Association between Working Hours and Anxiety/Depression of Medical Staff during Large-Scale Epidemic Outbreak of COVID-19: A Cross-Sectional Study

        Qin Lang,Xiaojing Liu,Ying He,Qin Lv,Sibo Xu 대한신경정신의학회 2020 PSYCHIATRY INVESTIGATION Vol.17 No.12

Objective This present study aimed to investigate the relationship between working hours and anxiety/depression mood of medical staff in China during COVID-19 epidemic. Methods The cross-sectional interview study was conducted during the period between February 14th and February 29th, 2020. A total of 291 Chinese medical professionals were recruited from 4 cities and participated in the study. Results In 291 participants, 116 (40.0%) medical staff experienced anxiety and 151 (51.8%) underwent depressed mood. In male, the level of GAD-7 and PHQ-9 scores increased with the elevation of working hours per day (WHPD) (β=0.579, p=0.003 and β=0.943; p=0.001) respectively. In female, nonlinear relationship mode was demonstrated. The levels of GAD-7 and PHQ-9 scores increased with the elevation of working hours when it was above 5 hours (β=1.432; p<0.001 and β=1.177; p<0.001), but it did not have a significant association with WHPD when it was less than 5 (p>0.05). Conclusion During the COVID-19 epidemic, we found a strong correlation between the psychological mood and WHPD. The correlation followed different modes in male and female medical workers. Enforcing an upper time limit of WHPD may help decrease the risk of pandemic-related psychological problems in medical workers.

Umbilical Cord Mesenchymal Stem Cell Treatment for Crohn’s Disease: A Randomized Controlled Clinical Trial

( Jian Zhang ),( Samei Lv ),( Xiaojing Liu ),( Bin Song ),( Liping Shi ) 대한간학회 2018 Gut and Liver Vol.12 No.1

Background/Aims: Stem cell therapy has been applied to treat a variety of autoimmune diseases, including Crohn’s disease (CD), but few studies have examined the use of umbilical cord mesenchymal stem cells (UC-MSCs). This trial sought to investigate the efficacy and safety of UC-MSCs for the treatment of CD. Methods: Eighty-two patients who had been diagnosed with CD and had received steroid maintenance therapy for more than 6 months were included in this study. Forty-one patients were randomly selected to receive a total of four peripheral intravenous infusions of 1×106 UC-MSCs/kg, with one infusion per week. Patients were followed up for 12 months. The Crohn’s disease activity index (CDAI), Harvey-Bradshaw index (HBI), and corticosteroid dosage were assessed. Results: Twelve months after treatment, the CDAI, HBI, and corticosteroid dosage had decreased by 62.5±23.2, 3.4±1.2, and 4.2±0.84 mg/day, respectively, in the UC-MSC group and by 23.6±12.4, 1.2±0.58, and 1.2±0.35 mg/day, respectively, in the control group (p<0.01, p<0.05, and p<0.05 for UC-MSC vs control, respectively). Four patients developed a fever after cell infusion. No serious adverse events were observed. Conclusions: UC-MSCs were effective in the treatment of CD and produced mild side effects. • Golgi Phosphoprotein 2 Is a Novel Regulator of IL-12 Production and Macrophage Polarization

Zhang, Wei,Kim, Hajeong,Lv, Jiyang,Zhao, Na,Ma, Xiaojing The American Association of Immunologists, Inc. 2018 JOURNAL OF IMMUNOLOGY Vol.200 No.4

<P>Golgi phosphoprotein 2 (GOLPH2), a widely expressed Golgi type II transmembrane protein, has been implicated in several important physiological and pathological processes, including virus infections, cancer cell proliferation, and metastasis. However, its biological functions and mechanisms, particularly in the immune system, remain highly obscure. In this study, we report the biochemical identification of GOLPH2 from B cell lymphoma culture supernatant and show that the secreted protein could inhibit IL-12 production by dendritic cells (DCs) and IL-12–induced IFN-γ production by activated T cells. Further molecular analysis revealed that GOLPH2’s IL-12–inhibiting activity was mediated through a proximal <I>IL12p35</I> promoter element involving a previously identified transcriptional repressor named GC-binding protein that is induced during phagocytosis of apoptotic cells by macrophages. We subsequently generated global <I>golph2</I> knockout mice, which exhibited little developmental abnormality but were more susceptible to LPS-induced endotoxic shock than were wild-type mice with elevated serum IL-12 levels. Furthermore, we found that GOLPH2 played a regulatory role in macrophage polarization toward the M2 type. A comprehensive analysis of gene expression profiles of activated wild-type and GOLPH2-deficient DCs by RNA sequencing uncovered mechanistic insights into the way GOLPH2 potentially modulates DC function during inflammatory insults. Our functional study of GOLPH2 helps advance the scientific understanding of the biological and pathogenic roles of this novel and intriguing molecule with great potential as a diagnostic and prognostic marker as well as a therapeutic target in many acute and chronic inflammatory disorders.</P>

