      • Risk Factors, Patterns, and Outcomes of Late Recurrence after Liver Resection for Patients with Hepatocellular Carcinoma (Analysis of a Multicenter Cohort over 15 Years)

        Aims: Late recurrence (> 2 years) after liver resection of hepatocellular carcinoma (HCC) is usually considered as multi-centric tumors or de novo cancer formation. We aimed to investigate risk factors, patterns and outcomes of late recurrence after HCC resection. Methods: From a multicenter database from 2001 to 2015, 734 patients who were alive and recurrence-free at 2 years after curative resection of initial HCC were enrolled into this retrospective study. Univariate and multivariate Cox-regression analysis were used to identify independent risk factors of late recurrence. Patterns, treatments and outcomes of late recurrence were investigated and analyzed. Results: During a median follow-up of 78.0 months after surgery, 303 patients (41.3%) developed late recurrence. Multivariate analysis revealed that cirrhosis, macroscopic vascular invasion, satellites, and tumor size > 5cm were independent risk factors of late recurrence. Among them, 273 (90.1%) were sole intrahepatic recurrence, 30 (9.9%) were concurrent intrahepatic and extrahepatic recurrence, and none of them was sole extrahepatic recurrence; 165 (54.4%) patients received curative treatments for recurrent HCC, including re-resection, transplantation and local ablation. Multivariate analysis showed regular postoperative surveillance and receiving curative treatments were two independent protective factors of prolonging survival for those patients with late recurrence. Conclusions: Late recurrence is correlated with cirrhosis and certain tumor-related characteristics of initial HCC. The patterns of late recurrence suggest that postoperative surveillance after 2 years of surgery could be adjusted and more targeted. Regular postoperative surveillance improves the probability to receive curative treatments again, yielding to better outcomes for patients with late recurrence.

      • Design of 1500V solar inverter stack beyond megawatt in NPC1 topology

        This paper describes a design concept of NPC1 power stack for 1500VDC megawatt level solar inverter. This stack uses three latest half-bridge IGBT modules with highest power density and operation junction temperature, which enable realization of power level beyond 1MW without paralleling. Critical design concept on loop inductance is explained. Dynamic characteristics are verified by double-pulse test. Thermal characteristics and output power limits are verified by thermal test. Temperature-sensitive component on PCB as output power constraint is identified. Different PCB repositioning solutions are tested to give the overall output power thermal derating curves, which enable output power of 1.15MW at TA=55°C with 15°C thermal margin. The power stack characteristic and performance change under different thermal environment is further analyzed.

        Development and Application of High-Performance Grouting Materials for Anchoring Pipe in Soft Rock Tunnel

        A high-performance grouting material is proposed to enhance the support effect of anchoring pipes in soft rock mass tunnels in this paper. Through mix proportion and strength tests, the initial setting time, compressive strength, and flowability of the grouting material were determined. Furthermore, based on grouting and pulling tests, the recommended parameters of the anchoringpipe were proposed. The results show that pure cement paste is an ideal grouting material for on-site application, demonstrating excellent anchoring, fluidity, high strength, and rapid hardening. Under optimal conditions of water-cement ratio at 0.46 and retarder proportion at 0.4‰, the initial settling time and the compressive strength of the grouting material are approximately 45 minutes and 28.5 MPa within 4 hours, respectively. The anchoring force of an anchoring pipe with a length of 6 m and a diameter of 76 mm can reach 448.9 kN within the same period. A field test involving anchoring pipes and pure cement paste was conducted during tunnel construction, resulting in a substantial reduction of maximum settlement and convergence of the tunnel by 57.53% and 40.48%, respectively.         Creep characteristics and instability analysis of concrete specimens with horizontal holes

        Uniaxial compressive strength test and uniaxial compression creep one were produced on four groups of twelve concrete specimens with different hole number by RLW-2000 rock triaxial rheology test system. The relationships between horizontal holes and instantaneous failure stress, the strain, and creep failure stress, the strain, and the relationships between stress level and instantaneous strain, creep strain were studied, and the relationship between horizontal holes and failure mode was determined. The results showed that: with horizontal hole number increasing, compressive strength of the specimens decreased whereas its peak strain increased, while both creep failure strength and its peak strain decreased. The relationships between horizontal holes and compressive strength of the specimens, the peak strain, were represented in quadratic polynomial, the relationships between horizontal holes and creep failure strength, the peak strain were represented in both linear and quadratic polynomial, respectively. Instantaneous strain decreased with stress level increasing, and the more holes in the blocks the less the damping of instantaneous strain were recorded. In the failure stress level, instantaneous strain reversally increased, creep strain showed three stages: decreasing, increasing, and sharp increasing; in same stress level, the less holes the less creep strain rate was recorded. The compressive-shear failure was produced along specimen diagonal line where the master surface of creep failure occurred, the more holes in a block, the higher chances of specimen failure and the more obvious master surface were.

