Malignant peritoneal mesothelioma is a rare neoplasm that arises from the mesothelium of a serosal cavity and is a rapidly fatal disease with a median survival of 4 to 12 months for untreated cases. Recently, we experienced a case with malignant peritoneal mesothelioma who was suspected hepatocelluar carcioma by abdominal CT scan and was confirmed by biopsy including immunohistochemical stain(calretinin) after surgery. We performed tumor excisions and wedge resection of the liver(segment Ⅷ)and inserted Tencoff catheter in abdominal cavity at 25th day of post-operation. We treated with intraperitoneal paclitaxel(25mg/m^(2)/day for 5 days) six courses monthly. She was well tolerable and is still living without any evidence of recurrence for 14th month of post-operation.

This study was conducted to evaluate the effect of dietary supplementation of anise flavor on the performance of lactating sows and suckling pigs at different weaning ages. A total of 120 sows (Landrace × Yorkshire) were used in this experiment. Sows were fed with a commercial lactation diet (60 sows) or diet with flavor (60 sows) during days 100 to 114 of gestation period and the whole lactation period. Piglets were allotted to 2 dietary treatments based on their initial body weight (BW) using a 2 × 2 factorial arrangement of treatments with 2 levels of anise flavor (0 or 0.5 g/kg) and weaning periods (weaned at 21 day or 28 day after birth). Sows fed with flavor supplemented diets had higher (p < 0.05) back fat and average daily feed intake (ADFI) at weaning, lower (p < 0.05) back fat loss than those fed with non-flavor diets. Sows weaned at 28 day had higher (p < 0.05) weaning back fat and lower (p < 0.05) back fat loss than those weaned on 21 day. During weaning, piglet average daily gain (ADG), ADFI, digestibility of dry matter (DM), nitrogen (N), and gross energy (GE) and fecal score were affected by anise flavor, respectively. In conclusion, dietary flavor supplementation could increase feed intake of lactating sows, improve growth performance and reduce weaning stress of piglets, especially effective in the early weanling piglets. Meanwhile, interactive effects were observed about the piglet performance between weaning ages and anise flavor supplementation after weaning.

Objective: The objective of this study was to investigate the effects of different degrees of maternal dietary energy restriction on lipid deposition in embryonic tissues during the medium laying period (37 to 39 weeks) in Arbor Acres (AA) broiler breeders. Methods: A single factor design was adopted, and 400 AA broiler breeders (20 weeks of age) with a similar weight were randomly allocated into four groups. The birds in the control group were fed a corn-soybean meal based diet, and those in trial groups were fed diets with 80%, 70%, and 50% energy levels of the basal diet. Incubated eggs from the medium laying period were collected. Samples of developing embryos at various stages were prepared for composition analysis. Results: The embryo weight in the 80% energy group was higher than those of the other groups on embryonic day (E) 13, but at 21 E, they were significantly decreased with decreasing energy intake of the broiler breeders (p<0.05). Additionally, the levels of crude fat in tissues in the restriction groups were significantly decreased (p<0.05). The long axis and area of adipocytes in breast muscle, thigh muscle and the liver were significantly decreased (p<0.05) at 21 E in the 80%, 70%, and 50% energy groups. Conclusion: The effects of the 80% maternal dietary energy restriction energy affects egg production performance, egg quality, and nutrient deposition in egg weights, which then directly impacts on the developmental process of embryos, especially on fat utilization and deposition.

Inflammatory factor such as Interleukin-1 play important roles in determining the fate of both acute and chronic neurological disorders. We investigated whether inhibitors of PKC or PTK can serve as pharmacological agents to reduce IL-I production and the mechanisms underlying their pharmacological effects in a mixed population of glia. Inhibitors of PKC such as H7, Go6976 and Ro31-8220 significantly reduced both the mRNA and protein levels of IL-1α and IL-β in lipopolysaccharide-activated primary glial cells. While the PTK inhibitor genistein also significantly reduced the production of these cytokines, it did not affect the expression of their mRNA. Taken together, inhibitors of PKC and PTK could serve as pharmacological agents to reduce IL-1 production. However, the mechanisms underlying their pharmacological effects are different. Our results provide evidence that inhibitors of protein kinases can serve as pharmacological agents to modulate IL-1 production in glial cell, and in turn, alleviate neuronal injury.

This study proposes a novel intelligent fault diagnosis method for rotating machinery using ant colony optimization (ACO) and possibilitytheory. The non-dimensional symptom parameters (NSPs) in the frequency domain are defined to reflect the features of the vibrationsignals measured in each state. A sensitive evaluation method for selecting good symptom parameters using principal componentanalysis (PCA) is proposed for detecting and distinguishing faults in rotating machinery. By using ACO clustering algorithm, the synthesizingsymptom parameters (SSP) for condition diagnosis are obtained. A fuzzy diagnosis method using sequential inference and possibilitytheory is also proposed, by which the conditions of the machinery can be identified sequentially. Lastly, the proposed method iscompared with a conventional neural networks (NN) method. Practical examples of diagnosis for a V-belt driving equipment used in acentrifugal fan are provided to verify the effectiveness of the proposed method. The results verify that the faults that often occur in V-beltdriving equipment, such as a pulley defect state, a belt defect state and a belt looseness state, are effectively identified by the proposedmethod, while these faults are difficult to detect using conventional NN.

