      • 人蔘 Total Saponin, Protopanaxadiol 및 Protopanaxatriol의 消炎作用에 關한 硏究

        Abstract-In an attempt to elucidate the anti-inflammatory action of ginseng total saponin, protopanaxadiol and protopanaxatriol, the anti-inflammatory activity of three compounds was investigated under various acute and chronic inflammatory models. The blood vessel permeability was slightly inhibited by total saponin and protopanaxatriol treatments. Granuloma formation induced by 2% carrageenan was inhibited by total saponin and protopanaxatriol. The cotton-pellet granuloma formation was significantly inhibited by intraperitoneal injection of total saponin. Total saponin and protopanaxadiol inhibited leukocyte emigration and protein exudation in CMC-induced pouch but protopanaxatriol increased leukocyte emigration. The swelling of rat hind paw induced by 1% carrageenan was significantly inhibited by total saponin, protopanaxadiol and protopanaxatriol both single and 2 weeks treatments. Total saponin, protopanaxadiol and protopanaxatriol decreased the anti-inflammatory activity in adrenalectomized rat. Our results suggest that total saponin, protopanaxadiol and protopanaxatriol have potent anti-inflammatory activity, this may be mediated in part through stimulation of adrenal glands.

      • 人蔘 Saponin成分이 흰쥐 膽膵液 分泌에 미치는 效果에 관한 硏究

        The effect of saponins including water extract of Panax Ginseng was studied on the pancreaticobiliary secretion and cholate output and amylase activity in rats. The saponins tested were crude saponin, panaxadiol (PDS), panaxatriol (PTS). After single or two weeks administration of the samples, pancreaticobiliary juice of rats were collected for 8 hrs and the results obtained were summarized as follows. (1) In animals with single administration, pancreaticobiliary juice volume(PJV) was not significanty increased as compared to the control group. However, there was a significant increase in PJV of animals from 4 to 6 hrs after two weeks administration of water extract (500mg / kg / day) or crude saponin(50mg / kg / day). (2) Cholate output was increased dose-dependently from 1 to 5 hrs after the single administration of PDS, PTS. On the other hand, after two weeks administration of crude saponin (50mg/kg/day), the cholate output of the animals was increased significantly. (3) amylase activity in the juice were intermittently increased in single administration of water extract, crude saponin and PTS(100mg / kg). After two weeks administration, there was a significant increase in amylase activity of animals treated with PTS as compared to the control group.

      • Ranitidine의 해명十二指腸切片에 대한 cholinergic效果

        In this experiment, the effect of ranitidine which is histamine H_2-recepter antagonist, were studied in aspect of the relationship between acetylcholine and histamine in isolated duodenum strips of guinea-pig. At the same time, the effect of the inhibition on the acethylcholinesterase activity in human serum were examined and the experimental results was summerized as following. 1) Ranitidine showed the synergistic effect with acetylcholine in the isolated duodenum strip and this effect was blocked by atropine. However, both physostigmine and DFP induced the synergistic effect with ranitidine. 2) Ranitidine potentiated the contractability of DMPP while a little effect on the contraction response due to pilocarpine, serotonin, tetragastrin and PGF_2α in the isolated duodenum strip. 3) Clonidine showed the inhibitory effect of histamine and acetylcholine contraction response of the strip but ranitidine and cimetidine showed no effects on this preparation. 4) ID_50 of ranitidine and cimetidine on serum acetylcholinesterase was 4 x 10 exp(-4)M and 9 x 10 exp(-3)M, respectively. From the above results it was suggusted that ranitidine might have cholinergic effect in guinea-pig duodenum strips and this effect partially might be due to the cholinesterase inhibition.

        Abstract-In order to investigate pharmacological properties of New Wonbang Woohwangchungsimwon Liquid(NSCL) and Wonbang Woohwangchungsimwon Liquid(SCL), the effects of NSCL and SCL on cerebral ischemia and central nervous system were compared. Cerebral ischemia insult was performed using unilateral carotid artery occlusion in mongolian gerbils. The histological observations showed a preventive effect of NSCL and SCL treatments with ischemia-induced brain damage. The ATP in brain tissue was decreased in vehicle-treated ischemic gerbils. This decrease was prevented by NSCL and SCL treatment. In contrast to what was seen with ATP, lipid peroxide were elevated in vehicle-treated ischemic gerbils. This elevation was inhibited by NSCL and SCL treatments. While NSCL and SCL had no effects on the hexobarbital-induced sleeping time, they showed sedative effect in rotarod and spontaneous activity test. NSCL and SCL prevented the seizures induced by electric shock and strychnine, but the effect of NSCL was less than that of SCL. Furthermore, NSCL and SCL showed anti-stress effect. Our findings suggest that the phamacological profiles of NSCL on cerebral ischemia and central nervous system are similar to those of SCL.

      • 中, 長鎖脂肪 添加食餌가 cholesterol 投與 흰쥐의 血中脂質에 미치는 影響

        The purpose of the study was to find and effect of medium chain triglyceride(MCT) diet on the serum lipids of spraque-Dawely rats when they were fed with 1% cholesterol. All experimental groups were fed with 1% cholesterol and at the same time, fed with different diet such as Soybean oil, Palm oil, Coconut oil, MCT mixed with Palm oil and MCT mixed Soybean oil respectively. After fed to each group, the levels of serum lipids among fed rats were examined and the data form the experiment were compared with a Control group. The result were as follows : (1) The body weight gain in MCT diet group and MCT mixed with Soybean diet group were lighter than that of the control group. (2) Total cholesterol levels in serum of all experimental diet groups were very lower than that of the Control group. (3) VLDL, LDL-cholesterol levels of all experimental diet groups were lower than that of the Control group. But the levels of VLDL, LDL-cholesterol in the MCT diet group was slightly higher than that of all the experimental diet groups. (4) HDL-cholesterol levels of all experimental groups were higher than that of the Control group. But the levels of HDL-cholesterol levels in the MCT diet group was slightly lower than that of all the experimental groups. (5) The ratio of VLDL, LDL-cholesterol to HDL-cholesterol, so called Atherogenic index in serum of all the experimental diet groups were significantly lower than that of the Control diet group. The levels of Atherogenic index in the MCT diet group was slightly higher than that of all the experimental diet groups. (6) Phospholipid levels in serum of MCT diet group was lower than that of all the experimental diet groups, but the ration of Total cholesterol to Phospholipid in serum was increased. (7) Triglyceride levels in serum of MCT diet group and mixed diet groups with MCT were significantly higher than that of all the experimental diet groups.
