        Enantiomeric separation of naproxen is necessary in drug evaluation processes as the pharmacological activity of naproxen resides mainly in the (S)-enantiomer, The enantioseparation of naproxen was carried out with three different stationary phases. The methods were validated with the terms of linearity, intraday and interday precision, accuracy, selectivity and recovery. Among the selected stationary phases, ChiroSil DMB column with hexane : t-butylmethylether : acetic acid (60:40:0.1, v/v/v) as mobile phase showed the best enantioseparation of the naproxen in the commercial preparations. The wavelength for detection was 270 nm and flow rate was 2.0 mL/min. The contents of (S)-naproxen in the commercial preparations were 92.7 - 98.3%, indicating that all tested preparations were satisfactory to current regulation. However, the contents of (R)-naproxen as impurity in the commercial preparations were varied from 0.08% to 1.08%.

        This study was performed to evaluation the skin and eye irritation of etofenprox 8% and spinosad 4% combination in male New Zealand White rabbits. In skin irritation test, the combination of etofenprox and spinosad did not showed any change of body weight but caused slight skin irritation with the P.I.I. (primary irritation index) value 0.25. The etofenprox and spinosad combination showed severe eye irritation 1 h, 24 h, 48 h, and 72 h after ocular treatment, having the A.O.I. (acute ocular irritation) value of 37.3. Therefore, these result indicated that the combination of etofenprox and spinosad might be irritant formulation on eye and skin.

      • Bicluster and Pathway Enrichment Analysis of HCV-induced Cirrhosis and Hepatocellular Carcinoma

        Background: Hepatocellular carcinoma (HCC) is the sixth most common cancer worldwide and the most common form of liver cancer. However, while it is associated frequently with hepatitis C virus (HCV) there is only an elementary understanding of its molecular pathogenesis. Methods: To gain insight into the molecular mechanisms of HCV-induced hepatocarcinogenesis, we performed microarray analysis on 75 surgical liver samples from 48 HCV-infected patients. Results: There were 395 differentially expressed geness between cirrhotic samples and HCC samples. Of these, 125 genes were up-regulated and 270 genes were down-regulated. We performed pathway enrichment analysis and screened as described previously. Conclusions: The differentially expressed genes might be involved in hepatocarcinogenesis through upregulating the pathways of ECM-receptor interaction, focal adhesion, cell adhesion molecules and other cancer-related pathways, and downregulating the pathways of "complement and coagulation cascades". We hope our results could aid in seeking of therapeutic targets for HCV-induced hepatocellular carcinoma.

        Random vibration analysis of structures by a time-domain explicit formulation method

        Non-stationary random vibration of linear structures with uncertain parameters is investigated in this paper. A time-domain explicit formulation method is first presented for dynamic response analysis of deterministic structures subjected to non-stationary random excitations. The method is then employed to predict the random responses of a structure with given values of structural parameters, which are used to fit the conditional expectations of responses with relation to the structural random parameters by the response surface technique. Based on the total expectation theorem, the known conditional expectations are averaged to yield the random responses of stochastic structures as the total expectations. A numerical example involving a frame structure is investigated to illustrate the effectiveness of the present approach by comparison with the power spectrum method and the Monte Carlo simulation method. The proposed method is also applied to non-stationary random seismic analysis of a practical arch bridge with structural uncertainties, indicating the feasibility of the present approach for analysis of complex structures.