Effects of different working fluids on the performance of a radial turbine in an organic Rankine cycle power system

        Ze-min Bo,Zhenkun Sang,Xiaojing Lv,Yi-wu Weng 대한기계학회 2018 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.32 No.9

Organic Rankine cycle (ORC) power systems are widely used for low-temperature heat recovery. The radial turbine is the most important component in the ORC system and its performance is significantly dependent on the working fluid. Therefore, the effect of different working fluid on the performance of radial turbine in ORC system for the low-temperature heat source (120~180 °C) was investigated. First, the feasibilities of different working fluids for 150 kW radial turbine using R600a as designed working fluid were investigated. The thermal aerodynamic calculation of R600a at design condition was conducted with evaporation temperature 95 °C and condensation temperature 38 °C. The operation parameters of 5 non-designed working fluids (R600, R245fa, R245ca, R123 and R601a) were selected in the same temperature variation range as R600a. Then, the performance of a radial turbine for six working fluids at design condition was analyzed, and their three-dimensional flow field performances were also obtained by CFD numerical simulation. Finally, to broaden the operation range of the radial turbine and investigate the matching relationship between operation parameters and different working fluids, the off-design performances of radial turbine using six working fluids were analyzed and the effect of rotation speed, inlet pressure and inlet temperature on the performance of radial turbine was obtained. Results show that the performance of the radial turbine using the designed working fluid R600a is the best with efficiency of 82.7 % and output power of 150.5 kW. For the non-designed working fluids, the comprehensive performance of R600 was also judged to be good owing to the highest efficiency (85.1 %) and a relatively high power output (115.8 kW) among the fluids tested. For other working fluids, the output power of the radial turbine decreased obviously. There exists an optimum rotation speed for different working fluids to ensure that the radial turbine has the highest efficiency. The inlet pressure has a larger effect on the performance of the radial turbine than that of the inlet temperature. The results can provide basic data for the optimization design and operation of radial turbines in the ORC system.

Antifreeze protein detection using Rhodamine B as photoluminescence label in porous silicon

        Hongyan Zhang,Zhenhong Jia,Xiaoyi Lv,Junwei Hou,Xiaojing Liu,Ji Ma,Jun Zhou 한국물리학회 2013 Current Applied Physics Vol.13 No.4

A novel method is demonstrated to detect Antifreeze proteins (AFPs) based on photoluminescence (PL)using porous silicon (PS) coated with silver as a substrate. Ag/PS substrate is obtained through immersion of PS in silver nitrate (AgNO3) solutions and is incubated with Rhodamine B (RB) as PL label. This substrate is easy to be fabricated and the pore size of PS is large enough for biological molecules to infiltrate, which is an ideal platform for biological molecule detection. Through functionalization used glutaraldehyde (GTA) and 4-(N-Maleimidomethyl) cyclohexane-1-carboxylicacid (Sulfo-SMCC) as crosslinkers separately, we test the role of the AFPs antibodies in selective capturing the AFPs antigen and explain the reason of the enhancement of PL intensity. The result shows a significant enhancement of the PL intensity of RB at around 590 nm due to the interaction of antibodyeantigen competitive binding with AFPs. Therefore, the PL corresponding to RB was selected to detect the target AFPs and the PL intensity of RB proportional to the AFPs concentration. The detection limit was found to be 1.65 mg/ml for AFPs when GTA was used as cross-linker, and the detection limit was 16.5 ng/ml with Sulfo-SMCC as cross-linker

Effects of a chitosan nanoparticles encapsulation on the properties of litchi polyphenols

        Xingan Cheng,Qiwen Zou,Hanhui Zhang,Jianwei Zhu,Murtaza Hasan,Fangyun Dong,Xin Liu,Junjie Li,Yuehua Wu,Xiaojing Lv,Keqiang Wang,Xiangling Deng,Zhanmei Liu,Xuhong Jiang 한국식품과학회 2023 Food Science and Biotechnology Vol.32 No.13

Litchi polyphenols have very specific biological activities. Nevertheless, the low and inconsistent oral bioavailability and instability hinder the further application of litchi polyphenols in food systems. This work prepared litchi polyphenols loaded chitosan nanoparticles (LP-CSNPs) by ionic gelation method to enhance the encapsulation on the properties of litchi polyphenols. The optimum conditions of formation via single factors and the Box–Behnken design were chitosan (CS) concentration 1.065 mg/mL, sodium tripolyphosphate (TPP) concentration 0.975 mg/mL, and the mass ratios of polyphenols and CS 1:1 with encapsulation efficiency (EE%) of 45.53%. LP-CSNPs presented the nanosized range of particle size (mean 170 nm), excellent polydispersity index (PDI) (0.156 ± 0.025), and zeta potential values (+ 35.44 ± 0.59). The in vitro release in simulated gastric fluid (pH 1.2) and intestinal fluid (pH 6.8) during 100 h was 58.34% and 81.68%, respectively. LP-CSNPs could effectively improve the storage stability and had great antibacterial activity compared with unencapsulated litchi polyphenols.
