        Inhibition of oxidative stress induced‑cytotoxicity by coptisine in V79‑4 Chinese hamster lung fibroblasts through the induction of Nrf‑2 mediated HO‑1 expression

        Background Coptisine is a natural alkaloid compound and is known to have multiple benefcial efects including antioxidant activity. However, whether it can protect lung fbroblasts from oxidative damage has not been studied yet. Objectives To investigate the potential inhibitory efect of coptisine against oxidative stress in V79-4 lung fbroblast cells. Methods V79-4 cells were treated with H2O2 (1 mM) in the presence or absence of coptisine (50 µg/ml), N-acetyl cysteine (NAC, 10 mM) or zinc protoporphyrin IX (ZnPP, 10 µM) for the indicated times. The alleviating efects of coptisine on cytotoxicity, cell cycle arrest, apoptosis, reactive oxygen species (ROS) production, DNA damage, mitochondrial dynamics, and inhibition of ATP production against H2O2 were investigated. Western blot analysis was used to analyze the expression levels of specifc proteins. Results Coptisine inhibited H2O2-induced cytotoxicity and DNA damage by blocking abnormal ROS generation. H2O2 treatment caused cell cycle arrest at the G2/M phase accompanied by increased expression of cyclin-dependent kinase (Cdk) inhibitor p21WAF1/CIP1 and decreased expression of cyclin B1 and cyclin A. However, these efects were attenuated in the presence of coptisine or NAC. Coptisine also prevented apoptosis by decreasing the rate of Bax/Bcl-2 expression in H2O2-stimulated cells and suppressing the loss of mitochondrial membrane potential and the cytosolic release of cytochrome c. In addition, the activation of nuclear factor-erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) was markedly promoted by coptisine in the presence of H2O2. However, zinc protoporphyrin IX, a potent inhibitor of HO-1, attenuated the ROS scavenging and anti-apoptotic efects of coptisine. Conclusions Based on current data, we suggest that coptisine can be used as a potential treatment for oxidative stress-related lung disease.

      • 수소 첨가에 의한 국내 식물성 유지 자원의 이화학적 변화

        This experiment was performed to study physical and chemical changes of edible five Korean oils during the hydrogenation. The condition used for hydrogenation was 0.05% Nickel catalyst under H2 pressure lkg/cm2 and temperature 180℃. Edible five Korean and corn oil. The purpose of this experiment was to suggest the possibility of practical usage of various Korean oils. Physicochemical change of the reaction products was examined by measuring iodine value, refractive index, melting point, solid fat index, fatty acid composition and trans isomer. The result obtained are as follows, 1.It was founds that soybean oil and corn oil could be replace by redpepper seed oil and ricebran oil under the same hydrogenation condition. 2.The result of this experiment could be used to find the desirable condition of hydrogention. 3.The erucic acid could not be hydrogenated therefore high content of erucic acid(43∼50%) in Korean rapeseed oil interfere hardening of rape seed oils in dead-end type laboratory reactor. The result of this experiment suggest that it is desirable to use rapeseed oil with low erucic acid contents.

        We estimated the self purification capacity of tidal flats in KanJin Bay, southwestern coast of Korea during flood tide and ebb tide in September, December, 1997 and March, 1998. The net flux of 17.8 gDIN/m²/yr with tidal current outflow to sea was estimated in September and 16.0 gDIN/m2/yr outflow to sea in December, while 0.57 gDIN/m²/yr influx into the tidal flat in March. The flux of 1.29 gDIP/m²/yr outflow to sea was estimated in September, while 2.64 gDIP/m2/yr and 5.90 gDIP/m²/yr influx into the tidal flat in December and in March, respectively. The SS flux of 0.849 kgSS/m²/yr and 5.28 kgSS/m²/yr outflow to sea were estimated in September and in March, while 12.0 kgSS/m²/yr inflow into the tidal flat in December. The net flux of 339 gCOD/m²/yr was inflow into the tidal flat was estimated in September, 254 gCOD/m²/yr in December and 63.7 gCOD/m²/yr in March, respectively. The capacity of COD purification of the tidal flats was 219 gCOD/m²/yr for a period of the diurnal tide.

