        Flocculant-producing microorganisms were screened from obtained strains in the laboratory using a pulp-wastewater treatment as the flocculating agent. K-111 strain that had high flocculating activity among them was selected and identified as Bacillus sp. K-111 16s rDNA sequencing. The favorable medium for the production of flocculant was glucose 1.5%, NH_4NO_3 0.2%, tryptone 0.01%, K_2HPO_4 0.08%, KH_2PO_4 0.06%, CaCO_3 0.03%, MgSO_4·7H_2O 0.005%, MnSO_4 0.005% in 1 liter of D.W. at initial pH 7.0. The optimum culture temperature and pH were 30℃ and pH 7.0, respectively. the flocculating activity was observed most highly after 36 to 48 hr of cultivation at the optimum conditions. the flocculating activity of produced biopolymer on optimum conditions was about 2.5-fold higher than that of screening medium.

        연 X-선 현미경을 이용한 금 나노입자 세포영상

        연 x-선 현미경은 ‘물의 창’ 영역(2.3~4.4 nm)의 파장을 이용하여, 수십 nm의 분해능으로 세포를 파괴하지 않고 살아있는 상태에서 세포의 내부구조를 관찰할 수 있어 가시광선현미경과 전자현미경을 단점을 보완하는 특징을 갖는 세포 생물학 연구에 적합한 현미경이다. 그러나 기존 연 x-선 현미경은 광원으로 방사선 가속기를 이용하기 때문에 사용이 제한적이었다. 이에, 본 연구에서는 2.88 nm의 연 x-선을 광원으로 사용하는 소형 연 x-선 현미경을 이용하여, 내포작용에 의해 금 나노입자를 포획한 HT1080과 MDA-MB 231 세포의 영상을 약 60 nm 분해능으로 획득하였다. 금 나노입자의 세포에 대한 독성을 제거하기 위하여 폴리에틸렌 글리콜을 캡핑하였고, 2.88 nm 파장의 연 x-선에 대하여 충분한 조영효과로 인하여 세포영상에서 뚜렷한 대조도를 나타내었다. 내포작용에 의해 액포에 포함되어 있는 다양한 크기의 금 나노입자 군집을 확인하였으며, 세포내부의 액포의 분포상태도 관찰할 수 있었다. 따라서 고분해능을 가진 소형 연 x-선 현미경을 이용하여 금 나노입자를 세포내의 미세기관이나 특정 단백질에 표지하면 연 x-선에 대한 조영효과의 증가에 의하여 더욱 유용한 정보를 획득할 수 있을 것으로 생각한다. A compact soft x-ray microscope operated in the ‘water window’ wavelength region (2.3~4.4 nm) was used for observing cells with nano-scale spatial resolution. To obtain cellular imaging captured with colloidal gold nanoparticles using a compact soft x-ray microscope. The colloidal gold nanoparticles showed higher contrast and lower transmission more than 7 times than that of cellular protein on the soft x-ray wavelength region. The structure and thickness of the cell membrane of the Coscinodiscus oculoides (diatome) and red blood cells were seen clearly. The gold nanoparticles within the HT1080 and MDA-MB 231 cells were seen clearly on the soft x-ray microscopy. The gold nanoparticles were aggregated within vesicles by endocytosis.

      • 여러가지 벽체조건을 가진 밀폐공간에서 열적쾌적에 대한 수치해석 연구

        This study is investigated for predicting a comfort region in the rectangular enclosure assumed Ondol cavity. Thermal environment is actually a combination of air temperature, mean radiant temperature, air velocity, humidity and two personal factors(clothing insulation and activity level) that will influence a person's perception of warmth or coolness. A comfort region is predicted by obtaining operative temperature(OT). The operative temperature is related with radiant and convective heat flux. The operative temperature of comfort region(thermal neutrality) is 20-24℃ at clo=0.1, met=1.2, RH=0%, -0.5<PMV<0.5 and PPD<10%. Radiant and convective heat flux is obtained for various adiabatic walls. This study is carried out a two dimensional numerical analysis by SIMPLER method. Radiant heat flux is obtained by a simplified zone analysis method. And shape factor is obtained by direct integral method.

