        WRKY22 Transcription Factor Mediates Dark-Induced Leaf Senescence in Arabidopsis

        Arabidopsis WRKY proteins are plant-specific transcrip-tion factors, encoded by a large gene family, which contain the highly conserved amino acid sequence WRKYGQK and the zinc-finger-like motifs, Cys_2His_2 or Cys_2HisCys. They can recognize and bind the TTGAC(C/T) W-box cis-elements found in the promoters of target genes, and are involved in the regulation of gene expression during pathogen defense, wounding, trichome development, and senescence. Here we investigated the physiological function of the Arabidopsis WRKY22 transcription factor during dark-induced senescence. WRKY22 transcription was suppressed by light and promoted by darkness. In addi-tion, AtWRKY22 expression was markedly induced by H_2O_2. These results indicated that AtWRKY22 was involved in signal pathways in response to abiotic stress. Dark-treated AtWRKY22 over-expression and knockout lines showed accelerated and delayed senescence phenotypes, respectively, and senescence-associated genes exhibited increased and decreased expression levels. Mutual regulation existed between AtWRKY22 and AtWRKY6, AtWR-KY53, and AtWRKY70, respectively. Moreover, AtWRKY22 could influence their relative expression levels by feedback regulation or by other, as yet unknown mechanisms in response to dark. These results prove that AtWRKY22 participates in the dark-induced senescence signal transduction pathway.

      • A Novel Mutant of Human Papillomavirus Type 18 E6E7 Fusion Gene and its Transforming Activity

        Background: Persistent human papillomavirus (HPV) infection, especially with high-risk types such as HPV16 and HPV18, has been identified as the primary cause of cervical cancer. E6 and E7 are the major onco-proteins of high-risk HPVs, which are consistently expressed in HPV infected tissues but absent in normal tissues and represent ideal therapeutic targets for immunotherapy of cervical cancer. Materials and Methods: In this study, the optimized fusion gene HPV18 E6E7 (HPV18 ofE6E7) was constructed according to genetic codon usage for human genes. At the same time, for safety future clinical application, a mutant of HPV18 ofE6E7 fusion gene was generated by site-directed mutagenesis at L52G for the E6 protein and C98G for the E7 protein. Results: HPV18-E6E7 mutant (HPV18 ofmE6E7) constructed in this work not only lost the transformation capability for NIH 3T3 cells and tumorigenicity in BALB/c nude mice, but also maintained very good stability and antigenicity. Conclusion: These results suggest that the mutant should undergo further study for application as a safe antigenspecific therapeutic vaccine for HPV18-associated tumors.

      • Immunotherapeutic Effects of Dendritic Cells Pulsed with a Coden-optimized HPV 16 E6 and E7 Fusion Gene in Vivo and in Vitro

        Background: Cervical cancer is the second most common cause of cancer related death of women. Persistent HPV infection, especially with high-risk types such as HPV16 and HPV18, has been identified to be the primary cause of cervical cancer. E6 and E7 are the major oncoproteins of high-risk HPVs, which are expressed exclusively in HPV infected tissues, and thereby represent ideal therapeutic targets for immunotherapy of cervical cancer. Materials and Methods: In this work, we used recombinant adenovirus expressing coden-optimized HPV16 E6 and E7 fusion protein (Ad-ofE6E7) to prime dendritic cells (DC-ofE6E7), to investigate the ability of primed DC vaccine in eliciting antitumor immunity in vitro and vivo. Results: Our results indicated that DC-ofE6E7 vaccine co-culturing with splenocytes could strongly induce a tumor-specific cytotoxic T lymphocyte (CTL) response and kill the TC-1 cells effectively in vitro. Moreover, DC-ofE6E7 vaccine induced protective immunity against the challenge of TC-1 cancer cells in vivo. Conclusions: The results suggested that the HPV16 ofE6E7 primed DC vaccine has potential application for cervical cancer immunotherapy.

        Bio-inspired leaf stent for direct treatment of cerebral aneurysms: design and finite element analysis

        Cerebral aneurysm is common lesion among adult population. Current methods for treating the disease have several limitations. Inspired by fern leaves, we have developed a new stent, called leaf stent, which can provide a tailored coverage at the neck of an aneurysm and thus prevent the blood from entering the aneurysm. It alone can be used to treat the cerebral aneurysm and therefore overcomes problems existing in current treating methods. The paper focuses on the numerical simulation of the leaf stents. The mechanical behaviour of the stent in various designs has been investigated using the finite element method. It has been found that certain designs provide adequate radial force and have excellent longitudinal flexibility. The performance of certain leaf stents is comparable and even superior to those of the commercially available cerebral stents such as the Neuroform stent and the Enterprise stent, commonly used for stent assisted coiling, while at the same time, providing sufficient coverage to isolate the aneurysm without using coils.

