        A general closed-form solution to a Timoshenko beam on elastic foundation under moving harmonic line load

        In this paper, a general closed-form solution for evaluating the dynamic behavior of a Timoshenko beam on elastic foundation under a moving harmonic line load is formulated in the frequency-wavenumber domain and in a moving coordinate system. It is found that the characteristic equation is quartic with real coefficients only, and its poles can be presented explicitly. This enables the substitution of these poles into Cauchy’s residue theorem, leading to the general closed-form solution. The solution can be reduced to seven existing closed-form solutions to different sub-problems and a new closed-form solution to the subproblem of a Timoshenko beam on an elastic foundation subjected to a moving quasi-static line load. Two examples are included to verify the solution.

      • Vibration-based delamination detection of composites using modal data and experience-based learning algorithm

        In this paper, a vibration-based method using the change ratios of modal data and the experience-based learning algorithm is presented for quantifying the position, size, and interface layer of delamination in laminated composites. Three types of objective functions are examined and compared, including the ones using frequency changes only, mode shape changes only, and their combination. A fine three-dimensional FE model with constraint equations is utilized to extract modal data. A series of numerical experiments is carried out on an eight-layer quasi-isotropic symmetric (0/-45/45/90)s composited beam for investigating the influence of the objective function, the number of modal data, the noise level, and the optimization algorithms. Numerical results confirm that the frequency-and-mode-shape-changes-based technique yields excellent results in all the three delamination variables of the composites and the addition of mode shape information greatly improves the accuracy of interface layer prediction. Moreover, the EBL outperforms the other three state-of-the-art optimization algorithms for vibration-based delamination detection of composites. A laboratory test on six CFRP beams validates the frequency-and-mode-shape-changesbased technique and confirms again its superiority for delamination detection of composites.

        Nonlinear identification of Bouc–Wen hysteretic parameters using improved experience-based learning algorithm

        In this paper, an improved experience-based learning algorithm (EBL), termed as IEBL, is proposed to solve the nonlinear hysteretic parameter identification problem with Bouc-Wen model. A quasi-opposition-based learning mechanism and new updating equations are introduced to improve both the exploration and exploitation abilities of the algorithm. Numerical studies on a single-degree-of-freedom system without/with viscous damping are conducted to investigate the efficiency and robustness of the proposed algorithm. A laboratory test of seven lead-filled steel tube dampers is presented and their hysteretic parameters are also successfully identified with normalized mean square error values less than 2.97%. Both numerical and laboratory results confirm that, in comparison with EBL, CMFOA, SSA, and Jaya, the IEBL is superior in nonlinear hysteretic parameter identification in terms of convergence and accuracy even under measurement noise.

      • Anti-metastasis Activity of Black Rice Anthocyanins Against Breast Cancer: Analyses Using an ErbB2 Positive Breast Cancer Cell Line and Tumoral Xenograft Model

        Background: Increasing evidence from animal, epidemiological and clinical investigations suggest that dietary anthocyanins have potential to prevent chronic diseases, including cancers. It is also noteworthy that human epidermal growth factor receptor 2 (ErbB2) protein overexpression or ErbB2 gene amplification has been included as an indicator for metastasis and higher risk of recurrence for breast cancer. Materials and Methods: The present experiments investigated the anti-metastasis effects of black rice anthocyanins (BRACs) on ErbB2 positive breast cancer cells in vivo and in vitro. Results: Oral administration of BRACs (150 mg/kg/day) reduced transplanted tumor growth, inhibited pulmonary metastasis, and decreased lung tumor nodules in BALB/c nude mice bearing ErbB2 positive breast cancer cell MDA-MB-453 xenografts. The capacity for migration, adhesion, motility and invasion was also inhibited by BRACs in MDA-MB-453 cells in a concentration dependent manner, accompanied by decreased activity of a transfer promoting factor, urokinase-type plasminogen activator (u-PA). Conclusions: Together, our results indicated that BRACs possess anti-metastasis potential against ErbB2 positive human breast cancer cells in vivo and in vitro through inhibition of metastasis promoting molecules.

        Gene microarray analysis revealed a potential crucial gene RACK1 in oral squamous cell carcinoma (OSCC)

        Oral squamous cell carcinoma (OSCC) is the sixth most common cancer worldwide, which appears as a consequence of multiple molecular genetic events in various chromosomes and genes. In order to unveil the possible mechanisms underlying OSCC tumorigenesis, the OSCC-related gene expression variance and the gene interaction network should be further investigated. Herein, we conducted the NimbleGen Human Gene Expression Microarray to analyze expression heterogeneity between OSCC primary tumor tissue and its adjacent normal tissue from two patients. A total number of 7872 out of 32,448 detected genes are differentially expressed in OSCC. Gene ontology (GO) analysis demonstrated that these differentially expressed transcripts were critical in a series of metabolic processes, cancer-related signal pathways, and biological regulations. KEGG signaling pathway enrichment suggested a number of pathways (metabolic process and immune response) which are frequently enrolled during cancer progression. 15 most differential regulated genes between OSCC tumor and non-tumor were confirmed by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Furthermore, the interaction network analysis of these confirmed genes by STRING database showed the two subunits of RACK1 had direct interaction with 14 differential proteins. This bioinformatics research lends support about the critical role of RACK1 which functions as a key node protein driving OSCC development.

