        With the arrival of the "post-epidemic era", the impact of multiple factors such as the epidemic and domestic and foreign economic weakness on the employment and entrepreneurship of graduates is still continuing and deepening, and youth employment has become a worldwide problem. Self-employment has gradually become a new employment channel, and self-employment of college students not only provides a large number of new jobs for the society, but also helps to alleviate the social employment pressure and the severe employment situation of college students. At present, exploring the factors influencing college students' entrepreneurial intention has become a hot topic in the field of entrepreneurship research. In this study, it is important to study the mechanism of each factor by constructing a model of the influencing factors of college students' entrepreneurial intention. The main conclusions from the study are as follows. First, all four personality traits have a significant positive influence on entrepreneurial intention, and college students with the above personality traits are more likely to have entrepreneurial intention, and the way of coping with employment pressure plays a mediating role in the relationship between personality traits and entrepreneurial intention. Based on this, the study provides valuable theoretical implications for research in this area and practical implications for how schools can improve students' entrepreneurial awareness.

        Systemic Inflammatory Biomarkers, Especially Fibrinogen to Albumin Ratio, Predict Prognosis in Patients with Pancreatic Cancer

        Purpose Systemic inflammatory response is a critical factor that promotes the initiation and metastasis of malignancies including pancreatic cancer (PC). This study was designed to determine and compare the prognostic value of neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), monocyte-to-lymphocyte ratio (MLR), and fibrinogen-to-albumin ratio (FAR) in resectable PC and locally advanced or metastatic PC. Materials and Methods Three hundred fifty-three patients with resectable PC and 807 patients with locally advan-ced or metastatic PC were recruited in this study. These patients were classified into a training set (n=758) and a validation set (n=402). Kaplan-Meier survival plots and Cox proportional hazards regression models were used to analyze prognosis. Results Overall survival (OS) was significantly better for patients with resectable PC with low preoperative PLR (p=0.048) and MLR (p=0.027). Low FAR, MLR, NLR (p < 0.001), and PLR (p=0.003) were significantly associated with decreased risk of death for locally advanced or metastatic PC patients. FAR (hazard ratio [HR], 1.522; 95% confidential interval [CI], 1.261 to 1.837; p < 0.001) and MLR (HR, 1.248; 95% CI, 1.017 to 1.532; p=0.034) were independent prognostic factors for locally advanced or metastatic PC. Conclusion The prognostic roles of FAR, MLR, NLR, and PLR in resectable PC and locally advanced or metastatic PC were different. FAR showed the most prognostic power in locally advanced or metastatic PC. Low FAR was positively correlated with OS in locally advanced or metastatic PC, which could be used to predict the prognosis.

        Effect of Sucrose Fatty Acid Esters with Different Hydrophiliclipophilic Balance Values on Pasting and Rheological Properties of Waxy Rice Flour

        Effects of sucrose fatty acid esters (SEs) with hydrophilic-lipophilic balance (HLB) values of 5 (S-570), 9 (S-970), and 15 (S-1570) on the pasting and rheological properties of waxy rice flour (WRF) were investigated. Rapid Visco Analyzer (RVA) showed that addition of SEs affected the pasting properties of WRF. Rheological experiment includes the steady shear flow characteristics, and dynamic viscoelastic properties were also determined using a controlled-stress rheometer. The steady shear tests demonstrated that the viscosity data fitted well with the power law model (R2≥0.976) and all WRF pastes exhibited typical pseudoplastic and shear-thinning properties. Dynamic rheological measurements revealed that the addition of S-970 and S-1570 significantly increased the values of G' and G'' of samples, whereas the addition of S-570 decreased these values. The addition of S-1570 reduced the value of tanδ, whereas S-570 and S-970 increased it.

        BMB Reports : Kaiso represses the expression of glucocorticoid receptor via a methylation-dependent mechanism and attenuates the anti-apoptotic activity of glucocorticoids in breast cancer cells

        Kaiso is a Pox Virus and Zinc Finger (POZ-ZF) transcription factor with bi-modal DNA-binding specificity. Here, we demonstrated that Kaiso expression is inversely correlated with glucocorticoid receptor (GR) expression in breast carcinomas. Knockdown of Kaiso increased GR expression, while overexpression of Kaiso inhibited GR expression in breast cancer cells. Furthermore, Kaiso repressed GR proximal promoter-reporter activity in a dose-dependent manner. Remarkably, ChIP experiments demonstrated that endogenous Kaiso was associated with the GR promoter sequence in a methylation-dependent manner. Since glucocorticoids inhibit chemotherapyinduced apoptosis and have been widely used as a co-treatment of patients with breast cancer, we assessed the role of Kasio in GR-mediated anti-apoptotic effects. We found that overexpression of Kaiso attenuated the anti-apoptotic effects of glucocorticoids in breast cancer cells. Our findings suggest that GR is a putative target gene of Kaiso and suggest Kaiso to be a potential therapeutic target in GC-combination chemotherapy in breast cancer.       • Downregulated MicroRNA-133a in Gastric Juice as a Clinicopathological Biomarker for Gastric Cancer Screening

        Circulatory miR-133a is a marker shared by several types of cancer. In this study we evaluated the feasibility of using miR-133a levels in gastric juice to screen for gastric cancer. A total of 204 samples of gastric juice and mucosa from gastric cancer, atrophic gastritis, gastric ulcer, superficial gastritis and healthy cases were collected by gastroscopy. The results showed that miR-133a levels in gastric juice and carcinoma tissues of patients with gastric cancer were significantly downregulated and positively correlated. Moreover, miR-133a in gastric juice has high operability, high reliability, high sensitivity, high specificity and relative stability, fit for clinical diagnosis of gastric cancer.

      • The structured multiparameter eigenvalue problems in finite element model updating problems

        The multiparameter eigenvalue method can be used to solve the damped finite element model updating problems. This method transforms the original problems into multiparameter eigenvalue problems. Comparing with the numerical methods based on various optimization methods, a big advantage of this method is that it can provide all possible choices of physical parameters. However, when solving the transformed singular multiparameter eigenvalue problem, the proposed method based on the generalised inverse of a singular matrix has some computational challenges and may fail. In this paper, more details on the transformation from the dynamic model updating problem to the multiparameter eigenvalue problem are presented and the structure of the transformed problem is also exposed. Based on this structure, the rigorous mathematical deduction gives the upper bound of the number of possible choices of the physical parameters, which confirms the singularity of the transformed multiparameter eigenvalue problem. More importantly, we present a row and column compression method to overcome the defect of the proposed numerical method based on the generalised inverse of a singular matrix. Also, two numerical experiments are presented to validate the feasibility and effectiveness of our method.
