        Effects of sweetener sucralose on diet preference, growth performance and hematological and biochemical parameters of weaned piglets

        Objective: Two experiments were conducted to investigate the effects of dietary sucralose on diet preference and growth performance of weaned piglets, and a third experiment was a 28-d safety study to examine if high-dose sucralose could affect the health state of weaned piglets. Methods: In experiment one, 48 piglets had free access to a corn-soybean based diet and the same diet supplemented with 150 mg/kg sucralose for 15 d. In experiment two, 180 piglets were blocked into 5 treatments with 6 replications. They were fed basal diets supplemented with 0, 75, 150, 225, and 300 mg/kg sucralose for 28 days. In experiment three, 108 piglets were randomly assigned to 3 treatments and fed diets supplemented with 0, 150 (suitable level), and 1,500 (ten-fold suitable level) mg/kg sucralose for 28 d. Results: The experiment 1 showed that piglets preferred (p<0.05) diets containing sucralose during experimental period. In experiment 2, piglets fed a diet supplemented with 150 mg/kg sucralose had a higher average daily gain (ADG) and average daily feed intake (ADFI) than pigs in the control group and other treatment groups during the experiment period. The concentrations of sucralose over 150 mg/kg may decrease feed intake. However, no difference in feed conversion ratio was observed. In experiment 3, piglets fed diet supplemented with 150 mg/kg sucralose had a higher ADG and ADFI than that of pigs in the control group and 1,500 mg/kg treatment groups during the experiment period. Clinical blood metabolites, organ index and histological morphology were not significantly different between sucralose treatments. Conclusion: Sucralose can promote feed intake and thereby improve growth performance of weaned piglets. Moreover, inclusion of 1,500 mg/kg sucralose was demonstrated to have no observed adverse effects. Supplementing 150 mg/kg sucralose for weaned piglets is recommended in this study.

        The genes involved in the protective effects of phytohormones in response to Verticillium dahliae infection in Gossypium hirsutum

        Verticillium wilt disease caused by soil-borne fungus Verticillium dahliae (V. dahliae), is a serious threats to cotton growth worldwide. Endogenous phytohormones, such as salicylic acid (SA), jasmonic acid (JA), ethylene (ET), and brassinosteroid (BR) have been reported to play important roles in defense response in plants. Based on highthroughput RNA sequencing analysis of resistant Gossypium hirsutum (G. hirsutum) variety Zhongzhimian KV-3, comparing the transcriptome with V. dahliae infection and non-infection, eight differentially expressed candidate genes functions indicated in SA (GhPUB17, GhTGA7 and GhPR1), JA (GhJAZ10 and GhbHLH18), ET (GhEBF1), cytokinine (GhE13L13) and BR (GhBZR1) signal pathways were investigated via virus-induced gene silencing (VIGS) using cotton leaf crumple virus (CLCrV) vector. The expression of eight genes was further validated through quantitative realtime PCR. The result showed that the expression level of GhPUB17, GhJAZ10, GhbHLH18, GhEBF1 and GhE13L13 increased faster in resistant variety Zhongzhimian KV-3 than in susceptible variety 86-1 upon V. dahliae infection. Silencing up-regulated genes GhPUB17, GhJAZ10, GhbHLH18 and GhEBF1 significantly enhanced resistant variety susceptibility to V. dahliae and silencing down-regulated genes GhTGA7 and GhBZR1 significantly increased susceptible variety resistance to V. dahliae. Taken together, these genes are important components in response to V. dahliae infection.

        INTEGRATED STABILITY CONTROL STRATEGY OF IN-WHEEL MOTOR DRIVEN ELECTRIC BUS

        Taking the independently actuated in-wheel motor driven electric bus as the research object, an integrated control strategy of yaw stability and roll stability is proposed. On the basis of the upper and lower layered structure, the stability monitoring layer is added to the control strategy, which is responsible for calculating the desired value of the vehicle\'s critical state and determining the vehicle stability control mode. The upper controller is the yaw moment decision layer. To improve the robustness, the sliding mode control is used to calculate the additional yaw moment required for yaw and roll stability, and the Lyapunov method is used to prove the convergence of the algorithm. The lower controller is the torque distribution layer, which uses the active-set algorithm and combines the motor torque and the braking torque of the braking system to optimally distribute the yaw moment. The proposed control strategy is verified through the co-simulation platform of Trucksim and Simulink. The results show that the control strategy can coordinate the various control modes well, improve the tracking ability of the vehicle, and reduce the risk of side slip and rollover of the vehicle.

        Numerical and experimental buckling of segmented spiral pressure hulls

        To optimally coordinate the operational safety, manoeuvrability, positioning reliability, and space expansibility of deep-sea stations, this study proposed a segmented spiral pressure hull with reference to the geometry of worm snails. The design addresses the disadvantages of existing ring-stiffened cylinder, segmented sphere, and circular toroid structures. This study numerically and experimentally examined the buckling performance of segmented spiral pressure hulls. The numerical, experimental, and analytical data exhibited satisfactory agreement. The proposed pressure hulls exhibited a favourable space expansibility and buckled in a nonlinear manner. The buckling loads varied slightly after the threshold number of segments. All the post buckling modes were local dents. The current findings may serve as a reference for the research and development of original deep-sea stations.

