Antiviral effects of Bovine antimicrobial peptide against TGEV in vivo and in vitro

        Xiuli Liang,Xiaojun Zhang,Kaiqi Lian,Xiuhua Tian,Mingliang Zhang,Shiqiong Wang,Cheng Chen,Cunxi Nie,Yun Pan,Fangfang Han,Zhanyong Wei,Wen-Ju Zhang 대한수의학회 2020 Journal of Veterinary Science Vol.21 No.5

Background: In suckling piglets, transmissible gastroenteritis virus (TGEV) causes lethal diarrhea accompanied by high infection and mortality rates, leading to considerable economic losses. This study explored methods of preventing or inhibiting their production. Bovine antimicrobial peptide-13 (APB-13) has antibacterial, antiviral, and immune functions. Objectives: This study analyzed the efficacy of APB-13 against TGEV through in vivo and in vitro experiments. Methods: The effects of APB-13 toxicity and virus inhibition rate on swine testicular (ST) cells were detected using 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT). The impact of APB-13 on virus replication was examined through the 50% tissue culture infective dose (TCID50). The mRNA and protein levels were investigated by real-time quantitative polymerase chain reaction and western blot (WB). Tissue sections were used to detect intestinal morphological development. Results: The safe and effective concentration range of APB-13 on ST cells ranged from 0 to 62.5 μg/mL, and the highest viral inhibitory rate of APB-13 was 74.1%. The log10TCID50 of 62.5 μg/mL APB-13 was 3.63 lower than that of the virus control. The mRNA and protein expression at 62.5 μg/mL APB-13 was significantly lower than that of the virus control at 24 hpi. Piglets in the APB-13 group showed significantly lower viral shedding than that in the virus control group, and the pathological tissue sections of the jejunum morphology revealed significant differences between the groups. Conclusions: APB-13 exhibited good antiviral effects on TGEV in vivo and in vitro.

Study on bidirectional fluid-solid coupling characteristics of reactor coolant pump under steady-state condition

        Xiuli Wang,Yonggang Lu,Rongsheng Zhu,Qiang Fu,Haoqian Yu,Yiming Chen 한국원자력학회 2019 Nuclear Engineering and Technology Vol.51 No.7

The AP1000 reactor coolant pump is a vertical shielded-mixed flow pump, is the most important coolant power supply and energy exchange equipment in nuclear reactor primary circuit system, whose steady-state and transient performance affect the safety of the whole nuclear island. Moreover, safety demonstration of reactor coolant pump is the most important step to judge whether it can be practiced, among which software simulation is the first step of theoretical verification. This paper mainly introduces the fluid-solid coupling simulation method applied to reactor coolant pump, studying the feasibility of simulation results based on workbench fluid-solid coupling technology. The study found that: for the unsteady calculations of the pure liquid media, the average head of the reactor coolant pump with bidirectional fluid-solid coupling decreases to a certain extent. And the coupling result is closer to the real experimental value. The large stress and deformation of rotor under different flow conditions are mainly distributed on impeller and idler, and the stress concentration mainly occurs at the junction of front cover plate and blade outlet. Among the factors that affect the dynamic stress change of rotor, the pressure load takes a dominant position.

A SERS-based lateral flow assay biosensor for highly sensitive detection of HIV-1 DNA

        Fu, Xiuli,Cheng, Ziyi,Yu, Jimin,Choo, Priscilla,Chen, Lingxin,Choo, Jaebum Elsevier Applied Science 2016 Biosensors & bioelectronics Vol. No.

Abstract

User-friendly lateral flow (LF) strips have been extensively used for point-of-care (POC) self-diagnostics, but they have some limitations in their detection sensitivity and quantitative analysis because they only identify the high cut-off value of a biomarker by utilizing color changes that are detected with the naked eye. To resolve these problems associated with LF strips, we developed a novel surface-enhanced Raman scattering (SERS)-based LF assay for the quantitative analysis of a specific biomarker in the low concentration range. Herein, human immunodeficiency virus type 1 (HIV-1) DNA was chosen as the specific biomarker. Raman reporter-labeled gold nanoparticles (AuNPs) were employed as SERS nano tags for targeting and detecting the HIV-1 DNA marker, as opposed to using bare AuNPs in LF strips. It was possible to quantitatively analyze HIV-1 DNA with high sensitivity by monitoring the characteristic Raman peak intensity of the DNA-conjugated AuNPs. Under optimized conditions, the detection limit of our SERS-based lateral flow assay was 0.24pg/mL, which was at least 1000 times more sensitive compared to colorimetric or fluorescent detection methods. These results demonstrate the potential feasibility of the proposed SERS-based lateral flow assay to quantitatively detect a broad range of genetic diseases with high sensitivity.