        Comparison of Two Site-Specifically <sup>18</sup>F-Labeled Affibodies for PET Imaging of EGFR Positive Tumors

        <P>The epidermal growth factor receptor (EGFR) serves as an attractive target for cancer molecular imaging and therapy. Our previous positron emission tomography (PET) studies showed that the EGFR-targeting affibody molecules <SUP>64</SUP>Cu-DOTA-Z<SUB>EGFR:1907</SUB> and <SUP>18</SUP>F-FBEM-Z<SUB>EGFR:1907</SUB> can discriminate between high and low EGFR-expression tumors and have the potential for patient selection for EGFR-targeted therapy. Compared with <SUP>64</SUP>Cu, <SUP>18</SUP>F may improve imaging of EGFR-expression and is more suitable for clinical application, but the labeling reaction of <SUP>18</SUP>F-FBEM-Z<SUB>EGFR:1907</SUB> requires a long synthesis time. The aim of the present study is to develop a new generation of <SUP>18</SUP>F labeled affibody probes (Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> and <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB>) and to determine whether they are suitable agents for imaging of EGFR expression. The first approach consisted of conjugating Z<SUB>EGFR:1907</SUB> with NOTA and radiolabeling with Al<SUP>18</SUP>F to produce Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB>. In a second approach the prosthetic group <SUP>18</SUP>F-labeled-2-cyanobenzothiazole (<SUP>18</SUP>F-CBT) was conjugated to Cys-Z<SUB>EGFR:1907</SUB> to produce <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB>. Binding affinity and specificity of Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> and <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB> to EGFR were evaluated using A431 cells. Biodistribution and PET studies were conducted on mice bearing A431 xenografts after injection of Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> or <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB> with or without coinjection of unlabeled affibody proteins. The radiosyntheses of Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> and <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB> were completed successfully within 40 and 120 min with a decay-corrected yield of 15% and 41% using a 2-step, 1-pot reaction and 2-step, 2-pot reaction, respectively. Both probes bound to EGFR with low nanomolar affinity in A431 cells. Although <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB> showed instability <I>in vivo</I>, biodistribution studies revealed rapid and high tumor accumulation and quick clearance from normal tissues except the bones. In contrast, Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> demonstrated high <I>in vitro</I> and <I>in vivo</I> stability, high tumor uptake, and relative low uptake in most of the normal organs except the liver and kidneys at 3 h after injection. The specificity of both probes for A431 tumors was confirmed by their lower uptake on coinjection of unlabeled affibody. PET studies showed that Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> and <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB> could clearly identify EGFR positive tumors with good contrast. Two strategies for <SUP>18</SUP>F-labeling of affibody molecules were successfully developed as two model platforms using NOTA or CBT coupling to affibody molecules that contain an N-terminal cysteine. Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> and <SUP>18</SUP>F-CBT-Z<SUB>EGFR:1907</SUB> can be reliably obtained in a relatively short time. Biodistribution and PET studies demonstrated that Al<SUP>18</SUP>F-NOTA-Z<SUB>EGFR:1907</SUB> is a promising PET probe for imaging EGFR expression in living mice.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/mpohbp/2014/mpohbp.2014.11.issue-11/mp5003043/production/images/medium/mp-2014-003043_0008.gif'></P>

      • A Simple Partial Discharge Detector for Low-Voltage Rotating Electrical Machines

        A new, simple partial discharge (PD) detector for low-voltage rotating electrical machines using acoustical emission (AE) technique is dseveloped in this study. Common electric components were used in the detector, which reduces the cost of inspection, comparing with the traditional PD detection methods that use expensive equipment costing from tens of thousands to several millions dollars. Experimental results by resonant type AE sensors (150 ㎑) which utilize power line-cycle in a microcontroller unit (MCU) as reference to measure the PDs generated in a low-voltage motor are presented. The AE signals are then amplified by a pre-amplifier (30 ㎑ ~ 300 ㎑, 34 ㏈). Since the resonant frequency of the sensor is much lower than that of the electromagnetic (EM) interferences around the motor, the effects of noise is substantially reduced by this method. In the mean time, the use of 150-㎑ resonant type AE sensor also avoids the disturbance of mechanical vibration noise. According to the experiment results, the measurement system developed in this study can be used to detect the PDs’ AE signals correctly. The AE measurement scheme proposed in this study provides an effective, low-cost method for PD measurements.
