        단클론 항체를 이용하여 정제한 톡소포자충 30 kDa 항원의 면역학적 특성

        The molecular weight 30 kDa membrane protein of Toxoplusma Sondii (Toxoplasma 30 kDa) apparently conserved in most strains of T. gondii and sera of infected hosts. The present study aimed to elucidate Toxoplasmc 30 kDa as a useful diagnotic antigen for serodiagnisis of toxoplasmosis by ELISA and for induction of protective immunity. Murine spleen cells immunized with the membrane antigen of T. gondii were fused with mouse Sp2/0-Ag 14 myeloma cells. Out of 8 clones selected, five were IgG2b, the others belonged to IgG 1 and IgG2a. The 30 kDa antigen was distributed mainly on the surface membrane of tachyzoites by indirect fluorescence method. Murine peritoneal macrophages which were activated by 30 kDa antigen produced more amounts of NO2 compared with crude antigen-treated group, however there were no significant differences in toxoplamacidal activity between the two groups. Higher specificity of Toxoplosma 30 kDa antigen was recognized for serodiagnosis of toxoplasmosis than the crude antigen. From these results, ToxopLasmo 30 kDa antigen enhances the cytotoxic effect of macrophages as well as a more reliable means for the serodiagnosis of toxoplasmosis by ELISA. Key words: Toxoplosma gondii, 30 kDa antigen (p30), mouse, serodiagnosis, macrophage, cytotoxicity. 톡소포자충(ToxopLasmn gondii)은 다양한 항원을 가지고 있으며, 이들 항원의 분석은 세 포매개성 면역반응 및 톡소포자충증의 면역학적 진단방법의 연구에 매우 중요하다 본 연구는 톡소포자충의 여러 단백질중 대부분의 충주(strain)에 존재하는 분자량 30 kDa의 단백질을 단클론 항체를 이용하여 분리한 후. 30 kDa 항원의 면역학적 특성을 초음파 추출 조항원과 비교 평가하였다. 톡소포자충의 세포막 항원으로 면역한 마우스 비장세포와 마우스 Sp2/0-Agl4 골수종세포를 융합하여 8개의 단클론 항체를 Western blot으로 확인하였다 이들 단클론 항체는 높은 특이성을 보였으며, $IgG_{2b}가{\;}5개,{\;}IgG^{1}이{\;}2개,{\;}IgG_{2a}$가 1개였다. 간접형광항체법으로 충체내 위치를 관찰한 결과. 30 kDa 항원은 tachyzoite의 표면 세포막에 주로 분포하였다. 단클론 항체와 CNBr-activated Sepharose 4B를 coupling하여 만든 immunoafrnity chromatography를 이용 하여 30 kDa 항원을 분리하였다. 분리한 30 kDa 항원으로 자극시킨 마우스 복강대식세포의 $NO_2^{-}$ 생산량은 초음파 추출 조항원 사용군에 비해 유의하게 증가하였으나 대식세포의 탐식능은 유의한 차이가 없었다. 또한 ELISA로 톡소포자충증을 진단시, 톡소포자충 30 kDa 항원 사용군은 조항원 사용군에 비해 민감도의 변화는 없었으나 특이성은 증가하였다 이상으로 보아 톡소포자충 30 kDa 항원은 감염 방어 면역 효과가 있었으며 진단에 이용시 특이성을 더 높일 수 있었다.

        To study fauna of Silla University located in Mt. Baek-yang, we conducted six surveys from 29 May 2011 to 28 April 2012. We found five phyla 41 orders 197 species of animals inhabiting in this area: four phyla 27 orders 163 species of invertebrates and in vertebrates one order six species of fishes, three orders nine species of amphibians and reptiles, six orders 14 species of aves, and four orders five species of mammals. Of these species, some Korean endemic species were included: a fish of Rhodeus uyekii in a pond near main gate of this university, a frog of Rana coreana in a creek near a dormitory, and a rabbit of Lepus coreanus and a water deer of Hydropotes inermis in a grass region near a main stadium. A kind of hawk, Falco tinnunculus as a natural monument (animal) of no. 323-8, was observed on a building of medical and life science. A blue frog of Hyla japonica as a variation was found near a spring resort. Therefore, this area is considered an ecologically important area having a high animal diversity.

        Toluene, Xylene, Trichloroethylene 투여가 흰쥐의 시상하부와 뇌하수체의 GnRH, GnRH Receptor, Pit-1 유전자 발현에 미치는 영향

