Purpose: the study aimed to compare the quality of life (QOL) and radiotherapy complications among Chinese nasopharyngeal carcinoma (NPC) patients at different 3-dimensional conformal radiotherapy (3DCRT) stages adjusting for other variables. Methods: 511 NPC patients at different 3DCRT stages were enrolled. They were interviewed regarding SF-36, complications and socio-demographic variables and cancer- or treatment-related variables. Analysis of covariance (ANCOVA) based on SF-36, complications scores as dependent variables, 3DCRT stages as independent variables, and other variables as covariate were established. Results: The influencing factors of PCS included 3DCRT stages and age group. The influencing factors of MCS included 3DCRT stages and income. Most QOL scores of NPC patients were significantly associated with 3DCRT stage, after accounting for other variables. QOL scores of the patients receiving 3DCRT were the lowest, QOL scores of people after 3DCRT gradually increased. PCS scores of people greater than 5 years after 3DCRT was improved to or even better than the level before 3DCRT. The complications with significantly different scores of patients at different 3DCRT status included xerostomia, throat ache, hypogeusia, caries, hearing loss, snuffles. Conclusions: Clinicians should pay more attention to older NPC patients and patients with lower income. When patients receive 3DCRT, measures should be taken to reduce radiation injury to improve the patients' QOL.
INVESTIGATION OF OCCUPANT LOWER EXTREMITY INJURES UNDER VARIOUS OVERLAP FRONTAL CRASHES

Objective: With widely usage of restraint system, fatal injuries to occupants have been largely limited while non-fatal lower extremity injuries have not been effectively improved. The present study aims to investigate occupant lower extremity injuries under realistic impact environments. Methods: A biofidelic lower extremity model, a dummy model and a car cab model were combined to set up a realistic impact environment. Three typical frontal impact groups were simulated. Occupant global lower kinematics, long bone axial force and bending moment were presented to in-depth investigate lower extremity injury mechanism and tolerance. Results: Various overlap frontal impacts cause totally different lower extremity kinematics in the combination of structural invasion and restraint system effects. The femur fracture occurred at a small axial force of 7.57 kN combing a substantial bending moment peak of 172 Nm. Ankle joint injuries were found in 100 % and 25 % overlap impacts that present large tibial axial force and joint rotation angle. Conclusions: Overall results indicate that a coupling threshold of femur axial force and bending moment is of rationality to predict global femur fracture. The ankle joint injury occurrence is significantly related to the coupling effects of tibia axial force and excessive self-kinematics.

A total of 29 simple sequence repeat (SSR) markers were used to analyze the genetic diversity of 150 accessions of cultivated rice (Oryza sativa L.) from Korea, China, and Japan. A total of 375 alleles were detected with an average of 12.9 per locus. The averaged values of gene diversity and polymorphism information content (PIC) for each SSR locus were 0.7001 and 0.6683, respectively. Alleles per locus in Korean rice were 8.8, whereas 8.1 and 7.2 alleles per locus were found in Chinese and Japanese rice, respectively. The mean gene diversity in Korean, Chinese, and Japanese rice was 0.6058, 0.6457, and 0.5174, respectively, whereas the mean PIC values for each SSR locus were 0.5759, 0.6138, and 0.4881, respectively. The genetic diversity of the Korean and Chinese cultivars was higher than that of the Japanese cultivars, and the genetic diversity of japonica was higher than that of indica. The model-based structure analysis revealed the presence of three subpopulations, which was basically consistent with clustering based on genetic distance. An AMOVA analysis showed that the between-population component of genetic variance was less than 22% in contrast to 78% for the within-population component. The overall F(ST) value was 0.2180, indicating a moderate differentiation among groups. The results could be used for designing effective breeding programs aimed at broadening the genetic bases of commercially grown varieties.

Vegetable insulating oils, with higher fire point, lower biological hazard and better availability compared to traditional mineral insulating oils, have enormous potential to become the substitute of the latter, given the trend of green development of the power equipment, for example, the transformers. However, the obviously different constitution of the vegetable oils makes the circumstance hard when considering their application on the insulation for actual transformers. One of the significant deficiencies of the vegetable oils in working condition is their insufficient lightning impulse endurance under partial high electric fields and large discharge gaps. In this article, three kinds of transformer insulating oils, rapeseed oil, FR3 and 25# mineral oil were chosen to investigate the performances of these insulation materials under long-gap lightning impulses with positive and negative polarity respectively. The variation rules of breakdown voltage and velocity with the gap length were obtained. And the difference of the lightning impulse voltage endurance between the vegetable and mineral insulating oils as well as its causes based on theories of streamer and ionization potential (IP) were analyzed, which led to the conclusion that the gap increase causes more threat to the positive lightning impulse breakdown of vegetable insulating oils, compared to its counterpart.
Geft is dispensable for the development of the second heart field

Geft is a guanine nucleotide exchange factor, which can specifically activate Rho family of small GTPase by catalyzing the exchange of bound GDP for GTP. Geft is highly expressed in the excitable tissue as heart and skeletal muscle and plays important roles in many cellular processes, such as cell proliferation, migration, and cell fate decision. However, the in vivo role of Geft remains unknown. Here, we generated a Geft conditional knockout mouse by flanking exons 5-17 of Geft with loxP sites. Cre-mediated deletion of the Geft gene in heart using Mef2c-Cre transgenic mice resulted in a dramatic decrease of Geft expression. Geft knockout mice develop normally and exhibit no discernable phenotype, suggesting Geft is dispensable for the development of the second heart field in mouse. The Geft conditional knockout mouse will be a valuable genetic tool for uncovering the in vivo roles of Geft during development and in adult homeostasis. (BMB reports 2012; 45(3): 153-158)