      • 노인들의 신체활동과 인지능력간의 관계

        , The relationship between physical activity and cognitive performance was studied in the elderly women. Fifty volunteers (60-74 yrs) agreed to participate and were placed in 2 groups; exercise group(n=25; 68.72±4.22 yrs) and Non-exercise group(n=25; 69.52±5.1 yrs). The subjects who joined in the exercise group were asked to participate in exercise program(3 times/week, 80 min/day) for 12 weeks. The subjects n both groups were tested on the cognitive psychomotor performance (finger simple reaction time(SRT) and choice reaction time(CRT), tapping, and eye-hand coordination) at the beginning and the end of the 12-weeks program. For data analysis, mean and standard deviation scores were calculated, and ANOVAs and MANOVAs were used. There was an interaction between Group(2 exercise and non-exercise groups) and Time (pre- and post-tests) on all cognitive psychomotor performance tasks. Especially, each group was significantly interacted with Times on the performance of eye-hand coordination. After the exercise program, the exercise group was significantly better than the inactive group on all dependent variables (SRT and CRT, number of tappings, and performance scores of eye-hand coordination).

      • RT-LAMP를 이용한 콩황화일반모자이크바이러스의 진단

        Soybean yellow common mosaic virus (SYCMV) has been recently reported, it has been occurred a lot with Soybean mosaic virus (SMV) and Soybean yellow mottle mosaic virus (SYMMV) in soybean field. SYCMV belongs to genus of Sobemovirus and induced viral symptoms with yellowing, mottle and mosaic. A reverse transcription loop-mediated isothermal amplification (RT-LAMP) method allowed one-step detection of gene amplification by simple procedure and needed only a simple incubator for isothermal template. This RT-LAMP method allowed direct detection of RNA from virus-infected plants without thermal cycling and gel electrophoresis. In this study, we designed RT-LAMP primers named SYCML-F3/B3/FIP/BIP from coat protein gene sequence of SYCMV. After the reaction of RTLAMP, products were identified by electrophoresis and with the detective fluorescent dye, SYBR Green I. under daylight and UV light. Optimal reaction condition was at 63 for 60min and the primers of RTLAMP showed the specificity for only SYCMV tested in this study.

        동시출현단어분석을 통한 다문화 교육 연구의 구조 분석

        The purpose of this study was to analyze a structure of multicultural education studies by using co-word analysis. Data were 4,695 keywords from 1,685 multicultural education studies. First, after refining, keywords that occurred more than 15 times were selected. Second, using the selected keywords, a co-occurrence matrix and cosine similarity matrix were made. Third, hierarchical cluster analysis and network analysis were conducted. Results of the study are as follows. First, 6 clusters were classified by area of multicultural education; Language education(Cluster1), Teacher education(Cluster2), Intercultural education and Education for international understanding(Cluster3), Lifelong education(Cluster4), Civic education(Cluster5), Literature education(Cluster6). Second, 6 clusters were classified by ecological system in 3 factors; 1)Microsystem includes Cluster1(Multicultural families), Cluster2(Teacher), Cluster6(Literacy and Literature education), 2)Exosystem includes Cluster3(Multicultural society and policy), 3)Macrosystem includes Cluster4(Multiculture), Cluster5(Ism). Third, there were 4 carrier keywords between clusters; 1)Literature education (Cluster6) and Korean language education(Cluster1). 2)Bilingual education(Cluster1) and Multicultural society(Cluster3), 3)Multicultural society(Cluster3) and Social integration(Cluster5). Based on the results, the character of each cluster and recommendations for multicultural education are discussed.

