        Cured becon meats were manufactured from bacon using dry curing method with varing levels of sodium chloride. To study the changes of moisture content, water activity, pH, salt concentration, extractability of proteins, SDS-PAGE and ATPase activity of protein were investigated during curing period at 3±1℃. During the process of curing, the moisture content and water activity in bacon meat decreased while the pH and salt concentration increased. Extractabilities of protein and SDS-PAGE shows that changes in extractability of water soluble protein are less noticeable while the extractability of salt soluble protein depended on curing period. These changes were rapid in the high level concentration of sodium chloride. The ATPase activity of myofibril from cured bacon with low level of salt concentration demonstrated a higher sensitivity to ionic strength than that of myofibril from cured bacon with high levels.

        Under the compressive loads, tensile stress fields are possibly induced according to the boundary condition of compressed body. At the crack tip, the possibility of occurance of tensile stress fields are strong even under the compressive loads. In this paper, the fracturing behavior of brittle materials containing crack are investigated and the compressive strength affected by crack clearance are inspected theoretically and experimentally. Upon the theoretical and experimental results, it is found that compressive strength is decreased as the crack clearance is increased.

        Nutritional evalution of proteins from soybean and soybean sprout and effect of methionine supplement to soybean sprout was studied by feeding albino rat. Sprague-Dawley strain of albino rats fed for 15 days with 5% (Group 1) and 10% (Group 2) level of protein from soybean, 10%(Group 3), 15% (Group 4) and 20%(Group 5) level of protein from soybean sprout, methionine supplement to soybean sprout (Group 6) and 10% level of protein form casein as a reference (Group 7). The results are as follows 1. Body weight gain was greater in soybean than soybean sprout in same protein level. Methionine supplementation increased weight gain significantly compared to soybean sprout alone. 2. By increasing protein level of soybean sprout, weight gain was increased significantly. 3. The more rat gained weight, the more rat in take food and there was significant correlationship between weight gain and food intake. 4. Protein intake was related to weight gain and there was significant positive correlationship between them. 5. PER was increased from 1.061 to 1.962 by adding methionine to soybean sprout, and PER increment was low by increasing the level of protein from soybean sprout in diet. There are no correlationship between PER and weight gain. 6. By adding methionine to soybean sprout, chemical score was increased. Chemical score showed significant positive correlationship with PER but not with weight gain. EAAI and RI was positively correlated significantly with PER.

      • 백화사설초 메탄올 추출물에 의한 HL-60 細胞 枯死過程에서의 cell cycle 關聯因子의 活性變化 硏究

        Objecives: Hedyotis diffusa is used to treat cancer in traditional Korea Medicine. So this study was carried out to examine the expression of cell cycle related genes in HL-60 cells undergoing apoptosis by the methanol extract of Hedyotis diffusa. Methods: 1. HL-60 cells were treated with various concentrations (from 200 to 50㎍/ml)of metnanol extract and H20 extract (200 ㎍/ml) of hedyotis diffusa. After 48 h later, the cells were tested for viability by MTT assay. 2. The HL-60 cells were treated with 200 ㎍/ml of methanol extract for the indicated periods. The whole cell lysates were prepared and analyzed by westem blotting using anti-p53 antibody. 3. The nuclear extract were prepared and analyed by western blotting using anti-p21 antibody, anti-p27 antibody, anti-cyclen A antibody, anti-cylin E antibody and anti-CDK2 antinbody. Results: 1. The methanol extract of Hedyotis diffusa induced the death of HL-60 cells in a dose dependent manner. 2. The methanol extract of Hedyotis diffusa makedly decreased the level of p21/Cipl and cyclin A in a time dependent manner. 3. The methanol extract of Hedyotis diffusa markedly increased tje ;eve; pf p27/Kip and cyclin E in a time dependent mammer. 4.The methanol extract of Hedyotis diffusa markedly did not affect the level of CDK2. Conclusions: These results provide evidence that expression of cell cycle related genes in HL-6- cells undergoing apoptosis by the methanol extract of Hedyotis diffusa mainly results from decreased level of p21/cipl and increased level of p27/Kipl of the cell cycle related genes.

      • 기체 방사성 물질 제거를 위한 첨착 활성탄 필터 시스템 성능에 관한 연구

        This paper presents an application of impregnated activated carbon and impregnated activated carbon filters. They are used for the removal of gaseous radioactive materials at radioactive material handling facilities such as nuclear power plants and research facilities. High adsorption efficiencies as well as stability are required for them to remove hazardous radioactive gases such as activated free iodine released during normal operation or accident condition. This research was initiated to enhance the independence of nuclear industry and raise the level of nuclear technology to that of the leading nuclear nations. By attaining the capability of design and development of HVAC system, national technological competitiveness can be achieved. Also, the ground for technology export will be hardened. Therefore, it is well timed to initiate the project to develope those filter systems domestically utilizing all the relevant technologies available.

      • 유방암에서 PCR을 이용한 p53 유전자의 Codon 175에서의 점돌연변이에 관한 연구

        The p53 gene, which is located in the short arm of chromosome 17, has been a constant source of fascination since its discovery at 1979. Allelic deletion coupled with mutation of the remaining allele is a theoretical hallmark of functional inactivation of tumor suppressor gene, and mutation and/or deletion of p53 gene is to be the most frequently observed genetic changes in breast cancer related to a single gene. In the present report, we have attempted to determine a point mutation at codon 175 which is known as one of four hot spots within highly conserved regions of the p53 gene where almost all of the reported mutations cluster A 304-bp fragment of exon 5 of p53 gene in fresh tissue of breast carcinoma was amplified by polymerase chain reaction (PCR), digested with Hha I restriction endonuclease, and electrophoresed. A point mutation was studied by observation of polymorphism with ethidium bromide staining under ultraviolet transillumination. The result revealed all of 14 carcinomas had wild-type p53 gene.

        Cured hams were manufactured using dry during method with 0, 1, 3, 5% levels of sodium chloride. The study was conducted to determine the effect of level of sodium chloride on moisture content, water activity, pH, salt concentration in cured meat, extractability of protein, SDS-PAGE and AT-Pase activity of myofibillar protein. During the process of curing, the moisture contents and water activity in cured ham meat decreased, while the pH and salt concentration incresed. The extractabilities of protein and SDS-PAGE shows that changes in extractactability of water soluble protein extracted from SSS-solution are less noticeable while the extractability of salt soluble proteins extracted from HS-and KI-solution increased. The ATPase activity of Myofibrillar protein from cured ham meat with low level of salt concentration demonstrated a higher sensitivity to ionic strength than that of myofibril from cured ham meat with high levels. These changes were rapid in the high level concentration of sodium chloride.
