      • Prognostic Implications for High Expression of MiR-25 in Lung Adenocarcinomas of Female Non-smokers

        Background: Adenocarcinoma (ADC) is the most common histological type of lung cancer and its proportion is rising, especially in Asian non-smoking women. Recent studies suggest miR-25 may have diverse effects on the pathogenesis of different types of cancer. However, the role of miR-25 in lung cancer is still unknown. The aim of this study was to investigate the potential clinical value of miR-25 in non-smoking women with lung ADC. Patients and Methods: Quantitative RT-PCR was performed to evaluate the expression of miR-25 in 100 lung ADC tumor tissues and matched plasma samples and Pearson correlation tests were used to analyze the relationship between values. Associations of miR-25 expression with clinicopathological features were determined using the Student's t-test. To determine prognostic value, overall survival (OS) was evaluated using the Kaplan-Meier method. Univariate and multivariate analyses were performed using the Cox proportional hazard model. Results: Expression of miR-25 in tissue was found to be associated with lymph node metastasis (P=0.021) and disease stage (P=0.012). Moreover, high miR-25 expression was also associated with poorer overall survival of women with lung ADC (P=0.008). Conclusion: Tissue miR-25 expression may be associated with tumor progression and have prognostic implications in female lung ADC patients.

        Gallbladder Tuberculosis: CT Findings with Histopathologic Correlation

        Objective: We wanted to describe the computed tomography (CT) fi ndings of gallbladder tuberculosis (TB) and to correlate them with pathologic fi ndings. Materials and Methods: There were seven patients (M:F = 3:4; mean age, 46.3 years; age range, 32 to 78 years) in whom gallbladder TB was eventually diagnosed. All of them underwent cross-sectional imaging with CT, a pathologic examination and a retrospective review. CT imaging evaluation was done in each case, including the fi ndings of a mass versus nodule, wall thickening (uniform or irregular) and the enhancement patterns (homogeneous or heterogeneous). Results: All the cases of gallbladder TB revealed the following three different CT fi ndings: micronodular lesion of the gallbladder wall (n = 1), a thickened wall (n = 4) and a gallbladder mass (n = 2). There were three cases of homogeneous enhancement of the lesions, including homogeneous enhancement with nodular lesion, homogeneous uniform thickness enhancement and homogeneous thickness enhancement in one case each, and these cases pathology showed tuberculous granuloma with a little caseating necrosis in one case and tuberculous granuloma with rich fi brous tissue, but little or no evident caseating necrosis in two cases. Four cases of heterogeneous enhancement of the lesions, including heterogeneous uniform-thickness enhancement in two cases, heterogeneous enhancement with a local mass lesion in one case and heterogeneous enhancement with a mass that replaced the gallbladder in one case; in these cases, pathology showed tuberculous granuloma with marked caseation or liquefaction necrosis in three cases and tuberculous granuloma by fi brous and calcifi cations accompanied by caseating necrosis in one case. Among the seven cases of gallbladder TB, six cases were accompanied by abdominal extra-gallbladder TB, including abdominal lymph node TB in fi ve cases and hepatic TB in four cases. Conclusion: Gallbladder TB has various CT manifestations, and the enhanced CT findings are well matched with pathological features. An irregularly thickened gallbladder wall or a gallbladder wall mass with multiple-focus necrosis or calcifi cations accompanied by the typical CT fi ndings of abdominal extra-gallbladder TB should suggest the diagnosis of gallbladder TB.

        Alien Chromosome-Specific PCR Markers for Selection of Powdery Mildew Resistance lntrogressed from Haynaldia villosa in Wheat

        In the introgression of the distinguished powdery mildew resistance from Haynaldia villosa to wheat, a focus task is to develop applicable molecular markers for tracing alien genetic substances and breeding selection. In an attempt to breed wheat breeding lines combining powdery mildew resistance of wheat stock 010714 conferred by alien 6V chromosome from H. villosa with thermo- photo-sensitive genic male sterility (TGMS) from wheat line C050S, a molecular marker RM874 was identified in PCR amplification using RAPD primer S2018. Batched sequencing indicated that the band contained large amounts of equal-length homologues amplified from Ty3-gypsy retrotransposon-like repetitive regions. Based on comparative cloning and bioinformatic analysis, it was converted into a more reproducible sequence characterized amplified region (SCAR) marker SM142. In wheat genetic background, the marker is 6V-specific and linked to powdery mildew resistance. The linkage of the marker is further verified in F5 generation. The approach of transforming repetitive regions-derived RAPD band into a specific SCAR marker can be referred for SCAR marker development of other genes. GISH (Genome in situ hybridization) analysis of 010714 and F7 translocation lines indicated that the resistance gene is conferred by the short arm of 6V.

