        The Aurora Kinase Inhibitor CYC116 Promotes the Maturation of Cardiomyocytes Derived from Human Pluripotent Stem Cells

        Human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) have great potential in applications such as regenerative medicine, cardiac disease modeling, and in vitro drug evaluation. However, hPSC-CMs are immature, which limits their applications. During development, the maturation of CMs is accompanied by a decline in their proliferative capacity. This phenomenon suggests that regulating the cell cycle may facilitate the maturation of hPSC-CMs. Aurora kinases are essential kinases that regulate the cell cycle, the role of which is not well studied in hPSC-CM maturation. Here, we demonstrate that CYC116, an inhibitor of Aurora kinases, significantly promotes the maturation of CMs derived from both human embryonic stem cells (H1 and H9) and iPSCs (induced PSCs) (UC013), resulting in increased expression of genes related to cardiomyocyte function, better organization of the sarcomere, increased sarcomere length, increased number of mitochondria, and enhanced physiological function of the cells. In addition, a number of other Aurora kinase inhibitors have also been found to promote the maturation of hPSC-CMs. Our data suggest that blocking aurora kinase activity and regulating cell cycle progression may promote the maturation of hPSC-CMs.

        Baseline Total Metabolic Tumor Volume and Total Lesion Glycolysis Measured on 18F-FDG PET-CT Predict Outcomes in T-Cell Lymphoblastic Lymphoma

        Purpose There is no optimal prognostic model for T-cell lymphoblastic lymphoma (T-LBL). Here, we discussed the predictive value of total metabolic tumor volume (TMTV) and total lesion glycolysis (TLG) measured on 18F-fluorodeoxyglucose positron emission tomography–computed tomography (PET-CT) in T-LBL.Materials and Methods Thirty-seven treatment naïve T-LBL patients with PET-CT scans were enrolled. TMTV was obtained using the 41% maximum standardized uptake value (SUVmax) threshold method, and TLG was measured as metabolic tumor volume multiplied by the mean SUV. Progression-free survival (PFS) and overall survival (OS) were analyzed by Kaplan-Meier curves and compared by the log-rank test.Results The optimal cutoff values for SUVmax, TMTV, and TLG were 12.7, 302 cm3, and 890, respectively. A high SUVmax, TMTV, and TLG indicated a shorten PFS and OS. On multivariable analysis, TMTV ≥ 302 cm3, and central nervous system (CNS) involvement predicted inferior PFS, while high SUVmax, TLG and CNS involvement were associated with worse OS. Subsequently, we generated a risk model comprising high SUVmax, TMTV or TLG and CNS involvement, which stratified the population into three risk groups, which had significantly different median PFS of not reached, 14 months, and 7 months for low-risk group, mediate-risk group, and high-risk group, respectively (p < 0.001). Median OS were not reached, 27 months, and 13 months, respectively (p < 0.001).Conclusion Baseline SUVmax, TMTV, and TLG measured on PET-CT are strong predictors of worse outcome in T-LBL. A risk model integrating these three parameters with CNS involvement identifies patients at high risk of disease progression.

        We investigate the homotopy category ${\mathcal{K}}_N(Inj{\mathfrak{A}})$ of N-complexes of injectives in a Grothendieck abelian category ${\mathfrak{A}}$ not necessarily locally noetherian, and prove that the inclusion ${\mathcal{K}}_N(Inj{\mathfrak{A}}){\rightarrow}{\mathcal{K}}({\mathfrak{A}})$ has a left adjoint and ${\mathcal{K}}_N(Inj{\mathfrak{A}})$ is well generated. We also show that the homotopy category ${\mathcal{K}}_N(PrjR)$ of N-complexes of projectives is compactly generated whenever R is right coherent.

