        Nodule Rich Protein 2 modulates nodule number in Medicago truncatula

        Symbiotic nitrogen fxation is beneft to sustainable agriculture and global nitrogen cycle. Many small peptides were identifed as regulators involving in the interaction between rhizobia and legume. Here we reported Nodule Rich Protein 2 (MtNRP2) encoding a small peptide in Medicago truncatula, belonged to a group of nodule rich protein restricted in legume species. MtNRP2 expressed highly in root nodule and its promoter was active during the initiation and development of root nodule and lateral root. To investigate the function of MtNRP2 in nodulation, we generated MtNRP2-overexpression and MtNRP2- knockdown transgenic Medicago. MtNRP2-overexpression transgenic lines performed normal nodulation phenotype compared with vector control. However, in the MtNRP2-RNAi transgenic plants, the decrease of MtNRP2 expression lead to the increase of infection threads number (7 day post inoculation) and nodules number (3 week post inoculation); meanwhile, the expression of MtRGF3 and MtPUB1 was inhibited. These results suggested that MtNRP2 negatively regulated nodulation in Medicago truncatula.

      • Effect of Baseline Resistance-associated Variants on SVR with the 3D Regimen with and without RBV in GT1a and GT1b-infected Patients

        Aims: The 3 direct-acting antiviral (3-DAA) regimen of ombitasvir, ritonavir-boosted paritaprevir and dasabuvir ± RBV is approved in the US and EU for treatment of hepatitis C virus (HCV) genotype (GT) 1 infection. Baseline resistance associated variants (RAVs) in HCV NS3 or NS5A can impact response to other DAA regimens; we assessed the prevalence and impact of RAVs on response to the 3-DAA regimen. Methods: Next-generation sequencing (Illumina MiSeq) assessed baseline samples from treatment-naive (PEARL-IV), -experienced (SAPPHIRE- II), or cirrhotic (TURQUOISE-II) GT1a patients who received 3-DAA + RBV, and treatment-experienced (PEARL-II) or cirrhotic (TURQUOISE-III) GT1b patients who received 3-DAA alone. Thresholds of 1 and 15%, respectively, detected the prevalence and impact of baseline RAVs; impact of RAVs conferring ≥ 5-fold resistance to components of the 3-DAA regimen on response was determined by com- paring SVR rates in patients with or without RAVs. Results: SVR rates were 96% and 100% in patients with GT1a and GT1b, respectively. One or more NS5A RAVs were present in 11% of treatment-experienced or cirrhotic GT1a patients, whereas NS5A RAVs were found in 19% of GT1b patients (15% threshold). Similar SVR rates were seen in GT1a patients with or without NS5A RAVs. All GT1b patients with NS5A RAVs, including at position Y93, achieved SVR. NS3 RAVs were uncommon (≤2%). NS3 RAVs were not seen in any of the 14 virologic failures and an NS5B RAV was seen in 1 virologic failure. The presence of the GT1a NS3 Q80K polymorphism had no impact on SVR. Conclusions: Understanding impact of baseline NS5A RAVs on treatment outcomes is important for relevant HCV therapies. Patients with HCV GT1a-infection treated with the 3-DAA regimen + RBV achieved high SVR rates, regardless of the presence of baseline RAVs. All GT1b patients treated with the 3-DAA regimen alone achieved SVR.

        LncRNA TMPO-AS1 promotes esophageal squamous cell carcinoma progression by forming biomolecular condensates with FUS and p300 to regulate TMPO transcription

        Esophageal squamous cell carcinoma (ESCC) is one of the most life- and health-threatening malignant diseases worldwide, especially in China. Long noncoding RNAs (lncRNAs) have emerged as important regulators of tumorigenesis and tumor progression. However, the roles and mechanisms of lncRNAs in ESCC require further exploration. Here, in combination with a small interfering RNA (siRNA) library targeting specific lncRNAs, we performed MTS and Transwell assays to screen functional lncRNAs that were overexpressed in ESCC. TMPO-AS1 expression was significantly upregulated in ESCC tumor samples, with higher TMPOAS1 expression positively correlated with shorter overall survival times. In vitro and in vivo functional experiments revealed that TMPO-AS1 promotes the proliferation and metastasis of ESCC cells. Mechanistically, TMPO-AS1 bound to fused in sarcoma (FUS) and recruited p300 to the TMPO promoter, forming biomolecular condensates in situ to activate TMPO transcription in cis by increasing the acetylation of histone H3 lysine 27 (H3K27ac). Targeting TMPO-AS1 led to impaired ESCC tumor growth in a patient-derived xenograft (PDX) model. We found that TMPO-AS1 is required for cell proliferation and metastasis in ESCC by promoting the expression of TMPO, and both TMPO-AS1 and TMPO might be potential biomarkers and therapeutic targets in ESCC.