Aims: ONYX-II is a phase 3, open-label study of 3-DAA regimen of OBV/PTV/r and DSV with RBV in treatment-naive and experienced patients with genotype 1b HCV infection and compensated cirrhosis in China, South Korea and Taiwan. SVR12 rate was 100% and the favourable safety profile was shown. The present analysis reports efficacy( SVR24) and safety results. Methods: Patients with chronic GT1b HCV infection and compensated cirrhosis received OBV/PTV/r + DSV + RBV for 12 weeks and will be followed for 48 weeks post-treatment. Efficacy was assessed by SVR12 and SVR24. Safety was assessed as the percentage of patients wi th treatment-emergent adverse events (TEAEs) and laboratory evaluation. Results: Total of 104 patients with chronic GT1b HCV infection (62% female, 100% Asian, 58% treatment-experienced) were enrolled from China (n=63), South Korea (n=21) and Taiwan (n=20). All patients received at least one dose of study drugs. The SVR24 rate was 100% (concordant with SVR12), with no patient relapsing between post-treatment week 12 and 24. Most TEAEs were mild in severity. The most common TEAEs (≥10%) were increased blood bilirubin levels (25%), pruritus (15%), anaemia (14%), asthenia (12%), bilirubin conjugated increased (12%), blood bilirubin unconjugated increased (12%), dizziness (11%) and fatigue (11%). Four patients had serious AEs and all were assessed as not being related to the 3-DAA regimen (one was assessed as being possibly related to RBV). One patient discontinued treatment due to TEAEs (elevations in alanine aminotransferase [ALT], aspartate aminotransferase [AST] and blood bilirubin) after 3 weeks of dosing but achieved SVR12 and SVR24. Laboratory abnormalities ≥ grade 3 were infrequent (ALT: 3%; AST 2%; total bilirubin: 7%). No grade 3 haemoglobin decrease was reported. Conclusions: SVR24 and SVR12 rates were concordant (100%) in HCV GT1b-infected Asian patients with compensated cirrhosis. The regimen was generally well tolerated with mostly mild TEAEs reported.

Aims: The combination of daclatasvir (DCV) plus asunaprevir (ASV)has demonstrated high sustained virologic response (SVR) rates andis generally well tolerated in clinical studies. This integrated analysisevaluated the safety profile of DCV (60mg once daily) and ASV (100mgsoftgel capsule or 200mg tablets twice daily in genotype 1b (GT1b)infected patients enrolled in four phase 3 and two phase 2 clinicalstudies conducted globally, including Asia.Methods: Integrated safety data from 1218 treatment-naive or treatment-experienced patients were analyzed for adverse events (AEs),serious AEs, discontinuations due to AEs and grade 3/4 AEs andlaboratory abnormalities reported on-treatment.Results: Patients were 58% female, median age was 58 years and23% had compensated cirrhosis. DCV+ASV was associated with infrequentserious AEs and discontinuations due to AEs (Table). Twelvepatients reported treatment-related serious AEs. The most commonAEs (any grade) were diarrhea, nausea, fatigue, and headache. Onepatient died due to coronary heart disease (not treatment-related).The most common grade 3/4 laboratory abnormalities were aminotransferaseelevations (more frequent among Japanese patients); however,all grade 3/4 laboratory abnormality occurred in <5% of patientsoverall. Grade 3/4 total bilirubin elevations were reported in <1%of patients. The DCV+ASV safety profile was similar in patients withor without cirrhosis.Conclusions: DCV+ASV was generally well tolerated across globalnon-Asian patient populations and in Asian patients from Japan, mainlandChina, Korea, and Taiwan.
Cloning and characterization of the cardiac-specific Lrrc10 promoter

Leucine-rich repeat containing protein 10 (LRRC10) is characterized as a cardiac-specific gene, suggesting a role in heart development and disease. A severe cardiac morphogenic defect in zebrafish morphants was recently reported but a contradictory result was found in mice, suggesting a more complicated molecular mechanism exists during mouse embryonic development. To elucidate how LRRC10 is regulated, we analyzed the 5`enhancer region approximately 3 kilo bases (kb) upstream of the Lrrc10 start site using luciferase reporter gene assays. Our characterization of the Lrrc10 promoter indicates it possesses complicated cis-and trans-acting elements. We show that GATA4 and MEF2C could both increase transcriptional activity of Lrrc10 promoter individually but that they do not act synergistically, suggesting that there exists a more complex regulation pattern. Surprisingly, knockout of Gata4 and Mef2c binding sites in the 5`enhancer region (-2,894/-2,889) didn`t change the transcriptional activity of the Lrrc10 promoter and the likely GATA4 binding site identified was located in a region only 100 base pair (bp) upstream of the promoter. Our data provides insight into the molecular regulation of Lrrc10 expression, which probably also contributes to its tissue-specific expression. [BMB reports 2011; 44(2): 123-128]
