      • The Use of Collagen Content as Determined by Spectral Domain Polarization-Sensitive Optical Coherence Tomography to Assess Colon Anastomosis Healing in a Rat Model

        <P>Abstract</P><P><B><I>Background/Purpose:</I></B> Many studies have been undertaken to prevent anastomosis leakage of the colon, and several methods have been used to assess anastomosis healing, such as measurement of bursting pressure or hydroxyproline (a marker of collagen) content at the anastomosis site. However, these methods are inappropriate for comparing anastomosis healing at two time points in the same animals. In the present study, we measured the collagen level by spectral domain polarization-sensitive optical coherence tomography (SD-PS-OCT) to assess anastomosis healing. <B><I>Methods:</I></B> Sprague-Dawley rats were divided into groups C (saline-administered controls; study group) and M [a 5-fluorouracil (5-FU)-administered experimental group]. Immediately after end-to-end anastomosis of the colon, SD-PS-OCT images of anastomoses were taken (baseline). Animals were administered saline or 5-FU for 7 days. On the 7th postoperative day, SD-PS-OCT images were acquired, a histopathologic exam was performed, and hydroxyproline levels as well as mRNA expressions of collagen-1 and collagen-3 were measured at the anastomosis site. <B><I>Results:</I></B> Fibroblast proliferation and inflammatory cell infiltration were greater in group C than in group M. The mRNA expressions of collagen-1 and collagen-3 were substantially higher in group C. Hydroxyproline levels were higher in group M than in group C. Though collagen levels measured by SD-PS-OCT at 7 days were elevated compared with baseline in group C, no such changes were observed for group M. <B><I>Conclusion:</I></B> Collagen levels at the colon anastomosis site, measured with SD-PS-OCT, were not increased at 7 days postoperatively versus baseline when 5-FU was injected, but were increased in saline-treated controls. The measurement of collagen content by SD-PS-OCT was found to provide a good means of assessing anastomosis healing, because it allows in situ assessment of collagen contents at baseline and during the postoperative period.</P><P>© 2014 S.       • Peroxiredoxin 6 expression is inversely correlated with nuclear factor-κB activation during Clonorchis sinensis infestation

        Clonorchis sinensis is a carcinogenic human liver fluke. Its infection promotes persistent oxidative stress and chronic inflammation environments in the bile duct and surrounding liver tissues owing to direct contact with worms and their excretory-secretory products (ESPs), provoking epithelial hyperplasia, periductal fibrosis, and cholangiocarcinogenesis. We examined the reciprocal regulation of two ESP-induced redox-active proteins, NF-κB and peroxiredoxin 6 (Prdx6), during C. sinensis infection. Prdx6 overexpression suppressed intracellular free-radical generation by inhibiting NADPH oxidase2 and inducible nitric oxide synthase activation in the ESP-treated cholangiocarcinoma cells, substantially attenuating NF-κB-mediated inflammation. NF-κB overexpression decreased Prdx6 transcription levels by binding to two κB sites within the promoter. This transcriptional repression was compensated for by other ESP-induced redox-active transcription factors, including erythroid 2-related factor 2 (Nrf2), hypoxia inducible factor 1α (HIF1α), and CCAAT/enhancer-binding protein β (C/EBPβ). Distribution of immunoreactive Prdx6 and NF-κB was distinct in the early stages of infection in mouse livers but shared concomitant localization in the later stages. The intensity and extent of their immunoreactive staining in infected mouse livers are proportional to lesion severity and infection duration. The constitutive elevations of Prdx6 and NF-κB during C. sinensis infection may be associated with more severe persistent hepatobiliary abnormalities mediated by clonorchiasis.

