        Valuation of ecosystem services in the organic carbon of the Pinus densiflora forest at Mt. Namsan, Seoul Metropolitan City

        Background: Standing biomass, litter production, rate of litter decomposition, amount of litter on forest floor, and amount of soil organic carbon distribution were investigated from March 2016 to November 2018 in order to evaluate the ecosystem value through organic carbon distribution in the Pinus densiflora forest at Namsan, Seoul Metropolitan City. Results: The amount of organic carbon in the Pinus densiflora forest was 261.09 ton C ha<sup>-1</sup> during the research period, and fixed organic carbon in plant through net photosynthesis was 3.2 ton C ha<sup>-1</sup> year<sup>-1</sup>. The organic carbon in plant was 62.77 ton C ha<sup>-1</sup> (24.04%), in litter on the forest floor was 3.65 ton C ha<sup>-1</sup> (1.40%), and in soil was 194.67 ton C ha<sup>-1</sup> (74.56%). The value of plant, litter on forest floor, and soil organic carbon distribution were each 6,277,000 won ha<sup>-1</sup>, 365, 000 won ha<sup>-1</sup>, and 19,467,000 won ha<sup>-1</sup>. The value per ton of fixed organic carbon in plant through net photosynthesis was 320,000 won ha<sup>-1</sup> year<sup>-1</sup> and the value of ecosystem services stored in carbon in the Namsan forest ecosystem was about 26.1 million won ha<sup>-1</sup> for 3 years. Conclusions: The results of this study suggest that the pine forests of Namsan rapidly decompose leaves due to the high atmospheric temperature and accumulate a large amount of organic carbon in the soil to provide climate control regulatory service function.

        남산의 소나무림, 신갈나무림, 아까시나무림의 낙엽생산과 토양유기탄소 동태

        남산의 소나무림, 신갈나무림 그리고 아까시나무림에서 2008년 1월부터 2009년 12월까지의 월별 낙엽생산량과 토양의 유기탄소량을 조사하였다. 소나무림은 2008년과 2009년 모두 10월에 낙엽생산량이 가장 많았으며, 2008년 1월과 2009년 12월에 낙엽생산량이 가장 적었다. 신갈나무림은 2008년과 2009년 모두 11월에 낙엽생산량이 가장 많았으며, 2월에 낙엽생산량이 가장 적었다. 아까시나무림은 2008년 11월과 2009년 10월에 낙엽생산량이 가장 많았으며, 2008년 1월과 2009년 12월에 낙엽생산량이 가장 적었다. 이것은 한반도 중부에서 상록성의 소나무림이 낙엽성의 참나무숲 보다 더 일찍 낙엽이 진다는 것을 의미한다. 소나무림과 신갈나무림, 아까시나무림의 2년 평균의 낙엽생산량은 7.07, 6.36, 5.02 ton ha-1로 소나무가 가장 많았다. 소나무림과 신갈나무림, 아까시나무림의 2년 평균의 토양유기탄소량은 76.2, 68.6, 72.5 ton C ha-1로 소나무림에서 가장 많았다. 이처럼 소나무숲에서 높게 나타난 것은 남산에서 신갈나무나 아까시나무를 벌목하여 임목밀도가 줄었기 때문이다. The objective of this study was to estimate dynamics of litter production and soil organic carbon of Pinus densiflora forest, Quercus mongolica forest, and Rhobina pseudo-acasia forest at Mt. Nam as a part of Korea National Long-Term Ecological Research (KNLTER) from 2008 to 2009. Litter production of P. densiflora forest was the highest in October 2008, 2009 and the lowest in January 2008 and December 2009. Litter production of Q. mongolica forest was the highest in November and the lowest in February in 2008 and 2009. Litter production of R. pseudo-acacia forest was the highest in November in 2008 and October in 2009 and the lowest in January in 2008 and December in 2009. It means that leaves of P. densiflora forest shed earlier than deciduous oak forests in Korean central region. An average of litter production for 2 years was 7.07, 6.36, 4.66 ton ha-1 in P. densiflora forest, Q. mongolica forest, R. psuedo-acacia forest, respectively. An average of soil organic carbon matter for 2 years was 88.3, 76.5, 84.2 ton ha-1 in P. densiflora forest, Q. mongolica forest, R. psuedo-acacia forest, respectively.