        A Day-Ahead Optimization Method of Source–Load Coordination for Power System Using Demand Response and Stackelberg Game

        The continuous expansion of renewable energy sources and their ongoing integration into existing power networks, alongside the emergence of new types of loads, has led to signifcant new challenges emerging for power grid source–load coordination scheduling in recent years. To fully harness the demand response (DR) potential of the load side of such systems, this paper proposes an optimal scheduling method based on DR and the Stackelberg game. A Stackelberg game model is established to formulate day-ahead source–load coordinated scheduling problem, taking account of the stochastic dynamic characteristics of the load response process and user electricity consumption satisfaction with electricity consumption. The model is essentially a strongly nonlinear optimization problem, however; thus, to avoid several of the limitations seen in traditional optimization methods, a data-model fusion-driven method is proposed, with electricity prices formulated by the data-driven method used as input for a model-driven method whose output is then used in turn to guide a data-driven formulation of electricity prices. Case studies on IEEE test systems are used to verify the feasibility and accuracy of the proposed method.

      • Sex Differences in Early and Late Recurrence after Liver Resection of Hepatocellular Carcinoma (A Multicenter Study from China)

        Aims: There is a striking sex difference in the incidence of hepatocellular carcinoma (HCC), with a strong predominance for males. However, the impact of sex on the incidence of recurrence after curative resection of HCC remains controversial. Herein, we assess sex differences in the risks of recurrence and mortality for patients undergone curative resection of HCC. Methods: Data from 1,435 HCC patients undergone curative resection (1,228 males and 207 females) between 2004 and 2014 at five institutions in China were retrospectively analyzed. Patients’ baseline characteristics, operative variables, and rates of early recurrence (≤ 2 years after resection), late recurrence (> 2 years) and cancer-specific mortality (CSM) were evaluated and compared. Multivariable competing-risks regression analyses were performed to identify predictors associated with CSM, early and late recurrence. Results: The early recurrence rates between males and females were similar (43.3% vs. 42.0%, P=0.728), but the late recurrence and CSM rates in males were higher when compared to females (17.2% vs. 11.2%, P=0.044; 42.8% vs. 34.3%, P=0.022). Multivariable competing-risks regression analyses revealed no sex difference in early recurrence; however, males had significantly higher late recurrence rate [hazard ratio (HR), 1.752; 95% CI, 1.145-2.682; P=0.010] and CSM rate (HR, 1.307; 95% CI, 1.015-1.683; P=0.038) than females. Conclusions: Males had significantly higher late recurrence and CSM rates after curative resection of HCC than females. This suggests postoperative surveillance for HCC recurrence be varied by sex, especially for patients without recurrence at 2 years after resection.

        Mitochondrial citrate accumulation drives alveolar epithelial cell necroptosis in lipopolysaccharide-induced acute lung injury

        Necroptosis is the major cause of death in alveolar epithelial cells (AECs) during acute lung injury (ALI). Here, we report a previously unrecognized mechanism for necroptosis. We found an accumulation of mitochondrial citrate (citratemt) in lipopolysaccharide (LPS)-treated AECs because of the downregulation of Idh3α and citrate carrier (CIC, also known as Slc25a1). shRNA- or inhibitor–mediated inhibition of Idh3α and Slc25a1 induced citratemt accumulation and necroptosis in vitro. Mice with AEC-specific Idh3α and Slc25a1 deficiency exhibited exacerbated lung injury and AEC necroptosis. Interestingly, the overexpression of Idh3α and Slc25a1 decreased citratemt levels and rescued AECs from necroptosis. Mechanistically, citratemt accumulation induced mitochondrial fission and excessive mitophagy in AECs. Furthermore, citratemt directly interacted with FUN14 domain-containing protein 1 (FUNDC1) and promoted the interaction of FUNDC1 with dynamin-related protein 1 (DRP1), leading to excessive mitophagy-mediated necroptosis and thereby initiating and promoting ALI. Importantly, necroptosis induced by citratemt accumulation was inhibited in FUNDC1-knockout AECs. We show that citratemt accumulation is a novel target for protection against ALI involving necroptosis.

        Multicomponent assessment and ginsenoside conversions of Panax quinquefolium L. roots before and after steaming by HPLC-MS<SUP>n</SUP>

        Background: The structural conversions in ginsenosides induced by steaming or heating or acidic condition could improve red ginseng bioactivities significantly. In this paper, the chemical transformations of red American ginseng from fresh Panax quinquefolium L. under steaming were investigated, and the possible mechanisms were discussed. Methods: A method with reversed-phase high-performance liquid chromatography coupled with linear ion trap mass spectrometry (HPLC-MS<SUP>n</SUP>)-equipped electrospray ionization ion source was developed for structural analysis and quantitation of ginsenosides in dried and red American ginseng. Results: In total, 59 ginsenosides of protopanaxadiol, protopanaxatriol, oleanane, and ocotillol types were identified in American ginseng before and after steaming process by matching the molecular weight and/or comparing MS<SUP>n</SUP> fragmentation with that of standards and/or known published compounds, and some of them were determined to be disappeared or newly generated under different steaming time and temperature. The specific fragments of each aglycone-type ginsenosides were determined as well as aglycone hydrated and dehydrated ones. The mechanisms were deduced as hydrolysis, hydration, dehydration, and isomerization of neutral and acidic ginsenosides. Furthermore, the relative peak areas of detected compounds were calculated based on peak areas ratio. Conclusion: The multicomponent assessment of American ginseng was conducted by HPLC-MS<SUP>n</SUP>. The result is expected to provide possibility for holistic evaluation of the processing procedures of red American ginseng and a scientific basis for the usage of American ginseng in prescription.