      • Boro-Nitriding 처리한 FC-25계 편상흑연주철의 표면특성에 관한 연구

        Surface properties and oxidation behaviors were studied for FC-25 gray cast iron (GCI), plasma nitrided FC-25 GCI (P-N GCI), powder boronized FC-25 GCI (B GCI) and powder boronized N GCI (B-N GCI). A boride (Fe2B, FeB) layer with a tooth shape was formed by boronizing GCI at temperatures of about 650℃. The thickness and hardness of the layers were proportional to the boronizing temperature and time. The hardness was decreased by increasing boronizing depth. The activation energy for boride formation was 73.8kJ/mol. The thickness of the graphite layer formed by boronizing was increased by the temperature and time. A silicon layer insoluble in borides was accumulated under the boride layer. The major phases were Fe4N, Fe3N and FeN in a nitride layer formed by plasma. The thickness and hardness of the layer were increased with nitriding temperatures and time under the Fe4N decomposition temperature of 680℃. The hardness was maximum at the surface and decreased with nitrided depth on B-N GCI at 700℃, above the decomposition temperature of the Fe4N. Hardness of B-N GCI at the layer depth of 10 ㎛ was ≒1000Hv, which was lower than ≒1500Hv of the B GCI at the same condition. Also the layer thickness was 50 ㎛; deeper than that of B GCI by 10 ㎛ The reasons were that the nitrogen given off by the decomposition made the layer porous and facilitated the diffusion of oxygen and other reactants. The samples were oxidated at 800℃ for 48hrs under atmospheric air. Oxidation rate was in decreasing order of N GCI, GCI, B GCI and G-N GCI. Oxidation rates were greatly influenced by the diffusion of nitrogen formed by Fe4N decomposition over the decomposition temperature of 680℃ and by the oxidation barrier of the less corrosive boride layer.

        The kinematics of the Seyfert 2 galaxy NGC 5728 circumnuclear region

        <P>ABSTRACT</P><P>We secured spectral images of the circumnuclear, 12 × 10 arcsec<SUP>2</SUP> region of the Seyfert 2 galaxy NGC 5728 with the Optically Adaptive System for Imaging Spectroscopy at the Canada–France–Hawaii Telescope 3.6-m telescope. The radial velocity distribution of the two bright zones, main central (C) core and north-west (NW) core, and strategically important points along the position angles, PA 33°, 90°, 123° and 143°, are analysed. The large-scale reconstructed monochromatic images of the Hα, Hβ, [O <SMALL>III</SMALL>] and [N <SMALL>II</SMALL>] lines show a bipolar morphology along PA ∼ 123°. The prominent elliptical ring is seen in the Hα and Hβ monochromatic images. Our analysis implies that this ring is rotating in the same clockwise direction as that of the galactic disc; this elliptical ring is in fact a projected result of a circular ring of radius ∼5.4 arcsec, with <I>i</I>∼ 50° (nearly aligned with the host galactic plane).</P><P>We used the [O <SMALL>III</SMALL>]5007 line profiles for a detailed analysis of kinematics in the circumnuclear region of the NGC 5728. The [O <SMALL>III</SMALL>]5007 line profiles reveal distinct kinematical structures. (1) The strongest component of Doppler peaks: two core components, consisting of the receding main C-core and the approaching NW-core with the radial velocities, <I>V</I><SUB>r</SUB>≃+250 and −240 (or −250) km s<SUP>−1</SUP>, respectively. These two cores consist of a roughly spherically symmetric distribution of the gas with radii of <I>r</I>∼ 1.0 arcsec (at <I>V</I><SUB>r</SUB>≃+240 km s<SUP>−1</SUP>) and 0.9 arcsec (at <I>V</I><SUB>r</SUB>≃−240 km s<SUP>−1</SUP>), respectively. Hints of the presence of inflows around these cores are observed. (2) The next strong line profile component: this component corresponds to the bipolar cone aligned PA ∼ 123°. (3) The reverse S-shaped kinematical boundary which runs through the double-peaked zone of 6–7 arcsec long in the galactic major axis direction is likely to be caused by a combination of the disc rotation of the host galaxy and the revolution of the two central cores.</P>

      • Enhancement of contraction and L-type Ca<sup>2+</sup> current by murrayafoline-A via protein kinase C in rat ventricular myocytes