      • 열교환기 구조 변화에 따른 NH₃와 R22의 성능특성연구

        Nowadays HCFCs refrigerant are restricted because it cause depletion of ozone layer. However, natural gases such as ammonia as an organic compound, propane and propylene as hydrocarbon are easy and cheap to obtain as well as environmental. Accordingly, this experiment apply the NH3 and R22 to study the performance characteristic from the superheat control and compare the energy efficiency of two refrigerants from the high performance. The condensing pressure of refrigeration system is increased from 15bar to 16bar and degree of superheat is increased from 0 to 10℃ at each condensing pressure. As the result of experiment, when comparing the each COP, we knew the NH₃ is suitable as the alternative refrigerant of the R22.

      • Vinblastine이 백서 배양 Type-1 별아교세포에 미치는 세포독성에 관한 연구

        It has been reported that vinblastine, anticancer drug, is a neuroteratogen inducing various neural malformations such as microcephaly and neural tube defect(NTD), but the study on the toxicity of neuroglial cells, especially type-1 astrocytes using culture system is not well established. In order to evaluate the cytotoxicity of vinblastine on cultured type-1 astrocytes of neonatal rat brain in vitro. MTT assay and also light and electron microscopic studies were carried out. The results were as follows: 1. MTT_90 and MTT_50 in cultured type-1 astrocytes were 1×10 exp (-1) μM and 1×10 exp (2)μM of vinblastine, respectively. 2. Vinblastine was highly toxic in cultured type-1 astrocytes(MTT_50 ≤ 100 μM). 3. In a light microscopy, cultured type-1 astrocytes showed decrease of cells in number, cytoplasmic perforation, vacuolization and cytoplasmic granulation after cultured type-1 astrocytes were treated with 1×10 exp (2)μM of vinblastine for 24 hours. Cultured type-1 astrocytes damaged by vinblastine showed degenerative changes of cells morphologically. 4. In an electron microscopy, increment of glial fibrillary acidic protein(GFAP), increased free-ribosomes, cisternal dilatation of rough endoplasmic reticulum (rER). few dense bodies and vacuoles were shown in cultured type-1 astrocytes treated with 1×10 exp (2)μM of vinblastine for 24 hours. The results indicate that vinblastine has markedly cytotoxic effect on the type-1 astrocytes of neonatal rat brain in vitro study.

      • 국산밀과 수입밀의 이화학적 특성

        To compare chemical characteristics of domestic wheat flour and imported one. we determined moisture. crude protein, ash, and crude lipid. In the case of moisture content, domestic crude wheat flour has shown 10.84%, the lowest value. while that of imported wheat flour has shown 13.33∼14.19%. In crude protein content, domestic fine wheat flour shown 11.69%. and domestic crude has shown 14.34%, while that of imported strong, medium, weak wheat flour has shown 14.30, 11.78, and 9.28%, respectively. In ash content. domestic crude wheat flour has shown 1.50%, and that of domestic fine one has shown 0.74%, while that of imported one has shown 9.85~11.12%. In color analysis. domestic fine wheat flour has shown similar lightness as that of imported strong wheat flour.