        Protection and Inheritance of Local Dramas in China - A Case Study of Liuqin Troupe in Pizhou City -

        본 연구는 중국 무형문화유산인 유금희(柳琴戲)의 발전 현상을 통하여, 그것이 중국 지방극의 보호 및 전승에 대해 갖는 시사점 분석에 목적을 둔다. 본 논문은 피저우시(邳州市)의 유금희 악단에 대한 사례 분석을 통하여, 총, ‘정부 성과’, ‘희극 본체’, ‘실천 주체’의 세 가지 측면으로 중국 지방극의 연구가 갖는 시사점에 대해 심층 분석하였으며, 극단의 관리자에 대한 인터뷰 조사를 통해 유금희가 지방에서 발전해온 면모를 전면적으로 재현하였다. 연구의 내용은 중국 특색 사회주의 제도에 기초하여 통일성을 갖는 반면, 한 걸음 더 나아가 직관적으로 중국 지방극의 발전에 있어, 보편적으로 존재하는 문제점을 반영하였다. 본 연구의 결과는, 첫째, ‘정부 성과’ 측면이다. 지방극은 중국 정부의 적극적인 추진 정책의 기초 위에서 왕성한 발전을 거두었다. 정부는 지방극의 발전 조건을 충족시키는 동시에 정부 간섭을 줄임으로써 발전 공간을 제공하였다. 둘째, ‘희극 본체’ 측면이다. 극의 창작은 지방극의 특색을 보전한다는 전제하에 최대한 ‘동질화(同質化)’ 형성을 피하고 연출 공간이 민간에서 활약되도록 하였다. 셋째, ‘실천 주체’ 측면이다. 이는 구체적으로, 원로 예술인의 발굴 및 보호, ‘사도전승(师徒传承)’으로의 회귀, 신예 예술인의 육성 강화 및 관중의 다양한 요구 만족 및 관중 모집 등을 포함한다. 본 연구의 의의는 중국 지방극에 대한 전승과 보호에서부터 심층 해석 및 확장 해석을 돕는다. 또한, 연구의 참고자료로서 ‘살아 있는 전승’인 무형 문화유산의 분석과 연구에 활용될 수 있다. This study aims to explore the enlightenment of Liuqin Drama on the protection and inheritance of Chinese local dramas by analyzing the development status of Liuqin Drama, China’s intangible cultural heritage. A case study of Pizhou Liuqin Troupe has been conducted from three aspects: “government actions”, “drama as the subject”, and “practice subject”. Besides, visit and interview against the management of the troupe have been conducted. Collectively, this has restored the appearance of Liuqin Drama in the local development. Based on the socialist system with Chinese characteristics, the development background of the research content is unified to directly reflect the widespread problems in the development of Chinese local drama. Research on Chinese local drama demonstrates that regarding “government actions”, Chinese local drama has developed vigorously on the basis of the government’s active policies. To satisfy its development, the government supports its development and reduces political interference. In terms of “drama as the subject”, the production of “homogenization” should be avoided while the performance space should be active in the folk for local dramas on the premise of retaining the characteristics of local dramas. Respecting “practice subject”, they try to discover and protect old artists, return to the mode of “inheritance from master to apprentice”, and accelerate the cultivation and training of new artists. Moreover, it should meet the diverse needs of the audience and cultivate the audience. The results of this study are bound to enable people to deeply interpret and expand the inheritance and protection of Chinese local dramas. It can be the reference materials for the research on the intangible cultural heritage of “living state” inheritance.