        Efficient photocatalytic degradation of 2-chloro-4,6-dinitroresorcinol in salty industrial wastewater using glass-supported TiO2

        2-chloro-4,6-dinitroresorcinol (CDNR) is detrimental to the environment and human health owing to its high toxicity and poor biodegradability. To demonstrate the feasibility of photocatalytic degradation of CDNR from industrial salty wastewater by borosilicate glass supported TiO2 under UV light irradiation, borosilicate glass supported TiO2 was prepared successfully by a novel sol-gel route via dip-coating method and characterized by XRD, SEM, FTIR and XPS analysis. The results showed that TiO2 catalyst has the anatase phase structure with crystallite size of 11.5 nm and coats uniformly on the borosilicate glass. Also, the effects of reaction time, pH value, TiO2 dosage, CDNR concentration, and Cl on the degradation efficiency of CDNR were investigated. The results indicated that at pH 2, reaction time 3.5 h, CDNR concentration 10mg/L, NaCl concentration 5.85% (w/w) and TiO2 dosage 1.0 g/L, 97.7% of CDNR was degraded in the presence of Cl, this corresponded to a rate constant of 1.05 h1, illustrating the feasibility of photocatalytic degradation process. This contribution provides a basic investigation regarding the potential application of borosilicate glass supported TiO2.

        Molecular genetic characterization of rice seed lipoxygenase 3 and assessment of its effects on seed longevity

        Lipoxygenases (LOXs) are enzymes involved in lipid peroxidation. Here we reported the identification, molecular and functional characterization of the gene encoding rice (Oryza sativa L.) seed LOX3 (sLOX3). Via a map-based cloning strategy we identified Os03g0700400 as the candidate gene encoding sLOX3. Further functional complementary test and biochemical characterization of the recombinant Os03g0700400 protein verified the identification. The sLOX3 gene was highly expressed in roots, moderately in embryos and very weakly in leaves, leaf sheaths and stems. Transient expression experiment (in rice protoplasts) and subsequent laser confocal microscopic analysis demonstrated that the sLOX3 protein was localized into the cytosol. We next showed that overexpression of sLOX3 in a japonica sLOX3-normal rice cultivar, Wuyunjing 7 accelerated the decrease of seed germination ability when the seeds were routinely stored, which demonstrated that sLOX3 had a negative effect on seed longevity (storability). Meanwhile, an increased occurrence of embryo decay was observed in the same transgenic seeds, suggesting that sLOX3 might negatively affect seed longevity by facilitating colonization of particular seed pathogens. Our result forwarded the understanding of the effects of 9-LOX on rice seed longevity.

        Dynamic QTL Analysis of Rice Protein Content and Protein Index Using Recombinant Inbred Lines

        Protein content (PC) and protein index (PI) play important roles in determining nutritional quality in rice (Oryza sativa L.). We used 71 lines derived from “Asominori/IR24” to analyze the developmental behavior of PC and PI through unconditional and conditional QTL mapping methods. In all, 10 unconditional QTLs and 6conditional QTLs for PC, and 11 unconditional QTLs and 9conditional QTLs for PI, were identified at four stages of grain filling. More were identified in the first three stages than at the final stage. Temporal patterns of gene expression for PC and PI differed over time, with several QTLs being expressed across two or three stages but many being expressed at only one stage. Some of these QTLs were closely linked with maturity QTLs reported previously. Many QTLs for PC and PI were co-localized, supporting the significant correlation found between PC and PI. Our results suggest that dynamic QTL mapping might be a valid means for revealing more genetic information about protein accumulations during seed development.

        A Critical Role of OsMADS1 in the Development of the Body of the Palea in Rice

        OsMADS1 plays crucial roles in the specification of floral organ identity and the determinacy of floral meristem in rice (Oryza sativa). Here we isolated a rice mutant cy15 with defective spikelets. Map-based cloning identified cy15 as a novel mutation in the C terminus of OsMADS1. Severe cy15 spikelets contained normal inner whorl organs but defective paleas, opposite to four abnormal whorls of Osmads1 flowers reported previously. In the cy15 spikelets, the size of the body was remarkably reduced in paleas, indicating a critical role of OsMADS1 in its differentiation. Moreover, the development of palea was impaired in the early stage of the cy15 spikelets. The sub-subcellular location of the mutant OsMADS1 was also affected in cy15. On the other hand, the truncated C terminus of the mutant OsMADS1 from cy15 still showed a significant transcriptional activator function. Meanwhile, though the mutant OsMADS1 retained intact M, I and K domains, its interactions with two highly homologous A-class proteins, OsMADS15 or OsMADS14, were weakened or abolished. Our data suggested that the C-terminal sequence of OsMADS1 might be required for maintaining normal morphology of spikelets in rice.

      • Residual Load Sway Reduction Control in Double-pendulum Overhead Cranes Using Motion Trajectory Based on Adams-Matlab Joint Simulation

        When the hook mass is greater than the load mass or the load has a distributed mass property, the load sway of the crane system can be considered as a double-pendulum. At this point, the crane system has two kinds of natural angular frequency which greatly increases the difficulty of the dynamic analysis and the controller design. To this end, the linear dynamics of a 2-D overhead crane with double-pendulum effect is derived based on a disturbance observer and is decoupled for controller design by modal analysis. Second, an S-shaped curve, which is widely used in the control fields, is applied to the linearized decoupling crane model. The trajectory parameters are obtained by solving the algebraic equation. Finally, the validity of the method is verified by simulation results.