        Cerium-loaded MnOx/attapulgite catalyst for the low-temperature NH3-selective catalytic reduction

        A series of MnO2/attapulgite (ATP) and n(Ce):n(Mn)/ATP (molar ratios) catalysts were prepared andinvestigated for the selective catalytic reduction of NO by NH3 (NH3-SCR) at low temperature. The resultsshowed that the 7 wt % MnO2/ATP exhibited the best NOx conversion (85% at 300 C) among all MnO2/ATPcatalysts of different mass ratios. The introduction of cerium enhanced the NOx conversion at lowtemperature, and so Ce–MnOx/ATP can reach the highest NOx conversion (95% at 300 C). Meanwhile, theas-prepared catalysts were characterized by XRD, TEM, BET, H2-TPR, NH3-TPD, and XPS. It can be deducedfrom TEM, XRD, and BET, MnOx nanorods in this work mainly existed in the b-MnO2, and cerium highlydispersed on the surface of ATP to form porous structure and thus improved the deNOx performance. Moreover, the study of SO2 tolerance demonstrated that cerium can effectively inhibit SO2 poison. XPSresults illustrated that Ce could enhance Mn4+ content on the surface of the catalyst and thus lead to highSCR activity. Therefore Mn(1):Ce(0.25)/ATP was proved to be an excellent catalyst for NH3-SCR.

        vfr, A Global Regulatory Gene, is Required for Pyrrolnitrin but not for Phenazine-1-carboxylic Acid Biosynthesis in Pseudomonas chlororaphis G05

        In our previous study, pyrrolnitrin produced in Pseudomonas chlororaphis G05 plays more critical role in suppression of mycelial growth of some fungal pathogens that cause plant diseases in agriculture. Although some regulators for pyrrolnitrin biosynthesis were identified, the pyrrolnitrin regulation pathway was not fully constructed. During our screening novel regulator candidates, we obtained a white conjugant G05W02 while transposon mutagenesis was carried out between a fusion mutant G05ΔphzΔprn::lacZ and E. coli S17- 1 (pUT/mini-Tn5Kan). By cloning and sequencing of the transposon-flanking DNA fragment, we found that a vfr gene in the conjugant G05W02 was disrupted with mini-Tn5Kan. In one other previous study on P. fluorescens, however, it was reported that the deletion of the vfr caused increased production of pyrrolnitrin and other antifungal metabolites. To confirm its regulatory function, we constructed the vfr-knockout mutant G05Δvfr and G05ΔphzΔprn::lacZΔvfr. By quantifying β-galactosidase activities, we found that deletion of the vfr decreased the prn operon expression dramatically. Meanwhile, by quantifying pyrrolnitrin production in the mutant G05Δvfr, we found that deficiency of the Vfr caused decreased pyrrolnitrin production. However, production of phenazine-1-carboxylic acid was same to that in the wild-type strain G05. Taken together, Vfr is required for pyrrolnitrin but not for phenazine-1-carboxylic acid biosynthesis in P. chlororaphis G05.

        Starch graft copolymer and polymer gel applied in Bohai oilfield for water plugging and profile control and their mechanisms

        Water plugging and profile control are becoming increasingly important in water-flooding oilfields. In this study, the experiments were conducted to determine the microscopic mechanisms and effects of water plugging, profile control, and ‘water plugging + profile control’. Initially, the starch graft copolymer and polymer gel used showed only slight changes in viscosity. However, within 12 h, the viscosities exceeded 100,000 mPa·s, which indicated that the two agents had good plugging effects. When the plugging agent was starch graft copolymer, the oil recovery with water plugging was 2% and 0.8% higher than the values for profile control with polymer gel on a heterogeneous core and parallel core, respectively. The recoveries with the ‘water plugging + profile control’ combination for the heterogeneous core and parallel core were 25.9% and 25.5%, respectively, which showed the superior enhanced oil recovery. The mechanism research showed that when the reservoir entered the middle and high water-cut development stages, the residual oil was mainly distributed in the middle- and low-permeability layers near the oil well. Thus, water plugging provided a greater increase in oil recovery. Under the actual demands of the Bohai oilfield, it would be better to adopt a combined water plugging and profile control operation.