      • Efficacy of Taxane-Based Regimens in a First-line Setting for Recurrent and/or Metastatic Chinese Patients with Esophageal Cancer

        Objective: To compare the efficacy of taxane-based regimens in the first line setting retrospectively in Chinese patients with recurrent and/or metastatic esophageal cancer. Methods: We analyzed 102 recurrent and/or metastatic esophageal cancer patients who received taxanes-based regimens in a first-line setting from January 2009 to December 2013. Sixteen (15.7%) patients were administered Nab-PTX based chemotherapy and 86 patients (84.3%) received paclitaxel (PTX) or docetaxel (DTX) based chemotherapy. Patients in the PTX/DTX group could be further divided into TP (71 patients) and TPF (15 patients) groups. Results: The objective response rate (ORR) of all patients was 20.6%, and the disease control rate (DCR) was 67.6%. The median overall survival (OS) was 10.5 months (95% CI 10.1-16.4) and the median progression-free survival (PFS) was 6.04 months (95% CI 5.09-7.91). The DCR was higher in the TPF group than the TP group (93.3% vs. 59.1%; p = 0.015 ). There were no significant differences in ORR, OS, and PFS among Nab-PTX, TPF and TP groups. Conclusions: The three regimens of Nab-PTX based, TP and TPF proved active in a first line setting of Chinese patients with recurrent and/or metastatic esophageal cancer, and should thus be regarded as alternative treatments.

      • Evolution of ALPPS: The Simpler, Safer and Effective One---TELPP

        Aims: The characteristic of associating liver partition and portal vein ligation for staged hepatectomy(ALPPS) carries high mortality and morbidity. There is room for improvement. We suggest Terminal Branches Portal Vein Embolization (TBPVE) as a way to compart the liver. As a result, only a single surgical operation is required.This method is termed Terminal branches portal vein Embolization Liver Partition Planned hepatectomy (TELPP). Methods: Patients with unresectable primary or metastatic liver tumor were performed with TELPP. The procedure of TELPP was that in addition to PVE, embolization agent was infused to the terminal branches of portal vein of S5,S8 or S4. CT scan was taken one or two weeks later, and standard liver volume(SLV), FLR and FLR/SLV are calculated. Two weeks later when the FLR and liver function is appropriate, open or laparoscopic hepatectomy is performed. Results: The study included 11patients including hepatocellular carcinoma: n =8, intrahepatic cholangiocarcinoma: n = 1, hilarcholangiocarcinoma: n =1, colorectal liver metastasis: n =1. After a waiting period of 14 days, the volume of theFLR had increased from 382mlto 578ml, representing a median volume increase of 51% (range =32.5%-86.7%). Of the 11patients with hepatectomy, right hemihepatectomy (n=2), extended right hemihepatectomy (n=5), right trisecmentectomy(2), extended left hemihepatectomy (n=1) and left trisecmentectomy(1). No patient died, and no serve perioperative morbidity occurred. Conclusions: ALPPS and all modifications need two-stage operations with a high morbidity and mortality rate. It seems that TELPP is very promising. It has the merit of ALPPS as extraordinarily rapid increasement of FLRvolume, yet the morbidity and mortality is much lower, owing to the fact that unlike ALPPS, there is no two liver raw surfaces left behind in the abdominal cavity to produce bile leak, as only single surgical operation is required

        Pokemon Inhibits Transforming Growth Factor β-Smad4-Related Cell Proliferation Arrest in Breast Cancer through Specificity Protein 1

        Purpose: Pokemon, also known as ZBTB7A, belongs to the POZ and Krüppel (POK) family of transcription repressors and is implicated in tumor progression as a key proto-oncogene. This present study aimed at determining the mechanism by which Pokemon inhibits transforming growth factor β (TGFβ)-Smad4 pathway-dependent proliferation arrest of breast cancer cells via specificity protein 1 (SP1). Methods: Over-expressing plasmid or small interfering RNA (siRNA) transfection was used to regulate Pokemon levels. The EdU incorporation assay, MTS assay, and clone formation were used to identify the inhibitory effect of Pokemon siRNA on cell proliferation. Quantitative real-time polymerase chain reaction assay confirmed that Pokemon deletion inhibited the expression of proliferation-associated genes. The dual-luciferase reporter assay, electrophoretic mobility shift assay, and co-immunoprecipitation assay were used to analyze binding between Pokemon, Smad4, and SP1. Results: Pokemon deletion induced proliferation arrest of breast cancer cells and inhibited the expression of proliferation-associated genes, especially Smad4. Pokemon bound with SP1 to interdict Smad4 promoter activity. Information on clinical samples was obtained from The Cancer Genome Atlas data, in which the Pokemon mRNA levels showed a negative correlation with Smad4 levels in different subtypes of breast cancer in two independent datasets. Conclusion: We demonstrated that Pokemon binds to SP1 to down-regulate Smad4 expression, thereby promoting proliferation of breast cancer cells. This suggests that Pokemon is a potential TGFβ-signaling participant in breast cancer progression.