      • 수축형 및 확장형 FACE-BOW에 의한 상악 제1대구치 견인시 치근주위 응력분포에 대한 광탄성법적 분석

        The purpose of this study was to investigate the effect of the Contraction and Expansion Face-Bows retraction of the maxillary first molars. The experimental models were made with the epoxy resin and the Face-Bows used in this study were standard type, contraction type and expansion type of the Ormco company. The maxillary first molars were retracted with each type of Face-Bow in the direction of straight pull and 500gm in total force. The obtained stress distributions around the roots were analysed by three dimensional photoelastic method. The results were as follows: 1. In the mesio-distal stress distribution, the maximum cervical stress of distal side of the Contraction and Expansion Face-Bows was approximately a half of the standard type case. 2. In the apical view, the ratio of the maximum stress distribution of the cervical one third is as follows: 1) In case of the standard type, distal and buccal maximum stress shows the ratio of 1 to 0.77. 2) In case of the contration type, distal and lingual maximum stress shows the ratio of 1 to 1.08. 3) In case of the expansion type, distal and lingual maximum stress shows the ratio of 1 to 1.46.

      • 男左·女右 理論에 따른 顔面神經痲痺의 臨床的 考察

        著者는 臨床에서 흔히 患者들로부터 구안와사의 豫候에 대해 "男左女右"와 關聯하여 질문을 받으므로 이에 대한 臨床的 妥當性을 알아보기 위해 1995年 9月 1日부터 1996年 8月 31日 까지 1年間 圓光大學校 附屬 全州韓方病院 外官科 外來에 來院하여 구안와사를 診斷받은 總 148명을 對象으로 發病動機 및 身體狀態, 年齡別 分布, 男女 左右別 發生率, 總 來院患者에 대한 治療率, 年齡別 發生率, 男女 左右에 따른 治療率을 각각 臨床 觀察하여 다음과 같은 결과를 얻었다. 1. 發病動機 및 身體狀態는 風寒과 연관되는 原因이 가장 많았으며, 肉體過勞, 精神過勞, 飮酒 등의 순으로 나타났고, 2가지 이상의 原因이 있는 경우는 精神過勞와 肉體過勞가 겸해오는 경우가 많았다. 2. 年齡別 分布는 50-59세가 가장 높았고, 40-49세, 30-39세, 20-29세, 60-69세, 70세 이상, 1-9세, 1세 이하의 순으로 각각 나타났다. 3. 男女 左右別 發病率은 右側 顔面神經이 痲痺된 男子 患者가 가장 많았으며, 右側 顔面神經이 痲痺된 女子 患者, 左側 顔面神經이 痲痺된 男子 患者, 左側 顔面神經이 痲痺된 女子 患者 순으로 나타났으나 發病率에 큰 차이는 없었다. 4. 總 來院患者 148명에 對한 治療率은 52.53%로 나타났다. 5. 年齡別 治療率은 1-9세가 가장 높았으며, 70세 이상, 50-59세, 40-49세, 60-69세, 20-29세, 30-39세, 10-19세의 순이었고, 주로 中·老年層이 治療率이 높게 나타났다. 6. 男女와 左右에 따른 治療率은 顔面神經痲痺의 女子 患者가 가장 높게 나타났고, 右側 顔面神經痲痺의 男子 患者, 左側 顔面神經痲痺의 女子 患者, 左側 顔面神經痲痺의 男子 患者의 순으로 나타났고, 男女 모두 右側에 發病한 患者의 治療率이 左側보다 높게 나타났으나, 특별한 有意性은 없었다. 以上의 結果로 보아 韓醫學的 生理 病理理論인 男左女右의 豫候와 治療率은 一致하지 않았으나 此後 좀더 많은 患者數와 오랜 期間의 臨床을 통한 硏究가 要求될 것으로 思料된다. The author analyzed 148cases of Facial Palsy, who were treated in the Jeon-ju Oriental Medical Hospital of Wonkwang University from September 1995 to August 1996. I've examined the 148 cases in the view of the etiologic distributions, the age, the relationship of the sex and paralytic side, and the ratio of recovery according to the age, the relations of sex and paralytic side. The following results are obtained. 1. The ratio of punghan(風寒) was 18.24%(27 cases), overwork was 14.86%(22 cases), stress was 12.84%(19 cases), excessive drinking was 2.70%(4 cases), teeth pain was 2.02%(3 cases), and idiopathy was 37.84(56) and etc. 2. The ratio of 5th decade were 20.95%(31 cases), 4th decade was 18.25%(27 cases), 3th decade was 17.57%(26 cases), 2th decade was 14.19%(21 cases), 6th decade was 12.16%(18 cases), over seventy was 10.81%(16 cases), teenagers was 4.05%(6 cases), under teenagers was 1.35%(2 cases), and infant was 0.67%(1 case). 3. The ratio of the male-right was 28.38%(42 cases), female-right was 25.68%(38 cases), male-left was 22.30%(33 cases), female-left was 21.62%(32 cases), female-both side was 1.35%(2 cases) and male-both side was 0.67%(1 case) in order. 4. When we examined the degree of recovery about the 148 patients, perfect cure was seen in 17.57%( 26 cases), excellent in 25.00%(37 cases), good in 22.30%(33 cases), fair in 20.27%(30 cases), poor in 14.86%(22 cases). The total remedial value of the 148 patients was revealed 52.53%. 5. When we examined the 148 patients the remedial value of the infant was 50.00%, under teenagers 62.50%, teenagers 41.67%, 2th decade 47.62%, 3th decade 44.23%, 4th decade 54.63%, 5th decade 58.87%, 6th decade 51.39%, over seventieth 60.94%. The remedial value of under teenagers, 4th decade, 5th decade, over seventieth were higher than the total remedial value(52.53%) 6. In the point of the relationship of the sex and the paralytic side, the remedial value of male-left was 51.47%, male-right 53.45%, male-both side 50.00%, femaled-left 47.62%, female-right 58.75% and female-both 37.50%. The remedial value of male-right(53.45%) and female-fight(58.75%) were higher than the total remedial value(52.53%). These results demonstrated that in the point of prognostic view, there was little concerned with the traditional rule of sex-paralytic relationship that man is awed left paralysis and woman right paralysis.