        The workplace exposure of chemicals has steadily increased, therefore the concern for subsequent effect on reproductive outcome has been an important issue in occupational medicine. In previous studies, higher rates of spontaneous abortion, reduced fertility and menstrual disorder among women, and an impairement of sperm quantity and quality among men have been associated with a wide variety of chemical agents. This study was conducted to evaluate the effects of toluene, xylene and trichloroethylene(TCE) injection on the mRNA levels of GnRH, GnRH receptor and Pit-1 genes in male rats hypothalamus and pituitary and the effects on the plasma levels of FSH, LH, prolactin and testosterone. Sprague-Dawley male rats were divided into five groups of five each according to concentration of toluene, xylene and TCE. The rats were injected subcutaneously to 0, 50, 100, 200, 400 mg/kg body weight/day of toluene, xylene and TCE, respectively for 6 days. Rat brains were excised and hypothalamus and pituitary were separated. Reverse transcription-polymerase chain reaction (RT-PCR) and RNase protection assay(RPA) were used to evaluate the GnRH, GnRH receptor and Pit-1 mRNA levels. Plasma concentrations of FSH, LH, prolactin and testosterone were assayed by radioimmunoassay(RIA). The results were as follows; 1. GnRH, GnRH receptor and Pit-1 mRNA levels in toluene and xylene injected groups, and GnRH receptor mRNA levels in TCE injected group were lowered dose-dependently. Especially, GnRH receptor and Pit-1 mRNA levels in 200mg/kg of toluene injected group, and GnRH, GnRH receptor and Pit-1 mRNA levels in 400mg/kg of toluene injected group were significantly lower than control group (p<0.05). GnRH receptor and Pit-1 mRNA levels in 400mg/kg of xylene injected group, and GnRH receptor mRNA levels in 400mg/kg of TCE injected group were significantly lower than control group(p<0.05). 2. The plasma levels of prolactin and testosterone in 400mg/kg of toluene injected group, and LH in 100, 200 and 400mg/kg of xylene injected group, and testosterone in 400 mg/kg of TCE injected group were significantly lower than control group(p<0.05). In conclusion, we speculated that toluene and xylene affected reproductive system secondarily through hypothalamus-pituitary axis, and TCE affected directly through steroidogenesis. And we recommended that further study for assessment of the reproductive toxicity of mixed organic solvent exposures should be conducted.

        산화아연 촉매 상에서 메탄올-요소 반응계로부터 DMC합성을 위한 표면적의 영향

        The synthesis of di-methyl-carbonate (DMC) with urea and methanol occurred by two step reactions. In first step, methyl-carbamate (MC) is produced by the reaction between urea and methanol. In second step, DMC is synthesized by the catalytic reaction between MC and methanol over the catalyst. In this study, ZnO was used as the catalyst for the synthesis of DMC and the reaction behavior for the synthesis of DMC over ZnO catalysts, which have a different surface area, was investigated. The surface area of ZnO was controlled with the change of calcination temperature and that calcined at with 400℃ was 27㎡ /g. Meanwhile, the yield of products was investigated with the reactionconditions, such as the temperature, the pressure, the time, the amount of catalyst and the agitation speed. The yield of DMC was changed with changing reaction conditions. It was also concluded that the production of N-methyl-methyl-carbamate (N-MMC), which is the by-product produced by the reaction between MC and DMC, taken affected in the yield of DMC. The maximum yield of DMC was obtained to approximately 23.9% in the test under the suitable reaction conditions, which was performed with 1 gram of catalyst at 175℃, 25 bar and 600 rpm for 10 h. The yield of DMC increased with the increase of catalyst’s surface area, while the changing surface area of catalyst was exhibited the little effect for the production of N-MMC.