        Examination of a Nurse-led Community-based Education and Coaching Intervention for Coronary Heart Disease High-risk Individuals in China

        Purpose: Early detection and management of coronary heart disease (CHD) are embedded into many community health service and primary care practices in western countries. The Framingham CHD risk score has been used to predict CHD and mortality for nearly 20 years, and it has predicted CHD event risk accurately in multiethnic populations. The aim of this study was to access the effect of a 6-month community-based intervention on CHD risk in individuals at high risk. Methods: A randomized controlled trial of individuals with a high 10-year CHD risk were recruited from two communities in China. Individuals in the intervention group (n ¼ 53) received a 3-month group education and a 3-month coaching session. Physical examination and self-report questionnaires were used to collect both pre- and postintervention data on blood pressure, glucose, cholesterol, body mass index, smoking, depression, and health-related quality of life (HRQoL). Results: A total of 102 participants (85.0%) completed the 6-month study. Compared with the usual care group, the intervention group had a 5 mmHg greater reduction in systolic blood pressure (t = 2.01, p = .047), larger declines in glucose (t = 2.49, p = .015), cholesterol (t = 2.44, p = .017), body mass index (t = 2.58, p = .011), and depression (t = 2.05, p = .043), and better reports of HRQoL (t = 3.36, p = .001). No significant group differences in smoking behaviors were reported. Conclusion: A 6-month community-based intervention in a CHD high-risk population improved diseaserelated risk factors, depression, and HRQoL. Results provide preliminary evidence for primary prevention of cardiovascular disease risk in a community high-risk population.

        Synthesis, Phase Behavior, and Simulated In vitro Degradation of Novel HTPB-b-PEG Polyurethane Copolymers

        Two types of polyurethanes with alternating and random block architectures, hydroxyl-terminated liquid polybutadiene and poly(ethylene glycol) block copolymers (HTPB-alt-PEG and HTPB-co-PEG), were synthesized using a coupling reaction route between the hydroxyl groups and the isocyanate groups. The chemical and crystal structures were characterized using Fourier transform-infrared spectroscopy (FTIR) and X-ray diffraction, while phase behavior was examined using scanning electron microscopy (SEM) and differential scanning calorimetry. The biodegradation in a simulated human body fluid was investigated through mass loss, SEM, and FTIR. The experimental results indicated that all of the polyurethane samples bore the microphase separation structure, and the separation degree depended on the sequence structure and molecular weight (MW) of PEG and further affected their in vitro degradation. The driving force was related to the restricted movement of the molecular segments, the crystallization of the soft/hard phases, and/or the hydrogen bonding interactions between the hard segments. The surface morphological change of the degraded samples further demonstrated that the degradation became serious as the PEG MW increased and that the random block copolymers decomposed more easily than the alternating copolymers. The block polymer materials are expected to be incorporated into specific applications in related biomedical fields.

        Fabrication and Characterization of Copper Nanoparticles in PVA/PAAm IPNs and Swelling of the Resulting Nanocomposites

        Well-dispersed copper nanoparticles were fabricated using poly(vinyl alcohol)/polyacrylamide interpenetrating polymer networks (PVA/PAAm IPNs) as a nanoreactor template. The synthesis of the IPNs hydrogels was achieved in the presence of glutaraldehyde and N,N'-methylene-bis-acrylamide. The resulting PVA/PAAm/Cu nanocomposite hydrogels were characterized, and the swelling and mechanical properties were investigated. The results indicated that the copper nanoparticles had a spherical shape with a size range from 10 to 20 nm. The complexation of PVA in PVA/PAAm IPNs with Cu 2+played an important role in avoiding the aggregation of copper nanoparticles and providing particle size and size distribution controllability and stability. Although the swelling capacity of the nanocomposite hydrogels was slightly lower than that of the control, they had better compression mechanical properties. The water uptake and mechanical properties can be easily tuned by changing the component ratios to meet the requirements of specific applications, such as drug controlled release or tissue engineering.

        An Optimal Energy Storage Operation Scheduling Algorithm for a Smart Home Considering Life Cost of Energy Storage System

        This paper presents an optimal operation scheduling algorithm for a smart home with energy storage system, electric vehicle and distributed generation. The proposed algorithm provides the optimal charge and discharge schedule of the EV and the ESS. In minimizing the electricity costs of the smart home, it considers not only the cost of energy purchase from the grid but also the life cost of batteries. The life costs of batteries are calculated based on the relation between the depth of discharge and life time of battery. As the life time of battery depends on the charge and discharge pattern, optimal charge and discharge schedule should consider the life cost of batteries especially when there is more than one battery with different technical characteristics. The proposed algorithm can also be used for optimal selection of size and type of battery for a smart home.

      • Study on Mutual Information Medical Image Registration Based on Ant Algorithm

        The mutual information has been combined with medical image registration wildly, in order to improve the registration precision and speed of medical image, the application of ant algorithm on it is studied in depth. Firstly, the basic conception of mutual information is analyzed; secondly, the basic procedure of medical image registration is discussed; thirdly, the basic idea of ant algorithm is analyzed, and then the mutual information medical image registration based on FA is designed. Finally, simulation is carried out, and results show that this method can get better medical image registration effect.