        Space Charge Behavior of Oil-paper Insulation Thermally Aged under Different Temperatures and Moistures

        Moisture and high temperature are the most important factors that lead to the ageing of oil-paper insulation, but the research about space charge characteristics of oil-paper insulation does not take the combined effect of ambient temperature, moisture and thermal ageing into account. The pulsed electroacoustic (PEA) method was used to investigate the influence of moisture and temperature on space charge characteristics of oil paper at different ageing stages. The results showed that moisture could speed up formation of space charge in oil paper when water concentration was low, but the formation was restrained if the water concentration was high. At the beginning of thermal ageing, heterogeneous charge accumulation had predominance, but it gradually changed to homogeneous charge injection with ageing. It was believed that moisture concentration could speed up ageing and enhance charge accumulation on one hand, and accelerate or slow down the establishment speed of space charge on the other hand, therefore, charge accumulation type changed with ageing. The more seriously the oil-paper insulation was thermally aged, the deeper the trap energy level was, hence more space charge was trapped, which could be speeded up by increasing the ageing temperature, but the effect of ambient temperature did not fit the Arrhenius law.

        Hemocyte Changes after the Extirpation of the Hemopoietic Organ-wing Disc Complexes in the Silkworm, Bombyx mori (Lepidoptera: Bombycidae)

        We successfully extirpated all four hemopoietic organ-wing disc complexes of the fifth instar larvae of Bombyx mori, and found that most of the treated silkworms could still develop into the moths. We investigated the changes of the circulating hemocytes and evaluated the effects of extirpation on the hemopoiesis. The results showed that proliferation of circulating hemocytes was sufficient to allow development of the silkworms which complexes were totally extirpated. We also found that hemopoietic organ-wing disc complexes extirpation might cause a certain hemopoietic compensation of the remainder complexes during early spinning. Exogenous hormones such as $20-{\beta}-hydroxyecdysone$ and juvenile hormone analog had a positive effect on hemocytes proliferation.

      • Full-scale test of dampers for stay cable vibration mitigation and improvement measures

        This paper reported test of full-scale cables attached with four types of dampers: viscous damper, passive Magneto-Rheological (MR) damper, friction damper and High Damping Rubber (HDR) damper. The logarithmic decrements of the cable with attached dampers were calculated from free vibration time history. The efficiency ratios of the mean damping ratios of the tested four dampers to theoretical maximum damping ratio were derived, which was very important for practical damper design and parameter optimization. Non-ideal factors affecting damper performance were discussed based on the test results. The effects of concentrated mass and negative stiffness were discussed in detail and compared theoretically. Approximate formulations were derived and verified using numerical solutions. The critical values for non-dimensional concentrated mass coefficient and negative stiffness were identified. Efficiency ratios were approximately 0.6, 0.6, and 0.3 for the viscous damper, passive MR damper and HDR damper, respectively. The efficiency ratio for the friction damper was between 0-1.0. The effects of concentrated mass and negative stiffness on cable damping were positive as both could increase damping ratio; the concentrated mass was more effective than negative stiffness for higher vibration modes.

        Forming a thick CdZnTe epitaxial film by sublimating closely spaced

        Thick Cadmium zinc telluride epitaxial films were grown on (100)GaAs substrate by Close-Spaced Sublimation. The growthrate was around 1 μm/min. SEM observations and XRD θ-2θ scan and ф scan revealed well-defined epitaxial features ofthe films. The films were found to be polycrystalline at the early stage of growth. As growth progressed, the films graduallybecame oriented until an epitaxial layer was formed. The polycrystalline to monocrystalline transition was achieved bypreferential lateral growth of the grains and with the aid of migration of atoms in the film. This growth mode differs fromthe well-established epitaxy where a suitable lattice fit is expected from the early stages of film growth. This mode can betterguide the growth mode of CZT thick films, and can grow large volume and high-quality thick films. High quality thick filmscan be better applied in fields such as solar cells, infrared and ultraviolet detection, and X-ray detection.