Highlights

We report a SERS-based lateral flow biosensor for the detection of HIV-1 DNA.

Quantitative analysis of HIV-1 DNA is possible using this SERS platform.

This approach provides new insights into the early diagnosis of a HIV-1 disease.

      • Research on Dynamic Compliance Analysis Technology of IEC 62351 Standard Based on Finite State Machine Theory

        Wang Chen,Ma Yuanyuan,Shao Zhipeng,Huang Xiuli,Fei Jiaxuan 보안공학연구지원센터 2016 International Journal of Multimedia and Ubiquitous Vol.11 No.10

With the development of power industry, communication security has become an important issue affecting the stability of power system. IEC 62351 standard provides security for substation communication system. However, different transmission protocols may lead to different manufacturers to produce different understanding, manufacturing is not compatible with the device. In order to avoid such a situation, the Finite State Machine (FSM) theory is used to model and analyze the transmission protocol of IEC 62351 standard. In this paper, the FSM model is simulated by the StateFlow tool, and it can be considered that the FSM theory is applicable to the analysis of compliance with IEC 62351 standard.

PM2.5 in poultry houses synergizes with Pseudomonas aeruginosa to aggravate lung inflammation in mice through the NF-κB pathway

        Meng Li,Xiuli Wei,Youzhi Li,Tao Feng,Linlin Jiang,Hongwei Zhu,Xin Yu,Jinxiu Tang,Guozhong Chen,Jianlong Zhang,Xingxiao Zhang 대한수의학회 2020 Journal of Veterinary Science Vol.21 No.3

Background: High concentrations of particulate matter less than 2.5 µm in diameter (PM2.5) in poultry houses is an important cause of respiratory disease in animals and humans. Pseudomonas aeruginosa is an opportunistic pathogen that can induce severe respiratory disease in animals under stress or with abnormal immune functions. When excessively high concentrations of PM2.5 in poultry houses damage the respiratory system and impair host immunity, secondary infections with P. aeruginosa can occur and produce a more intense inflammatory response, resulting in more severe lung injury. Objectives: In this study, we focused on the synergistic induction of inflammatory injury in the respiratory system and the related molecular mechanisms induced by PM2.5 and P. aeruginosa in poultry houses. Methods: High-throughput 16S rDNA sequence analysis was used for characterizing the bacterial diversity and relative abundance of the PM2.5 samples, and the effects of PM2.5 and P. aeruginosa stimulation on inflammation were detected by in vitro and in vivo. Results: Sequencing results indicated that the PM2.5 in poultry houses contained a high abundance of potentially pathogenic genera, such as Pseudomonas (2.94%). The lung tissues of mice had more significant pathological damage when co-stimulated by PM2.5 and P. aeruginosa, and it can increase the expression levels of interleukin (IL)-6, IL-8, and tumor necrosis factor-α through nuclear factor (NF)-κB pathway in vivo and in vitro. Conclusions: The results confirmed that poultry house PM2.5 in combination with P. aeruginosa could aggravate the inflammatory response and cause more severe respiratory system injuries through a process closely related to the activation of the NF-κB pathway.

      • A Study on the Influence of Subjective Exercise Experience on Exercise Persistence of the Elderly

Zijian Zhao,Xiuli Zhang,Yue Chen 아시아건강운동학회 2019 Journal of Asian Society for Health & Exercise Vol.1 No.1

PURPOSE: This paper explores the influence of subjective exercise experience on the persistence of exercise in the elderly in China, and helps to make decisions on how to further improve the persistence of the elderly group to participate in physical exercise. METHODS: The data is randomly collected from a sample of 100 elderly people engaged in physical exercise who were invited to participate in the interview and fill in questionnaires; and the data is processed and analyzed according to two scales, which are "Subjective Exercise Experience Scale" and "Exercise Adherence Scale" in various regions of Henan Province, China. RESULTS: The positive well-being dimensions in the subjective exercise experience of the elderly are positively correlated with the five dimensions of persistence, exercise interest, value judgment, cognitive choice and exercise effort in exercise persistence. There is a significant negative correlation between the psychological distress dimension and the persistence, exercise interest, value judgment, and cognitive choice dimensions in exercise persistence(p <0.01); fatigue dimension is significantly negative correlative with the persistence dimension (p < 0.05) of the exercise persistence; the subjective exercise experience of the elderly has a significant predictive power for their exercise persistence, but there are differences in the predictive power of the various dimensions of exercise persistence. CONCLUSIONS: Positive well-being can positively predict the five dimensions of exercise persistence; psychological distress can negatively predict persistence, exercise interest, value judgment and cognitive choice dimension in exercise persistence. Therefore, by improving positive well-being and reducing psychological annoyance, it can effectively improve the persistence of exercise for the elderly, and thus promote the healthy development of the elderly.
