        Cellulase 에 의한 목재당화에 (木材糖化) 관한 연구 - (Ⅰ) 기질 (基質) 처리의 효과 -

        In this study, enzymatic hydrolysis of the holocellulose from Alnus hirsuta (Spach) Rupr. (8-14 yr`s) was investigated using crude cellulase preparations of Trichoderma viride Pers. ex. Fr. SANK 16374. And conducted on the optimum condition of the treated substrate for saccharification. A strain of Trichoderma viride Pers. ex. Fr. SANK 16374 was found to be highly efficient for the cellulase productivity, especially in the submerged culture process. The culture medium used in this experiment was prepared from an extract of wheat bran consisting also of KH₂ PO₄ 10, (NH₄)₂ SO₄ 3, NaNO₃ 3, and MgSO₄7H₂O 0.5g/1. Cellulose powder (Toyo filter paper, 60 mesh) was found to be an importent factar for inducing the cellulase formation. And the cellulase produced in the culture fluid was salted out quantitatively by the use of ammonium sulfate (Fig. 1) Reducing sugar was determined by the Dinitrosalicylic acid (DNS) method, using reagents prepared according to the method of Sumner (1925). The results obtained were summerized as follows; 1. The method of delignification were treated by the Peracetic acid (PA) method, according to the method of Toyama (1970). The yield of holocellulose were decreased in accordance with increasing concentration of Peracetic acid solution; delignification of Alnus hirsuta Rupr. with 20% Peracetic acid was satisfied for 48 hours and 40%∼60% peracetic acid was satisfied for 24 hrs: 2. The substrate (holocellulose) was changed easely into fine powder with enzymatic hydrolysis and cellulase exhibits optimum activity on the reducing sugar formation from substrate at the range of 60-100 mesh. 3. The reducing sugar formation increased in accordance with increasing dry temperature on holocellulose substrate was found to be 190±5℃. 4. The optimal heat treated time of holocellulose substrate was found to be 45 min. for the reducing sugar formation showed the best products. The reducing sugar formation did not show statisticaly significent diflerences at 5% levels by heat treated time for 45 min. and 60 min.

        Purification and Characterization of Helicobacter pylori ${\gamma}$-Glutamyltranspeptidase

        Gamma-glutamyltranspeptidase (GGT) was purified to electrophoretic homogeneity from the cell extract of H. pylori. The purified enzyme consisted of heavy and light subunits with molecular weights of 38 kDa and 21 kDa, respectively. N-terminal amino acid sequence of heavy and light subunits revealed that H. pylori GGT was processed into 3 parts for a signal peptide of 27 amino acid residues, a heavy subunit of 352 residues, and a light subunit of 188 residues during translation. The reaction rate for hydrolysis of ${\gamma}$-GpNA was 84.4 ${\mu}mol/min$ per milligram of protein, and that for the ${\gamma}$-glutamyl transfer from ${\gamma}$-GpNA to gly-gly was 23.8 ${\mu}mol/min$ per milligram of protein. The apparent Km values of H. pylori GGT for ${\gamma}$-glutamyl compounds were on the order of $10^{-3}$ to $10^{-4}$ M and those for acceptor peptides and amino acids were on the order of $10^{-1}$ to $10^{-2}$ M. The GGT protein kept approximately 80% of the initial enzymatic activity on incubation at $60^{\circ}C$ for 15 min. The optimum temperature and pH for reactions of both hydrolysis and transpeptidation were $40^{\circ}C$ and 9.0, respectively. The transpeptidation and hydrolysis reactions catalyzed by H. pylori GGT were strongly inhibited by L-Gln and moderately inhibited by L-Ala, L-Ser, ${\beta}$-chloro-L-Ala, and L-Glu. These results demonstrated that the biochemical properties of H. pylori GGT are different from those of other bacterial GGTs. Further, H. pylori GGT might degrade glutathione in the gastric mucous layer of humans if the enzyme could be secreted in the bacterial niches.