        보조생식술(ART)치료를 받은 환자들에서 자궁근종이 임신에 미치는 영향

        목적 : 본 연구는 장막하근종, 근육층내의 근종, 또는 점막하 근종등의 출현이 보조생식술후 임신율과 착상율에 어떠한 영향을 미치는가를 보기 위해 시행되었다. 연구방법 : 1998년 1월 1일 부터 1998년 11월 30일 까지 본원에서 보조생식술(ART)을 시행받은 불임환자들 중 양극단의 연령을 제외한 26-45세 사이의 환자들을 대상으로 하였으며, 자궁근종이 진단된 환자들의 278예와 근종이 없는 환자들의 1180예를 비교하였다. 결과 : 이 연령범위내 전체적인 비교에서는 대조군(비근종군)과 근종군에서의 임신율이 각각 41.78%, 31.65%, 그리고 착상율은 각각 15.7%, 11.48%로 통계적으로 유의하게 근종군에서 낮았다. 그러나 평균연령이 대조군(33.1세)에서 보다 근종군(35.5세)에서 유의하게 높음을 알게 되었고, 이에 따라서 연령군들(26-30, 31-35, 36-4, 41-45)에 따른 근종군과 대조군을 각각 비교하였다. 그 결과 동일 연령군에서 근종군(장막하 근종SS, 근육층내 근종IM)과 대조군, 그리고 근종의 그룹들(SS, IM)간의 임신율과 착상율의 비교에서는 서로간에 유의한 차이를 볼 수 없었다. 그리고 점막하 근종그룹의 경우는 통계적 비교를 하기에는 그 예와 수가 너무 적어 결과를 알 수 없었다. 결론 : 일반적으로 근종군에서 상대적으로 낮은 임신율을 보고한 여러 논문에서와 달리, 본 연구에서는 동일한 각각의 연령군에서의 근종군과 대조군간의 임신율과 착상율의 차이가 없었다. 또한 근종의 위치에 따른 그룹별 임신율 및 착상율의 비교에서도 유의한 차이가 없음을 보였는데 이것은 근종이 근육층을 어느정도의 기준치 이상으로 침범했을 때에만 IM으로 분류한 국외의 일부 보고와 달리 근육층을 조금이라도 침범하면 IM군으로 분류한 본 연구에서의 차이 때문일 것으로 생각되며, 장막하 근종그룹과 근육층내 근종그룹의 분류 기준을 달리한다면 다른 결과를 보일 수도 있으리라 여겨진다. 또한, 근종이 임신에 미치는 영향이 다양함을 고려할 때, 근종의 크기 및 위치에 따른 SS군과 IM군 등의 체계적 분류 후 이에 따르는 전향적 연구가 진행된다면 임신에 대한 근종의 영향평가에 많은 도움이 될 것으로 본다. Objective : The purpose of this study was to examine whether the uterine leiomyomas [subserosal(SS), intramural(IM), submucosal(SM)] influence on the pregnancy rate(PR) and implantation rate(IR) of ART program. Methods : This study was done by the review of medical records of 1458 ART cycles from Jan. 1 of 1998 to Nov. 30 of 1998 at Infertility Medical Center, CHA General Hospital. We compared the PR and IR between 278 cycles of myoma group and 1180 cycles of control(non-myoma) group. Results : The mean age of patients with myomas was significantly higher than that of control group. Accordingly, we set the age range(24 - 46yrs) and divided it into four parts(26-30, 31-35, 36-40, 41-45) that did not show the significant difference in PR and IR. As a result, there were no significant differences of PR and IR among control, SS and IM within same age range. (We didn't know the result in SM myoma group because the number of SM myoma cases were very small.) Conclusion : To the contrary of our expectation, there was no significant difference in PR and IR between control and myoma groups. On the other hand, the result of no significant difference in PR and IR between myoma groups may will show different result if we apply different classification in categorizing SS and IM myomas.

        Serum Globotriaosylceramide Assay as a Screening Test for Fabry Disease in Patients with ESRD on Maintenance Dialysis in Korea

        Background/Aims: Fabry disease is an X-linked recessive and progressive disease caused by α-galactosidase A (α-GaL A) deficiency. We sought to assess the prevalence of unrecognized Fabry disease in dialysis-dependent patients and the efficacy of serum globotriaosylceramide (GL3) screening. Methods: A total of 480 patients of 1,230 patients among 17 clinics were enrolled. Serum GL3 levels were measured by tandem mass spectrometry. Additionally, we studied the association between increased GL3 levels and cardiovascular disease, cerebrovascular disease, or left ventricular hypertrophy. Results: Twenty-nine patients had elevated serum GL3 levels. The α-GaL A activity was determined for the 26 patients with high GL3 levels. The mean α-GaL A activity was 64.6 nmol/hr/mg (reference range, 45 to 85), and no patient was identified with decreased α-GaL A activity. Among the group with high GL3 levels, 15 women had a α-GaL A genetics analysis. No point mutations were discovered among the women with high GL3 levels. No correlation was observed between serum GL3 levels and α-GaL A activity; the Pearson correlation coefficient was 0.01352 (p = 0.9478). No significant correlation was observed between increased GL3 levels and the frequency of cardiovascular disease or cerebrovascular disease. Conclusions: Fabry disease is very rare disease in patients with end-stage renal disease. Serum GL3 measurements as a screening method for Fabry disease showed a high false-positive rate. Thus, serum GL3 levels determined by tandem mass spectrometry may not be useful as a screening method for Fabry disease in patients with end stage renal disease.         A Thermal Noise Measurement System for Noise Temperature Standards in <tex> $W$</tex>-Band

        <P>A W-band waveguide noise measurement system has been developed for noise temperature (NT) standards. The system consists of two standard noise sources and a waveguide radiometer. A horn-type cryogenic noise source operating at the liquid nitrogen temperature and an ambient temperature termination have been employed as standard noise sources. To measure noise power radiated from the noise sources including a device under test, a total-power waveguide radiometer has been constructed and its performance has been evaluated. Measured NT of a commercial waveguide noise source is presented with the measurement uncertainty of 0.10-0.13 dB (k = 2) in W-band.</P>

      • 난소암 환자 복수내의 면역복합체로부터 새로운 암관련항원 존재 확인

        Immune complexes were isolated from ascitic fluid of patients with ovarian carcinoma by using protein-A Sepharose CL-4B affinity chromatography, and antigen-antibody complexes then dissociated by ion exchange chromatography in the presence of 8M urea. Only one antigen fraction was obtained by the ion exchange chromatography, but three antigenic bands were demonstrated by SDS-PAGE. The molecular weights of the three protein bands were 70, 41, and 38 kDa, respectively. Of the three proteins the 38 kDa band has already been known, but 70 and 41 kDa bands seems to represent new ovarian tumorassociated antigens. This study may suggest the involvement of multiple antigens in ovarial tumor which can be included in producing tumor-directed monoclonal antibodies