Significance

The canonical targeting signal for degrading proteins by the ubiquitin (Ub) system—a chain composed of multiple Ub moieties—has remained a mystery. The structure of the proteasome, the enzyme that recognizes the signal and degrades the target substrate cannot explain why such a long chain is needed. To better understand this problem, we synthesized α-globin to which chains with different number of Ubs were attached. In long adducts, the proximal Ub remains on the substrate, likely securing its attachment to the proteasome, and is degraded with it. The distal Ub protects the proximal from removal by deubiquitinating enzymes and is then removed and recycled. In short adducts, the Ub moieties are rapidly removed, and the substrate remains stable.

One of the enigmas in the ubiquitin (Ub) field is the requirement for a poly-Ub chain as a proteasomal targeting signal. The canonical chain appears to be longer than the distance between the two Ub-binding proteasomal receptors. Furthermore, genetic manipulation has shown that one receptor subunit is sufficient, which suggests that a single Ub can serve as a degradation signal. To shed light on this mystery, we chemically synthesized tetra-Ub, di-Ub (K48-based), and mono-Ub adducts of HA-α-globin, where the distal or proximal Ub moieties were tagged differentially with either Myc or Flag. When incubated in a crude cell extract, the distal Ub moiety in the tetra-Ub adduct was mostly removed by deubiquitinating enzymes (DUBs) and reconjugated to other substrates in the extract. In contrast, the proximal moiety was most likely degraded with the substrate. The efficacy of degradation was proportionate to the chain length; while tetra-Ub globin was an efficient substrate, with mono-Ub globin, we observed rapid removal of the Ub moiety with almost no degradation of the free globin. Taken together, these findings suggest that the proximal moieties are necessary for securing the association of the substrate with the proteasome along the proteolytic process, whereas the distal moieties are important in protecting the proximal moieties from premature deubiquitination. Interestingly, when the same experiment was carried out using purified 26S proteasome, mono- and tetra-Ub globin were similarly degraded, highlighting the roles of the entire repertoire of cellular DUBs in regulating the degradation of proteasomal substrates.

This study focused on the enzymatic biotransformation of the major ginsenoside Rb1 into Rd for the mass production of minor ginsenosides using a novel recombinant β-glucosidase from Flavobacterium johnsoniae. The gene (bglF3) consisting of 2,235 bp (744 amino acid residues) was cloned and the recombinant enzyme overexpressed in Escherichia coli BL21(DE3) was characterized. This enzyme could transform ginsenoside Rb1 and gypenoside XVII to the ginsenosides Rd and F2, respectively. The glutathione S-transferase (GST) fused BglF3 was purifi ed with GST-bind agarose resin and characterized. The kinetic parameters for β-glucosidase had apparent Km values of 0.91±0.02 and 2.84±0.05 mM and Vmax values of 5.75±0.12 and 0.71±0.01 μmol·min-1·mg of protein-1 against p-nitrophenyl-β-D-glucopyranoside and Rb1, respectively. At optimal conditions of pH 6.0 and 37°C, BglF3 could only hydrolyze the outer glucose moiety of ginsenoside Rb1 and gypenoside XVII at the C-20 position of aglycon into ginsenosides Rd and F2, respectively. These results indicate that the recombinant BglF3 could be useful for the mass production of ginsenosides Rd and F2 in the pharmaceutical or cosmetic industry.

In this study, Pd-SnS nanoparticles (NPs) decorated with layered-double-hydroxide (LDH) nanosheetswere found to display an enhanced photocatalytic activity toward the degradation of aqueous rhodamineB (RhB, dye) and chromium VI (Cr(VI)), which was superior to that of pure SnS, LDH, and binary composites. The morphological and structural properties of the as-prepared photocatalysts were characterizedthrough various techniques including XRD, FT-IR, SEM, TEM, and BET surface areas, and their RhB andCr(VI) degradation activity were evaluated under visible light irradiation. The Pd-SnS/LDH heterojunctionsextended the lifetime of the photo-induced electrons and promoted the efficient interfacial transferof charge carriers. Moreover, the strong interactions between Pd-SnS NPs and LDHs nanosheets furtherimproved the photocatalytic activity. In particular, Pd-SnS/LDH heterojunctions with SnS and Pd contentsof 15 and 1 wt%, which showed the highest degradation performance of 99 % within 40 min under illumination, and superior stability during successive experimental progress. We believe that this study providesnovel mechanistic insights into the design and preparation of highly efficient photocatalysts withapplications in energy conversion and environmental remediation.