      • 장기간 요가운동이 알코올의존자의 면역글로블린에 미치는 영향

        The purpose of this study was to investigate the effect of Yoga exercise on Immunoglobulin(IgE, IgM, IgD, IgG, IgA) in the alcoholism. The subjects involved in this study consists of 14 people who were confirmed to have alcohol-related problems by DSM-Ⅳ(1994) of American Psychiatric Association and were diagnosed as alcoholism in hospital. The subjects were classified into yoga exercise group(7 people) and control group(7 people). The yoga exercise group participated in the exercise programs named Hatayoga and Asana. This treatment was performed an hour per day, three days a week for 16 weeks while the control group watched TV for the same time. A paired t-test was used to examine differences between pre, post 8weeks and post 16weeks test scores. The results are as follows. After 16 week yoga exercise, there was a significant increase(p<.01) of Immunoglobulin (IgM, IgA(p<.05) & IgG) in the experiment group, compared with the control group. In terms of periodic change, the experiment group showed a significant higher result(p<.01) than the control group after 16 weeks, rather than after the 8 weeks. The result of this study above indicates that the long-term low intensity exercise can activate Immunoglobulint in alcoholism.

        A new device, muscle clamping system was developed to facilitate exact quatifying technique A strabismus surgery, and reduce the risk of complications. The device is composed of a lower fixing body with three jaws and an upper supporting body. They are used to clip an extraocular muscle and fix it to the sclera with a single bite. Superior rectus recession on 16 rabbit eyes were performed with this new device. Conjunctival injection, muscle adhesion strength, and light microscopic findings were examined at 1, 2, 4 and 8 weeks postoperatively. The Conjunctival injection were minimal, adhesion power ranged from 420 to 600 gram gravity, which is sufficient in withstand the normal pull of human extraocular muscle. In microscopic exam, some Inflammatory cells and fibrosis were found. The new device was technically easy, fast, and accurate, so it may be useful in stabismus surgery.

        Bluetooth is the name given to a new technology standard using short-range radio links, intended to replace the cable(s) connecting portable and/or fixed electronic devices. The standard defines a uniform structure for a wide range of devices to communicate with each other, with minimal user effort. Its key features are robustness, low complexity, low power and low cost. The standard is aimed at achieving global acceptance such that any Bluetooth device, anywhere in the world, can connect to other Bluetooth devices in its proximity , regardless of brand. Bluetooth enabled electronic devices connect and communicate wirelessly via short-range, ad hoc networks called piconets. Each unit can simultaneously communicate with up to seven other units per piconet. Moreover, each unit can simultaneously belong to several piconets. These piconets are established dynamically and automatically as Bluetooth devices enter and leave the radio proximity.

        This Study investigated a possibility of lateral movement in the reconstruction field of Nongso bridge at Nongso-ri, Joochon-myeon, Kimhae. The methods applied in this study were the judgement formula and the Finite Element Method(FEM). The conclusions are shown in the following. 1) The application of the FEM, as well as the judgement formula resulted in the lateral movement. 2) Bridge abutment and foundation must be constructed after completing consolidation. 3) The occurrence possibility of lateral movement should be judged by the FEM that can re-present field conditions. 4) Lateral movement will take place in the case. Hence, settlement of proper countermeasure method should be followed by the construction.

        In this study, additions of aluminium foil eching characteristics were compared with sulfuric acid for capacitor. sulfuric acid and ethylene glycol is used as addition, characteristics of funtional group from addition were guessed by comparing of aluminium foil eching characteristics. When sulfuric acid was added in HCI etching solution, corrosion of aluminum metal was inhibited by the chemical adsorption of sulfate ion, when ethylen glycol was added in HCI etching solution, corrosion inhibited by the chemical adsorption of OH­^ion.

      • 생물변환 천연식용색소 생산공정기술 개발 : Ⅰ. 홍화 황색소의 안정성에 미치는 pH 및 온도의 영향 Ⅰ. Effect of pH and Temperature on the Stability of the Yellow Pigment from Carthamus tinctorious L.