        Most patients with aplastic anemia who do not respond to immunosuppressive treatment or are not candidates for bone marrow transplantation die of infection or bleeding. The neutropenia in acute leukemia, aplastic anemia, or occurring subsequently to chemotherapy and bone marrow transplantation increases susceptibility to infection. In general, the number of infectious episodes correlate with the degree and duration of neutropenia. Global immunosuppression produced by conditioning for bone marrow transplantation or graft-versus-host disease, is associated with unusual bacterial and fungal pathogens, or serious viral and protozoan infections. In addition, repeated treatment with broad-spectrum antibiotics is associated with the emergence of resistant organisms and fungal diseases because of the altered microbial microenvironment of the host. The incidence of invasive fungal infection caused by Saccharomycetes eerevisiae in immunosuppressed patients is very rare, compared with that of infection by candida or aspcrgillus species. Cases of Saccharomycetes cerevisiae fungemia occurring in the course of treatment with broad-spectrum antibiotics are reported in patients with extensive burn or with prosthetic valve endocarditis. We experienced a case of urinary tract infection by Saccharomycetes cerevisiae in a 27-year old female patient with severe aplastic anemia. We report the case with a review of relevant literatures.

      • 진행성 폐 종양 환자에서 종양 내 OK-432 주입술의 효과

        Background: Intratumoral injection of OK-432 produced significant antitumor effects. There are reports that OK-432 was injected directly into tumor tissue on gastrointestinal tracts and HCC tissue, producing decrease in serum alpha-fetoprotein level. We adopted this technique for treating locally advanced lung cancer. The object of this study is to evaluate the local therapeutic efficacy, side effects of injection for treating symptomatic mass. Methods: Twenty lung cancer patients (13 males and 7 females) underwent sonography guided OK-432 injection. We analyzed relationships between response of OK-432 and response of skin test, histology and age. Pain that patients feel was evaluated by VAS (Visual Analog scale). Results: A significant decrease of tumor size was observed. The mean pre-injection diameter of the lesions was 54.7±11.3mm, PR rate were 15.0% (3/20) and minimal change (50%<tumor size decreased.) patients were 35.0% (7/20). The VAS score before injecting OK-432 into the lesion was 5±3, this was decreased to 2.5±1.5 (p=0.04). Object response was significantly correlated with skin response, adenocarcinoma, and age. Conclusion: A significant decrease of tumor size and pain was observed. So OK-432 is a valuable treatment option for patients with symptomatic lesion from advanced lung cancer who are not amenable to further therapy.

      • 고속철도 터널출구로부터 방출되는 충격성 소음에 관한 모형실험

        As a railway train enters a tunnel at high speed, a compression wave is formed in front of the train and propagates along the tunnel. The compression wave subsequently emerges from the exit of the tunnel and generates a pulse wave, which causes an impulsive noise. In order to clarify the acoustic characteristics of the noises, in the present study experiment was carried out by using the pulse waves emitted from an open end of a shock tube. Various pulse waves were obtained by controlling the compression wave formed in the tube. The results show that as the compression wave formed at tunnel exit becomes strong, the pulse wave emitted propagates with strong directional characteristics forward the axial direction of the tube, and that attenuation of the pulse wave propagating a ground surface can be reasonably predicted by a linear acoustic theory. Furthermore, the impulsive noise at far-field from the tube exit results in a sound level reduction of 6dB with a doubling of the propagating distance.

        The incidence of hematologic disorders in patients with Down's syndrome is significantly increased, about 14∼30 - fold higher than in general population and includes neonatal transient abnormal myelopoieis and acute leukemias. The age of onset of leukemia in Down's syndrome is peaking first in the newborn period and then under 4 years of age. Down's syndrome with acute leukemia above the age of 20 is very rare and it's treatment oucome is unclear. The treatment of Down's syndrome with leukemia has been controversial because of toxicity and associated congenital cardiac and other abnormalities. But if treated adequately, children with Down's syndrome show a favorable response to anti-leukemia therapy. A 24-year-old man with Down's syndrome was first seen for the evaluation of anemia and thrombocytopenia. The peripheral blood morphology and bone marrow study revealed acute myelogenous leukemia, cytogenetic study of bone marrow showed trisomy 21. Beacuse of his sicioeconomic condition and medical abnormalities including deafness, visual loss, he was treated with low dose subcutaneous cytosine arabinoside(Ara-C) for 11 days. Complete remission was obtained after 37 days. The complete remission lasted for 5 months. He subsequently relapsed, and died 6 months later.