        단계적 온도 하강법을 이용한 췌도세포 냉동보존법

        연구배경:최근 당뇨병의 새로운 치료법으로 시도되고 있는 췌도이식은 충분한 췌도수의 확보와 췌도생존율을 높이기 위한 면역억제제 사용이 제한점이 되고 있다. 본 연구의 목적은 이식 전 충분한 췌도 수의 확보를 위해 분리한 췌도를 냉동보존하는 방법을 확립하고 냉동보존한 백서 췌도세포의 시험관내 그리고 생체내 기능을 조사하였다. 방법:분리한 백서의 췌장소도를 48시간 배양한 후 한 시험관당 췌도세포 1000개씩 나누었다. 냉동보존은 6개의 시험관에 DMSO를 첨가한 후 초 냉각(supercooling), 핵화(nucleation)단계를 거친 후 99% isopropanol과 액체질소가 들어있는 dewer를 이용하여-0.25℃/분의 냉각속도로 -40℃까지 단계적으로 얼린후-70℃ 액체질소 탱크에 보관했다. 해동은 냉동시킨 vial들을 액체 질소 태으에서 꺼내 37℃ 항온조에 담가 급격히 해동시킨 후, 원심분리하여 상층액을 제거하고 각 vial에 0.75M sucrose 용액을 가한 후, 10% fetal calf serum이 함유된 RPMI 1640 media에서 배양하였다 각각 6개의 시험관에서 해동한 췌도들을 광학현미경 및 형광현미경하에서 췌도의 모양과 생존율에 대해 조사하고 인슐린 정적반응을 알아보았다. 또한 분리한 췌도를 냉동보존하지 않고 이식한 경우를 대조군(6마리)과 생체내 기능을 비교하였다. 결과:① 냉동보존후 획득한 췌도의 수와 생존율 해동후 획득한 췌도의 수는 해동시킨 당일날이 902±21, 24시간 배양 후에는 857±16, 72시간 후에는 817±18개로 점차 감소되었다. AO/PI 염색상 각 췌도의 생존율은 냉동 전을 100으로 하였을 때 해동당일, 24시간 후, 72시간 후가 각각 60±5, 80±5, 90±5%로 해동후 3일간 배양하였을 때 냉동전의 수준으로 회복하였다. ② 냉동보존후 췌도의 포도당에 대한 정적 인슐린 분비능:냉동직후 감소된 경향을 보였으나 해동후 3일간 배양한 췌도의 인슐린 분비는 냉동전과 통계적으로 의미있는 차이가 없이 냉동보존 전의 수준으로 회복되었다. ③ 냉동보존후 췌도의 포도당에 대한 동적 인슐린 분비능:냉동보존한 췌도를 해동후 3일째의 인슐린 동적 분비능은 냉동 전과 마찬가지로 자극 인슐린의 반응의 제1기와 2기가 잘 관찰되었다. ④ 냉동보존한 췌도세포 이식 후 혈당 변화:스트렙토조토신으로 유도된 당뇨병 쥐에 췌도이식 후 혈당은 냉동보존한 췌도이식군에서 대조군에 비해 혈당의 조절효과가 더 오래 지속되었다. 결론:소동물에서 단계적 온도 하강법을 이용한 췌도세포 냉동보존법을 확립하였으며 이는 기능, 구조 및 생존율에 큰 이상을 보이지 않았으므로 장차 사람의 췌도세포 동종이식시 부족한 췌도세포수를 극복하고 면역반응을 줄일 수 있는 매우 유용한 방법이 될 것으로 판단된다. Background : Although islet transplantation has been attempted to reverse the state of diabetes, achieving a critical number of islets and modulating the immune response limit the success of islet transplantation. Cryo-preservation of islets offers many important benefits for islet transplantation by collecting islets with a wide variety of HLA phenotypes and islet MHC expression. The aims of this study was to determine the optimal conditions for cryo-preservation by using a controlled cooling method and to evaluate in vitro and in vivo functional properties of the cryo-preserved islets. Methods : Collagenase-isolated, Ficoll-purified islets were cultured for 48 hours. They were aliquoted into freezing tubes (1000 islets per tube), equilibrated with 2M dimethyl sulfoxide (DMSO) in three steps, supercooled, nucleated, and controll-cooled at rate of 0.25℃/min to - 40℃ prior to storage at - 196℃. Rapid thawing and removal of DMSO with 0.75 M sucrose preceded 48 hour of culture and the morphology, viability, glucose-induced insulin secretion, and in vivo function of rats transplanted with cryopreserved islets was reexamined. Results : ① Recovery was 90.2±0.2%, 85.7±0.1% and 81.7±0.1% immendiately after, 24 hours and 72 hours after thawing respectively. The viability was 60±5%, 80±5%, 90±5% immediately after, 24 hours and 72 hours after thawing respectively. ② The glucose-stimulated-insulin secretion (GSIS) tended to decrease immediately after thawing, but GSIS increase to the level of pre-cryopreservation 72 hours after thawing. ③ The in dynamic GSIS, the first and the second phase of insulin secretion were well preserved in islets cultured for 72 hours after thawing. ④ The cryopreserved islets were cultured for 3 days and transplanted into renal sub-capsular space of streptozotocin (STZ) induced diabetic rats. The duration of normoglycemia in the STZ-induced diabetic rats transplanted with cryopreserved islets was significantly longer than of the fresh islets. Conclusion : The optimal condition of cryopreservation using the controlled cooling method was established in rat pancreatic islets. This cryopreservation method can be a feasible approach for human islet transplantation (J Kor Diabetes Asso 26:64~74, 2002).