        홍화로부터 열에 의한 안정성을 검토하였다. 황색소의 분해반응은 산성, 중성, 염기성 조건에서 UV/Vis spectral measurement에 의해 조사되었다. 시간변화에 다른 흡광도를 측정해 본 결과 온도가 상승함에 따라 황색소의 분해되는 정도가 점차적으로 증가하는 것을 확인 할 수 있었다. 40℃에서 황색소의 분해 반감기는 pH3.0, pH7.0, 그리고 pH12.0에서 각각 112.1, 39.3 과 25.3 시간이었다. 이것은 황색소가 pH3.0에서 비교적 더 안정함을 말한다. pH3.0, 7.0 그리고 12.0에서 황색소의 activation energy는 각각 519, 15.0 그리고 12.3 kJ/mol이었다 We investigated thermal stability of yellow pigment from Carthamus tinctorious L. Decomposition reactions of yellow pigment were monitored at acidic, neutral and alkaline conditions by UV/Vis spectral measurement. Decomposition of yellow pigment increased as temperature increased Decomposition half lives of yellow pigment at 40℃ were 112.1 39.3, and 25.3 hrs at pH 3.0 , pH 7.0, and pH 12.0, respectively, indicating that yellow pigment is more stable at pH 3.0. The activation energies of yellow pigment at pH 3.0, pH 7.0, and pH 12.0 were 51.9, 15.0, and 12. 3 kJ/mol, respectively.

      • 사상체질 유형에 따른 손바닥문, 손금의 특징

        In Sasang constitutional medicine, the classification of Sasang constitutional types (Tae-Yang, Tae-Eum, So-Yang, and SO-Eum) is importnat for the treatment. There are some problems with the methods used for classifying Sasang constitutional types ; old methods such as pulse-palpation are not considered objective and recent methods such as immunohematology are considered expensive, painful and time-consuming. To overcome these problems, a body measurement and finger prints analysis were performed. The purpose of this study was to determine whether the palm prints and the palm creases could be helpful in classifying Sasang constitutional types. Thus, we looked for characteristics of the palm prints and the palm creates according to Sasang constitutional types. Before analyzing the palm prints and palm creases, 760 Koreans (465 males, 295 females) were surveyed using two kinds of questionnaires for classifying Sasang constitutional types. As there were no Tae-Yang individuals, we were only able to identify the characteristics of the palm prints and the palm creases for Tae-Eum (288 persons), So-Yang(193 persons), and So-Eum (279 persons) individuals. In this study, the terminal point 11 and closed crease were frequent in Tae-Eum individuals; whereas, terminal point 7 and open crease were frequent in So-Eum individuals. Therefore, the palm prints and the palm creases seem to contribute to the classification of Sasang constitutional types.

      • CDMA 시각 동기용 GPS 시스템의 고주파부 설계 및 제작

        When GPS(Global Positioning System) is used as synchronous signal in CDMA digital cellular base station system and high speed digital synchronous communication network, antenna cable lengh is increased, comparing with other GPS application such as positioning or car navigation. In this paper, it is proposed that a type of new GPS antenna system including RF stage for reduction of cable loss in case of long cable. The antenna system with TMPA(Truncated-corners Microstrip Patch Antenna) is designed and fabricated because GPS signal has RHCP(Right-Hand Circular Polarization), consequently antenna size can be made small size. NAN(Low Noise Amplifier) is designed by using HEMT(High Electron Mobility Transistor) which has lower noise figure and better AGC characteristics at low voltage than GaAs FET, and we equipped mixer, in GPS antenna unit, which converts from 1575.42MHz to 75.42MHz. As result of comparing between typical system and proposed system when cable length is 60m, 63dB, 55dB and 25dB gain are obtained for RG-316/U, RG-58C/U and RG-213/U, and better characteristics are achieved than typical system as far as cable length is longer.