        <P>We previously reported that murrayafoline-A (1-methoxy-3-methyl-9H-carbazole, Mu-A) increases the contractility of ventricular myocytes, in part, via enhancing Ca2+ influx through L-type Ca2+ channels, and that it increases the Ca2+ transients by activation of protein kinase C (PKC). In the present study, we further examined the cellular mechanisms for the enhancement of contractility and L-type Ca2+ current (I-ca,I-L) by Mu-A. Cell shortening and I-Ca,I-L were measured in rat ventricular myocytes using a video edge detection method and perforated patch-clamp technique, respectively. We found that the positive inotropic effect of Mu-A was not affected by pre-exposure to the beta-adrenoceptor antagonist propranolol, the protein kinase A (PIGS) inhibitors KT5720 or H-89, or the phospholipase C inhibitor U73122. Interestingly, the Mu-A-mediated increases in cell shortening and in the rate of contraction were completely suppressed by pre-treatment with the PKC inhibitor GF109203X. The stimulatory effect of Mu-A on I-Ca,I-L was not altered by inhibition of PKA (KT5720), G-protein coupled receptors (suramin), or alpha(1)-adrenoceptor (prazosin). However, pre-exposure to the PKC inhibitor, GF109203X or chelerythrine, abolished the Mu-A-induced increase in I-Ca,I-L. Pre-exposure to the Ca2+-calmodulin-dependent protein kinase II (CaMKII) inhibitor KN93 slightly reduced the stimulatory effects on contraction and I-Ca,I-L by Mu-A. Phosphorylation of PKC was enhanced by Mu-A in ventricular myocytes. These data suggest that Mu-A increases contraction and I-Ca,I-L via PKC in rat ventricular myocytes, and that the PKC-mediated responses in the presence of Mu-A may be partly mediated by CaMKII.         Purpose: This study aimed to know whether there is a comparison of perception, knowledge and health promoting behaviors about particular matter between nursing students and non-health departments college students. Methods: A quantitative research is used. Data were collected from August 1 to 19. The study sample was nursing students and non-health departments students. For the similar condition of samples, this study excluded whose first language is not Korean. Independent T-test and pearson’s correlation was used for this study. Results: A significant positive correlation did not existed between difference of major and perception(t=0.017, p=0.986), knwledge(t=0.652, p=.516) and health promoting behaviors(t=-0.114, p=.910) about particular matters. Conclusion: Based on the study results, there is no significant correlation of perception and actions about particular matters between nursing students and non-health department college students.

        韓牛 牝牛의 Body Condition Score가 繁殖形質에 미치는 影響

        Data from five Hanwoo breeding farms were collected to analyze the effects of parity, season, nutrition and reproduction on body conditions of heifers and cows and to analyze the relationships of body conditions with reproductive performances and body growth. Analytic models included farms and parity which were run through SAS to estimate least squares means and correlation coefficients between traits - body condition scores(BCS) of 1(very emaciated) through 9(very obese) scale at service and at calving, calving interval, days to rebreeding, gestation length, number of services per conception. Overall averages were 4.55 for BCS at service, 5.42 for BCS at calving, 406.7days of calving interval, 287.7days of gestation length, 66.2days from calving to first service and 1.78 times of services per conception. Number of services per conception was 1.50~1.74 times for the cows of BCS 5 or under and 2.00~3.00 for those of BCS 6 or higher. Body weight of calves from cows observed averaged 23.3㎏ at birth and 70.7㎏ at weaning. 57 percent of cows showed BCS 4~5 at service while 46 percent of those showed 5~6 at calving. Extimated phenotypic correlation coefficients of BCS at service with BCS at calving, calving interval, gestation length, number of services per conception were low but positive: 0.16, 0.26, 0.08 and 0.06, respectively. Phenotypic correlations of BCS at calving with calving interval, gestation length and number of services per conception were estimated to be also low and positive: 0.10, 0.13 and 0.10, respectively. However, phenotypic correlation between calving interval and gestation length, and between gestation length and number of services per conception were negative but low as - 0.11 and -0.13 each.