        Analysis the role of arabidopsis CKRC6/ASA1 in auxin and cytokinin biosynthesis

        The crosstalk between auxin and cytokinin (CK) is important for plant growth and development, although the underlying molecular mechanisms remain unclear. Here, we describe the isolation and characterization of a mutant of Arabidopsis Cytokinin-induced Root Curling 6 (CKRC6), an allele of ANTHRANILATE SYNTHASE ALPHA SUBUNIT 1 (ASA1) that encodes the á-subunit of AS in tryptophan (Trp) biosynthesis. The ckrc6 mutant exhibits root gravitropic defects and insensitivity to both CK and the ethylene precursor 1-aminocyclopropane-1-carboxylicacid (ACC) in primary root growth. These defects can be rescued by exogenous indole-3-acetic acid (IAA) or tryptophan (Trp) supplementation. Furthermore, our results suggest that the ckrc6 mutant has decreased IAA content, differential expression patterns of auxin biosynthesis genes and CK biosynthesis isopentenyl transferase (IPT) genes in comparison to wild type. Collectively, our study shows that auxin controls CK biosynthesis based on that CK sensitivity is altered in most auxin-resistant mutants and that CKs promote auxin biosynthesis but inhibit auxin transport and response. Our results also suggest that CKRC6/ASA1 may be located at an intersection of auxin, CK and ethylene metabolism and/or signaling.

      • Analysis of Indoleamine 2-3 Dioxygenase (IDO) and EGFR Co-expression in Breast Cancer Tissue by Immunohistochemistry

        Background: To determine the amount of co-expression of IDO and EGFR in breast cancer patients. Materials and Methods:In order to obtain the distribution of co-expression of IDO and EGFR in breast cancer, we tested 110 breast cancer paraffin tissue blocks with immunohistochemical methods. Then we investigated the relationship between the diagnostic and pathologic characteristics (tumor size, lymph node status, histologic grade, the gene expression of ER, PR, HER2, p53, Ki67 and PCNA) with the situation of co-expression of IDO and EGFR by reviewing the medical records of 32 breast cancer patients. Results: Among 110 breast cancers, 32 cases demonstrated IDO and EGFR co-expression (29.1%), IDO and EGFR synchronous co-expression being found in 19.1% and asynchronous in 10.0%. Conclusions: IDO and EGFR were co-expressed in breast cancer, including synchronous and asynchronous co-expression. The results suggest that considering IDO and EGFR as two indicators for breast cancer treatment or prognosis analysis provides a potential option of individual treatment for the portion of breast cancer patients with co-expression of IDO and EGFR.

        SAMD4B, a novel SAM-containing protein, inhibits AP-1-, p53- and p21-mediated transcriptional activity

        The sterile alpha motif (SAM) is a putative protein interaction domain involved in a wide variety of biological processes. Here we report the identification and characterization of a novel gene, SAMD4B, which encodes a putative protein of 694 amino acids with a SAM domain. Northern blot and RT-PCR analysis showed that SAMD4B is widely expressed in human embryonic and adult tissues. Transcriptional activity assays show SAMD4B suppresses transcriptional activity of L8G5-luciferase. Over-expression of SAMD4B in mammalian cells inhibited the transcriptional activities of activator protein-1 (AP-1), p53 and p21, and the inhibitory effects can be relieved by siRNA. Deletion analysis indicates that the SAM domain is the main region for transcriptional suppression. The results suggest that SAMD4B is a widely expressed gene involved in AP-1-, p53-and p21-mediated transcriptional signaling activity. [BMB reports 2010; 43(5): 355-361]

        Leaf Wilting Movement Can Protect Water-Stressed Cotton (Gossypium hirsutum L.) Plants Against Photoinhibition of Photosynthesis and Maintain Carbon Assimilation in the Field

        Under severe water stress, leaf wilting is quite general in higher plants. This passive movement can reduce the energy load on a leaf. This paper reports an experimental test of the hypothesis that leaf wilting movement has a protective function that mitigates against photoinhibition of photosynthesis in the field. The experiments exposed cotton (Gossypium hirsutum L.) to two water regimes: waterstressed and well-watered. Leaf wilting movement occurred in water-stressed plants as the water potential decreased to −4.1 MPa, reducing light interception but maintaining comparable quantum yields of photosystem II (PS II; Yield for short) and the proportion of total PS II centers that were open (qP). Predrawn Fv/Fm (potential quantum yield of PS II) as an indicator of overnight recovery of PS II from photoinhibition was higher than or similar to that in wellwatered plants. Compared with water-stressed cotton leaves for which wilting movement was permitted, water-stressed cotton leaves restrained from such movement had significantly increased leaf temperature and instantaneous CO2assimilation rates in the short term, but reduced Yield, qP,and Fv/Fm. In the long term, predrawn Fv/Fm and CO2assimilation capacity were reduced in water-stressed leaves restrained from wilting movement. These results suggest that, under water stress, leaf wilting movement could reduce the incident light on leaves and their heat load, alleviate damage to the photosynthetic apparatus due to photoinhibition,and maintain considerable carbon assimilation capacity in the long term despite a partial loss of instantaneous carbon assimilation in the short term.