      • 강선 이음형 PC 보-기둥 내부 접합부의 구조적 거동에 관한 실험적 연구

        In this study, new moment-resisting precast concrete beam-column joint is proposed for moderate seismic regions. it is attempt to devise connection detail which makes workability improve and confirm effective stress transfer in the region of connection. It has the connection details which help working process simplified and construction cost reduced. as replace 9.3㎜, 7strand for reinforcement. Fore specimens for partial tension test and three specimens for beam-column interior joint test were producted. An experimental study was carried out to investigate the joint behavior subjected to reverse cyclic loading and constant axial compressive load. The test variables was transverse reinforcement in joint. Structural performance of joint is evaluated on the basis of connection strength, stiffness, energy dissipation, and drift capacity. Based on the test results, the precast connection is capable of matching or exceeding the performance of the monolithic connection and thereby provides the sufficient moment-resisting behavior to be applicable for moderate seismic regions.

      • 대장암종에서 암관련 유전자들의 Promoter Methylation과 암종 발생 및 진행과의 관게에 대한 연구

        The colorectal corcinoma is one of the most common malignant tumor in Korea and its carcinogenic mechanism has not been elucidated exactly yet. So far, the most common carcinogenic mechanism has been known to be multiple genetic changes. But, the CpG island hypermethylation has been revealed to be one of the most important carcinogenic mechanism second to the genetic changes. The epigenetic changes characteristically imprint and transfer to the next generation without changes of nucleotide sequences. When the CpG island of promoter or 5'- exon 1 of a specific gene is methylated by DNA methyltrasferases (DNMTs), the gene expression is inhibited. Carcinogenic mechansisms by promoter methylation has been well known, but the progression of the cancer has not been elucidated yet. The molecular researches on the carcinogenic mechanisms largely depended on the tumor tissue containing various components of cell, due to difficulties in harvesting pure homogeneous tumor cells. Promoter methylatonof the cancer related genes as APc, hMLH2, p16, DAP-K and MGMT in 50 cases of sporadic colorectal carcinoma was studied. The pure normal colorectal epithelial cells, and cancer cells from superficial and deep invasive portion were harvested respectively by laser captured microdissection(LCM), and DNA was extracted from them. The promoter methylation was studied by methylation specific poymerase reaction (MSP) after bisulfite modification of the DNA and the expression of the genes by immunohistochemistry. The results suggests that promoter methylation of the cancer related genes are closely related with the colorectal carcinogenesis and it occurrs from early phase of carcinogenesis, progressing constantly to the late stage. And the promoter methylation of normal epithelial cells increased by age. The p15, known to be the genes related to hematologic malignancy, was closely related to the colorectal carcinoma as well.

      • Esmolo과 lidocaine의 혼합사용시 기관내 삽관에 따른 혈압과 심박수에 미치는 영향

        Background: Ultrashort acting β-adrenergic antagonist, esmolol and local anesthetics, lidocaine are well known about effects of hemodynamic stabilization to tracheal intubation. But, the effects of these drugs are still controversial. The aim of the present study is to assess whether a combination of both drugs are more effective than either drug alone. Methods: Forty eight patients, ASA physical status Ⅰ or Ⅱ, were randomly assigned to one of four groups (n=12 each): control (Group S), esmolol 1 ㎎/㎏ (Group E), lidocaine 1 ㎎/㎏ (Group L) and esmolol-lidocaine combination (Group M). Anesthesia was induced with thiopental 5 ㎎/㎏ intravenously, and then saline, esmolol, lidocaine and combination drug were administered as an intravenous bolus, and immediately followed by succinylcholine 1.5 ㎎/㎏. Tracheal intubation was done 90 seconds after intravenous injection of drugs, respectively. Systolic and diastolic blood pressure and heart rate were measured before induction and every minute for 5 minutes after tracheal intubation. Results: There was a significant attenuation in systolic, diastolic arterial pressure and heart rates after tracheal intubation in esmolol-lidocaine group compared to other three groups. Conclusions: Combination of esmolol 1 ㎎/㎏ with lidocaine 1 ㎎/㎏ seems to be useful for attenuation of the increases in blood pressure and heart rates compare to either drug alone after tracheal intubation.