        기억력 저하를 호소하는 노인들을 대상으로 INM176를 3개월간 임상시험한 후의 치료성적 및 부작용 평가 보고

        Objectives : We examined the effects of INM 176 (K-l107) compared with placebo on the cognitive functions of 92 old aged subjects with cognitive impairment. Methods : This was a prospective, 12 week, double-blind, Placebo-controlled clinical trial. The elderly who achieved a score of less than 25 points on the K-MMSE or showed a high risk of Alzheimer's disease from the 7-Minute Neurocognitive Screening Battery were considered to have objective impairment and were selected as subjects for this study. The subjects were randomized to placebo or INM 176 group. The outcome measures were from the Alzheimer's Disease Assessment Scale-cognitive subscale (ADAS-Cog), the Instrumental Activities ofDaily Living (IADL) and the Korean Geriatric Depression Scale (KGDS) and two kinds of computerized priming tests. After setting the total error score in the Alzheimer's Disease Assessment Scale-Cognitive section (ADAS-cog) as the repeated measurement factor, an analysis of variance of the combined factor design was done between the placebo and INM 176 group. Results : The interaction effect of time (pre- and post-trial) and group (placebo and INM 176 group) was significant in the analysis of the ADAS-cog's total error score. The INM 176 group's total error score in the ADAS-cog decreased significantly (p<0.01), whereas the placebo group showed a slight increase. The mean changes in IADL and GDS from baseline scores favored in the INM 176 group than in the placebo group. Outcome changes ofADAS-cog, IADL, KGDS scores during the 12 week clinical trail ofINM 176 and placebo demonstrated favorable responses in the INM176 administered group. Conclusions : This is a preliminary clinical trial result of INM176 as a memory pill. Based on these results, INM176 may be a candidate molecule for the improvement of cognitive functions, including memory, Further clinical trial should demonstrate its efficacy.

      • Drosophila short neuropeptide F signalling regulates growth by ERK-mediated insulin signalling

        Insulin and insulin growth factor have central roles in growth, metabolism and ageing of animals, including Drosophila melanogaster. In Drosophila, insulin-like peptides (Dilps) are produced by specialized neurons in the brain. Here we show that Drosophila short neuropeptide F (sNPF), an orthologue of mammalian neuropeptide Y (NPY), and sNPF receptor sNPFR1 regulate expression of Dilps. Body size was increased by overexpression of sNPF or sNPFR1. The fat body of sNPF mutant Drosophila had downregulated Akt, nuclear localized FOXO, upregulated translational inhibitor 4E-BP and reduced cell size. Circulating levels of glucose were elevated and lifespan was also extended in sNPF mutants. We show that these effects are mediated through activation of extracellular signal-related kinases (ERK) in insulin-producing cells of larvae and adults. Insulin expression was also increased in an ERK-dependent manner in cultured Drosophila central nervous system (CNS) cells and in rat pancreatic cells treated with sNPF or NPY peptide, respectively. Drosophila sNPF and the evolutionarily conserved mammalian NPY seem to regulate ERK-mediated insulin expression and thus to systemically modulate growth, metabolism and lifespan.
